The purpose of this study was to analyze the usefulness of PCR for the detection of leptospiral carriers in sheep under tropical field conditions. Two flocks, previously reported as seroreactive (A) and seronegative (B), were selected for this study. From those, the totality of animals of each flock, urine and vaginal fluid (VF)/semen were collected for bacteriological culture and PCR, as well as serum samples for serology. Serology confirmed the previous status of the two flocks. Culture was negative for all the samples. In PCR, animals of Flock A presented 26.7% (VF), 33.3% (semen) and 38.9% (urine) of positivity. Flock B presented 40.0% (VF), 33.3% (semen) and 5.6% (urine) of positivity by PCR. In conclusion, PCR was important to identify carriers of leptospires, including animals from a seronegative flock, what reinforces the advantages of the usage of this tool for the detection of carriers in sheep as part of control programs of leptospirosis under tropical field conditions.O objetivo do presente estudo foi analisar a aplicabilidade da PCR na detecção de ovinos carreadores de Leptospira em ambiente tropical.
The suspension of classroom classes from March 2020, determined by the Portuguese government in response to the CoViD-19 pandemic, imposed on educational institutions an unprecedented urgency of innovative pedagogical practices facilitated by technology. Among the many challenges, teachers had to adapt and reinvent their teaching strategies and methodologies. Having additional difficulties such as the lack of access to technologies that facilitate online classes and a concern about the possible decrease in the quality of learning due to the sudden switch to emergency remote learning (ERE). This communication proposal is an integral part of a broader study, still in progress, and from which we will present the results regarding the use of technologies for synchronous and asynchronous communication, in Portuguese higher education institutions, during the period of social confinement. The main objective of the investigation was to understand how higher education professors and students experienced the digital transition to the ERE, the pedagogical practices and the communication strategies adopted during this period. The investigation used mixed methods and, in order to respond to the proposed objectives and describe the pedagogical practices implemented and the communication strategies, specific instruments were built for data collection. A questionnaire was developed, aimed at students, and interviews aimed at both students and teachers, focusing on the technological, pedagogical and emotional dimensions, as well as on the evaluation of the experience. Data collection was carried out after the first confinement, at the end of the first semester of 2020, and took place in eight Portuguese higher education institutions (four Universities and four Polytechnic Institutes). In this communication, we will present a preliminary analysis of the results obtained and that are related to the pedagogical strategies adopted and the use of technology in synchronous and asynchronous online communication. We will also discuss the impacts of remote teaching on teaching work, evaluating whether they can bring positive and/or negative consequences to the pedagogical activity.info:eu-repo/semantics/publishedVersio

Integrative transcriptomic and metabolomic approaches to unravel the resistance profile of Kawisari coffee against Hemileia vastatrix
29th Conference of Association for the Science and Information on Coffee, 11 Sept. - 14 Sept. 2023 Hanoi, Vietnaminfo:eu-repo/semantics/publishedVersio
Characterization of the transcriptional signatures associated with resistance and susceptibility to Hemileia vastatrix in the Kawisari coffee hybrid
Coffee leaf rust (CLR), a disease caused by the biotrophic fungus Hemileia vastatrix (Hv), is the main threat to the worldwide production of Arabica
coffee. The gradual breakdown of resistance in coffee varieties in the last years has highlighted the need for novel sources of resistance to CLR.
This work aimed to unveil the cellular and molecular resistance profile of the Kawisari hybrid (C. arabica x C. liberica), a genotype used as a
resistance donor in Arabica breeding programs in India. This coffee genotype was inoculated with two Hv races that triggered either resistance or
susceptibility. Progress of infection was monitored using light microscopy. Simultaneously, we conducted a time-course RNA-seq characterization
of the transcriptional responses. The microscopic studies showed that the post-haustorial resistance of Kawisari was associated with the
hypersensitive response, accumulation of phenolic-like compounds and haustorium encasement with callose. The transcriptomic analysis suggest
the downregulation of host primary metabolism genes at the early onset of infection, followed later by activation of genes functionally associated
with multiple plant defense responses, including salicylic acid and jasmonate hormonal signaling. Resistance was also accompanied by the
differential regulation of genes associated with phenylpropanoid metabolism and lignin biosynthesis. Our results, further validated by qPCR,
provide important new insight into the molecular mechanisms underpinning resistance against CLR in this coffee genotype.Foundation for Science and Technology (FCT) and FEDER funds through PORNorte under the project CoffeeRES PTDC/ASPPLA/

Morphological and Postural changes in the foot during pregnancy and puerperium : a longitudinal study
The aim of this study is to observe the morphological and postural changes to the foot that take place during pregnancy and the puerperium. Method: In this descriptive, observational, longitudinal study, we analysed 23 pregnant women, with particular attention to morphological and postural aspects of the foot, at three time points during and after pregnancy: in weeks 9-13 of gestation, weeks 32-35 of gestation and weeks 4-6 after delivery. The parameters considered were changes in foot length, the Foot Posture Index (FPI) and the Hernández Corvo Index, which were analysed using a pedigraph and taking into account the Body Mass Index (BMI). The same procedure was conducted in each review. Results: The statistical analyses obtained for each foot did not differ significantly between the three measurement times. A pronator-type footprint was most frequently observed during the third trimester of pregnancy; it was predominantly neutral during the postpartum period. Statistically significant differences between the measurement times were obtained in the right foot for cavus vs. neutral foot type (between the first and third trimesters and also between the first trimester and the puerperium) (in both cases, p < 0.0001). Conclusions: Foot length increases in the third trimester and returns to normal in the puerperium. According to FPI findings, the third trimester of pregnancy is characterised by pronation, while the posture returns to neutrality during the postpartum period. During pregnancy, the plantar arch flattens, and this persists during the puerperium. The incidence of cavus foot increases significantly in the third trimester and in the puerperium
