    Measurement of residual stresses induced by sequential weld buttering and cladding operations involving a 2.25Cr-1Mo substrate material

    Dissimilar metal welds are necessary in high-pressure subsea systems and in cases where forged components must be welded to pipelines. F22 (2.25Cr-1Mo) steel is often used in such forged steel components and, since this steel cannot enter service without undergoing post-weld heat treatment (PWHT), the components are usually prepared for field welds through the application of a buttering layer. Furthermore, a weld overlay is deposited for the purpose of mitigating corrosion. This combination of multiple welding tasks and dissimilar materials leads to the possibility of developing substantial residual stresses. This study aims to provide insights to the evolution of residual stresses at each stage of the welding operation. The assessment has been undertaken on laboratory-scale weld mock-ups using the contour method for residual stress measurement, and incremental centre hole drilling. It was found that both buttering and cladding introduce near-yield levels of tensile residual stresses, but that these stresses are successfully relieved upon PWHT

    Changes in Arctic Ocean Climate Evinced through Analysis of IPY 2007–2008 Oceanographic Observations

    Full-depth hydrographical surveys conducted in 2007–2009 during the International Polar Year (IPY) collaboration provide an accurate snapshot of the Arctic Ocean (AO) hydrography at a time when the Arctic Ocean Oscillation (AOO) index was highest in recent record. We construct pan-Arctic temperature and salinity (T/S) reference states from these data using variational optimal interpolation and discuss some key differences between the 2007–2009 state and a similarly constructed climatology from historical 1950–1994 Russian archives. These data provide a recent, known reference state for both qualitative and quantitative future AO climate change studies. Furthermore, we present an analysis of sea-surface height (SSH) and upper-layer circulation constructed from the IPY data via 4DVar data assimilation and use them to examine circulation and freshwater source changes visible during IPY

    CASSETTE—clindamycin adjunctive therapy for severe Staphylococcus aureus treatment evaluation: Study protocol for a randomised controlled trial

    Background Exotoxins are important virulence factors in Staphylococcus aureus. Clindamycin, a protein synthesis inhibitor antibiotic, is thought to limit exotoxin production and improve outcomes in severe S. aureus infections. However, randomised prospective data to support this are lacking. Methods An open-label, multicentre, randomised controlled trial (RCT) will compare outcome differences in severe S. aureus infection between standard treatment (flucloxacillin/cefazolin in methicillin-susceptible S. aureus; and vancomycin/daptomycin in methicillin-resistant S. aureus) and standard treatment plus an additional clindamycin given for 7 days. We will include a minimum of 60 participants (both adult and children) in the pilot study. Participants will be enrolled within 72 h of an index culture. Severe infections will include septic shock, necrotising pneumonia, or multifocal and non-contiguous skin and soft tissue/osteoarticular infections. Individuals who are immunosuppressed, moribund, with current severe diarrhoea or Clostridiodes difficile infection, pregnant, and those with anaphylaxis to β-lactams or lincosamides will be excluded. The primary outcomes measure is the number of days alive and free (1 or 0) of systemic inflammatory response syndrome (SIRS) within the first 14 days post randomisation. The secondary outcomes measure will include all-cause mortality at 14, 42, and 90 days, time to resolution of SIRS, proportion with microbiological treatment failure, and rate of change of C-reactive protein over time. Impacts of inducible clindamycin resistance, strain types, methicillin susceptibility, and presence of various exotoxins will also be analysed. Discussion This study will assess the effect of adjunctive clindamycin on patient-centred outcomes in severe, toxin-mediated S. aureus infections. The pilot study will provide feasibility for a much larger RCT. Trial registration Australian New Zealand Clinical Trials Registry, ACTRN12617001416381p. Registered on 6 October 2017

    Detecting Microscopic Black Holes with Neutrino Telescopes

    If spacetime has more than four dimensions, ultra-high energy cosmic rays may create microscopic black holes. Black holes created by cosmic neutrinos in the Earth will evaporate, and the resulting hadronic showers, muons, and taus may be detected in neutrino telescopes below the Earth's surface. We simulate such events in detail and consider black hole cross sections with and without an exponential suppression factor. We find observable rates in both cases: for conservative cosmogenic neutrino fluxes, several black hole events per year are observable at the IceCube detector; for fluxes at the Waxman-Bahcall bound, tens of events per year are possible. We also present zenith angle and energy distributions for all three channels. The ability of neutrino telescopes to differentiate hadrons, muons, and possibly taus, and to measure these distributions provides a unique opportunity to identify black holes, to experimentally constrain the form of black hole production cross sections, and to study Hawking evaporation.Comment: 20 pages, 9 figure

    Discursive constructions of language and identity: parents' competing perspectives in London Turkish complementary schools

    In this paper, I draw on interview data to explore parents' constructions of language and identity in two London Turkish complementary schools. I examine parents' evaluative talk about standard Turkish, Cypriot-Turkish and other regional varieties of Turkish, the cultural values they attach to them and images of personhood these invoke. I demonstrate how parents brought up in Turkey and Cyprus tend to privilege standard Turkish and acknowledge the crucial role Turkish complementary schools play as one of the key vehicles for its spread and promotion. Some parents, however, especially those politically active in promoting Cypriot-Turkish language, culture and history to the British-born generation, voice an alternative discourse where Cypriot-Turkish is intimately linked to their sense of self. Moreover, I investigate parents' perspectives of their children and of their own Turkish language competence in the case of parents brought up in the UK. I illustrate how their Turkish and their children's are compared against a ‘native’ speaker norm firmly located in the countries of origin. The parents' accounts show how different self- and other-ascriptions of proficiency in Turkish are linked to claims of or lack of ‘Turkishness’, largely shaped by their migration histories and narratives, personal and professional transnational experiences

    Cosmic Neutrinos from the Sources of Galactic and Extragalactic Cosmic Rays

    Although kilometer-scale neutrino detectors such as IceCube are discovery instruments, their conceptual design is very much anchored to the observational fact that Nature produces protons and photons with energies in excess of 10^20 eV and 10^13 eV, respectively. The puzzle of where and how Nature accelerates the highest energy cosmic particles is unresolved almost a century after their discovery. From energetics considerations we anticipate order 10~100 neutrino events per kilometer squared per year pointing back at the source(s) of both galactic and extragalactic cosmic rays. In this context, we discuss the results of the AMANDA and IceCube neutrino telescopes which will deliver a kilometer-square-year of data over the next 3 years.Comment: 8 pages, 4 figure

    Patient-specific Alzheimer-like pathology in trisomy 21 cerebral organoids reveals BACE2 as a gene dose-sensitive AD suppressor in human brain

    A population of >6 million people worldwide at high risk of Alzheimer’s disease (AD) are those with Down Syndrome (DS, caused by trisomy 21 (T21)), 70% of whom develop dementia during lifetime, caused by an extra copy of β-amyloid-(Aβ)-precursor-protein gene. We report AD-like pathology in cerebral organoids grown in vitro from non-invasively sampled strands of hair from 71% of DS donors. The pathology consisted of extracellular diffuse and fibrillar Aβ deposits, hyperphosphorylated/pathologically conformed Tau, and premature neuronal loss. Presence/absence of AD-like pathology was donor-specific (reproducible between individual organoids/iPSC lines/experiments). Pathology could be triggered in pathology-negative T21 organoids by CRISPR/Cas9-mediated elimination of the third copy of chromosome-21-gene BACE2, but prevented by combined chemical β and γ-secretase inhibition. We found that T21-organoids secrete increased proportions of Aβ-preventing (Aβ1-19) and Aβ-degradation products (Aβ1-20 and Aβ1-34). We show these profiles mirror in cerebrospinal fluid of people with DS. We demonstrate that this protective mechanism is mediated by BACE2-trisomy and cross-inhibited by clinically trialled BACE1-inhibitors. Combined, our data prove the physiological role of BACE2 as a dose-sensitive AD-suppressor gene, potentially explaining the dementia delay in ~30% of people with DS. We also show that DS cerebral organoids could be explored as pre-morbid AD-risk population detector and a system for hypothesis-free drug screens as well as identification of natural suppressor genes for neurodegenerative diseases

    Search for the standard model Higgs boson decaying into two photons in pp collisions at sqrt(s)=7 TeV

    A search for a Higgs boson decaying into two photons is described. The analysis is performed using a dataset recorded by the CMS experiment at the LHC from pp collisions at a centre-of-mass energy of 7 TeV, which corresponds to an integrated luminosity of 4.8 inverse femtobarns. Limits are set on the cross section of the standard model Higgs boson decaying to two photons. The expected exclusion limit at 95% confidence level is between 1.4 and 2.4 times the standard model cross section in the mass range between 110 and 150 GeV. The analysis of the data excludes, at 95% confidence level, the standard model Higgs boson decaying into two photons in the mass range 128 to 132 GeV. The largest excess of events above the expected standard model background is observed for a Higgs boson mass hypothesis of 124 GeV with a local significance of 3.1 sigma. The global significance of observing an excess with a local significance greater than 3.1 sigma anywhere in the search range 110-150 GeV is estimated to be 1.8 sigma. More data are required to ascertain the origin of this excess.Comment: Submitted to Physics Letters
