    Influence of drought stress and PGPR inoculation on essential oil yield and volatile organic compound emissions in Mentha piperita

    Considering that inoculation with beneficial rhizobacteria promotes biomass and secondary metabolite biosynthesis and also the fact that drought stress can increase their production, we determined the effects when both of these situations were combined in EO yield. In addition, the levels of endogenous phytohormones and the gene expression of enzymes related to monoterpenes biosynthesis were measured. The experimental results showed that inoculation with PGPR in stressed plants produced the same effects on trichome density, the EO main components and total yield as in plants only inoculated or with moderate stress alone. In addition, the VOC emissions of inoculated stressed plants did not vary the release amount in relation to uninoculated stressed ones. The results observed have suggested a way of improving aromatic plant productivity, particularly that of peppermint, by managing drought stress through the inoculation of plants with PGPR. This inoculation under moderate drought stress is appropriate when the purpose of the crops is to obtain plants with a high secondary metabolites yield.Fil: Chiappero, Julieta.     Volatile organic compounds from rhizobacteria increase the biosynthesis of secondary metabolites and improve the antioxidant status in mentha Piperita L. Grown under salt stress

    Salinity is a major abiotic stress factor that affects crops and has an adverse effect on plant growth. In recent years, there has been increasing evidence that microbial volatile organic compounds (mVOC) play a significant role in microorganism plant interactions. In the present study, we evaluated the impact of microbial volatile organic compounds (mVOC) emitted by Bacillus amyloliquefaciens GB03 on the biosynthesis of secondary metabolites and the antioxidant status in Mentha piperita L. grown under 0, 75 and 100 mM NaCl. Seedlings were exposed to mVOCs, avoiding physical contact with the bacteria, and an increase in NaCl levels produced a reduction in essential oil (EO) yield. Nevertheless, these undesirable effects were mitigated in seedlings treated with mVOCs, resulting in an approximately a six-fold increase with respect to plants not exposed to mVOCs, regardless of the severity of the salt stress. The main components of the EOs, menthone, menthol, and pulegone, showed the same tendency. Total phenolic compound (TPC) levels increased in salt-stressed plants but were higher in those exposed to mVOCs than in stressed plants without mVOC exposure. To evaluate the effect of mVOCs on the antioxidant status from salt-stressed plants, the membrane lipid peroxidation was analyzed. Peppermint seedlings cultivated under salt stress and treated with mVOC showed a reduction in malondialdehyde (MDA) levels, which is considered to be an indicator of lipid peroxidation and membrane damage, and had an increased antioxidant capacity in terms of DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity in relation to plants cultivated under salt stress but not treated with mVOCs.     How do plants protect themselves from insects? A practical laboratory exercise to illustrate the defence mechanisms of the plant through secondary metabolites

    Despite the growing awareness of the importance of plant secondary metabolites in insect-plant interactions, undergraduate degree content in agronomy and biology generally does not provide a clear concept to students in relation to secondary metabolite induction of plant defences, implying that students do not obtain a good understanding of the secondary metabolism or its functions. To address this deficiency, we have designed a practical exercise where students determine the phytochemical induction of secondary metabolites in aromatic plants subjected to herbivory. This approach involves an experimental laboratory class in which students evaluate the phenolic compounds and main essential oil compound induction in peppermint damaged by an armyworm. By the end of this exercise, based on the results and findings, students will: have a better comprehension of plant defence responses to herbivores; be able to illustrate the consequences of insect herbivory in relation to plant secondary metabolites induction; acquire lab skills related to the use of a spectrophotometer; be able to understand and analyse a GC chromatogram report. Authentic research experiences in the classroom are considered valuable elements for promoting science at undergraduate level, as well as providing motivation for the student and linking research with teaching.Fil: Palermo, Tamara Belen.     Analysis of Plant Growth-Promoting Effects of Fluorescent Pseudomonas Strains Isolated from Mentha piperita Rhizosphere and Effects of Their Volatile Organic Compounds on Essential Oil Composition

    Many species or strains of the genus Pseudomonas have been characterized as plant growth promoting rhizobacteria (PGPR). We used a combination of phenotypic and genotypic techniques to analyze the community of fluorescent Pseudomonas strains in the rhizosphere of commercially grown Mentha piperita (peppermint). Biochemical techniques, Amplified rDNA Restriction Analysis (ARDRA), and 16S rRNA gene sequence analysis revealed that the majority of the isolated native fluorescent strains were P. putida. Use of two Repetitive Sequence-based PCR (rep-PCR) techniques, BOX-PCR and ERIC-PCR, allowed us to evaluate diversity among the native strains and to more effectively distinguish among them. PGPR activity was tested for the native strains and reference strain P. fluorescens WCS417r. Micropropagated M. piperita plantlets were exposed to microbial volatile organic compounds (mVOCs) emitted by the bacterial strains, and plant biomass parameters and production of essential oils (EOs) were measured. mVOCs from 11 of the native strains caused an increase in shoot fresh weight. mVOCs from three native strains (SJ04, SJ25,SJ48) induced changes in M. pierita EO composition. The mVOCs caused a reduction of metabolites in the monoterpene pathway, for example menthofuran, and an increase in menthol production. Menthol production is the primary indicator of EO quality. The mVOCs produced by native strains SJ04, SJ25,SJ48 and strain WCS417r were analyzed. The obtained mVOC chromatographic profiles were unique for each of the three native strains analyzed, containing varying hydrocarbon, aromatic, and alogenic compounds. The differential effects of the strains were most likely due to the specific mixtures of mVOCs emitted by each strain, suggesting a synergistic effect occurs among the compounds present

    Improving phenolic total content and monoterpene in mentha x piperita by using salicylic acid or methyl jasmonate combined with rhizobacteria inoculation

    The effects of plant inoculation with plant growth-promoting rhizobacteria (PGPR) and those resulting from the exogenous application of salicylic acid (SA) or methyl jasmonte (MeJA) on total phenolic content (TPC) and monoterpenes in Mentha x piperita plants were investigated. Although the PGPR inoculation response has been studied for many plant species, the combination of PGPR and exogenous phytohormones has not been investigated in aromatic plant species. The exogenous application of SA produced an increase in TPC that, in general, was of a similar level when applied alone as when combined with PGPR. This increase in TPC was correlated with an increase in the activity of the enzyme phenylalanine ammonia lyase (PAL). Also, the application of MeJA at different concentrations in combination with inoculation with PGPR produced an increase in TPC, which was more relevant at 4 mM, with a synergism effect being observed. With respect to the main monoterpene concentrations present in peppermint essential oil (EO), it was observed that SA or MeJA application produced a significant increase similar to that of the combination with rhizobacteria. However, when plants were exposed to 2 mM MeJA and inoculated, an important increase was produced in the concentration on menthol, pulegone, linalool, limonene, and menthone concentrations. Rhizobacteria inoculation, the treatment with SA and MeJA, and the combination of both were found to affect the amount of the main monoterpenes present in the EO of M. piperita. For this reason, the expressions of genes related to the biosynthesis of monoterpene were evaluated, with this expression being positively affected by MeJA application and PGPR inoculation, but was not modified by SA application. Our results demonstrate that MeJA or SA application combined with inoculation with PGPR constitutes an advantageous management practice for improving the production of secondary metabolites from M. piperita.Fil: Cappellari, Lorena del Rosario.     Biofilm-Forming Ability of Phytopathogenic Bacteria: A Review of its Involvement in Plant Stress

    Phytopathogenic bacteria not only affect crop yield and quality but also the environment. Understanding the mechanisms involved in their survival is essential to develop new strategies to control plant disease. One such mechanism is the formation of biofilms; i.e., microbial communities within a three-dimensional structure that offers adaptive advantages, such as protection against unfavorable environmental conditions. Biofilm-producing phytopathogenic bacteria are difficult to manage. They colonize the intercellular spaces and the vascular system of the host plants and cause a wide range of symptoms such as necrosis, wilting, leaf spots, blight, soft rot, and hyperplasia. This review summarizes up-to-date information about saline and drought stress in plants (abiotic stress) and then goes on to focus on the biotic stress produced by biofilm-forming phytopathogenic bacteria, which are responsible for serious disease in many crops.     The role of plant growth promoting bacteria on arsenic removal: a review of existing perspectives

    Phytobial remediation is an innovative tool that uses plants and microbes to mitigate Arsenic (As) contamination of the environment. Recently, plant growth-promoting bacteria (PGPB) that assists phytoremediation has been highly touted for both improving plant metal tolerance and promoting plant growth while achieving the goal of large-scale removal of As. This review focuses on the PGPB characteristics influencing plants and the mechanisms in which they function to overcome/lessen As-induced adversities. Several recent examples of mechanisms responsible for increasing the availability of As to plants and coping with As stresses facilitated by PGPB will be reviewed. Although drawbacks to phytoremediation have been reported, encouraging results have been developed with regular monitoring. Introducing PGPB-assisted phytoremediation of As in a field requires an assessment of the environmental effects of PGPB, especially with respect to the impacts on indigenous bacteria

    Microbial Volatile Organic Compounds Produced by Bacillus amyloliquefaciens GB03 Ameliorate the Effects of Salt Stress in Mentha piperita Principally Through Acetoin Emission

    We investigated the effects of microbial volatile organic compounds (mVOC) emitted by Bacillus amyloliquefaciens GB03on Mentha piperita growing under different levels of NaCl stress, by evaluating their growth-promoting potential and abilityto increase salt tolerance effects. Plants were exposed to bacterial VOCs without having any physical contact with therhizobacteria. The VOCs emitted by the rhizobacteria (mVOCs) were analyzed using SPME fibers. An increase in the levelof salt concentration led to a decrease in plant growth. However, these negative effects of salinity were inhibited in theplants exposed to mVOCs. Plants grown in a saline media and exposed to GB03 VOCs had significantly better morphologicalcharacteristics and higher total chlorophyll content compared to controls. The level of endogenous jasmonic acid (JA),salicylic acid, and abscisic acid increased in salt-stressed plants compared to controls. The level of JA did not show anychange in plants grown in a saline media either exposed to mVOCs or not. In contrast, the amount of salicylic acid increasedremarkably in salt-stressed plants exposed to mVOCs compared to controls (salt-stressed plants not exposed to mVOCs),but the levels of abscisic acid decreased in salt-stressed plants exposed to mVOCs. The chromatographic analyses of themVOCs produced by salt-stressed GB03 bacteria were similar, regardless of the concentration of salt in the media wherethe bacteria were grown, although it was observed that the relative percentage of acetoin increased with salt concentration.After determining that acetoin was the main VOCs compound, we exposed plants to acetoin, which demonstrated that acetoincaused similar effects on plants grown under salt stress conditions as those exposed to GB03 mVOCs.     As salt stress has a negative impact on plant growth and crop yield, it is very important to identify and develop any available biotechnology which can improve the salt tolerance of plants. Inoculation with plant-growth-promoting rhizobacteria (PGPR) is a proven environmentally friendly biotechnological resource for increasing the salt stress tolerance of plants and has a potential in-field application. In addition, bacterial volatile organic compounds (mVOCs) are signal molecules that may have beneficial roles in the soil–plant–microbiome ecosystem. We investigated the effects of mVOCs emitted by Pseudomona putida SJ46 and SJ04 on Mentha piperita grown under different levels of NaCl stress by evaluating their growth-promoting potential and capacity to increase salt tolerance effects. Furthermore, we evaluated under control and salt stress conditions the biocontrol ability of VOCs emitted by both these strains to inhibit the growth of Alternaria alternata and Sclerotium rolfsii. The VOCs emitted by both strains under control conditions did not lead to an significant improvement in peppermint growth. However, under salt stress conditions (75 or 100 mM NaCl), an amelioration of its physiological status was observed, with this effect being greater at 100 mM NaCl. This led to an enhancement of the number of leaves and nodes and, increased the shoot fresh and root dry weight by approximately twice in relation to control stressed plants. Moreover, the VOCs released by the two bacteria grown in control or saline media showed a significant reduction in the mycelial growth of A. alternata. In contrast, S. rolfsii growth was reduced 40% by the mVOCs released only under control conditions, with no effects being observed under salt stress. We also explored the composition of the bacterial volatile profiles by means of a solid-phase microextraction/gas chromatography–mass spectrometry (SPME/GC–MS) analysis. From the headspace of SJ46, three VOCs were identified: n-octanol, decane and tetradecane. The emission of SJ04 had the same chromatographic profile, with the addition of two more compounds: 1-(N-phenyl carbamyl)-2-morpholino cyclohexene and tridecane. Only compounds that were not present in the headspace of the control groups were recorded. The salt stress conditions where the bacteria were grown did not qualitatively modify the mVOC emissions. Taken together, our results suggest that plant-associated rhizobacterial VOCs play a potentially important role in modulating plant salt tolerance and reducing fungal growth. Thus, biological resources represent novel tools for counteracting the deleterious effects of salt stress and have the potential to be exploited in sustainable agriculture. Nevertheless, future studies are necessary to investigate technological improvements for bacterial VOC application under greenhouse and open field conditions
