    Parachlamydia spp. and Related Chlamydia-like Organisms and Bovine Abortion

    Chlamydophila abortus and Waddlia chondrophila cause abortion in ruminants. We investigated the role of Parachlamydia acanthamoebae in bovine abortion. Results of immunohistochemical analyses were positive in 30 (70%) of 43 placentas from which Chlamydia-like DNA was amplified, which supports the role of Parachlamydia spp. in bovine abortion

    Protochlamydia naegleriophila as Etiologic Agent of Pneumonia

    Using ameba coculture, we grew a Naegleria endosymbiont. Phenotypic, genetic, and phylogenetic analyses supported its affiliation as Protochlamydia naegleriophila sp. nov. We then developed a specific diagnostic PCR for Protochlamydia spp. When applied to bronchoalveolar lavages, results of this PCR were positive for 1 patient with pneumonia. Further studies are needed to assess the role of Protochlamydia spp. in pneumonia

    Paying attention to the spiritual flourishing of young children in church toddler groups: a scoping study evaluating the feasibility of a research study in this context

    Attention to the spiritual flourishing of young children contributes to their well-being, both their being and their becoming, but much-needed research in this area poses many challenges. The National Institute for Christian Education Research is undertaking a funded research project investigating the spiritual flourishing of young children in church toddler groups in England. This article reports on a scoping study which investigated the feasibility of a major research study in this area. The study was multifaceted, it involved clarifying terminology, evaluating previous research studies and early years research methods, and led to the development of a potential research framework. The article defines spiritual flourishing as a dynamic state of being, revealed in nurturing the right relationship with self, others, creation, and the transcendent. The findings propose an understanding of children’s spirituality as relational, based in the here and now, and shaped by the environment. Secondly, an exploration of midweek church toddler groups, a common feature across the denominations, through the lens of belonging, behaving, and believing reveals a potential critical contribution to young children’s spiritual flourishing or faith development. Thirdly, following an evaluation of potential effective research methods, the discussion highlights the value of an interdisciplinary study, the potential challenges and proposes a research framework. The article draws attention to the urgent need for more research to understand what contributes to young children’s spiritual flourishing within a Christian context

    Molecular mechanisms of cell death: recommendations of the Nomenclature Committee on Cell Death 2018.

    Over the past decade, the Nomenclature Committee on Cell Death (NCCD) has formulated guidelines for the definition and interpretation of cell death from morphological, biochemical, and functional perspectives. Since the field continues to expand and novel mechanisms that orchestrate multiple cell death pathways are unveiled, we propose an updated classification of cell death subroutines focusing on mechanistic and essential (as opposed to correlative and dispensable) aspects of the process. As we provide molecularly oriented definitions of terms including intrinsic apoptosis, extrinsic apoptosis, mitochondrial permeability transition (MPT)-driven necrosis, necroptosis, ferroptosis, pyroptosis, parthanatos, entotic cell death, NETotic cell death, lysosome-dependent cell death, autophagy-dependent cell death, immunogenic cell death, cellular senescence, and mitotic catastrophe, we discuss the utility of neologisms that refer to highly specialized instances of these processes. The mission of the NCCD is to provide a widely accepted nomenclature on cell death in support of the continued development of the field

    Given the low sensitivity of amoebal coculture, we developed a specific real-time PCR for the detection of Parachlamydia. The analytical sensitivity was high, and the inter- and intrarun variabilities were low. When the PCR was applied to nasopharyngeal aspirates, it was positive for six patients with bronchiolitis. Future studies should assess the role of Parachlamydia in bronchiolitis

    Tissue microarray technology was used to establish immunohistochemistry protocols and to determine the specificity of new antisera against various Chlamydia-like bacteria for future use on formalin-fixed and paraffin-embedded tissues. The antisera exhibited strong reactivity against autologous antigen and closely related heterologous antigen, but no cross-reactivity with distantly related species
