    Trypanocidal and leishmanicidal activity of six limonoids

    Six limonoids [kotschyienone A and B (1, 2), 7-deacetylgedunin (3), 7-deacetyl-7-oxogedunin (4), andirobin (5) and methyl angolensate (6)] were investigated for their trypanocidal and leishmanicidal activities using bloodstream forms of Trypanosoma brucei and promastigotes of Leishmania major. Whereas all compounds showed anti-trypanosomal activity, only compounds 1–4 displayed anti-leishmanial activity. The 50% growth inhibition (GI 50) values for the trypanocidal and leishmanicidal activity of the compounds ranged between 2.5 and 14.9 μM. Kotschyienone A (1) was found to be the most active compound with a minimal inhibition concentration (MIC) value of 10 μM and GI 50 values between 2.5 and 2.9 μM. Only compounds 1 and 3 showed moderate cytotoxicity against HL-60 cells with MIC and GI 50 values of 100 μM and 31.5–46.2 μM, respectively. Compound 1 was also found to show activity against intracellular amastigotes of L. major with a GI 50 value of 1.5 μM. The results suggest that limonoids have potential as drug candidates for the development of new treatments against trypanosomiasis and leishmaniasis

    Sensibilidad y especificidad del método inmunocromatográfico utilizado para el diagnóstico de rotavirus

    El rotavirus (RV) es el principal agente viral causante de diarrea aguda en niños menores de 5 años y es responsable de aproximadamente el 6% de las muertes en este grupo etáreo, lo que conlleva la necesidad de utilizar métodos de diagnósticos rápidos y confiables.El objetivo del estudio es evaluar la sensibilidad y especificidad del método inmunocromatográfico (ICG), en el que se basan muchos kits comerciales utilizados para el diagnóstico de rotavirus grupo A, tomando como referencia el método de electroforesis en gel de poliacrilamida (PAGE). Se seleccionaron muestras de heces de 317 pacientes con diarrea aguda que concurrieron a un laboratorio privado de mayo a noviembre del 2006, con pedido de análisis de rotavirus y todos los datos de los pacientes fueron manejados de manera confidencial Se utilizaron kits de las marcas comerciales Operónsimple (n=154) o SD Bioline rotavirus (n=163), siguiendo estrictamente las instrucciones del fabricante. Las muestras fueron conservadas en frío y remitidas al IICS para la realización de los estudios moleculares. La sensibilidad obtenida por el método ICG fue de 97,8% y la especificidad de 84%. La concordancia absoluta fue del 90%. Las muestras que dieron resultados discrepantes entre ICG y PAGE, fueron confirmadas por nRT-PCR,resultados que coincidieron con los obtenidos por PAGE. La sensibilidad del método ICG es muy buena, si bien la especificidad es moderada el método puede ser utilizado como screening para el diagnóstico rápido de rotavirus y sería aconsejable utilizar métodos más específicos como los moleculares para estudios epidemiológicos

    Self-similar extinction for a diffusive Hamilton-Jacobi equation with critical absorption

    International audienceThe behavior near the extinction time is identified for non-negative solutions to the diffusive Hamilton-Jacobi equation with critical gradient absorption ∂_t u − ∆_p u + |∇u|^{p−1} = 0 in (0, ∞) × R^N , and fast diffusion 2N/(N + 1) < p < 2. Given a non-negative and radially symmetric initial condition with a non-increasing profile which decays sufficiently fast as |x| → ∞, it is shown that the corresponding solution u to the above equation approaches a uniquely determined separate variable solution of the form U (t, x) = (T_e − t)^{1/(2−p)} f_* (|x|), (t, x) ∈ (0, T_e) × R^N , as t → T_e , where T_e denotes the finite extinction time of u. A cornerstone of the convergence proof is an underlying variational structure of the equation. Also, the selected profile f_* is the unique non-negative solution to a second order ordinary differential equation which decays exponentially at infinity. A complete classification of solutions to this equation is provided, thereby describing all separate variable solutions of the original equation. One important difficulty in the uniqueness proof is that no monotonicity argument seems to be available and it is overcome by the construction of an appropriate Pohozaev functional

    Non-deterministic reef fish community assembly in an upwelling-influenced transitional subprovince of the southwestern Atlantic

    There are two major theories for setting up ecological communities, the Niche Theory and the Neutral Theory. Both seek to explain the main factors that form a community, which is a great challenge, since each community has its particularities and the environment has different ways to manifest. We devised a process-oriented study that sought to establish the role of environmental niche driven by coastal upwelling in the assembly of reef fish communities from exposed and sheltered environments a few kilometers apart, in the region of Arraial do Cabo (southwestern Atlantic). A multivariate hierarchical generalized linear mixed model fitted with Bayesian inference was applied to abundance and presence-absence data from visual census, together with environmental data from satellite and reanalysis. We found a stronger contribution of random effects to abundance variance with 24% for sites and 20.7% for sheltered vs. exposed locations, and weaker environmental effects with 7.1% for surface chlorophyll-a concentration (SCC) and 5.4% for sea surface temperature (SST). Environmental effects had a stronger contribution in the presence-absence model, with 20.1% for SCC and 14.6% for SST. The overall influence of the upwelling environment across all species was negative, e.g., Gymnothorax moringa and Canthigaster figueiredoi showing negative responses to SCC and Parablennius pilicornis and Malacoctenus delalandii to SST. The joint action of migration-niche mechanisms is inferred from the dominance of spatio-temporal structure, limited influence of life history traits and phylogeny, explaining around 95% of species niches in the abundance model. Our results bring new evidence for the importance of different filters for community assembly other than the environment, such as phylogenetic history and dispersal. We also discuss the balance between niche (environment) and neutral (stochasticity) processes for the assembly of reef fish communities in a tropical-subtropical transition zone

    Mutations of the GLA gene in young patients with stroke: the PORTYSTROKE study-screening genetic conditions in Portuguese young stroke patients

    BACKGROUND AND PURPOSE: Fabry disease is an X-linked monogenic disorder caused by mutations in the GLA gene. Recent data suggest that stroke in young adults may be associated with Fabry disease. We aimed to ascertain the prevalence of this disorder among young adult patients with stroke in Portugal by GLA genotyping. METHODS: During 1 year, all patients aged 18 to 55 years with first-ever stroke, who were admitted into any of 12 neurology hospital departments in Portugal, were prospectively enrolled (n=625). Ischemic stroke was classified according to Trial of Org 10172 in Acute Stroke Treatment criteria. Alpha-galactosidase activity was further assayed in all patients with GLA mutations. RESULTS: Four hundred ninety-three patients (mean age, 45.4 years; 61% male) underwent genetic analyses: 364 with ischemic stroke, 89 with intracerebral hemorrhage, 26 with subarachnoid hemorrhage, and 14 with cerebral venous thrombosis. Twelve patients had missense GLA mutations: 9 with ischemic stroke (p.R118C: n=4; p.D313Y: n=5), including 5 patients with an identified cause of stroke (cardiac embolism: n=2; small vessel disease: n=2; other cause: n=1), 2 with intracerebral hemorrhage (p.R118C: n=1; p.D313Y: n=1), and one with cerebral venous thrombosis (p.R118C: n=1). Leukocyte alpha-galactosidase activity was subnormal in the hemizygous males and subnormal or low-normal in the heterozygous females. Estimated prevalence of missense GLA mutations was 2.4% (95% CI, 1.3% to 4.1%). CONCLUSIONS: Despite a low diagnostic yield, screening for GLA mutations should probably be considered in different types of stroke. Restricting investigation to patients with cryptogenic stroke may underestimate the true prevalence of Fabry disease in young patients with stroke

    Observation of an Excited Bc+ State

    Using pp collision data corresponding to an integrated luminosity of 8.5 fb-1 recorded by the LHCb experiment at center-of-mass energies of s=7, 8, and 13 TeV, the observation of an excited Bc+ state in the Bc+π+π- invariant-mass spectrum is reported. The observed peak has a mass of 6841.2±0.6(stat)±0.1(syst)±0.8(Bc+) MeV/c2, where the last uncertainty is due to the limited knowledge of the Bc+ mass. It is consistent with expectations of the Bc∗(2S31)+ state reconstructed without the low-energy photon from the Bc∗(1S31)+→Bc+γ decay following Bc∗(2S31)+→Bc∗(1S31)+π+π-. A second state is seen with a global (local) statistical significance of 2.2σ (3.2σ) and a mass of 6872.1±1.3(stat)±0.1(syst)±0.8(Bc+) MeV/c2, and is consistent with the Bc(2S10)+ state.     Bose-Einstein correlations of same-sign charged pions in the forward region in pp collisions at √s=7 TeV

    Bose-Einstein correlations of same-sign charged pions, produced in protonproton collisions at a 7 TeV centre-of-mass energy, are studied using a data sample collected by the LHCb experiment. The signature for Bose-Einstein correlations is observed in the form of an enhancement of pairs of like-sign charged pions with small four-momentum difference squared. The charged-particle multiplicity dependence of the Bose-Einstein correlation parameters describing the correlation strength and the size of the emitting source is investigated, determining both the correlation radius and the chaoticity parameter. The measured correlation radius is found to increase as a function of increasing charged-particle multiplicity, while the chaoticity parameter is seen to decreas

    Measurement of the inelastic pp cross-section at a centre-of-mass energy of 13TeV

    The cross-section for inelastic proton-proton collisions at a centre-of-mass energy of 13TeV is measured with the LHCb detector. The fiducial cross-section for inelastic interactions producing at least one prompt long-lived charged particle with momentum p &gt; 2 GeV/c in the pseudorapidity range 2 &lt; η &lt; 5 is determined to be ϭ acc = 62:2 ± 0:2 ± 2:5mb. The first uncertainty is the intrinsic systematic uncertainty of the measurement, the second is due to the uncertainty on the integrated luminosity. The statistical uncertainty is negligible. Extrapolation to full phase space yields the total inelastic proton-proton cross-section ϭ inel = 75:4 ± 3:0 ± 4:5mb, where the first uncertainty is experimental and the second due to the extrapolation. An updated value of the inelastic cross-section at a centre-of-mass energy of 7TeV is also reported
