    Le giunzioni metalliche incollate offrono diversi vantaggi nei confronti dei tradizionali collegamenti meccanici (rivettatura, bullonatura, ecc.), tuttavia richiedono un’estensiva caratterizzazione meccanica al fine di garantire un’adeguata affidabilità delle strutture in cui vengono inserite. In questo lavoro si presentano i risultati delle prove sperimentali e delle simulazioni numeriche condotte nell’ambito della riprogettazione di una giunzione rivettata di un componente reale, sfruttando la tecnologia dell’incollaggio. L’adesivo usato ù una resina epossidica bicomponente, caratterizzata attraverso una campagna di prove sperimentali e simulazioni numeriche, su provini di tipo single-lap e T-peel. Si presenta quindi un confronto, sia numerico che sperimentale, delle prestazioni meccaniche della giunzione rivettata con l’analoga giunzione incollata. I risultati hanno evidenziato un sostanziale miglioramento in termini di rigidezza e resistenza con l’uso della tecnica dell’incollaggio

    Genetic Association and Altered Gene Expression of CYBB in Multiple Sclerosis Patients

    Multiple sclerosis (MS) is a chronic neurological disorder characterized by inflammation, demyelination, and axonal damage. Increased levels of reactive oxygen species (ROS), produced by macrophages and leading to oxidative stress, have been implicated as mediators of demyelination and axonal injury in both MS and experimental autoimmune encephalomyelitis, the murine model of the disease. On the other hand, reduced ROS levels can increase susceptibility to autoimmunity. In this work, we screened for association with MS 11 single nucleotide polymorphisms (SNPs) and two microsatellite markers in the five genes (NCF1, NCF2, NCF4, CYBA, and CYBB) of the nicotinamide adenine dinucleotide phosphate (NADPH) oxidase (NOX2) system, the enzymatic pathway producing ROS in the brain and neural tissues, in 347 Finnish patients with MS and 714 unaffected family members. This analysis showed suggestive association signals for NCF1 and CYBB (lowest p = 0.038 and p = 0.013, respectively). Functional relevance for disease predisposition was further supported for the CYBB gene, by microarray analysis in CD4+/− mononuclear cells of 21 individuals from five Finnish multiplex MS families, as well as by real-time RT-PCRs performed on RNA extracted from peripheral blood mononuclear cells of an Italian replication cohort of 21 MS cases and 21 controls. Our results showed a sex-specific differential expression of CYBB, suggesting that this gene, and more in general the NOX2 system, deserve to be further investigated for their possible role in MS.Peer reviewe

    Microbial diversity of traditional Sicilian cheeses

    Traditional Sicilian cheeses are manufactured in small size farms with raw milk from animals of indigenous breeds and without the addition of starter cultures. In order to transform milk into cheese, the presence of lactic acid bacteria (LAB) is required. The main sources of desirable LAB are generally the milk, the rennet, the equipment used during processing and the dairy environment. In the last years, the microbial characterisation of traditional Sicilian cheeses, such as Caciocavallo Palermitano, Protected Designation of Origin (PDO) Pecorino Siciliano and PDO Vastedda della valle del BelĂŹce have been the object of different studies conducted by our research group. To this purpose, the aim of the present study was to describe the microbial population of traditional Sicilian cheeses. The analysis of the microbial diversity of these cheeses revealed the presence of several dairy LAB at dominant levels, while the undesired microorganisms, including coliforms and coagulase-positive staphylococci (CPS), were at very low densities. Overall, the presence of the pathogenic bacteria Listeria monocytogenes and Salmonella spp. was never detected. This may be due to the quality of milk, the optimal maintenance of wooden vats and on following good production practices. Furthermore, the technological characterisation of the LAB found in these cheeses and in raw materials showed interesting dairy properties, including acidification capacity, diacetyl formation, autolytic properties and the ability to inhibit undesired bacteria

    AGN Populations in Large Volume X-ray Surveys: Photometric Redshifts and Population Types found in the Stripe 82X Survey

    Multi-wavelength surveys covering large sky volumes are necessary to obtain an accurate census of rare objects such as high luminosity and/or high redshift active galactic nuclei (AGN). Stripe 82X is a 31.3 deg2^2 X-ray survey with ChandraChandra and XMMXMM-Newton observations overlapping the legacy Sloan Digital Sky Survey (SDSS) Stripe 82 field, which has a rich investment of multi-wavelength coverage from the ultraviolet to the radio. The wide-area nature of this survey presents new challenges for photometric redshifts for AGN compared to previous work on narrow-deep fields because it probes different populations of objects that need to be identified and represented in the library of templates. Here we present an updated X-ray plus multi-wavelength matched catalog, including SpitzerSpitzer counterparts, and estimated photometric redshifts for 5961 (96% of a total of 6181) X-ray sources, which have a normalized median absolute deviation, σnmad\sigma_{\rm nmad} = 0.06 and an outlier fraction, η\eta = 13.7%. The populations found in this survey, and the template libraries used for photometric redshifts, provide important guiding principles for upcoming large-area surveys such as eROSITAeROSITA and 3XMMXMM (in X-ray) and the Large Synoptic Survey Telescope (LSST; optical).Comment: Accepted for publication by The Astrophysical Journal (33 pages, 20 figures, 13 tables).     The 31 Deg2^2 Release of the Stripe 82 X-ray Survey: The Point Source Catalog

    We release the next installment of the Stripe 82 X-ray survey point-source catalog, which currently covers 31.3 deg2^2 of the Sloan Digital Sky Survey (SDSS) Stripe 82 Legacy field. In total, 6181 unique X-ray sources are significantly detected with {\it XMM-Newton} (>5σ>5\sigma) and {\it Chandra} (>4.5σ>4.5\sigma). This catalog release includes data from {\it XMM-Newton} cycle AO 13, which approximately doubled the Stripe 82X survey area. The flux limits of the Stripe 82X survey are 8.7×10−168.7\times10^{-16} erg s−1^{-1} cm−2^{-2}, 4.7×10−154.7\times10^{-15} erg s−1^{-1} cm−2^{-2}, and 2.1×10−152.1\times10^{-15} erg s−1^{-1} cm−2^{-2} in the soft (0.5-2 keV), hard (2-10 keV), and full bands (0.5-10 keV), respectively, with approximate half-area survey flux limits of 5.4×10−155.4\times10^{-15} erg s−1^{-1} cm−2^{-2}, 2.9×10−142.9\times10^{-14} erg s−1^{-1} cm−2^{-2}, and 1.7×10−141.7\times10^{-14} erg s−1^{-1} cm−2^{-2}. We matched the X-ray source lists to available multi-wavelength catalogs, including updated matches to the previous release of the Stripe 82X survey; 88\% of the sample is matched to a multi-wavelength counterpart. Due to the wide area of Stripe 82X and rich ancillary multi-wavelength data, including coadded SDSS photometry, mid-infrared {\it WISE} coverage, near-infrared coverage from UKIDSS and VHS, ultraviolet coverage from {\it GALEX}, radio coverage from FIRST, and far-infrared coverage from {\it Herschel}, as well as existing ∌\sim30\% optical spectroscopic completeness, we are beginning to uncover rare objects, such as obscured high-luminosity AGN at high-redshift.     Inhibition of Ubc13-mediated ubiquitination by GPS2 regulates multiple stages of B cell development

    Non-proteolytic ubiquitin signaling mediated by Lys63 ubiquitin chains plays a critical role in multiple pathways that are key to the development and activation of immune cells. Our previous work indicates that GPS2 (G-protein Pathway Suppressor 2) is a multifunctional protein regulating TNF signaling and lipid metabolism in the adipose tissue through modulation of Lys63 ubiquitination events. However, the full extent of GPS2-mediated regulation of ubiquitination and the underlying molecular mechanisms are unknown. Here, we report that GPS2 is required for restricting the activation of TLR and BCR signaling pathways and the AKT/FOXO1 pathway in immune cells based on direct inhibition of Ubc13 enzymatic activity. Relevance of this regulatory strategy is confirmed in vivo by B cell-targeted deletion of GPS2, resulting in developmental defects at multiple stages of B cell differentiation. Together, these findings reveal that GPS2 genomic and non-genomic functions are critical for the development and cellular homeostasis of B cells

    The circular RNA landscape in multiple sclerosis: Disease-specific associated variants and exon methylation shape circular RNA expression profile

    BACKGROUND: Circular RNAs (circRNAs) are a class of non-coding RNAs increasingly emerging as crucial actors in the pathogenesis of human diseases, including autoimmune and neurological disorders as multiple sclerosis (MS). Despite several efforts, the mechanisms regulating circRNAs expression are still largely unknown and the circRNA profile and regulation in MS-relevant cell models has not been completely investigated. In this work, we aimed at exploring the global landscape of circRNA expression in MS patients, also evaluating a possible correlation with their genetic and epigenetic background. METHODS: We performed RNA-seq experiments on circRNA-enriched samples, derived from peripheral blood mononuclear cells (PBMCs) of 10 MS patients and 10 matched controls and performed differential circRNA expression. The genetic background was evaluated using array genotyping, and an expression quantitative trait loci (eQTL) analysis was carried out. RESULTS: Expression analysis revealed 166 differentially expressed circRNAs in MS patients, 125 of which are downregulated. One of the top dysregulated circRNAs, hsa_circ_0007990, derives from the PGAP3 gene, encoding a protein relevant for the control of autoimmune responses. The downregulation of this circRNA was confirmed in two independent replication cohorts, suggesting its implementation as a possible RNA-based biomarker. The eQTL analysis evidenced a significant association between 89 MS-associated loci and the expression of at least one circRNA, suggesting that MS-associated variants could impact on disease pathogenesis by altering circRNA profiles. Finally, we found a significant correlation between exon methylation and circRNA expression levels, supporting the hypothesis that epigenetic features may play an important role in the definition of the cell circRNA pool. CONCLUSION: We described the circRNA expression profile of PBMCs in MS patients, suggesting that MS-associated variants may tune the expression levels of circRNAs acting as circ-QTLs , and proposing a role for exon-based DNA methylation in regulating circRNA expression

    The Hunt for Red Quasars : Luminous Obscured Black Hole Growth Unveiled in the Stripe 82 X-Ray Survey

    We present results of a ground-based near-infrared campaign with Palomar TripleSpec, Keck NIRSPEC, and Gemini GNIRS to target two samples of reddened active galactic nucleus (AGN) candidates from the 31 deg(2) Stripe 82 X-ray survey. One sample, which is similar to 89% complete to K 4, Vega). The fainter sample (K > 17, Vega) represents a pilot program to follow-up four sources from a parent sample of 34 that are not detected in the single-epoch SDSS catalog and have WISE quasar colors. All 12 sources are broad-line AGNs (at least one permitted emission line has an FWHM exceeding 1300 km s(-1)) and span a redshift range 0.59 0.5), and a greater percentage have high X-ray luminosities (L-X,L- full > 10(44) erg s(-1)). Such outflows and high luminosities may be consistent with the paradigm that reddened broad-line AGNs represent a transitory phase in AGN evolution as described by the major merger model for black hole growth. Results from our pilot program demonstrate proof of concept that our selection technique is successful in discovering reddened quasars at z > 1 missed by optical surveys.Peer reviewe

X-ray sources are matched to FIRST and Herschel sources using a nearest neighbor matching. Spectroscopic redshifts obtained from dedicated ground-based follow-up are also published in this catalog.
