    Biotinidase deficiency: Genotype-biochemical phenotype association in Brazilian patients

    [EN] The association between the BTD genotype and biochemical phenotype [profound biotinidase deficiency (BD), partial BD or heterozygous activity] is not always consistent. This study aimed to investigate the genotype-biochemical phenotype association in patients with low biotinidase activity. Methods All exons, the 5'UTR and the promoter of the BTD gene were sequenced in 72 Brazilian individuals who exhibited low biotinidase activity. For each patient, the expected biochemical phenotype based on the known genotype was compared with the observed biochemical phenotype. Additional non-genetic factors that could affect the biotinidase activity were also analysed. Most individuals were identified by neonatal screening (n = 66/72). When consecutive results for the same patient were compared, age, prematurity and neonatal jaundice appeared to affect the level of biotinidase activity. The biochemical phenotype at the time of the second blood collection changed in 11/22 patients compared to results from the first sample. Three novel variants were found: c.1337T>C (p.L446P), c.1466A>G (p.N489S) and c.962G>A (p.W321*). Some patients with the same genotype presented different biochemical phenotypes. The expected and observed biochemical phenotypes agreed in 68.5% of cases (concordant patients). The non-coding variants c.-183G>A, c.-315A>G and c.-514C>T were present in heterozygosis in 5/17 discordant patients. In addition, c.- 183G>A and c.-514C>T were also present in 10/37 concordant patients. The variants found in the promoter region do not appear to have a strong impact on biotinidase activity. Since there is a disparity between the BTD genotype and biochemical phenotype, and biotinidase activity may be affected by both genetic and non-genetic factors, we suggest that the diagnosis of BD should be based on more than one measurement of plasma biotinidase activity.     Impact of interaction between chronic variable stress and moderate intensity physical exercise on antibody production in Wistar rats

    Stress in its chronic form has been an important factor in the onset of depression. In addition, it can lead to immunosuppression. On the other hand, physical exercise has a protective action on the immune system and reduces the signs and symptoms of stress. Studies on the effects of chronic variable stress (CVS) and moderate intensity physical exercise (MIPE) on humoral immunity are scarce. Thus, in this study, we investigated the interaction between the effects of CVS and MIPE on antibody production. Wistar rats were divided into four groups: control (C); physical exercise (P); stress (S); and physical exercise and stress (PS). The P and PS groups were trained in MIPE for six weeks. From the fourth week of the study, concomitant with the MIPE, the S and PS groups were subjected to CVS. To evaluate the production of antibodies, all groups were immunized. Regarding antibody production, it was observed that females in the C and S groups presented higher IgM antibody production in relation to males. Furthermore, the production of IgM was potentiated in males of the PS group. On the other hand, no significant differences were observed in relation to the production of IgG1 and IgG2a. We conclude that CVS prevented the increase in IgM production in male rats and MIPE was effective in reversing the effects of stress on the production of IgM in male rats. On the other hand, the production of IgG1 and IgG2a was not affected by MIPE or CVS

    Avaliação de Características Químicas do Solo Durante a Decomposição Cadavérica e suas AplicaçÔes à Química Forense

    The estimation of the Post-Mortem Interval (PMI) is a primordial parameter in the investigation of crimes against life. In tropical climate areas, the cadaveric decomposition occurs in an accelerated form, preventing the PMI estimation by usual methods. Understanding how different circumstances affect the cadaveric composition is very important to develop methods that enable the PMI estimation in several climatic conditions. One of the alternatives, however not much explored in Brazil, refers to the use of soil chemistry for the determination of longstanding PMIs. In this research, variations in soil chemical properties were monitored during swine carcasses cadaverous decomposition in a tropical forest area in order to recognize patterns and evaluate the applicability of these by the criminal expertise. Among the evaluated characteristics are the levels of pH, inorganic nitrogen (ammonia and nitrate), total nitrogen, carbon nitrogen proportion (C: N), exchangeable aluminum, available phosphorus and macro and micronutrients. The results showed distinct variations according to the decomposition phases observed and also to the way the carcasses were deposited in the study environment (above or below the soil), evidencing the possibility of using these variations in the development of models for the prediction of postmortem intervals lasting up to 67 days. © 2018 Secretaria Regional do Rio de Janeiro da Sociedade Brasileira de Quimica.     The objective of this work was to determine the proximate composition, yield, the influence of frozen storage time on the quality, and the cut type x shelf life relation of rib, tenderloin and flitch cuts of tambaqui (Colossoma macropomum) reared in a fish farm, during 180 days of cold storage. The cuts were stored in polyethylene bags and frozen at -25oC. Sensory, physicochemical and microbiological analyses were carried out. The pH, total volatile basic nitrogen, and thiobarbituric acid-reactive substance values, as well as the results of the microbiological analyses, proved to be good indexes for freshness evaluation along the storage time. The cut rib was more susceptible to oxidation than the other cuts. However, the cuts remained appropriate for consumption during the experiment, under correct handling and good hygienic practices between harvest and storage

    Measurement of the Bs0→J/ψKS0B_s^0\to J/\psi K_S^0 branching fraction

    The Bs0→J/ψKS0B_s^0\to J/\psi K_S^0 branching fraction is measured in a data sample corresponding to 0.41fb−1fb^{-1} of integrated luminosity collected with the LHCb detector at the LHC. This channel is sensitive to the penguin contributions affecting the sin2ÎČ\beta measurement from B0→J/ψKS0B^0\to J/\psi K_S^0 The time-integrated branching fraction is measured to be BF(Bs0→J/ψKS0)=(1.83±0.28)×10−5BF(B_s^0\to J/\psi K_S^0)=(1.83\pm0.28)\times10^{-5}. This is the most precise measurement to date

    Measurement of the CP-violating phase \phi s in Bs->J/\psi\pi+\pi- decays

    Measurement of the mixing-induced CP-violating phase phi_s in Bs decays is of prime importance in probing new physics. Here 7421 +/- 105 signal events from the dominantly CP-odd final state J/\psi pi+ pi- are selected in 1/fb of pp collision data collected at sqrt{s} = 7 TeV with the LHCb detector. A time-dependent fit to the data yields a value of phi_s=-0.019^{+0.173+0.004}_{-0.174-0.003} rad, consistent with the Standard Model expectation.     Measurement of the ratio of branching fractions BR(B0 -> K*0 gamma)/BR(Bs0 -> phi gamma) and the direct CP asymmetry in B0 -> K*0 gamma

    The ratio of branching fractions of the radiative B decays B0 -> K*0 gamma and Bs0 phi gamma has been measured using an integrated luminosity of 1.0 fb-1 of pp collision data collected by the LHCb experiment at a centre-of-mass energy of sqrt(s)=7 TeV. The value obtained is BR(B0 -> K*0 gamma)/BR(Bs0 -> phi gamma) = 1.23 +/- 0.06(stat.) +/- 0.04(syst.) +/- 0.10(fs/fd), where the first uncertainty is statistical, the second is the experimental systematic uncertainty and the third is associated with the ratio of fragmentation fractions fs/fd. Using the world average value for BR(B0 -> K*0 gamma), the branching fraction BR(Bs0 -> phi gamma) is measured to be (3.5 +/- 0.4) x 10^{-5}. The direct CP asymmetry in B0 -> K*0 gamma decays has also been measured with the same data and found to be A(CP)(B0 -> K*0 gamma) = (0.8 +/- 1.7(stat.) +/- 0.9(syst.))%. Both measurements are the most precise to date and are in agreement with the previous experimental results and theoretical expectations.Comment: 21 pages, 3 figues, 4 table
