    Population and Virulence Factor Analysis of Staphylococcus aureus from Bovine Mastitis.

    End of Project ReportsStaphylococcus aureus is a major cause of bovine mastitis and the disease is responsible for substantial economic losses in the dairy industry world-wide. A large number of commonly accepted virulence factors are associated with S. aureus but it is yet to be elucidated which of these are important for infection of the bovine udder. A rational and effective strategy for the control of intramammary infections may need to be directed against clones of S. aureus that commonly cause disease. The objective of this study was to characterise the genetic variance of S. aureus isolate populations from infected udders in Ireland using RAPD-PCR, ribotyping and multilocus enzyme electrophoresis (MLEE). Similar S. aureus isolates collected in the USA were also typed in order to compare strain differences in staphylococcal populations in a different environment. Phenotypic diversity based on a number of presumed virulence factors together with antibiotic sensitivity was examined and correlations between phenotype and genotype were identified, if present. In addition, a pathogenicity island encoding multiple superantigens was completely sequenced and characterised. Knockout mutants of these superantigens were also constructed and in vitro functional analysis performed. Laboratory animal experiments (mice and rabbits) were used to study the relative pathogenicity of individual staphylococcal strains (mice) and also to measure the immunological responses after prolonged exposure to the predominant strains (rabbits)

    The High-Pressure Structural Evolution of Olivine along the Forsterite–Fayalite Join

    Structural refinements from single-crystal X-ray diffraction data are reported for olivine with a composition of Fo100 (forsterite Mg2SiO4, synthetic), Fo80 and Fo62 (~Mg1.6Fe0.4SiO4 and ~Mg1.24Fe0.76SiO4, both natural) at room temperature and high pressure to ~8 GPa. The new results, along with data from the literature on Fo0 (fayalite Fe2SiO4), were used to investigate the previously reported structural mechanisms which caused small variations of olivine bulk modulus with increasing Fe content. For all the investigated compositions, the M2 crystallographic site, with its bonding configuration and its larger polyhedral volume, was observed to control the compression mechanisms in olivine. From Fo100 to Fo0, the compression rates for M2\u2013O and M1\u2013O bond lengths were observed to control the relative polyhedral volumes, resulting in a less-compressible M1O6 polyhedral volume, likely causing the slight increase in bulk modulus with increasing Fe content

    Temperature dependence and mechanisms for vortex pinning by periodic arrays of Ni dots in Nb films

    Pinning interactions between superconducting vortices in Nb and magnetic Ni dots were studied as a function of current and temperature to clarify the nature of pinning mechanisms. A strong current dependence is found for a square array of dots, with a temperature dependent optimum current for the observation of periodic pinning, that decreases with temperature as (1-T/Tc)3/2. This same temperature dependence is found for the critical current at the first matching field with a rectangular array of dots. The analysis of these results allows to narrow the possible pinning mechanisms to a combination of two: the interaction between the vortex and the magnetic moment of the dot and the proximity effect. Moreover, for the rectangular dot array, the temperature dependence of the crossover between the low field regime with a rectangular vortex lattice to the high field regime with a square configuration has been studied. It is found that the crossover field increases with decreasing temperature. This dependence indicates a change in the balance between elastic and pinning energies, associated with dynamical effects of the vortex lattice in the high field range.Comment: 12 text pages (revtex), 6 figures (1st jpeg, 2nd-6th postscript) accepted in Physical Review

    A multi-objective evolutionary algorithm fitness function for case-base maintenance.

    Case-Base Maintenance (CBM) has two important goals. On the one hand, it aims to reduce the size of the case-base. On the other hand, it has to improve the accuracy of the CBR system. CBM can be represented as a multi-objective optimization problem to achieve both goals. Multi-Objective Evolutionary Algorithms (MOEAs) have been recognised as appropriate techniques for multi-objective optimisation because they perform a search for multiple solutions in parallel. In the present paper we introduce a fitness function based on the Complexity Profiling model to perform CBM with MOEA, and we compare its results against other known CBM approaches. From the experimental results, CBM with MOEA shows regularly good results in many case-bases, despite the amount of redundant and noisy cases, and with a significant potential for improvement

    The uses and abuses of power: teaching school leadership through children's literature

    There are relatively few studies of how representations of teachers, schools and educational administrators in popular films and television might be, and are, used in leadership preparation. This paper seeks to add to this small body of work; it reports on an exploratory study of the representation of headteachers in contemporary children's fiction. Thirty-one texts are analysed to ascertain key themes and the major characterisations. The paper draws on children's literature scholars to argue that both the historical school story and its contemporary counterpart focus heavily on the power of the head to control the micro-world of the school. Because these fictional accounts deal with issues of power and justice more openly than many mainstream educational administration texts, this makes them particularly useful in the preparation of potential school leaders

    Studies of dust devils, and their impact on society, are reviewed. Dust devils have been noted since antiquity, and have been documented in many countries, as well as on the planet Mars. As time-variable vortex entities, they have become a cultural motif. Three major stimuli of dust devil research are identified, nuclear testing, terrestrial climate studies, and perhaps most significantly, Mars research. Dust devils present an occasional safety hazard to light structures and have caused several deaths

    Scientific communication is facilitated by a data-driven, scientifically sound taxonomy that considers the end-user¿s needs and established successful practice. In 2013, the Fusarium community voiced near unanimous support for a concept of Fusarium that represented a clade comprising all agriculturally and clinically important Fusarium species, including the F. solani species complex (FSSC). Subsequently, this concept was challenged in 2015 by one research group who proposed dividing the genus Fusarium into seven genera, including the FSSC described as members of the genus Neocosmospora, with subsequent justification in 2018 based on claims that the 2013 concept of Fusarium is polyphyletic. Here, we test this claim and provide a phylogeny based on exonic nucleotide sequences of 19 orthologous protein-coding genes that strongly support the monophyly of Fusarium including the FSSC. We reassert the practical and scientific argument in support of a genus Fusarium that includes the FSSC and several other basal lineages, consistent with the longstanding use of this name among plant pathologists, medical mycologists, quarantine officials, regulatory agencies, students, and researchers with a stake in its taxonomy. In recognition of this monophyly, 40 species described as genus Neocosmospora were recombined in genus Fusarium, and nine others were renamed Fusarium.     We show the distribution of SARS-CoV-2 genetic clades over time and between countries and outline potential genomic surveillance objectives. We applied three available genomic nomenclature systems for SARS-CoV-2 to all sequence data from the WHO European Region available during the COVID-19 pandemic until 10 July 2020. We highlight the importance of real-time sequencing and data dissemination in a pandemic situation. We provide a comparison of the nomenclatures and lay a foundation for future European genomic surveillance of SARS-CoV-2.Peer reviewe
