    Two of the many factors that influence soil properties and energy consumption are soil moisture and soil composition. The correlations derived from our theoretical and experimental research allow to evaluate physical and mechanical properties of soil such as density, hardness, friction, adhesion, and to assess the draft resistance of soil tillage machines (ploughs, cultivators) depending on the value of the moisture of soil and its composition, as well as on the design parameters and operating speed of the machines, and to determine the optimum range of soil moisture when the energy capacity of tillage is the lowest

    ENERGETICAL, ECONOMICAL AND ECOLOGICAL ASPECTS OF SOIL TILLAGE MINIMISATION

    The work sums up and briefly discusses solutions of soil tillage minimisation technologies and machinery, their agronomic, energetic, mechanic, economical and ecological aspects. Tillage minimisation is performed in directions: reducing number of passes, as well as tillage depth and intensity, joining operations, improving machine design and aggregation, using advanced more suitable technologies and machines, conducting tillage in optimal terms. Minimization of soil tillage is agronomically acceptable, energy, labour and cost saving action. Improvements in the machine design and use for the traditional soil tillage technologies allow to save 24- 36 % of energy (46-110 kWh/ha, which corresponds 12-27 kg/ha of fuel), to reduce labour consumption by 16- 22 %, as well as to cat tillage costs by 14-26 % (10-20 USD/ha). Soil tillage minimisation with ploughing reduces these indices up to two times, without ploughing – up to six times. Besides these actions there is preservation of soil and environment

    Technical, Economical and Ecological Estimation of Up-To-Date Combined Soil Tillage, Fertilising and Sowing Machines

    Comparative studies have been carried out to estimate the possibilities, purposefulness and efficiency of combine soil tollage, fertilising and sowing machines and units for field crops used on peasant farms under the Baltic conditions. It is found out that most widely used machines and units are aggregates combining pre-sowing tillage of soil (combined cultivators, or revolving power harrow), mineral fertiliser application (pneumatic distributors) and grain sowing (pneumatic, or mechanical sowing machines). As to the agrotechnical terms, the ploughing-sowing aggregates may have limited application. The most suitable are tractor-mounted machines. For combined aggregates preference should be given to more powerful tractors having frontally mounted equipment as well. In contrast to single-operational machines, the use of combined aggregates removes the weather risk, improves the quality of work, raises their efficiency, reduces the consumption of energy and fuel (accordingly, the amount of harmful exhaust gases) and cuts the total costs by 10-18 %. For agrotechnical, energetic, economical and ecological considerations the up-to-date combined aggregates of soil tillage; fertiliser application and sowing are efficient and should be widely introduced on the farms of Latvia

    The operationalisation of sustainability: Sustainable aquaculture production as defined by certification schemes

    Sustainability certification has become an increasingly important feature in aquaculture production, leading to a multitude of schemes with various criteria. However, the large number of schemes and the complexity of the standards creates confusion with respect to which sustainability objectives are targeted. As a result, what is meant by ‘sustainability’ is unclear. In this paper, we examine the operationalisation of the concept from the vantage point of the certifying authorities, who devise standards and grant or withhold certification of compliance. We map the criteria of eight widely-used certification schemes using the four domains of the Wheel of Sustainability, a reference model designed to encompass a comprehensive understanding of sustainability. We show that, overall, the sustainability certifications have an overwhelming focus on environmental and governance indicators, and only display scattered attempts at addressing cultural and economic issues. The strong focus on governance indicators is, to a large degree, due to their role in implementing and legitimising the environmental indicators. The strong bias implies that these certification schemes predominantly focus on the environmental domain and do not address sustainability as a whole, nor do they complement each other. Sustainability is by definition and by necessity a comprehensive concept, but if the cultural and economic issues are to be addressed in aquaculture, the scope of certification schemes must be expanded.     Antimicrobial use in European acute care hospitals: results from the second point prevalence survey (PPS) of healthcare-associated infections and antimicrobial use, 2016 to 2017

    Antimicrobial agents used to treat infections are life-saving. Overuse may result in more frequent adverse effects and emergence of multidrug-resistant microorganisms. In 2016-17, we performed the second point-prevalence survey (PPS) of healthcare-associated infections (HAIs) and antimicrobial use in European acute care hospitals. We included 1,209 hospitals and 310,755 patients in 28 of 31 European Union/European Economic Area (EU/EEA) countries. The weighted prevalence of antimicrobial use in the EU/EEA was 30.5% (95% CI: 29.2-31.9%). The most common indication for prescribing antimicrobials was treatment of a community-acquired infection, followed by treatment of HAI and surgical prophylaxis. Over half (54.2%) of antimicrobials for surgical prophylaxis were prescribed for more than 1 day. The most common infections treated by antimicrobials were respiratory tract infections and the most commonly prescribed antimicrobial agents were penicillins with beta-lactamase inhibitors. There was wide variation of patients on antimicrobials, in the selection of antimicrobial agents and in antimicrobial stewardship resources and activities across the participating countries. The results of the PPS provide detailed information on antimicrobial use in European acute care hospitals, enable comparisons between countries and hospitals, and highlight key areas for national and European action that will support efforts towards prudent use of antimicrobials

    The dynamics of root cap sloughing in Arabidopsis is regulated by peptide signalling

    The root cap protects the stem cell niche of angiosperm roots from damage. In Arabidopsis, lateral root cap (LRC) cells covering the meristematic zone are regularly lost through programmed cell death, while the outermost layer of the root cap covering the tip is repeatedly sloughed. Efficient coordination with stem cells producing new layers is needed to maintain a constant size of the cap. We present a signalling pair, the peptide IDA-LIKE1 (IDL1) and its receptor HAESA-LIKE2 (HSL2), mediating such communication. Live imaging over several days characterized this process from initial fractures in LRC cell files to full separation of a layer. Enhanced expression of IDL1 in the separating root cap layers resulted in increased frequency of sloughing, balanced with generation of new layers in a HSL2-dependent manner. Transcriptome analyses linked IDL1-HSL2 signalling to the transcription factors BEARSKIN1/2 and genes associated with programmed cell death. Mutations in either IDL1 or HSL2 slowed down cell division, maturation and separation. Thus, IDL1-HSL2 signalling potentiates dynamic regulation of the homeostatic balance between stem cell division and sloughing activity

    Large variation in ESBL-producing Escherichia coli carriers in six European countries including Russia

    We investigated the faecal carriage prevalence of extended-spectrum β-lactamase production in Escherichia coli (EP-EC) and/or Klebsiella pneumoniae (EP-KP) and risk factors associated with carriage among adult study subjects in Finland, Germany, Latvia, Poland, Russia and Sweden (partner countries). The aim was to get indicative data on the prevalence of ESBL-carriage in specific populations in the region. Faecal samples were collected from four study populations and screened on ChromID-ESBL and ChromID-OXA-48 plates. Positive isolates were further characterised phenotypically. Our results show a large variation in carrier prevalence ranging from 1.6% in Latvia to 23.2% in Russia for EP-EC. For the other partner countries, the prevalence of EP-EC were in increasing numbers, 2.3% for Germany, 4.7% for Finland, 6.6% for Sweden, 8.0% for Poland and 8.1% for all partner countries in total. Carriers of EP-KP were identified only in Finland, Russia and Sweden, and the prevalence was < 2% in each of these countries. No carriers of carbapenemase-producing isolates were identified. This is the first study reporting prevalence of carriers (excluding traveller studies) for Finland, Latvia, Poland and Russia. It contributes with important information regarding the prevalence of EP-EC and EP-KP carriage in regions where studies on carriers are limited

    Gene expression profiles of breast biopsies from healthy women identify a group with claudin-low features

    Background Increased understanding of the variability in normal breast biology will enable us to identify mechanisms of breast cancer initiation and the origin of different subtypes, and to better predict breast cancer risk. Methods Gene expression patterns in breast biopsies from 79 healthy women referred to breast diagnostic centers in Norway were explored by unsupervised hierarchical clustering and supervised analyses, such as gene set enrichment analysis and gene ontology analysis and comparison with previously published genelists and independent datasets. Results Unsupervised hierarchical clustering identified two separate clusters of normal breast tissue based on gene-expression profiling, regardless of clustering algorithm and gene filtering used. Comparison of the expression profile of the two clusters with several published gene lists describing breast cells revealed that the samples in cluster 1 share characteristics with stromal cells and stem cells, and to a certain degree with mesenchymal cells and myoepithelial cells. The samples in cluster 1 also share many features with the newly identified claudin-low breast cancer intrinsic subtype, which also shows characteristics of stromal and stem cells. More women belonging to cluster 1 have a family history of breast cancer and there is a slight overrepresentation of nulliparous women in cluster 1. Similar findings were seen in a separate dataset consisting of histologically normal tissue from both breasts harboring breast cancer and from mammoplasty reductions. Conclusion This is the first study to explore the variability of gene expression patterns in whole biopsies from normal breasts and identified distinct subtypes of normal breast tissue. Further studies are needed to determine the specific cell contribution to the variation in the biology of normal breasts, how the clusters identified relate to breast cancer risk and their possible link to the origin of the different molecular subtypes of breast cancer

    Expression levels of uridine 5'-diphospho-glucuronosyltransferase genes in breast tissue from healthy women are associated with mammographic density

    Introduction Mammographic density (MD), as assessed from film screen mammograms, is determined by the relative content of adipose, connective and epithelial tissue in the female breast. In epidemiological studies, a high percentage of MD confers a four to six fold risk elevation of developing breast cancer, even after adjustment for other known breast cancer risk factors. However, the biologic correlates of density are little known. Methods Gene expression analysis using whole genome arrays was performed on breast biopsies from 143 women; 79 women with no malignancy (healthy women) and 64 newly diagnosed breast cancer patients, both included from mammographic centres. Percent MD was determined using a previously validated, computerized method on scanned mammograms. Significance analysis of microarrays (SAM) was performed to identify genes influencing MD and a linear regression model was used to assess the independent contribution from different variables to MD. Results SAM-analysis identified 24 genes differentially expressed between samples from breasts with high and low MD. These genes included three uridine 5'-diphospho-glucuronosyltransferase (UGT) genes and the oestrogen receptor gene (ESR1). These genes were down-regulated in samples with high MD compared to those with low MD. The UGT gene products, which are known to inactivate oestrogen metabolites, were also down-regulated in tumour samples compared to samples from healthy individuals. Several single nucleotide polymorphisms (SNPs) in the UGT genes associated with the expression of UGT and other genes in their vicinity were identified. Conclusions Three UGT enzymes were lower expressed both in breast tissue biopsies from healthy women with high MD and in biopsies from newly diagnosed breast cancers. The association was strongest amongst young women and women using hormonal therapy. UGT2B10 predicts MD independently of age, hormone therapy and parity. Our results indicate that down-regulation of UGT genes in women exposed to female sex hormones is associated with high MD and might increase the risk of breast cancer

    No evidence that protein truncating variants in BRIP1 are associated with breast cancer risk: implications for gene panel testing.

    BACKGROUND: BRCA1 interacting protein C-terminal helicase 1 (BRIP1) is one of the Fanconi Anaemia Complementation (FANC) group family of DNA repair proteins. Biallelic mutations in BRIP1 are responsible for FANC group J, and previous studies have also suggested that rare protein truncating variants in BRIP1 are associated with an increased risk of breast cancer. These studies have led to inclusion of BRIP1 on targeted sequencing panels for breast cancer risk prediction. METHODS: We evaluated a truncating variant, p.Arg798Ter (rs137852986), and 10 missense variants of BRIP1, in 48 144 cases and 43 607 controls of European origin, drawn from 41 studies participating in the Breast Cancer Association Consortium (BCAC). Additionally, we sequenced the coding regions of BRIP1 in 13 213 cases and 5242 controls from the UK, 1313 cases and 1123 controls from three population-based studies as part of the Breast Cancer Family Registry, and 1853 familial cases and 2001 controls from Australia. RESULTS: The rare truncating allele of rs137852986 was observed in 23 cases and 18 controls in Europeans in BCAC (OR 1.09, 95% CI 0.58 to 2.03, p=0.79). Truncating variants were found in the sequencing studies in 34 cases (0.21%) and 19 controls (0.23%) (combined OR 0.90, 95% CI 0.48 to 1.70, p=0.75). CONCLUSIONS: These results suggest that truncating variants in BRIP1, and in particular p.Arg798Ter, are not associated with a substantial increase in breast cancer risk.
