    Überprüfung der Messmethodenübereinstimmung der gängigen manuellen GP Methode mit der automatischen BoneXpert Methode nach GP zur Bestimmung des Knochenalters bei Kindern mit Pubertas praecox

    Zusammenfassung: Das Knochenalter von Kinderhänden wurde bisher meist durch manuelle Befundung von Röntgenbildern ermittelt. Diese Methode bringt diverse Ungenauigkeiten mit sich. Zumeist ist es innerhalb einer Klinik geregelt, nach welchem manuellen Verfahren ( z.B. Greulich – Pyle und Tanner – Whitehouse) befundet wird, jedoch variieren die Ergebnisse trotzdem (Intra- und Interobservervariabilität). Das vollautomatische Programm BoneXpert (BX) verspricht eine akkurate, einheitlichere Knochenalterbestimmung. Das Ziel dieser Arbeit war es, die Performance von BX bei Kindern mit Pubertas praecox zu untersuchen. Hierfür verglichen wir die Messergebnisse des vollautomatischen Programms BoneXpert (BX) mit den radiologischen Vorbefunden. Diese Befunde wurden anhand der Methode nach Greulich – Pyle befundet. Bei Differenzen größer als 1,5 Jahre zwischen den beiden Verfahren befundeten drei Experten unserer Klinik die Bilder blind nach. In die Studie wurden 126 Kinder integriert (103 Mädchen sowie 13 Jungen). Diese wiesen ein chronologisches Alter zwischen 0,3 – 14,8 Jahren zum Zeitpunkt der Röntgendiagnostik auf. Das Knochenalter war im Durchschnitt um 2,3 Jahre, mit einer Standardabweichung (SD) von 1,3 Jahren, akzeleriert. Die 745 Röntgenbilder wurden innerhalb des angegebenen Zeitraums in der Radiologischen Abteilung des Universitätsklinikums Tübingen erstellt, befundet und dann durch BX analysiert. 736 Bilder konnten befundet werden. 9 Bilder mussten verworfen werden, da BX Knochenalterstadien, welche außerhalb der gegebenen Analysegrenzen des Programms, lagen, bzw. fehlerhafte Handpositionen nicht erkannte. Von den analysierten BIldern stammten 97 Aufnahmen (13,2 %) von Jungenhänden. Die restlichen 639 Bilder (86,8 %) stammten von Mädchenhänden. Bis auf 43 Bilder lagen alle innerhalb des gewählten Äquivalenzbereiches von + / - 1,5 Jahren (Differenz). Der Mittelwert der Differenz (manueller Befund – BX Befund) betrug 0,18 Jahre bei einer Standardabweichung von 0,77 Jahren. Das 95%-Konfidenzintervall lag bei 0,12 – 0,23 Jahre, bei einer Varianz von 0,594. Der rms, welcher als Quadrat der mittleren Abweichung der Werte von der Kurve des „Linear Fit“ definiert ist, lag bei 0,68 Jahren. Von den o.g. 43 Ausreißern lagen nach der dreifachen blinden Rebefundung noch 5 Bilder außerhalb des Korridors. Bei allen 5 Bildern analysierte das Programm BX die Bilder korrekt, vergab jedoch ein jüngeres Knochenalter als der manuelle Vorbefund. So konnte in der Arbeit gezeigt werden, dass die automatische Knochenalterbestimmung durch BX verlässlich, effizient und präzise, auch bei Kinderhänden von Kindern mit Pubertas praecox war

    RNase A Treatment Interferes With Leukocyte Recruitment, Neutrophil Extracellular Trap Formation, and Angiogenesis in Ischemic Muscle Tissue

    Background: RNase A (the bovine equivalent to human RNase 1) and RNase 5 (angiogenin) are two closely related ribonucleases. RNase 5 is described as a powerful angiogenic factor. Whether RNase A shares the same angiogenic characteristic, or interferes with vessel growth as demonstrated for arteriogenesis, has never been investigated and is the topic of this present study. Methods and Results: To investigate whether RNase A shows a pro‐ or anti-angiogenic effect, we employed a murine hindlimb model, in which femoral artery ligation (FAL) results in arteriogenesis in the upper leg, and, due to provoked ischemia, in angiogenesis in the lower leg. C57BL/6J male mice underwent unilateral FAL, whereas the contralateral leg was sham operated. Two and seven days after the surgery and intravenous injection of RNase A (50 μg/kg dissolved in saline) or saline (control), the gastrocnemius muscles of mice were isolated from the lower legs for (immuno-) histological analyses. Hematoxylin and Eosin staining evidenced that RNase A treatment resulted in a higher degree of ischemic tissue damage. This was, however, associated with reduced angiogenesis, as evidenced by a reduced capillary/muscle fiber ratio. Moreover, RNase A treatment was associated with a significant reduction in leukocyte infiltration as shown by CD45+ (pan-leukocyte marker), Ly6G+ or MPO+ (neutrophils), MPO+/CitH3+ [neutrophil extracellular traps (NETs)], and CD68+ (macrophages) staining. CD68/MRC1 double staining revealed that RNase A treated mice showed a reduced percentage of M1-like polarized (CD68+/MRC1−) macrophages whereas the percentage of M2-like polarized (CD68+/MRC1+) macrophages was increased. Conclusion: In contrast to RNase 5, RNase A interferes with angiogenesis, which is linked to reduced leukocyte infiltration and NET formation

    Epstein–Barr virus-encoded microRNA miR-BART2 down-regulates the viral DNA polymerase BALF5

    MicroRNAs (miRNAs) have been implicated in sequence-specific cleavage, translational repression or deadenylation of specific target mRNAs resulting in post-transcriptional gene silencing. Epstein–Barr virus (EBV) encodes 23 miRNAs of unknown function. Here we show that the EBV-encoded miRNA miR-BART2 down-regulates the viral DNA polymerase BALF5. MiR-BART2 guides cleavage within the 3′-untranslated region (3′UTR) of BALF5 by virtue of its complete complementarity to its target. Induction of the lytic viral replication cycle results in a reduction of the level of miR-BART2 with a strong concomitant decrease of cleavage of the BALF5 3′UTR. Expression of miR-BART2 down-regulates the activity of a luciferase reporter gene containing the BALF5 3′UTR. Forced expression of miR-BART2 during lytic replication resulted in a 40–50% reduction of the level of BALF5 protein and a 20% reduction of the amount of virus released from EBV-infected cells. Our results are compatible with the notion that EBV-miR-BART2 inhibits transition from latent to lytic viral replication

    A picture paints a thousand words: Heart drawings reflect acute distress and illness perception and predict posttraumatic stress symptoms after acute myocardial infarction

    The aim of this study was to examine whether heart drawings of patients with acute myocardial infarction reflect acute distress symptoms and negative illness beliefs and predict posttraumatic stress symptoms 3 months post-myocardial infarction. In total, 84 patients aged over 18 years drew pictures of their heart. The larger the area drawn as damaged, the greater were the levels of acute distress (r = 0.36; p < 0.05), negative illness perceptions (r = 0.42, p < 0.05), and posttraumatic stress symptoms (r = 0.54, p < 0.01). Pain drawings may offer a tool to identify maladaptive cognitions and thus patients at risk of posttraumatic stress disorder

    Suppressed basal melting in the eastern Thwaites Glacier grounding zone

Much of the ice sheet within the catchment of Thwaites Glacier is grounded below sea level on bedrock that deepens inland4, making it susceptible to rapid and irreversible ice loss that could raise the global sea level by more than half a metre2,3,5. The rate and extent of ice loss, and whether it proceeds irreversibly, are set by the ocean conditions and basal melting within the grounding-zone region where Thwaites Glacier first goes afloat3,6, both of which are largely unknown. Here we show—using observations from a hot-water-drilled access hole—that the grounding zone of Thwaites Eastern Ice Shelf (TEIS) is characterized by a warm and highly stable water column with temperatures substantially higher than the in situ freezing point. Despite these warm conditions, low current speeds and strong density stratification in the ice–ocean boundary layer actively restrict the vertical mixing of heat towards the ice base7,8, resulting in strongly suppressed basal melting. Our results demonstrate that the canonical model of ice-shelf basal melting used to generate sea-level projections cannot reproduce observed melt rates beneath this critically important glacier, and that rapid and possibly unstable grounding-line retreat may be associated with relatively modest basal melt rates.Publisher PDFPeer reviewe

    Epizootic Emergence of Usutu Virus in Wild and Captive Birds in Germany

    This study aimed to identify the causative agent of mass mortality in wild and captive birds in southwest Germany and to gather insights into the phylogenetic relationship and spatial distribution of the pathogen. Since June 2011, 223 dead birds were collected and tested for the presence of viral pathogens. Usutu virus (USUV) RNA was detected by real-time RT-PCR in 86 birds representing 6 species. The virus was isolated in cell culture from the heart of 18 Blackbirds (Turdus merula). USUV-specific antigen was demonstrated by immunohistochemistry in brain, heart, liver, and lung of infected Blackbirds. The complete polyprotein coding sequence was obtained by deep sequencing of liver and spleen samples of a dead Blackbird from Mannheim (BH65/11-02-03). Phylogenetic analysis of the German USUV strain BH65/11-02-03 revealed a close relationship with strain Vienna that caused mass mortality among birds in Austria in 2001. Wild birds from lowland river valleys in southwest Germany were mainly affected by USUV, but also birds kept in aviaries. Our data suggest that after the initial detection of USUV in German mosquitoes in 2010, the virus spread in 2011 and caused epizootics among wild and captive birds in southwest Germany. The data also indicate an increased risk of USUV infections in humans in Germany

    High-Throughput Sequencing of Arabidopsis microRNAs: Evidence for Frequent Birth and Death of MIRNA Genes

    In plants, microRNAs (miRNAs) comprise one of two classes of small RNAs that function primarily as negative regulators at the posttranscriptional level. Several MIRNA genes in the plant kingdom are ancient, with conservation extending between angiosperms and the mosses, whereas many others are more recently evolved. Here, we use deep sequencing and computational methods to identify, profile and analyze non-conserved MIRNA genes in Arabidopsis thaliana. 48 non-conserved MIRNA families, nearly all of which were represented by single genes, were identified. Sequence similarity analyses of miRNA precursor foldback arms revealed evidence for recent evolutionary origin of 16 MIRNA loci through inverted duplication events from protein-coding gene sequences. Interestingly, these recently evolved MIRNA genes have taken distinct paths. Whereas some non-conserved miRNAs interact with and regulate target transcripts from gene families that donated parental sequences, others have drifted to the point of non-interaction with parental gene family transcripts. Some young MIRNA loci clearly originated from one gene family but form miRNAs that target transcripts in another family. We suggest that MIRNA genes are undergoing relatively frequent birth and death, with only a subset being stabilized by integration into regulatory networks

In this Letter we focus on the Click to view the MathML source channel which potentially holds spectroscopic information on the unbound nucleus 10Li. The experimental excitation energy spectrum and angular distribution are compared with CCBA calculations. These calculations clearly support the existence of a low-lying (s) virtual state, with a (negative) scattering length of the order asnot, vert, similar1324 fm and a p1/2 resonance with an energy of Ersimilar, equals0.38 MeV and a width of Γsimilar, equals0.2 MeV

    Differential cross section measurements for the production of a W boson in association with jets in proton–proton collisions at √s = 7 TeV

    Measurements are reported of differential cross sections for the production of a W boson, which decays into a muon and a neutrino, in association with jets, as a function of several variables, including the transverse momenta (pT) and pseudorapidities of the four leading jets, the scalar sum of jet transverse momenta (HT), and the difference in azimuthal angle between the directions of each jet and the muon. The data sample of pp collisions at a centre-of-mass energy of 7 TeV was collected with the CMS detector at the LHC and corresponds to an integrated luminosity of 5.0 fb[superscript −1]. The measured cross sections are compared to predictions from Monte Carlo generators, MadGraph + pythia and sherpa, and to next-to-leading-order calculations from BlackHat + sherpa. The differential cross sections are found to be in agreement with the predictions, apart from the pT distributions of the leading jets at high pT values, the distributions of the HT at high-HT and low jet multiplicity, and the distribution of the difference in azimuthal angle between the leading jet and the muon at low values.United States.
