    Determination of Additives in Cigarettes Utilizing Evolved Gas Analysis (EGA)

    While additives to consumer products must be listed in order by amount, it is not required to report the actual amounts of these additives. Given that impacts on human health are dose dependent, knowledge of additive amounts would aid discussions of the safety of these compounds for inhalation. Additives for cigarettes include flavors, such as menthol, and propylene glycol, a humectant. Rapid determination of the amounts of menthol and propylene glycol additives in cigarettes was accomplished utilizing evolved gas analysis (EGA) – gas chromatography (GC) – flame ionization detection (FID). Careful preparation of samples and appropriate sample storage are critical components to this study. Menthol amounts in cigarettes were consistent with previous studies and this represents the first report of propylene glycol amounts

    Powering Triton’s recent geological activity by obliquity tides: Implications for Pluto geology

    We investigate the origins of Triton’s deformed and young surface. Assuming Triton was captured early in solar system history, the bulk of the energy released during capture will have been lost, and cannot be responsible for its present-day activity. Radiogenic heating is sufficient to maintain a long-lived ocean beneath a conductive ice shell, but insufficient to cause convective deformation and yielding at the surface. However, Triton’s high inclination likely causes a significant ( 0.7) obliquity, resulting in large heat fluxes due to tidal dissipation in any subsurface ocean. For a 300 km thick ice shell, the estimated ocean heat production rate (0.3 TW) is capable of producing surface yielding and mobile-lid convection. Requiring convection places an upper bound on the ice shell viscosity, while the requirement for yielding imposes a lower bound. Both bounds can be satisfied with an ocean temperature 240 K for our nominal temperature-viscosity relationship, suggesting the presence of an antifreeze such as NH3. In our view, Triton’s geological activity is driven by obliquity tides, which arise because of its inclination. In contrast, Pluto is unlikely to be experiencing significant tidal heating. While Pluto may have experienced ancient tectonic deformation, we do not anticipate seeing the kind of young, deformed surfaces seen at Triton

    The Role of Trust in Public Attitudes toward Invasive Species Management on Guam: A Case Study

    Public attitudes toward invasive alien species management and trust in managers’ ability to effectively manage non-native species can determine public support for conservation action. Guam has experienced widespread species loss and ecosystem transformation due to invasive species. Despite Guam’s long history with invasives and efforts to eradicate them, we know little about the sociological context of invasive species. Using focused group discussions, we explore public attitudes toward invasive species management. Respondents expressed support for management activities and a desire to participate directly in conservation actions. Participants also expressed frustration with government institutions and lack of confidence in managers’ abilities to control invasive species. Perceptions of managers’ trustworthiness, communication with managers, and positive personal experiences with managers were related to positive attitudes about management and support for existing initiatives

    Unique spectral signatures of the nucleic acid dye acridine orange can distinguish cell death by apoptosis and necroptosis

    Cellular injury and death are ubiquitous features of disease, yet tools to detect them are limited and insensitive to subtle pathological changes. Acridine orange (AO), a nucleic acid dye with unique spectral properties, enables real-time measurement of RNA and DNA as proxies for cell viability during exposure to various noxious stimuli. This tool illuminates spectral signatures unique to various modes of cell death, such as cells undergoing apoptosis versus necrosis/ necroptosis. This new approach also shows that cellular RNA decreases during necrotic, necroptotic, and apoptotic cell death caused by demyelinating, ischemic, and traumatic injuries, implying its involvement in a wide spectrum of tissue pathologies. Furthermore, cells with pathologically low levels of cytoplasmic RNA are detected earlier and in higher numbers than with standard markers including TdT-mediated dUTP biotin nick-end labeling and cleaved caspase 3 immunofluorescence. Our technique highlights AO-labeled cytoplasmic RNA as an important early marker of cellular injury and a sensitive indicator of various modes of cell death in a range of experimental models

    Maternal Obesity and Western-Style Diet Impair Fetal and Juvenile Offspring Skeletal Muscle Insulin-Stimulated Glucose Transport in Nonhuman Primates

    Infants born to mothers with obesity have a greater risk for childhood obesity and metabolic diseases; however, the underlying biological mechanisms remain poorly understood. We used a Japanese macaque model to investigate whether maternal obesity combined with a Western-style diet (WSD) impairs offspring muscle insulin action. Adult females were fed a control or WSD prior to and during pregnancy through lactation, and offspring subsequently weaned to a control or WSD. Muscle glucose uptake and signaling were measured ex vivo in fetal (n = 5-8/group) and juvenile (n = 8/group) offspring. In vivo signaling was evaluated after an insulin bolus just prior to weaning (n = 4-5/group). Maternal WSD reduced insulin-stimulated glucose uptake and impaired insulin signaling at the level of Akt phosphorylation in fetal muscle. In juvenile offspring, insulin-stimulated glucose uptake was similarly reduced by both maternal and postweaning WSD and corresponded to modest reductions in insulin-stimulated Akt phosphorylation relative to controls. We conclude that maternal WSD leads to a persistent decrease in offspring muscle insulin-stimulated glucose uptake even in the absence of increased offspring adiposity or markers of systemic insulin resistance. Switching offspring to a healthy diet did not reverse the effects of maternal WSD on muscle insulin action, suggesting earlier interventions may be warranted
