    Antimicrobial resistance and virulence factors in Escherichia coli from swedish dairy calves

    <p>Abstract</p> <p>Background</p> <p>In Sweden, knowledge about the role of enteropathogenic <it>Escherichia coli </it>in neonatal calf diarrhea and the occurrence of antimicrobial resistance in <it>E. coli </it>from young calves is largely unknown. This has therapeutic concern and such knowledge is also required for prudent use of antimicrobials.</p> <p>Methods</p> <p>In a case control study <it>Esherichia coli </it>isolated from faecal samples from dairy calves were phenotyped by biochemical fingerprinting and analyzed for virulence genes by PCR. Antimicrobial susceptibility was tested by determination of minimum inhibitory concentration (MIC). Farm management data were collected and Fisher's exact test and univariable and multivariable logistic regression analysis were performed.</p> <p>Results</p> <p>Of 95 <it>E. coli </it>tested for antimicrobial susceptibility 61% were resistant to one or more substances and 28% were multi-resistant. The virulence gene F5 (K99) was not found in any isolate. In total, 21 out of 40 of the investigated virulence genes were not detected or rarely detected. The virulence genes <it>espP, irp</it>, and <it>fyuA </it>were more common in resistant <it>E. coli </it>than in fully susceptible isolates (<it>P </it>< 0.05). The virulence gene <it>terZ </it>was associated with calf diarrhea (<it>P </it>≤ 0.01).</p> <p>The participating 85 herds had a median herd size of 80 lactating cows. Herds with calf diarrhea problems were larger (> 55 cows; P < 0.001), had higher calf mortality (P ≤ 0.01) and calf group feeders were more in use (<it>P </it>< 0.05), compared to herds without calf diarrhea problems.</p> <p>There was no association between calf diarrhea and diversity of enteric <it>E. coli</it>.</p> <p>Conclusions</p> <p>Antimicrobial resistance was common in <it>E. coli </it>from pre-weaned dairy calves, occurring particularly in calves from herds experiencing calf diarrhea problems. The results indicate that more factors than use of antimicrobials influence the epidemiology of resistant <it>E. coli</it>.</p> <p>Enteropathogenic <it>E. coli </it>seems to be an uncommon cause of neonatal calf diarrhea in Swedish dairy herds. In practice, calf diarrhea should be regarded holistically in a context of infectious agents, calf immunity, management practices etc. We therefore advice against routine antimicrobial treatment and recommend that bacteriological cultures, followed by testing for antimicrobial susceptibility and for virulence factors, are used to guide decisions on such treatment.</p

    Some important Ectoparasites of Alpaca (Vicugna pacos) and Llama (Lama glama)

    Abstract The most important ectoparasites infesting/infecting alpaca (Vicugna pacos) and llama (Lama glama) are reviewed. The clinical manifestation and the diagnosis of the different parasitic infections/infestations of Sarcoptes scabiei, Chorioptes sp., Psoroptes sp., Damalinia (Bovicola) breviceps and Microthoracius spp. are described as well as therapies against them. Demonstrating S. scabiei and Chorioptes sp. with available diagnostic methods are challengingly often due to the relatively small numbers of mites that may elicit clinical disease. In Chorioptes sp. infestations it has been shown that alpacas are often subclinically infested. Predilection places are between the toes and in the axillae. The variable response to modern acaricidal treatments emphazises the need of more evidence based studies. The lack of lanolin in the fibres of South American camelids may explain the poor response to topical applications of modern insecticidal/acaricidal products used on other animals. Pharmacokinetic studies of such substances are limited. Few products are licenced although several products that are used and are available for other animals are used off-label. Applying a combination of systemic and topical treatments may produce optimal results. The need to apply treatments against the mange mites more frequently and with higher dosages of some of the acaricidal substances than recommended for other livestock is indicated. Lice infestations are often easier to deal with. Systemic treatment should be applied against suckling lice and topical against the biting lice. All animals in affected herds should be treated at the same time and stringent biosecurity measures following treatment is recommended to avoid re-infections/infestations

    Extenders for alpaca epididymal spermatozoa: Comparison of INRA96 and andromed

    Artificial insemination would be a useful technique for alpaca breeders to use as an aid to breeding to increase fleece quality. The technique, however, is not well developed in alpacas, partly because of the viscous nature of their seminal plasma. Castration conducted for husbandry purposes can provide a source of epididymal spermatozoa to test semen extenders or handling regimens, thus circumventing the problem of the viscous ejaculate. In this experiment, two semen extenders (Andromed and INRA96) developed for other species (bovine and equine, respectively) were tested with alpaca spermatozoa derived from the cauda epididymis. Sperm total motility (mean ± SEM A: 29.1 ± 4.8 % compared with I: 35.4 ± 4.8 %; NS), membrane integrity (A: 58 ± 9% compared with I: 56 ± 9%; NS) and acrosome integrity (A: 65 ± 7% compared with I: 54 ± 7%; NS) were not different between the two extenders. Progressive motility with use of INRA96 was greater after incubating for 30 min than after incubating for 10 min (35 ± 4% vs. 12 ± 4%, respectively; P = 0.03). In conclusion, viable epididymal spermatozoa could be extracted from the castrated organs after overnight transport. There were no differences in sperm quality between the two extenders; therefore, it appears that either extender could be used for alpaca spermatozoa. These results could help in the development of a technique for artificial insemination in alpacas

    Testicular length as an indicator of the onset of sperm production in alpacas under Swedish conditions

    Background: The popularity of alpacas (Vicugna pacos) is increasing in Sweden as well as in other countries; however, knowledge about optimal management practices under Swedish conditions is still limited. The wide age range reported when the onset of puberty can occur, between 1 and 3years of age, makes management decisions difficult and may be influenced by the conditions under which the alpacas are kept. The aim of this study was to find out when Swedish alpacas can be expected to start producing sperm, by using testicular length and body condition score as a more precise indirect indicator than age. Results: This study suggests that animals with a testicular length ≥3.8cm would be producing sperm; however, if it is crucial to know that there is no sperm production for management purposes, the threshold level for testicular length used to differentiate between sperm-producing and non-sperm producing animals should be ≤1.6cm instead. If only one variable is considered, testicular length appears to better than age alone to predict sperm production. Body condition score together with testicular length explains the individual onset of puberty and better guide management recommendations. Conclusions: Using a combination of these parameters (testicular length, body condition score and age) as a tool for decision making for alpaca husbandry under Swedish conditions is suggested

    Prevalence of footrot in Swedish slaughter lambs

    <p>Abstract</p> <p>Background</p> <p>Footrot is a world-wide contagious disease in sheep and goats. It is an infection of the epidermis of the interdigital skin, and the germinal layers of the horn tissue of the feet. The first case of footrot in Swedish sheep was diagnosed in 2004. Due to difficulties in distinguishing benign footrot from early cases of virulent footrot and because there is no possibility for virulence testing of strains of <it>Dichelobacter nodosus </it>in Sweden, the diagnosis is based of the presence or absence of clinical signs of footrot in sheep flocks. Ever since the first diagnosed case the Swedish Animal Health Service has worked intensively to stop the spread of infection and control the disease at flock level. However, to continue this work effectively it is important to have knowledge about the distribution of the disease both nationally and regionally. Therefore, the aims of this study were to estimate the prevalence of footrot in Swedish lambs at abattoirs and to assess the geographical distribution of the disease.</p> <p>Methods</p> <p>A prevalence study on footrot in Swedish lambs was performed by visual examination of 2000 feet from 500 lambs submitted from six slaughter houses. Each foot was scored according to a 0 to 5 scoring system, where feet with score ≥2 were defined as having footrot. Moreover, samples from feet with footrot were examined for <it>Dichelobacter nodosus </it>by culture and PCR.</p> <p>Results</p> <p>The prevalence of footrot at the individual sheep level was 5.8%, and <it>Dichelobacter nodosus </it>was found by culture and PCR in 83% and 97% of the samples from feet with footrot, respectively. Some minor differences in geographical distribution of footrot were found in this study.</p> <p>Conclusions</p> <p>In a national context, the findings indicate that footrot is fairly common in Swedish slaughter lambs, and should be regarded seriously.</p

    Diseases and causes of death among alpacas in Sweden: a retrospective study

    Background: Due to increasing popularity in Sweden during the last decade, alpacas are frequently encountered by practising veterinarians and pathologists. Knowledge regarding their health and diseases under Swedish conditions is, however, limited. Objectives: To improve knowledge about the health of alpacas in Sweden by collecting information on diseases and health status. Design: A retrospective study was made of 93 necropsies conducted on alpacas in Sweden during the period 2001–2013. Setting Data were obtained from the two major veterinary pathology centres in Sweden. The alpacas were hobby or farm animals and they were submitted by veterinarians in local practices or at a national animal healthcare organisation. Results: The digestive system was most frequently affected (29 per cent), with parasitic gastroenteritis (17 per cent) and hepatic disease being especially prevalent (15 per cent fascioliasis and 7 per cent hepatitis). Cardiovascular conditions (9 per cent), systemic diseases (7 per cent) and perinatal deaths were also common, including abortions (10 per cent) and fatal septicaemia (4 per cent). Wasting/emaciation was a frequent finding (26 per cent). Other diagnoses included dermatitis (8 per cent), diseases of the central nervous system (8 per cent), traumatic injuries (7 per cent), neoplasia (5 per cent), pneumonia (5 per cent) and nephritis (3 per cent). Conclusions: This study identified areas of concern regarding diagnostic and pathological procedures, for which specific measures have been recommended. One particular cause for concern was the number of deaths from emaciation in weanling alpacas during late winter or early spring. For adult alpacas, infectious and noninfectious causes of death were approximately equally frequent. Many of the diseases were considered clinically acute but pathology often showed them to be chronic conditions that had eventually deteriorated and presented as acute cases in the late stages. This study revealed similarities in the health/disease status reported in other European countries and in North America. The results can be used by alpaca keepers and veterinary practitioners to improve management, diagnosis and treatment of alpacas

    Risk portfolio on modern finance has become increasingly technical, requiring the use of sophisticated mathematical tools in both research and practice. Since companies cannot insure themselves completely against risk, as human incompetence in predicting the future precisely that written in Al-Quran surah Luqman verse 34, they have to manage it to yield an optimal portfolio. The objective here is to minimize the variance among all portfolios, or alternatively, to maximize expected return among all portfolios that has at least a certain expected return. Furthermore, this study focuses on optimizing risk portfolio so called Markowitz MVO (Mean-Variance Optimization). Some theoretical frameworks for analysis are arithmetic mean, geometric mean, variance, covariance, linear programming, and quadratic programming. Moreover, finding a minimum variance portfolio produces a convex quadratic programming, that is minimizing the objective function ðð¥with constraintsð ð 𥠥 ðandð´ð¥ = ð. The outcome of this research is the solution of optimal risk portofolio in some investments that could be finished smoothly using MATLAB R2007b software together with its graphic analysis

    Differential cross section measurements for the production of a W boson in association with jets in proton–proton collisions at √s = 7 TeV

    Measurements are reported of differential cross sections for the production of a W boson, which decays into a muon and a neutrino, in association with jets, as a function of several variables, including the transverse momenta (pT) and pseudorapidities of the four leading jets, the scalar sum of jet transverse momenta (HT), and the difference in azimuthal angle between the directions of each jet and the muon. The data sample of pp collisions at a centre-of-mass energy of 7 TeV was collected with the CMS detector at the LHC and corresponds to an integrated luminosity of 5.0 fb[superscript −1]. The measured cross sections are compared to predictions from Monte Carlo generators, MadGraph + pythia and sherpa, and to next-to-leading-order calculations from BlackHat + sherpa. The differential cross sections are found to be in agreement with the predictions, apart from the pT distributions of the leading jets at high pT values, the distributions of the HT at high-HT and low jet multiplicity, and the distribution of the difference in azimuthal angle between the leading jet and the muon at low values.United States.
