Flight test evaluation of a method to determine the level flight performance of a propeller-driven aircraft
The overall drag of the aircraft is expressed in terms of the measured increment of power required to overcome a corresponding known increment of drag, which is generated by a towed drogue. The simplest form of the governing equations, D = delta D SHP/delta SHP, is such that all of the parameters on the right side of the equation can be measured in flight. An evaluation of the governing equations has been performed using data generated by flight test of a Beechcraft T-34B. The simplicity of this technique and its proven applicability to sailplanes and small aircraft is well known. However, the method fails to account for airframe-propulsion system
Implementation of A Least Squares Method To A Navier-Stokes Solver
The Navier-Stokes equations are used to model fluid flow. Examples include fluid structure interactions in the heart, climate and weather modeling, and flow simulations in computer gaming and entertainment. The equations date back to the 1800s, but research and development of numerical approximation algorithms continues to be an active area. To numerically solve the Navier-Stokes equations we implement a least squares finite element algorithm based on work by Roland Glowinski and colleagues. We use the deal.II academic library , the C++ language, and the Linux operating system to implement the solver. We investigate convergence rates and apply the least squares solver to the lid driven cavity problem and discuss results
Centrosome defects cause microcephaly by activating the 53BP1-USP28-TP53 mitotic surveillance pathway
Mutations in centrosome genes deplete neural progenitor cells (NPCs) during brain development, causing microcephaly. While NPC attrition is linked to TP53-mediated cell death in several microcephaly models, how TP53 is activated remains unclear. In cultured cells, mitotic delays resulting from centrosome loss prevent the growth of unfit daughter cells by activating a pathway involving 53BP1, USP28, and TP53, termed the mitotic surveillance pathway. Whether this pathway is active in the developing brain is unknown. Here, we show that the depletion of centrosome proteins in NPCs prolongs mitosis and increases TP53-mediated apoptosis. Cell death after a delayed mitosis was rescued by inactivation of the mitotic surveillance pathway. Moreover, 53BP1 or USP28 deletion restored NPC proliferation and brain size without correcting the upstream centrosome defects or extended mitosis. By contrast, microcephaly caused by the loss of the non-centrosomal protein SMC5 is also TP53-dependent but is not rescued by loss of 53BP1 or USP28. Thus, we propose that mutations in centrosome genes cause microcephaly by delaying mitosis and pathologically activating the mitotic surveillance pathway in the developing brain
Phylogeography of an endangered disjunct herb: long-distance dispersal, refugia and colonization routes
Quaternary glacial cycles appear to have had a consistent role in shaping the genetic diversity and
structure of plant species. Despite the unusual combination of the characteristics of the western Mediterranean–
Macaronesian area, there are no studies that have specifically examined the effects of palaeoclimatic and palaeogeographic
factors on the genetic composition and structure of annual herbs. Astragalus edulis is a disjunct endemic
found in the easternmost Canary Islands and the semi-arid areas of north-eastern Africa and south-eastern Iberian
Peninsula. This endangered species shows no evident adaptations to long-distance dispersal. Amplified fragment
length polymorphism (AFLP) data and plastid DNA sequences were analysed from a total of 360 individuals distributed
throughout the range of this species. The modelled potential distribution of A. edulis under current conditions
was projected over the climatic conditions of the Last Interglacial (130 ka BP) and Last Glacial Maximum (21
ka BP) to analyse changes in habitat suitability and to look for associations between the modelling and genetic
results. Amplified fragment length polymorphism analysis showed clear phylogeographic structure with four distinct
genetic clusters. Approximate Bayesian computation (ABC) models based on plastid DNA sequences indicated a
Middle Pleistocene long-distance dispersal event as the origin of the populations of the Canary Islands. The models
also suggested south-western Morocco as the ancestral area for the species, as well as subsequent colonization of
north-eastern Morocco and the Iberian Peninsula. The data compiled indicated the possibility of the presence of refuge
areas at favourable locations around the High Atlas and Anti-Atlas mountain ranges. Moreover, palaeodistribution
models strongly support the events inferred by ABC modelling and show the potential distribution of the species
in the past, suggesting a putative colonization route.This work has been financed by the Spanish Ministerio

Consistent phenological shifts in the making of a biodiversity hotspot: the Cape flora
Background
The best documented survival responses of organisms to past climate change on short (glacial-interglacial) timescales are distributional shifts. Despite ample evidence on such timescales for local adaptations of populations at specific sites, the long-term impacts of such changes on evolutionary significant units in response to past climatic change have been little documented. Here we use phylogenies to reconstruct changes in distribution and flowering ecology of the Cape flora - South Africa's biodiversity hotspot - through a period of past (Neogene and Quaternary) changes in the seasonality of rainfall over a timescale of several million years.
Results
Forty-three distributional and phenological shifts consistent with past climatic change occur across the flora, and a comparable number of clades underwent adaptive changes in their flowering phenology (9 clades; half of the clades investigated) as underwent distributional shifts (12 clades; two thirds of the clades investigated). Of extant Cape angiosperm species, 14-41% have been contributed by lineages that show distributional shifts consistent with past climate change, yet a similar proportion (14-55%) arose from lineages that shifted flowering phenology.
Conclusions
Adaptive changes in ecology at the scale we uncover in the Cape and consistent with past climatic change have not been documented for other floras. Shifts in climate tolerance appear to have been more important in this flora than is currently appreciated, and lineages that underwent such shifts went on to contribute a high proportion of the flora's extant species diversity. That shifts in phenology, on an evolutionary timescale and on such a scale, have not yet been detected for other floras is likely a result of the method used; shifts in flowering phenology cannot be detected in the fossil record
Possible relationship between Seismic Electric Signals (SES) lead time and earthquake stress drop
Stress drop values for fourteen large earthquakes with MW ≥ 5.4 which occurred in Greece during the period 1983–2007 are available. All these earthquakes were preceded by Seismic Electric Signals (SES). An attempt has been made to investigate possible correlation between their stress drop values and the corresponding SES lead times. For the stress drop, we considered the Brune stress drop, ΔσB, estimated from far field body wave displacement source spectra and ΔσSB derived from the strong motion acceleration response spectra. The results show a relation may exist between Brune stress drop, ΔσB, and lead time which implies that earthquakes with higher stress drop values are preceded by SES with shorter lead time
Stakeholder engagement in the city branding process
This paper explores perceptions of stakeholder engagement in the city branding process from the perspective of two post-industrial cities: Sheffield, UK and Essen, Germany. This qualitative research utilises a multi case study approach, which allowed for semi-structure interviews and semiotics to be used. Preliminary findings highlight that there are four stakeholder ‘levels’. Each of these stakeholder groupings is involved in the city branding process to some extend. Findings suggest that the degree of involvement strongly depends on the primary stakeholders, who are seen as key decision-makers in the branding process. These primary stakeholders select other stakeholders that ‘can’ be involved in the branding process. Although this may be beneficial it is vital to provide more opportunities and incorporate stakeholders that are willing to participate in the branding process. Alienating stakeholders may also lead to losing parts of an identity that is based on heritage. The focus is on two cities with a highly industrialised background, thus findings may not be applicable to cities without this heritage. The paper looks at both stakeholder engagement and city branding, thereby proposing four layers of stakeholder involvement in the city branding process
Modulation of apoptosis in human hepatocellular carcinoma (HepG2 cells) by a standardized herbal decoction of Nigella sativa seeds, Hemidesmus indicus roots and Smilax glabra rhizomes with anti- hepatocarcinogenic effects
<p>Abstract</p> <p>Background</p> <p>A standardized poly-herbal decoction of <it>Nigella sativa </it>seeds, <it>Hemidesmus indicus </it>roots and <it>Smilax glabra </it>rhizomes used traditionally in Sri Lanka for cancer therapy has been demonstrated previously, to have anti-hepatocarcinogenic potential. Cytotoxicity, antioxidant activity, anti-inflammatory activity, and up regulation of p53 and p21 activities are considered to be some of the possible mechanisms through which the above decoction may mediate its anti-hepatocarcinogenic action. The main aim of the present study was to determine whether apoptosis is also a major mechanism by which the decoction mediates its anti-hepatocarcinogenic action.</p> <p>Methods</p> <p>Evaluation of apoptosis in HepG2 cells was carried out by (a) microscopic observations of cell morphology, (b) DNA fragmentation analysis, (c) activities of caspase 3 and 9, as well as by (d) analysis of the expression of pro-apoptotic (Bax) and anti-apoptotic (Bcl-2) proteins associated with cell death.</p> <p>Results</p> <p>The results demonstrated that in HepG2 cells, the decoction can induce (a) DNA fragmentation and (b) characteristic morphological changes associated with apoptosis (nuclear condensation, membrane blebbing, nuclear fragmentation and apoptotic bodies). The decoction could also, in a time and dose dependent manner, up regulate the expression of the pro-apoptotic gene <it>Bax </it>and down regulate expression of anti-apoptotic <it>Bcl-2 </it>gene (as evident from RT-PCR analysis, immunohistochemistry and western blotting). Further, the decoction significantly (<it>p </it>< .001) enhanced the activities of caspase-3 and caspase-9 in a time and dose dependent manner.</p> <p>Conclusions</p> <p>Overall findings provide confirmatory evidence to demonstrate that the decoction may mediate its reported anti-hepatocarcinogenic effect, at least in part, through modulation of apoptosis.</p
Phenotyping pipeline reveals major seedling root growth QTL in hexaploid wheat
Seedling root traits of wheat (Triticum aestivum L.) have been shown to be important for efficient establishment and linked to mature plant traits such as height and yield. A root phenotyping pipeline, consisting of a germination paper-based screen combined with image segmentation and analysis software, was developed and used to characterize seedling traits in 94 doubled haploid progeny derived from a cross between the winter wheat cultivars Rialto and Savannah. Field experiments were conducted to measure mature plant height, grain yield, and nitrogen (N) uptake in three sites over 2 years. In total, 29 quantitative trait loci (QTLs) for seedling root traits were identified. Two QTLs for grain yield and N uptake co-localize with root QTLs on chromosomes 2B and 7D, respectively. Of the 29 root QTLs identified, 11 were found to co-localize on 6D, with four of these achieving highly significant logarithm of odds scores (>20). These results suggest the presence of a major-effect gene regulating seedling root vigour/growth on chromosome 6D
