SPITZER survey of dust grain processing in stable discs around binary post-AGB stars
Aims: We investigate the mineralogy and dust processing in the circumbinary
discs of binary post-AGB stars using high-resolution TIMMI2 and SPITZER
infrared spectra. Methods: We perform a full spectral fitting to the infrared
spectra using the most recent opacities of amorphous and crystalline dust
species. This allows for the identification of the carriers of the different
emission bands. Our fits also constrain the physical properties of different
dust species and grain sizes responsible for the observed emission features.
Results: In all stars the dust is oxygen-rich: amorphous and crystalline
silicate dust species prevail and no features of a carbon-rich component can be
found, the exception being EPLyr, where a mixed chemistry of both oxygen- and
carbon-rich species is found. Our full spectral fitting indicates a high degree
of dust grain processing. The mineralogy of our sample stars shows that the
dust is constituted of irregularly shaped and relatively large grains, with
typical grain sizes larger than 2 micron. The spectra of nearly all stars show
a high degree of crystallinity, where magnesium-rich end members of olivine and
pyroxene silicates dominate. Other dust features of e.g. silica or alumina are
not present at detectable levels. Temperature estimates from our fitting
routine show that a significant fraction of grains must be cool, significantly
cooler than the glass temperature. This shows that radial mixing is very
efficient is these discs and/or indicates different thermal conditions at grain
formation. Our results show that strong grain processing is not limited to
young stellar objects and that the physical processes occurring in the discs

Early pneumothorax as a feature of response to crizotinib therapy in a patient with ALK rearranged lung adenocarcinoma.
Background: Single arm phase 1 and 2 studies on Crizotinib in ALK-positive patients so far have shown rapid and durable responses. Spontaneous pneumothoraces as a result of response to anti-cancer therapy are rare in oncology but have been documented in a number of tumour types including lung cancer. This includes cytotoxic chemotherapy as well as molecular targeted agents such as gefitinib and Bevacizumab. These often require chest drain insertion or surgical intervention with associated morbidity and mortality. They have also been associated with response to treatment. This is the first report we are aware of documenting pneumothorax as response to crizotinib therapy.Case presentation: A 48-year-old Caucasian male presented with a Stage IV, TTF1 positive, EGFR wild-type adenocarcinoma of the lung. He received first line chemotherapy with three cycles of cisplatin-pemetrexed chemotherapy with a differential response, and then second-line erlotinib for two months before further radiological evidence of disease progression. Further analysis of his diagnostic specimen identified an ALK rearrangement by fluorescence in situ hybridization (FISH). He was commenced on crizotinib therapy 250 mg orally twice daily. At his 4-week assessment he had a chest radiograph that identified a large left-sided pneumothorax with disease response evident on the right. Chest CT confirmed a 50% left-sided pneumothorax on a background of overall disease response. A chest tube was inserted with complete resolution of the pneumothorax that did not recur following its removal.Conclusion: Our case demonstrates this potential complication of crizotinib therapy and we therefore recommend that pneumothorax be considered in patients on crizotinib presenting with high lung metastatic burden and with worsening dyspnoea. © 2013 Gennatas et al.; licensee BioMed Central Ltd

Toward understanding of differences in current cloud retrievals of ARM ground-based measurements
Accurate observations of cloud microphysical properties are needed for evaluating
and improving the representation of cloud processes in climate models and better estimate
of the Earth radiative budget. However, large differences are found in current cloud
products retrieved from ground-based remote sensing measurements using various retrieval
algorithms. Understanding the differences is an important step to address uncertainties
in the cloud retrievals. In this study, an in-depth analysis of nine existing ground-based
cloud retrievals using ARM remote sensing measurements is carried out. We place
emphasis on boundary layer overcast clouds and high level ice clouds, which are the focus
of many current retrieval development efforts due to their radiative importance and
relatively simple structure. Large systematic discrepancies in cloud microphysical
properties are found in these two types of clouds among the nine cloud retrieval products,
particularly for the cloud liquid and ice particle effective radius. Note that the differences
among some retrieval products are even larger than the prescribed uncertainties reported by
the retrieval algorithm developers. It is shown that most of these large differences have
their roots in the retrieval theoretical bases, assumptions, as well as input and constraint
parameters. This study suggests the need to further validate current retrieval theories and
assumptions and even the development of new retrieval algorithms with more observations
under different cloud regimes
Retrospective evaluation of whole exome and genome mutation calls in 746 cancer samples
Funder: NCI U24CA211006Abstract: The Cancer Genome Atlas (TCGA) and International Cancer Genome Consortium (ICGC) curated consensus somatic mutation calls using whole exome sequencing (WES) and whole genome sequencing (WGS), respectively. Here, as part of the ICGC/TCGA Pan-Cancer Analysis of Whole Genomes (PCAWG) Consortium, which aggregated whole genome sequencing data from 2,658 cancers across 38 tumour types, we compare WES and WGS side-by-side from 746 TCGA samples, finding that ~80% of mutations overlap in covered exonic regions. We estimate that low variant allele fraction (VAF < 15%) and clonal heterogeneity contribute up to 68% of private WGS mutations and 71% of private WES mutations. We observe that ~30% of private WGS mutations trace to mutations identified by a single variant caller in WES consensus efforts. WGS captures both ~50% more variation in exonic regions and un-observed mutations in loci with variable GC-content. Together, our analysis highlights technological divergences between two reproducible somatic variant detection efforts
Fluorescence In Situ Hybridization for MDM2 Amplification as a Routine Ancillary Diagnostic Tool for Suspected Well-Differentiated and Dedifferentiated Liposarcomas: Experience at a Tertiary Center
Background. The assessment of MDM2 gene amplification by fluorescence in situ hybridization (FISH) has become a routine ancillary tool for diagnosing atypical lipomatous tumor (ALT)/well-differentiated liposarcoma and dedifferentiated liposarcoma (WDL/DDL) in specialist sarcoma units. We describe our experience of its utility at our tertiary institute. Methods. All routine histology samples in which MDM2 amplification was assessed with FISH over a 2-year period were included, and FISH results were correlated with clinical and histologic findings. Results. 365 samples from 347 patients had FISH for MDM2 gene amplification. 170 were positive (i.e., showed MDM2 gene amplification), 192 were negative, and 3 were technically unsatisfactory. There were 122 histologically benign cases showing a histology:FISH concordance rate of 92.6%, 142 WDL/DDL (concordance 96.5%), and 34 cases histologically equivocal for WDL (concordance 50%). Of 64 spindle cell/pleomorphic neoplasms (in which DDL was a differential diagnosis), 21.9% showed MDM2 amplification. Of the cases with discrepant histology and FISH, all but 3 had diagnoses amended following FISH results. For discrepancies of benign histology but positive FISH, lesions were on average larger, more frequently in “classical” (intra-abdominal or inguinal) sites for WDL/DDL and more frequently core biopsies. Discrepancies of malignant histology but negative FISH were smaller, less frequently in “classical” sites but again more frequently core biopsies. Conclusions. FISH has a high correlation rate with histology for cases with firm histologic diagnoses of lipoma or WDL/DDL. It is a useful ancillary diagnostic tool in histologically equivocal cases, particularly in WDL lacking significant histologic atypia or DDL without corresponding WDL component, especially in larger tumors, those from intra-abdominal or inguinal sites or core biopsies. There is a significant group of well-differentiated adipocytic neoplasms which are difficult to diagnose on morphology alone, in which FISH for MDM2 amplification is diagnostically contributory
Country differences in the relationship between leadership and employee engagement: A meta-analysis
Leadership is frequently related to important organizational outcomes such as follower engagement. However, to date we have little insight into the degree to which this relation is contingent upon (a) types of leadership style and (b) national culture. These two issues are addressed in a meta-analysis of 209 independent (257 effect sizes), mainly cross-sectional studies (79%), involving 82,386 participants from 45 countries. The findings show that whereas abusive supervision was negatively associated with work engagement, several leadership styles (e.g., servant, empowering, ethical, and charismatic leadership) have positive correlations with subordinate engagement; some dimensions of national culture (e.g., gender egalitarianism, human orientation, performance orientation, future orientation, and power distance) moderate the leadership–employee engagement relationship. However, the correlations between servant, ethical, and transactional leadership and subordinate engagement are less likely to vary across national cultural characteristics. Notwithstanding the proliferation of leadership–employee engagement literature with more than 200 published articles, a strong reliance on cross-sectional designs have impeded it to gain any solid conclusions about causality due to endogeneity biases. We conclude by providing a detailed future research agenda and discussing how our results can stimulate future leadership research and inform practices with regards to leader development
