    Nonlinear optical properties of a channel waveguide produced with crosslinkable ferroelectric liquid crystals

    A binary mixture of ferroelectric liquid crystals (FLCs) was used for the design of a channel waveguide. The FLCs possess two important functionalities: a chromophore with a high hyperpolarizability β\beta and photoreactive groups. The smectic liquid crystal is aligned in layers parallel to the glass plates in a sandwich geometry. This alignment offers several advantages, such as that moderate electric fields are sufficient to achieve a high degree of polar order. The arrangement was then permanently fixed by photopolymerization which yielded a polar network possessing a high thermal and mechanical stability which did not show any sign of degradation within the monitored period of several months. The linear and nonlinear optical properties have been measured and all four independent components of the nonlinear susceptibility tensor dˉ\bar d have been determined. The off-resonant dd-coefficients are remarkably high and comparable to those of the best known inorganic materials. The alignment led to an inherent channel waveguide for p-polarized light without additional preparation steps. The photopolymerization did not induce scattering sites in the waveguide and the normalized losses were less than 2 dB/cm.     True phase-matched second-harmonic generation in a waveguide of crosslinkable ferroelectric liquid crystals is demonstrated. These materials allow the formation of macroscopically polar structures whose order can be frozen by photopolymerization. Homeotropic alignment was chosen which offers decisive advantages compared to other geometries. All parameters contributing to the conversion efficiency are maximized by deliberately controlling the supramolecular arrangement.     Increase of the peroxidase activity of cytochrome c-550 by the interaction with detergents

    The effect was studied of detergents on the peroxidase activity of the heme-containing electron-transport protein, cytochrome c-550 from Paracoccus versutus (cytc550). Cytc550 does not interact with non-ionic or zwitterionic detergents, but its peroxidase activity is significantly enhanced in the presence of both anionic and cationic detergents. The increase in peroxidase activity is caused by (partial) unfolding of cytc550, resulting in weakening of the bond between the axial methionine ligand and the heme-iron. With sodium dodecyl sulfate (SDS) and dodecyltrimethylammonium bromide (DTAB), the activity increases roughly 10- and 100-fold, respectively. The detergent concentration required to enhance the peroxidase activity of cytc550 consistently coincides with the critical micellisation concentration (CMC). When cytc550 carries substantial opposite charge to that of the detergent headgroup, the protein-detergent complex is formed at lower detergent concentration than the critical micellisation concentration, i.e. at acid pH for anionic detergent and at alkaline pH for cationic detergent. It is concluded that in the presence of ionic detergents, cytc550 can acquire considerable peroxidase activity. This may be exploited by applying cytc550 as an oxidative catalyst in detergent-rich environments, where regular peroxidases rapidly lose their catalytic potential. (C) 2003 Elsevier B.V.     Exosomes released by EBV-infected nasopharyngeal carcinoma cells convey the viral Latent Membrane Protein 1 and the immunomodulatory protein galectin 9

    BACKGROUND: Nasopharyngeal carcinomas (NPC) are consistently associated with the Epstein-Barr virus (EBV). Their malignant epithelial cells contain the viral genome and express several antigenic viral proteins. However, the mechanisms of immune escape in NPCs are still poorly understood. EBV-transformed B-cells have been reported to release exosomes carrying the EBV-encoded latent membrane protein 1 (LMP1) which has T-cell inhibitory activity. Although this report suggested that NPC cells could also produce exosomes carrying immunosuppressive proteins, this hypothesis has remained so far untested. METHODS: Malignant epithelial cells derived from NPC xenografts – LMP1-positive (C15) or negative (C17) – were used to prepare conditioned culture medium. Various microparticles and vesicles released in the culture medium were collected and fractionated by differential centrifugation. Exosomes collected in the last centrifugation step were further purified by immunomagnetic capture on beads carrying antibody directed to HLA class II molecules. Purified exosomes were visualized by electron microscopy and analysed by western blotting. The T-cell inhibitory activities of recombinant LMP1 and galectin 9 were assessed on peripheral blood mononuclear cells activated by CD3/CD28 cross-linking. RESULTS: HLA-class II-positive exosomes purified from C15 and C17 cell supernatants were containing either LMP1 and galectin 9 (C15) or galectin 9 only (C17). Recombinant LMP1 induced a strong inhibition of T-cell proliferation (IC50 = 0.17 nM). In contrast recombinant galectin 9 had a weaker inhibitory effect (IC50 = 46 nM) with no synergy with LMP1. CONCLUSION: This study provides the proof of concept that NPC cells can release HLA class-II positive exosomes containing galectin 9 and/or LMP1. It confirms that the LMP1 molecule has intrinsic T-cell inhibitory activity. These findings will encourage investigations of tumor exosomes in the blood of NPC patients and assessment of their effects on various types of target cells

    A search for the standard model Higgs boson in the H to ZZ to 2l 2nu decay channel, where l = e or mu, in pp collisions at a center-of-mass energy of 7 TeV is presented. The data were collected at the LHC, with the CMS detector, and correspond to an integrated luminosity of 4.6 inverse femtobarns. No significant excess is observed above the background expectation, and upper limits are set on the Higgs boson production cross section. The presence of the standard model Higgs boson with a mass in the 270-440 GeV range is excluded at 95% confidence level.Comment: Submitted to JHE

    Measurement of the t t-bar production cross section in the dilepton channel in pp collisions at sqrt(s) = 7 TeV

    The t t-bar production cross section (sigma[t t-bar]) is measured in proton-proton collisions at sqrt(s) = 7 TeV in data collected by the CMS experiment, corresponding to an integrated luminosity of 2.3 inverse femtobarns. The measurement is performed in events with two leptons (electrons or muons) in the final state, at least two jets identified as jets originating from b quarks, and the presence of an imbalance in transverse momentum. The measured value of sigma[t t-bar] for a top-quark mass of 172.5 GeV is 161.9 +/- 2.5 (stat.) +5.1/-5.0 (syst.) +/- 3.6(lumi.) pb, consistent with the prediction of the standard model.Comment: Replaced with published version.     Search for anomalous t t-bar production in the highly-boosted all-hadronic final state

    A search is presented for a massive particle, generically referred to as a Z', decaying into a t t-bar pair. The search focuses on Z' resonances that are sufficiently massive to produce highly Lorentz-boosted top quarks, which yield collimated decay products that are partially or fully merged into single jets. The analysis uses new methods to analyze jet substructure, providing suppression of the non-top multijet backgrounds. The analysis is based on a data sample of proton-proton collisions at a center-of-mass energy of 7 TeV, corresponding to an integrated luminosity of 5 inverse femtobarns. Upper limits in the range of 1 pb are set on the product of the production cross section and branching fraction for a topcolor Z' modeled for several widths, as well as for a Randall--Sundrum Kaluza--Klein gluon.
