    Parental diet, pregnancy outcomes and offspring health:metabolic determinants in developing oocytes and embryos

    The periconceptional period, embracing the terminal stages of oocyte growth and post-fertilisation development up to implantation, is sensitive to parental nutrition. Deficiencies or excesses in a range of macro- and micronutrients during this period can lead to impairments in fertility, fetal development and long-term offspring health. Obesity and genotype-related differences in regional adiposity are associated with impaired liver function and insulin resistance, and contribute to fatty acid-mediated impairments in sperm viability and oocyte and embryo quality, all of which are associated with endoplasmic reticulum stress and compromised fertility. Disturbances to maternal protein metabolism can elevate ammonium concentrations in reproductive tissues and disturb embryo and fetal development. Associated with this are disturbances to one-carbon metabolism, which can lead to epigenetic modifications to DNA and associated proteins in offspring that are both insulin resistant and hypertensive. Many enzymes involved in epigenetic gene regulation use metabolic cosubstrates (e.g. acetyl CoA and S-adenosyl methionine) to modify DNA and associated proteins, and so act as 'metabolic sensors' providing a link between parental nutritional status and gene regulation. Separate to their genomic contribution, spermatozoa can also influence embryo development via direct interactions with the egg and by seminal plasma components that act on oviductal and uterine tissues

    Current achievements and future research directions in ovarian tissue culture, in vitro follicle development and transplantation: implications for fertility preservation

    Female cancer patients are offered 'banking' of gametes before starting fertility-threatening cancer therapy. Transplants of fresh and frozen ovarian tissue between healthy fertile and infertile women have demonstrated the utility of the tissue banked for restoration of endocrine and fertility function. Additional methods, like follicle culture and isolated follicle transplantation, are in development. Specialist reproductive medicine scientists and clinicians with complementary expertise in ovarian tissue culture and transplantation presented relevant published literature in their field of expertise and also unpublished promising data for discussion. As the major aims were to identify the current gaps prohibiting advancement, to share technical experience and to orient new research, contributors were allowed to provide their opinioned expert views on future research. Normal healthy children have been born in cancer survivors after orthotopic transplantation of their cryopreserved ovarian tissue. Longevity of the graft might be optimized by using new vitrification techniques and by promoting rapid revascularization of the graft. For the in vitro culture of follicles, a successive battery of culture methods including the use of defined media, growth factors and three-dimensional extracellular matrix support might overcome growth arrest of the follicles. Molecular methods and immunoassay can evaluate stage of maturation and guide adequate differentiation. Large animals, including non-human primates, are essential working models. Experiments on ovarian tissue from non-human primate models and from consenting fertile and infertile patients benefit from a multidisciplinary approach. The new discipline of oncofertility requires professionalization, multidisciplinarity and mobilization of funding for basic and translational research

    From crisis to meaning: creativity in the Biblical narrative of Eve and the inversion by F. Kafka

    Abstract. The possibilities of creativity in human life depend on discovering the meaning of life. Franz Kafka’s story Before the Law evokes how every attempt at finding this meaning, represented as the law, comes with a crisis and can result in failure. The secret of human existence is all about intimate personal encounters. The other than myself, which is also deeply present inside myself, is not the impersonal thing that it is oftentimes made out to be. In an already published article, we presented a close-reading of Kafka’s text. We only refer to the results in the first paragraph. In this article, we read the biblical narrative on Adam and Eve as a text that is structured by the same economy – the intertwinedness of the I-Thou relation with the I-It relation. But in opposition to the Kafka narrative, the crisis is overcome and results in a new creative perspective on the human condition: the discovery of the ultimate intersubjective meaning of life. The meaning makes love and labour possible. It means that the creativity of becoming human is the result of interaction. The context of our research is a long project on developing a philosophical reading of Biblical texts that was developed at the KU Leuven, Leuven, Belgium. The methodology is inspired by the Talmudic readings of Emmanuel Levinas and by the philosophical interpretations of the Bible of Paul Ricœur. In this way, we introduce a non-confessional way of understanding religious texts on the creativity of becoming human.
