    Agrobacterium-mediated transformation and regeneration of transgenic 'Chancellor' wine grape plants expressing the tfdA gene

    The establishment of grape vineyards in the Midwestern USA is greatly hampered by the use of the herbicide 2,4-D in grain crop fields. The overall goal of this study was to transform 'Chancellor' with the plant expressible bacterial tfdA gene to make it tolerant to 2,4-D. Embryogenic callus was infected and cocultivated with an Agrobacterium construct (LBA4404: pAL4404::tfdA); transformed callus was selected with kanamycin; kanamycin resistant callus was bulked and transformed cell lines identified by PCR. Three PCRpositive embryogenic callus lines were used to regenerate transgenic plants. Analysis of the plant lines for the presence of the tfdA gene by PCR and southern hybridization confirmed its stable integration in their genomes. Transgenic 'Chancellor' grape plants regenerated from these calli proved to be resistant to up to 10 kg·ha-1 of a commercial ester-formulation of 2,4-D, indicating positive expression of the tfdA gene and affording them protection from 2,4-D injury

    Inhibiting the Thermal Gelation of Copolymer Stabilized Nonaqueous Dispersions and the Synthesis of Full Color PMMA Particles

    Polymeric particle dispersions have numerous potential applications; currently one of the most relevant is their use as inks in electrophoretic displays. These colloidal particles are synthesized from the appropriate monomer using nonaqueous dispersion (NAD) polymerization in a nonpolar solvent, which requires a stabilizer to control particle size and morphology. We have previously reported the facile synthesis of poly(methyl methacrylate)-block-poly(octadecyl acrylate) (PMMA-b-PODA) by atom transfer radical polymerization (ATRP), and its use in the NAD polymerization of MMA in hexane/dodecane solvent mixtures. Here we report the synthesis of monodisperse PMMA particles in dodecane following a standard “industrial” procedure using these PMMA-b-PODA stabilizers. However, it was observed that the particle suspensions solidified when they were left at temperatures below ?18 °C yet redispersed upon being heated. Differential scanning calorimetry, dynamic light scattering, and rheological studies demonstrated that this thermoresponsive behavior was due to a liquid–gel transition occurring at 17.5 °C as a consequence of the upper critical solution temperature of PODA in dodecane being traversed. Consequently, new copolymers were synthesized by ATRP with an ethylhexyl acrylate (EHA) co-monomer incorporated into the lyophilic (dodecane compatible) block. Dispersions stabilized by these PMMA-b-P(ODA-co-EHA) polymers with high EHA contents exhibited lower gelation temperatures because of the greater solvent compatibility with dodecane. The use of a PMMA65-b-(ODA10-co-EHA45) copolymer stabilizer (with the highest EHA content) gave PMMA dispersions that showed no gelation down to 4 °C and monodisperse cross-linked PMMA particles containing organic dyes (cyan, magenta, red, and black) giving colored particles across the size range of approximately 100–1300 nm

    Nutrition education in medical schools (NEMS) project: Promoting clinical nutrition in medical schools – Perspectives from different actors

    Background & aims: Nutrition education is not adequately represented in the medical curriculum, and this prompted the European Society for Clinical Nutrition and Metabolism (ESPEN) to launch the Nutrition Education in Medical Schools (NEMS) Project in 2017. The aim of this original paper was to describe the perspectives of different actors in the promotion of nutrition education in medical schools. Methods: On 11 November 2021, an online meeting was held on this topic, where nine representatives from different backgrounds participated in the scientific programme. More than 640 participants registered to this webinar. Results: The different models of Nutrition Education in Medical Schools were introduced by Prof. Cristina Cuerda (Spain) and Prof. Maurizio Muscaritoli (Italy). The students' perspective was given by Ms. Alexandra Archodoulakis (Germany) and Ms. Sila Gürbüz (Turkey), representing the European Medical Students' Association. The dietitian's perspective was given by Dr. Kirsten Berk (The Netherlands), whereas Dr. Matti Aapro (Switzerland) gave the medical doctor (oncology)'s perspective. Ms. Clare Farrand (Australia) gave the WHO perspective and Dr. Kristiina Patja (Finland) explained the healthy lifestyle teaching to medical students. Lastly, Prof. Michael Chourdakis (Greece) and Prof. Zeljko Krznaric (Croatia) hosted the round-table discussion. Conclusions: There was strong agreement among the representatives from different settings joining this ESPEN initiative that increasing nutritional knowledge and skills of young doctors is now possible and will launch a virtuous cycle that will proactively involve all the other healthcare professionals working in the nutritional field

    Spectral, mineralogical, and geochemical variations across Home Plate, Gusev Crater, Mars indicate high and low temperature alteration

    Over the last ~ 3 years in Gusev Crater, Mars, the Spirit rover observed coherent variations in color, mineralogy, and geochemistry across Home Plate, an ~ 80 m-diameter outcrop of basaltic tephra. Observations of Home Plate from orbit and from the summit of Husband Hill reveal clear differences in visible/near-infrared (VNIR) colors between its eastern and western regions that are consistent with mineralogical compositions indicated by Mössbauer spectrometer (MB) and by Miniature Thermal Emission Spectrometer (Mini-TES). Pyroxene and magnetite dominate the east side, while olivine, nanophase Fe oxide (npOx) and glass are more abundant on the western side. Alpha Particle X-Ray Spectrometer (APXS) observations reveal that eastern Home Plate has higher Si/Mg, Al, Zn, Ni, and K, while Cl and Br are higher in the west. We propose that these variations are the result of two distinct alteration regimes that may or may not be temporally related: a localized, higher temperature recrystallization and alteration of the east side of Home Plate and lower temperature alteration of the western side that produced npOx

    Cost and Outcome of Behavioural Activation versus Cognitive Behavioural Therapy for Depression (COBRA): a randomised, controlled, non-inferiority trial

    Background Depression is a common, debilitating, and costly disorder. Many patients request psychological therapy, but the best-evidenced therapy—cognitive behavioural therapy (CBT)—is complex and costly. A simpler therapy—behavioural activation (BA)—might be as effective and cheaper than is CBT. We aimed to establish the clinical efficacy and cost-effectiveness of BA compared with CBT for adults with depression. Methods In this randomised, controlled, non-inferiority trial, we recruited adults aged 18 years or older meeting Diagnostic and Statistical Manual of Mental Disorders IV criteria for major depressive disorder from primary care and psychological therapy services in Devon, Durham, and Leeds (UK). We excluded people who were receiving psychological therapy, were alcohol or drug dependent, were acutely suicidal or had attempted suicide in the previous 2 months, or were cognitively impaired, or who had bipolar disorder or psychosis or psychotic symptoms. We randomly assigned participants (1:1) remotely using computer-generated allocation (minimisation used; stratified by depression severity [Patient Health Questionnaire 9 (PHQ-9) score of <19 vs ≥19], antidepressant use, and recruitment site) to BA from junior mental health workers or CBT from psychological therapists. Randomisation done at the Peninsula Clinical Trials Unit was concealed from investigators. Treatment was given open label, but outcome assessors were masked. The primary outcome was depression symptoms according to the PHQ-9 at 12 months. We analysed all those who were randomly allocated and had complete data (modified intention to treat [mITT]) and also all those who were randomly allocated, had complete data, and received at least eight treatment sessions (per protocol [PP]). We analysed safety in the mITT population. The non-inferiority margin was 1·9 PHQ-9 points. This trial is registered with the ISCRTN registry, number ISRCTN27473954. Findings Between Sept 26, 2012, and April 3, 2014, we randomly allocated 221 (50%) participants to BA and 219 (50%) to CBT. 175 (79%) participants were assessable for the primary outcome in the mITT population in the BA group compared with 189 (86%) in the CBT group, whereas 135 (61%) were assessable in the PP population in the BA group compared with 151 (69%) in the CBT group. BA was non-inferior to CBT (mITT: CBT 8·4 PHQ-9 points [SD 7·5], BA 8·4 PHQ-9 points [7·0], mean difference 0·1 PHQ-9 points [95% CI −1·3 to 1·5], p=0·89; PP: CBT 7·9 PHQ-9 points [7·3]; BA 7·8 [6·5], mean difference 0·0 PHQ-9 points [–1·5 to 1·6], p=0·99). Two (1%) non-trial-related deaths (one [1%] multidrug toxicity in the BA group and one [1%] cancer in the CBT group) and 15 depression-related, but not treatment-related, serious adverse events (three in the BA group and 12 in the CBT group) occurred in three [2%] participants in the BA group (two [1%] patients who overdosed and one [1%] who self-harmed) and eight (4%) participants in the CBT group (seven [4%] who overdosed and one [1%] who self-harmed). Interpretation We found that BA, a simpler psychological treatment than CBT, can be delivered by junior mental health workers with less intensive and costly training, with no lesser effect than CBT. Effective psychological therapy for depression can be delivered without the need for costly and highly trained professionals. Funding National Institute for Health Research

    Alumina+Silica+/-Germanium Alteration in Smectite-Bearing Marathon Valley, Endeavour Crater Rim, Mars

    Mars Exploration Rover Opportunity has been exploring Mars for 12+ years, and is presently investigating the geology of a western rim segment of 22 kilometers diameter, Noachian- aged Endeavour crater. The Alpha Particle X-ray Spectrometer has determined the compositions of a pre-impact lithology, the Matijevic fm., and polymict impact breccias ejected from the crater, the Shoemaker fm. Opportunity is now investigating a region named Marathon Valley that cuts southwest-northeast through the central portion of the rim segment and provides a window into the lower stratigraphic record. (Geographic names used here are informal.) At the head of Marathon Valley, referred to here as Upper Marathon Valley, is a shallow, ovoid depression approximately 2535 millimeters in size, named Spirit of Saint Louis. Layering inside Spirit of Saint Louis appears continuous with the Upper Marathon Valley rocks outside, indicating they are coeval. Spirit of Saint Louis is partly bounded by approximately 10-20 centimeters wide zone containing reddish altered rocks (red zone). Red zones also form prominent curvilinear features in Marathon Valley. Compact Reconnaissance Imaging Spectrometer for Mars (CRISM) spectra provide evidence for a really extensive Fe-Mg smectite in the Marathon Valley region, indicating distinct styles of aqueous alteration. The CRISM detections of smectites are based on metal-OH absorptions at approximately 2.3 and 2.4 micron that are at least two times the background noise level

    Santorini, Another Meteorite on Mars and Third of a Kind

    The Mars Exploration Rover (MER) Opportunity has been studying Meridiani Planum for five years. On sol 1634 of its mission, Opportunity left Victoria crater after investigating it for approximately 682 sols [1] and is now on a journey towards Endeavour, a 24 km diameter crater about 12 km southeast of Victoria. A priority along the way is the investigation of cobbles, which in the jargon of the MER science team denotes any loose rock fragment larger than a couple of centimeters. Cobbles investigated thus far are of diverse origin [2] and provide the only means to investigate material other than the ubiquitous sulfate-rich outcrop, basaltic sand or hematiterich spherules dubbed blueberries. Some of these cobbles are meteorites [3]. Meteorites on Mars are not just a curiosity that make Mars a more Earth-like planet. Metallic iron in meteorites, for example, may be used as a more sensitive tracer for volatile surface interactions compared to igneous minerals [4]. Between sols 1713 and 1749, including the period of Mars solar conjunction, Opportunity investigated a cobble informally named Santorini. Its chemical and mineralogical composition is very similar to Barberton and Santa Catarina, two cobbles that were identified as meteorites and which are probably related to each other [3]. Santorini was investigated with the rover s Panoramic Camera (Pancam), Microscopic Imager (MI), Alpha-Particle X-ray Spectrometer (APXS) and Moessbauer (MB) spectrometer. The miniature Thermal Emission Spectrometer (mini-TES) was not operational at the time. The Rock Abrasion Tool (RAT) could not be used to brush off potential dust coatings because of unfavorable geometry

    Mineralogy and chemistry of cobbles at Meridiani Planum, Mars, investigated by the Mars Exploration Rover Opportunity

    Numerous loose rocks with dimensions of a few centimeters to tens of centimeters and with no obvious physical relationship to outcrop rocks have been observed along the traverse of the Mars Exploration Rover Opportunity. To date, about a dozen of these rocks have been analyzed with Opportunity&rsquo;s contact instruments, providing information about elemental chemistry (Alpha Particle X‐ray Spectrometer), iron mineralogy and oxidation states (M&ouml;ssbauer Spectrometer) and texture (Microscopic Imager). These &ldquo;cobbles&rdquo; appear to be impact related, and three distinct groups can be identified on the basis of chemistry and mineralogy. The first group comprises bright fragments of the sulfate‐rich bedrock that are compositionally and texturally indistinguishable from outcrop rocks. All other cobbles are dark and are divided into two groups, referred to as the &ldquo;Barberton group&rdquo; and the &ldquo;Arkansas group,&rdquo; after the first specimen of each that was encountered by Opportunity. Barberton group cobbles are interpreted as meteorites with an overall chemistry and mineralogy consistent with a mesosiderite silicate clast composition. Arkansas group cobbles appear to be related to Meridiani outcrop and contain an additional basaltic component. They have brecciated textures, pointing to an impact‐related origin during which local bedrock and basaltic material were mixed

    A feasibility study of educational tools for osteomalacia

    Many people in the UK, particularly people of South Asian origin, are advised to supplement their vitamin D intake, yet most do not. This suggests an unmet educational need. The osteomalacia mind map was developed to meet this need. The mind map contains culturally sensitive images, translated into Urdu and made interactive on a DVD. This study explores the feasibility of a randomised controlled study to measure the effect of education on improving vitamin D knowledge and adherence. This was a pilot and feasibility study. Cluster randomisation was used to avoid inter person contamination. Two South Asian women’s groups were recruited to receive information about osteomalacia either by interactive DVD or an Arthritis Research UK leaflet. Knowledge and compliance were tested before and after the educational interventions via a knowledge questionnaire and the measurement of vitamin D and parathormone levels. The groups were found to be mismatched for knowledge, educational attainment and language at baseline. There were also organisational difficulties and possible confounding due to different tutors and translators. The DVD group had high knowledge at baseline which did not improve. The leaflet group had low knowledge at baseline that did improve. The DVD group had lower parathormone which did not change. The leaflet group had an increase in vitamin D but parathormone remained high. Performing a randomised study with this population utilising an educational intervention was difficult to execute. If cluster randomisation is used, extreme care must be taken to match the groups at baseline

    Noachian Impact Ejecta on Murray Ridge and Pre-impact Rocks on Wdowiak Ridge, Endeavour Crater, Mars: Opportunity Observations

    Mars Exploration Rover Opportunity has been exploring Meridiani Planum since January 2004, and has completed 4227% of its primary mission. Opportunity has been investigating the geology of the rim of 22 km diameter Endeavour crater, first on the Cape York segment and now on Cape Tribulation. The outcrops are divided York; (ii) the Shoemaker fm, impact breccias representing ejecta from the crater; into three formations: (i) the lower Matijevic fm, a pre-impact lithology on Cape and (iii) the upper Grasberg fm, a post-impact deposit that drapes the lower portions of the eroded rim segments. On the Cape Tribulation segment Opportunity has been studying the rocks on Murray Ridge, with a brief sojourn to Wdowiak Ridge west of the rim segment. team member Thomas Wdowiak, who died in 2013.) One region of Murray Ridge has distinctive CRISM spectral characteristics indicating the presence of a small concentration of aluminous smectite based on a 2.2 micron Al-OH combination band (hereafter, the Al-OH region)
