    Effects of some rhizosphere bacteria for the biocontrol of nematodes of the genus Meloidogyne with Arthrobotrys oligospora

    A été étudiée l'action d'isolats bactériens au champignon nématophage #Arthrobotrys oligospora ORS 18692 S7 isolé au Sénégal sur le développement du champignon (croissance saprophytique et activité prédatrice), la multiplication d'une population de #Meloidogyne mayaguensis sur des plants de tomate, et la croissance de la plante hôte. Trois isolats bactériens ont été capables de stimuler in vitro l'activité prédatrice du champignon et de réduire le développement d'une population de #M. mayaguensis$ sur des plants de tomate cultivés en pots. Ces bactéries ont été nommées bactéries auxiliaires de champignons nématophages (BAN). Les critères permettant de distinguer ces organismes des autres bactéries rhizosophériques n'ont pu être mis en évidence. L'utilisation des BAN pour le contrôle biologique des nématodes est discutée.     Amidoxime Functionalization of Algal/Polyethyleneimine Beads for the Sorption of Sr(II) from Aqueous Solutions

    International audienceThere is a need for developing new sorbents that incorporate renewable resources for the treatment of metal-containing solutions. Algal-polyethyleneimine beads (APEI) (reinforced with alginate) are functionalized by grafting amidoxime groups (AO-APEI). Physicochemical characteristics of the new material are characterized using FTIR, XPS, TGA, SEM, SEM-EDX, and BET. AO-APEI beads are tested for the recovery of Sr(II) from synthetic solutions after pH optimization (≈ pH 6). Uptake kinetics is fast (equilibrium ≈ 60-90 min). Sorption isotherm (fitted by the Langmuir equation) shows remarkable sorption capacity (≈ 189 mg Sr g −1). Sr(II) is desorbed using 0.2 M HCl/0.5 M CaCl 2 solution; sorbent recycling over five cycles shows high stability in terms of sorption/desorption performances. The presence of competitor cations is studied in relation to the pH; the selectivity for Sr(II) is correlated to the softness parameter. Finally, the recovery of Sr(II) is carried out in complex solutions (seawater samples): AO-APEI is remarkably selective over highly concentrated metal cations such as Na(I), K(I), Mg(II), and Ca(II), with weaker selectivity over B(I) and As(V). AO-APEI appears to be a promising material for selective recovery of strontium from complex solutions (including seawater)

    CenomanianTuronian transition in a shallow water sequence of the Sinai, Egypt

    Environmental and depositional changes across the Late Cenomanian oceanic anoxic event (OAE2) in the Sinai, Egypt, are examined based on biostratigraphy, mineralogy, δ13C values and phosphorus analyses. Comparison with the Pueblo, Colorado, stratotype section reveals the Whadi El Ghaib section as stratigraphically complete across the late Cenomanianearly Turonian. Foraminifera are dominated by high-stress planktic and benthic assemblages characterized by low diversity, low-oxygen and low-salinity tolerant species, which mark shallow-water oceanic dysoxic conditions during OAE2. Oyster biostromes suggest deposition occurred in less than 50 m depths in low-oxygen, brackish, and nutrient-rich waters. Their demise prior to the peak δ13C excursion is likely due to a rising sea-level. Characteristic OAE2 anoxic conditions reached this coastal region only at the end of the δ13C plateau in deeper waters near the end of the Cenomanian. Increased phosphorus accumulations before and after the δ13C excursion suggest higher oxic conditions and increased detrital input. Bulk-rock and clay mineralogy indicate humid climate conditions, increased continental runoff and a rising sea up to the first δ13C peak. Above this interval, a dryer and seasonally well-contrasted climate with intermittently dry conditions prevailed. These results reveal the globally synchronous δ13C shift, but delayed effects of OAE2 dependent on water depth

    Characteristic cohomology of pp-form gauge theories

    The characteristic cohomology Hchark(d)H^k_{char}(d) for an arbitrary set of free pp-form gauge fields is explicitly worked out in all form degrees k<n1k<n-1, where nn is the spacetime dimension. It is shown that this cohomology is finite-dimensional and completely generated by the forms dual to the field strengths. The gauge invariant characteristic cohomology is also computed. The results are extended to interacting pp-form gauge theories with gauge invariant interactions. Implications for the BRST cohomology are mentioned.Comment: Latex file, no figures, 44 page

    Transcriptomic properties of her2+ ductal carcinoma in situ of the breast associate with absence of immune cells

    SIMPLE SUMMARY: Tumor-infiltrating lymphocytes (TILs) are likely to play a role in the biological behavior of HER2+ ductal carcinoma in situ (DCIS). To prevent invasiveness, the potential of targeted immune-modulating treatment of HER2+ DCIS has been explored. We identified a 29-gene expression profile that was associated with the density of TILs. These genes included CCND3, DUSP10 and RAP1GAP, which may guide towards more rationalized choices with respect to immune-mediated therapy in HER2+ DCIS, such as targeted vaccine therapy. ABSTRACT: The identification of transcriptomic alterations of HER2+ ductal carcinoma in situ (DCIS) that are associated with the density of tumor-infiltrating lymphocytes (TILs) could contribute to optimizing choices regarding the potential benefit of immune therapy. We compared the gene expression profile of TIL-poor HER2+ DCIS to that of TIL-rich HER2+ DCIS. Tumor cells from 11 TIL-rich and 12 TIL-poor DCIS cases were micro-dissected for RNA isolation. The Ion AmpliSeq Transcriptome Human Gene Expression Kit was used for RNA sequencing. After normalization, a Mann–Whitney rank sum test was used to analyze differentially expressed genes between TIL-poor and TIL-rich HER2+ DCIS. Whole tissue sections were immunostained for validation of protein expression. We identified a 29-gene expression profile that differentiated TIL-rich from TIL-poor HER2+ DCIS. These genes included CCND3, DUSP10 and RAP1GAP, which were previously described in breast cancer and cancer immunity and were more highly expressed in TIL-rich DCIS. Using immunohistochemistry, we found lower protein expression in TIL-rich DCIS. This suggests regulation of protein expression at the posttranslational level. We identified a gene expression profile of HER2+ DCIS cells that was associated with the density of TILs. This classifier may guide towards more rationalized choices regarding immune-mediated therapy in HER2+ DCIS, such as targeted vaccine therapy

    The ALMA Spectroscopic Survey in the Hubble Ultra Deep Field: Evolution of the Molecular Gas in CO-selected Galaxies

    We analyze the interstellar medium properties of a sample of 16 bright CO line emitting galaxies identified in the ALMA Spectroscopic Survey in the Hubble Ultra Deep Field (ASPECS) Large Program. This CO−selected galaxy sample is complemented by two additional CO line emitters in the UDF that are identified based on their MultiUnit Spectroscopic Explorer (MUSE) optical spectroscopic redshifts. The ASPECS CO−selected galaxies cover a larger range of star formation rates (SFRs) and stellar masses compared to literature CO emitting galaxies at z > 1 for which scaling relations have been established previously. Most of ASPECS CO-selected galaxies follow these established relations in terms of gas depletion timescales and gas fractions as a function of redshift, as well as the SFR–stellar mass relation (“galaxy main sequence”). However, we find that ∼30% of the galaxies (5 out of 16) are offset from the galaxy main sequence at their respective redshift, with ∼12% (2 out of 16) falling below this relationship. Some CO-rich galaxies exhibit low SFRs, and yet show substantial molecular gas reservoirs, yielding long gas depletion timescales. Capitalizing on the well-defined cosmic volume probed by our observations, we measure the contribution of galaxies above, below, and on the galaxy main sequence to the total cosmic molecular gas density at different lookback times. We conclude that main-sequence galaxies are the largest contributors to the molecular gas density at any redshift probed by our observations (z ∼ 1−3). The respective contribution by starburst galaxies above the main sequence decreases from z ∼ 2.5 to z ∼ 1, whereas we find tentative evidence for an increased contribution to the cosmic molecular gas density from the passive galaxies below the main sequenc

    Phosphorothioate antisense oligonucleotides induce the formation of nuclear bodies

    Antisense oligonucleotides are powerful tools for the in vivo regulation of gene expression. We have characterized the intracellular distribution of fluorescently tagged phosphorothioate oligodeoxynucleotides (PS-ONs) at high resolution under conditions in which PS-ONs have the potential to display antisense activity. Under these conditions PS-ONs predominantly localized to the cell nucleus where they accumulated in 20-30 bright spherical foci designated phosphorothioate bodies (PS bodies), which were set against a diffuse nucleoplasmic population excluding nucleoli. PS bodies are nuclear structures that formed in cells after PS-ON delivery by transfection agents or microinjection but were observed irrespectively of antisense activity or sequence. Ultrastructurally, PS bodies corresponded to electron-dense structures of 150-300 nm diameter and resembled nuclear bodies that were found with lower frequency in cells lacking PS-ONs. The environment of a living cell was required for the de novo formation of PS bodies, which occurred within minutes after the introduction of PS-ONs. PS bodies were stable entities that underwent noticeable reorganization only during mitosis. Upon exit from mitosis, PS bodies were assembled de novo from diffuse PS-ON pools in the daughter nuclei. In situ fractionation demonstrated an association of PS-ONs with the nuclear matrix. Taken together, our data provide evidence for the formation of a nuclear body in cells after introduction of phosphorothioate oligodeoxynucleotides

    Pasireotide, a multireceptor-targeted somatostatin analog with highest affinity for somatostatin receptor subtype (SST) 5, has demonstrated superior efficacy over the SST2-preferential somatostatin analogs octreotide and lanreotide. The safety profile is similar to those of octreotide and lanreotide, except for a higher frequency and degree of hyperglycemia. This analysis investigated baseline characteristics and occurrence and management of hyperglycemia during pasireotide treatment in patients with acromegaly treated in two prospective clinical studies, SOM230C2305 (C2305) and SOM230C2402 (C2402; PAOLA)

    Are biological systems poised at criticality?

    Many of life's most fascinating phenomena emerge from interactions among many elements--many amino acids determine the structure of a single protein, many genes determine the fate of a cell, many neurons are involved in shaping our thoughts and memories. Physicists have long hoped that these collective behaviors could be described using the ideas and methods of statistical mechanics. In the past few years, new, larger scale experiments have made it possible to construct statistical mechanics models of biological systems directly from real data. We review the surprising successes of this "inverse" approach, using examples form families of proteins, networks of neurons, and flocks of birds. Remarkably, in all these cases the models that emerge from the data are poised at a very special point in their parameter space--a critical point. This suggests there may be some deeper theoretical principle behind the behavior of these diverse systems.Comment: 21 page

    AD51B in Familial Breast Cancer

    Common variation on 14q24.1, close to RAD51B, has been associated with breast cancer: rs999737 and rs2588809 with the risk of female breast cancer and rs1314913 with the risk of male breast cancer. The aim of this study was to investigate the role of RAD51B variants in breast cancer predisposition, particularly in the context of familial breast cancer in Finland. We sequenced the coding region of RAD51B in 168 Finnish breast cancer patients from the Helsinki region for identification of possible recurrent founder mutations. In addition, we studied the known rs999737, rs2588809, and rs1314913 SNPs and RAD51B haplotypes in 44,791 breast cancer cases and 43,583 controls from 40 studies participating in the Breast Cancer Association Consortium (BCAC) that were genotyped on a custom chip (iCOGS). We identified one putatively pathogenic missense mutation c.541C&gt;T among the Finnish cancer patients and subsequently genotyped the mutation in additional breast cancer cases (n = 5259) and population controls (n = 3586) from Finland and Belarus. No significant association with breast cancer risk was seen in the meta-analysis of the Finnish datasets or in the large BCAC dataset. The association with previously identified risk variants rs999737, rs2588809, and rs1314913 was replicated among all breast cancer cases and also among familial cases in the BCAC dataset. The most significant association was observed for the haplotype carrying the risk-alleles of all the three SNPs both among all cases (odds ratio (OR): 1.15, 95% confidence interval (CI): 1.11–1.19, P = 8.88 x 10−16) and among familial cases (OR: 1.24, 95% CI: 1.16–1.32, P = 6.19 x 10−11), compared to the haplotype with the respective protective alleles. Our results suggest that loss-of-function mutations in RAD51B are rare, but common variation at the RAD51B region is significantly associated with familial breast cancer risk
