    Is there a linear relationship between the dose of ruminant trans-fatty acids and cardiovascular risk markers in healthy subjects: results from a systematic review and meta-regression of randomised clinical trials

    International audienceThe effects of ruminant (R) trans-fatty acids (TFA) on the risk of CVD are still under debate. It could be argued that the lack of the effect of R-TFA may be the result of the small amount of their intake. Taking into consideration the growing available data from intervention studies, we carried out a systematic review and meta-regression to assess the impact of R-TFA intake levels on changes in the total cholesterol: HDL-cholesterol (TC: HDL-C) ratio. A systematic review of the literature was conducted and thirteen randomised clinical trials were included, yielding a total of twenty-three independent experimental groups of subjects. A univariate random-effects meta-regression approach was used to quantify the relationship between the dose of R-TFA and changes in the TC: HDL-C ratio. To consider several potential modifiers such as subject and dietary characteristics, a multivariate regression analysis was performed. We found no relationship between R-TFA intake levels of up to 4.19% of daily energy intake (EI) and changes in cardiovascular risk factors such as TC: HDL-C and LDL-cholesterol (LDL-C): HDL-C ratios. In addition, a multivariate regression analysis that included other dietary variables, as well as subject baseline characteristics, confirmed that doses of R-TFA did not significantly influence the changes in the lipid ratio. Our findings showed that doses of R-TFA did not influence the changes in the ratios of plasma TC: HDL-C and LDL-C: HDL-C. These data suggest that TFA from natural sources, at least at the current levels of intake and up to 4.19% EI, have no adverse effects on these key CVD risk markers in healthy people

    Response to Cysteamine in Osteoclasts Obtained from Patients with Nephropathic Cystinosis: A Genotype/Phenotype Correlation

    Bone complications of cystinosis have been recently described. The main objectives of this paper were to determine in vitro the impact of CTNS mutations and cysteamine therapy on human osteoclasts and to carry out a genotype-phenotype analysis related to osteoclastic differentiation. Human osteoclasts were differentiated from peripheral blood mononuclear cells (PBMCs) and were treated with increasing doses of cysteamine (0, 50, 200 ”M) and then assessed for osteoclastic differentiation. Results are presented as median (min-max). A total of 17 patients (mainly pediatric) were included, at a median age of 14 (2–61) years, and a eGFR of 64 (23–149) mL/min/1.73 m2. Most patients (71%) were under conservative kidney management (CKM). The others were kidney transplant recipients. Three functional groups were distinguished for CTNS mutations: cystinosin variant with residual cystin efflux activity (RA, residual activity), inactive cystinosin variant (IP, inactive protein), and absent protein (AP). PBMCs from patients with residual cystinosin activity generate significantly less osteoclasts than those obtained from patients of the other groups. In all groups, cysteamine exerts an inhibitory effect on osteoclastic differentiation at high doses. This study highlights a link between genotype and osteoclastic differentiation, as well as a significant impact of cysteamine therapy on this process in humans

    Recent advances in dairy ingredients and cardiovascular diseases with special reference to milk fat components

    Cardiovascular diseases are the first cause of mortality in the world and their incidence has been related to intake of saturated fats. Current dietary recommendations recognize the contribution of milk and dairy products to a healthy diet because consumption of them produces an increase in the levels of many high-quality nutrients. However, this consensus does not apply where milk fat is considered because of its high saturated fatty acid content; therefore, it has been advised to limit or suppress its consumption. This chapter analyzes the relation between consumption of dairy foods and cardiovascular diseases. In the view of current scientific results and after years of controversy, the negative image of milk fat is weakening. From the scientific evidence available it is possible to conclude that consumption of milk and a balanced diet that contains dairy foods can have a neutral effect or an inverse association with cardiovascular risk in healthy adolescents or adults.Peer reviewe

    Reigitherium (Meridiolestida, Mesungulatoidea) an Enigmatic Late Cretaceous Mammal from Patagonia, Argentina: Morphology, Affinities, and Dental Evolution

    New dental and dentary fossils collected in the Upper Cretaceous La Colonia Formation in central Patagonia provide new evidence on the morphology, feeding ecology, and relationships of the enigmatic mammal Reigitherium. The newly discovered specimens described here include elements of the upper dentition and several partial dentaries, elucidating fundamental questions of serial homology and postcanine dental formula (four premolars and three molars). This new evidence supports a nested position of Reigitherium within the advanced meridiolestidan clade Mesungulatoidea. Apomorphic features of the upper and lower molariform elements include intense enamel crenulation circumscribed within the primary trigon and trigonid, elevated cingulids, and the neomorphic appearance of cusps/cuspulids, all of which increase overall crown complexity. A Dental Topography Analysis comparing Reigitherium and its sister taxon Peligrotherium to Cretaceous and Cenozoic therians demonstrates functional similarity between the mesungulatoids and South American marsupial taxa that succeed them in the small-to medium-sized herbivore niche during the Paleocene. Previous taxonomic attributions of Reigitherium are discussed and comparisons with other meridiolestidans highlight the remarkable radiation of this group in the Cretaceous of South America.Fil: Harper, Tony.
