    Women in Educational Leadership: Finding Common Ground

    The purpose of this research project was to engage in self-reflective analysis of leadership development as an ongoing process of social action towards democratizing edlucation. Four White women connected by their work as educational leaders, teachers and administrators, engaged this topic by conducting a dialogical analysis of their experiences in leadership. They dialogued from what were technically different positions in the hierarchy at their University and implemented a research process to speak across or marginalize those technical differences to produce a text that explored the rich terrain of leading in which they shared experiences of growth, the conceptual frameworks that guide their leading, and their differing interpretations of gender\u27s role in the leadership process

    The effects of serious gaming on risk perceptions of climate tipping points

    A growing body of research indicates that effective science-policy interactions demand novel approaches, especially in policy domains with long time horizons like climate change. Serious games offer promising opportunities in this regard, but empirical research on game effects and games’ effectiveness in supporting science-policy engagement remains limited. We investigated the effects of a role-playing simulation game on risk perceptions associated with climate tipping points among a knowledgeable and engaged audience of non-governmental observers of the international climate negotiations and scientists. We analysed its effects on concern, perceived seriousness, perceived likelihood and psychological distance of tipping points, using pre- and post-game surveys, debriefing questions and game observations. Our findings suggest that the game reduced the psychological distance of tipping points, rendering them more ‘real’, proximate and tangible for participants. More generally, our findings indicate that role-playing simulation games, depending on their design and future orientation, can provide effective science-policy engagement tools that allow players to engage in future thinking and corresponding meaning making.publishedVersio

    (Re) defining salesperson motivation: current status, main challenges, and research directions

    The construct of motivation is one of the central themes in selling and sales management research. Yet, to-date no review article exists that surveys the construct (both from an extrinsic and intrinsic motivation context), critically evaluates its current status, examines various key challenges apparent from the extant research, and suggests new research opportunities based on a thorough review of past work. The authors explore how motivation is defined, major theories underpinning motivation, how motivation has historically been measured, and key methodologies used over time. In addition, attention is given to principal drivers and outcomes of salesperson motivation. A summarizing appendix of key articles in salesperson motivation is provided

    Sortase A Substrate Specificity in GBS Pilus 2a Cell Wall Anchoring

    Streptococcus agalactiae, also referred to as Group B Streptococcus (GBS), is one of the most common causes of life-threatening bacterial infections in infants. In recent years cell surface pili have been identified in several Gram-positive bacteria, including GBS, as important virulence factors and promising vaccine candidates. In GBS, three structurally distinct types of pili have been discovered (pilus 1, 2a and 2b), whose structural subunits are assembled in high-molecular weight polymers by specific class C sortases. In addition, the highly conserved housekeeping sortase A (SrtA), whose main role is to link surface proteins to bacterial cell wall peptidoglycan by a transpeptidation reaction, is also involved in pili cell wall anchoring in many bacteria. Through in vivo mutagenesis, we demonstrate that the LPXTG sorting signal of the minor ancillary protein (AP2) is essential for pilus 2a anchoring. We successfully produced a highly purified recombinant SrtA (SrtAΔN40) able to specifically hydrolyze the sorting signal of pilus 2a minor ancillary protein (AP2-2a) and catalyze in vitro the transpeptidation reaction between peptidoglycan analogues and the LPXTG motif, using both synthetic fluorescent peptides and recombinant proteins. By contrast, SrtAΔN40 does not catalyze the transpeptidation reaction with substrate-peptides mimicking sorting signals of the other pilus 2a subunits (the backbone protein and the major ancillary protein). Thus, our results add further insight into the proposed model of GBS pilus 2a assembly, in which SrtA is required for pili cell wall covalent attachment, acting exclusively on the minor accessory pilin, representing the terminal subunit located at the base of the pilus

    Recent research on Gulf War illness and other health problems in veterans of the 1991 Gulf War: Effects of toxicant exposures during deployment

    Veterans of Operation Desert Storm/Desert Shield - the 1991 Gulf War (GW) - are a unique population who returned from theater with multiple health complaints and disorders. Studies in the U.S. and elsewhere have consistently concluded that approximately 25-32% of this population suffers from a disorder characterized by symptoms that vary somewhat among individuals and include fatigue, headaches, cognitive dysfunction, musculoskeletal pain, and respiratory, gastrointestinal and dermatologic complaints. Gulf War illness (GWI) is the term used to describe this disorder. In addition, brain cancer occurs at increased rates in subgroups of GW veterans, as do neuropsychological and brain imaging abnormalities. Chemical exposures have become the focus of etiologic GWI research because nervous system symptoms are prominent and many neurotoxicants were present in theater, including organophosphates (OPs), carbamates, and other pesticides; sarin/cyclosarin nerve agents, and pyridostigmine bromide (PB) medications used as prophylaxis against chemical warfare attacks. Psychiatric etiologies have been ruled out. This paper reviews the recent literature on the health of 1991 GW veterans, focusing particularly on the central nervous system and on effects of toxicant exposures. In addition, it emphasizes research published since 2008, following on an exhaustive review that was published in that year that summarizes the prior literature (RACGWI, 2008). We conclude that exposure to pesticides and/or to PB are causally associated with GWI and the neurological dysfunction in GW veterans. Exposure to sarin and cyclosarin and to oil well fire emissions are also associated with neurologically based health effects, though their contribution to development of the disorder known as GWI is less clear. Gene-environment interactions are likely to have contributed to development of GWI in deployed veterans. The health consequences of chemical exposures in the GW and other conflicts have been called "toxic wounds" by veterans. This type of injury requires further study and concentrated treatment research efforts that may also benefit other occupational groups with similar exposure-related illnesses

    Background The immunological escape of tumors represents one of the main ob- stacles to the treatment of malignancies. The blockade of PD-1 or CTLA-4 receptors represented a milestone in the history of immunotherapy. However, immune checkpoint inhibitors seem to be effective in specific cohorts of patients. It has been proposed that their efficacy relies on the presence of an immunological response. Thus, we hypothesized that disruption of the PD-L1/PD-1 axis would synergize with our oncolytic vaccine platform PeptiCRAd. Methods We used murine B16OVA in vivo tumor models and flow cytometry analysis to investigate the immunological background. Results First, we found that high-burden B16OVA tumors were refractory to combination immunotherapy. However, with a more aggressive schedule, tumors with a lower burden were more susceptible to the combination of PeptiCRAd and PD-L1 blockade. The therapy signifi- cantly increased the median survival of mice (Fig. 7). Interestingly, the reduced growth of contralaterally injected B16F10 cells sug- gested the presence of a long lasting immunological memory also against non-targeted antigens. Concerning the functional state of tumor infiltrating lymphocytes (TILs), we found that all the immune therapies would enhance the percentage of activated (PD-1pos TIM- 3neg) T lymphocytes and reduce the amount of exhausted (PD-1pos TIM-3pos) cells compared to placebo. As expected, we found that PeptiCRAd monotherapy could increase the number of antigen spe- cific CD8+ T cells compared to other treatments. However, only the combination with PD-L1 blockade could significantly increase the ra- tio between activated and exhausted pentamer positive cells (p= 0.0058), suggesting that by disrupting the PD-1/PD-L1 axis we could decrease the amount of dysfunctional antigen specific T cells. We ob- served that the anatomical location deeply influenced the state of CD4+ and CD8+ T lymphocytes. In fact, TIM-3 expression was in- creased by 2 fold on TILs compared to splenic and lymphoid T cells. In the CD8+ compartment, the expression of PD-1 on the surface seemed to be restricted to the tumor micro-environment, while CD4 + T cells had a high expression of PD-1 also in lymphoid organs. Interestingly, we found that the levels of PD-1 were significantly higher on CD8+ T cells than on CD4+ T cells into the tumor micro- environment (p < 0.0001). Conclusions In conclusion, we demonstrated that the efficacy of immune check- point inhibitors might be strongly enhanced by their combination with cancer vaccines. PeptiCRAd was able to increase the number of antigen-specific T cells and PD-L1 blockade prevented their exhaus- tion, resulting in long-lasting immunological memory and increased median survival

    Playing Telephone: On the Negotiation and Mediation of Climate Science Communication

    In this thesis, I investigate the effects of social and political context on the process and outcomes of science communication in two different settings, using Dietram Scheufele’s interpretation of science communication as political communication. In the first setting, I examine the communication of climate tipping points at the United Nations Framework Convention on Climate Change (UNFCCC) using 26 semi-structured interviews and 271 surveys administered to members of the UNFCCC policy community. Survey results revealed that only a small minority (14.3%) of policymakers defined climate tipping points consistently with the scientific community. Interview responses revealed that many policymakers believed they were not responsible for incorporating new scientific advice into their work on negotiations, and that this was the responsibility of scientists. Scientists interviewed expressed frustration that policymakers were not willing to hear scientific information they saw as irrelevant to their work on the negotiations. Policymakers responding to interviews were also unwilling to defy social norms by introducing a topic they saw as “complicated” into negotiations. Interview respondents who believed climate tipping points should be discussed within formal negotiations also noted that they interpreted the effects of climate change as temporally or spatially immediate to themselves. In the second setting, I examine how the United States print media incorporated discussion of climate change into coverage of the 2017 hurricane season via a content analysis of hurricane coverage in six major US newspapers. Conservative papers and liberal papers displayed significant differences in frequency and directness of references to climate change, as well as a significant difference in the references to climate denial messages, climate consensus messages, and use of proximity cues. However, the conservative paper near a 2017 hurricane consistently displayed significant differences in coverage from the other conservative papers. This paper frequently used social norms in messaging to shift narratives of acceptability of climate change discussion among conservatives. Both conservative and liberal papers near a 2017 hurricane used proximity cues to indicate the effects of climate change are both physically and temporally near at greater rates than elite and regional papers not near a 2017 hurricane. Taken together, these results reveal that three major factors influenced climate change communication in these two settings. First, power to define direction and content of science communication explains the lack of communication about climate tipping points at the UNFCCC. Policymakers’ hold legitimate power over science communication. This power is codified within UNFCCC structure. Policymakers’ expert power is also interpreted as more relevant to negotiations processes than scientists’ expert power; meaning policymakers are free to define what information is “policy relevant” and therefore, what is communicated. Second, social norms influenced how and whether communication occurred. Social norms prohibiting behavior disruptive to consensus building influenced policymaker definitions of “policy relevant.” Social norms among US conservatives prohibiting serious discussion of climate tipping points were also apparent. Finally, perceptions of climate change as immediate and nearby seemed related to willingness to defy social norms around climate change communication
