    So round the spiral again: a reflective participatory research project with children and young people

    Historically the voices of children in research have been silent. They are often seen as victims or beneficiaries of research rather than co-researchers or partners. This is beginning to change with rowing awareness that involving children in the design, delivery and evaluation of services can make services more accessible to them and their peers. This article reviews the processes involved n a research project commissioned by Children’s Fund, which investigated the use and non-use of services within a local area. The involvement of children was paramount and resulted in the recruitment f nine young researchers between the ages of 7–13. Various cycles of participatory action research evolved throughout the project and this article focuses specifically on two—recruiting the researcher and training young researchers. We consider the cycles of reflection and action crucial to any participatory project and discuss how lessons were learned to inform further stages of the process.     The genome of influenza A viruses (IAV) is split into eight viral RNAs (vRNAs) that are encapsidated as viral ribonucleoproteins. The existence of a segment-specific packaging mechanism is well established, but the molecular basis of this mechanism remains to be deciphered. Selective packaging could be mediated by direct interaction between the vRNA packaging regions, but such interactions have never been demonstrated in virions. Recently, we showed that the eight vRNAs of a human H3N2 IAV form a single interaction network in vitro that involves regions of the vRNAs known to contain packaging signals in the case of H1N1 IAV strains. Here, we show that the eight vRNAs of an avian H5N2 IAV also form a single network of interactions in vitro, but, interestingly, the interactions and the regions of the vRNAs they involve differ from those described for the human H3N2 virus. We identified the vRNA sequences involved in five of these interactions at the nucleotide level, and in two cases, we validated the existence of the interaction using compensatory mutations in the interacting sequences. Electron tomography also revealed significant differences in the interactions taking place between viral ribonucleoproteins in H5N2 and H3N2 virions, despite their canonical '7 + 1' arrangement

    Monitoring ten insect pests in selected orchards in three Azorean Islands : The project CUARENTAGRI

    BACKGROUND: The data we present are part of the CUARENTAGRI project, which involves all archipelagos of the Macaronesia (Azores, Madeira, Canary Islands and Cabo Verde). The project aims to: i) identify and evaluate the risks associated with the introduction of new arthropod pests; ii) study the population dynamics of selected arthropod pest species currently responsible for the damage of key target crops and iii) develop monitoring systems, based on prediction and/or population dynamics of the crop pests, creating warnings and a phytosanitary prevention system. In this contribution, we compile data for three Azorean Islands (Terceira, São Jorge and São Miguel Islands), where pheromone-baited traps were placed in pastures, potato fields and several orchards’ types (apples, banana, chestnuts, olives, orange and strawberry), during three consecutive years (2020, 2021 and 2022). NEW INFORMATION: A total of 114,827 specimens of insects (Arthropoda, Insecta) were collected, belonging to four orders, six families and ten recorded pest species. A total of eight species are considered introduced (Cosmopolites sordidus (Germar, 1824), Drosophila suzukii (Matsumura, 1931), Bactrocera oleae (Rossi, 1790), Ceratitis capitata (Wiedemann, 1824), Phthorimaea operculella (Zeller, 1873), Cydia pomonella (Linnaeus, 1758), Cydia splendana (Hübner, 1799) and Grapholita molesta (Busck, 1916); n = 84,986 specimens) and two native non-endemic (Mythimna unipuncta (Haworth, 1809) and Spodoptera littoralis (Boisduval, 1833); n = 17,465 specimens). This study intended to contribute to a better knowledge of the arthropods pests that can affect the Azorean crops and will serve as a baseline for future monitoring actions, pest risk assessments and prevention systems.This work was financed under the project CUARENTAGRI by Cooperation Programs INTERREG V A (Spain-Portugal) and MAC 2014-2020.     Influenza A viruses alter the stability and antiviral contribution of host E3-ubiquitin ligase Mdm2 during the time-course of infection

    International audienceThe interplay between influenza A viruses (IAV) and the p53 pathway has been reported in several studies, highlighting the antiviral contribution of p53. Here, we investigated the impact of IAV on the E3-ubiquitin ligase Mdm2, a major regulator of p53, and observed that IAV targets Mdm2, notably via its non-structural protein (NS1), therefore altering Mdm2 stability, p53/Mdm2 interaction and regulatory loop during the time-course of infection. This study also highlights a new antiviral facet of Mdm2 possibly increasing the list of its many p53-independent functions. Altogether, our work contributes to better understand the mechanisms underlining the complex interactions between IAV and the p53 pathway, for which both NS1 and Mdm2 arise as key players

    Rescue of a H3N2 Influenza Virus Containing a Deficient Neuraminidase Protein by a Hemagglutinin with a Low Receptor-Binding Affinity

    Influenza viruses possess at their surface two glycoproteins, the hemagglutinin and the neuraminidase, of which the antagonistic functions have to be well balanced for the virus to grow efficiently. Ferraris et al. isolated in 2003–2004 viruses lacking both a NA gene and protein (H3NA- viruses) (Ferraris O., 2006, Vaccine, 24(44–46):6656-9). In this study we showed that the hemagglutinins of two of the H3NA- viruses have reduced affinity for SAα2.6Gal receptors, between 49 and 128 times lower than that of the A/Moscow/10/99 (H3N2) virus and no detectable affinity for SAα2.3Gal receptors. We also showed that the low hemagglutinin affinity of the H3NA- viruses compensates for the lack of NA activity and allows the restoration of the growth of an A/Moscow/10/99 virus deficient in neuraminidase. These observations increase our understanding of H3NA- viruses in relation to the balance between the functional activities of the neuraminidase and hemagglutinin

    Youth social behaviour and network therapy (Y-SBNT) : adaptation of a family and social network intervention for young people who misuse alcohol and drugs – a randomised controlled feasibility trial

    Background: Family interventions appear to be effective at treating young people’s substance misuse. However, implementation of family approaches in UK services is low. This study aimed to demonstrate the feasibility of recruiting young people to an intervention based on an adaptation of adult social behaviour and network therapy. It also sought to involve young people with experience of using substance misuse services in the research process. Objectives: To demonstrate the feasibility of recruiting young people to family and social network therapy and to explore ways in which young people with experience of using substance misuse services could be involved in a study of this nature. Design: A pragmatic, two-armed, randomised controlled open feasibility trial. Setting: Two UK-based treatment services for young people with substance use problems, with recruitment taking place from May to November 2014. Participants: Young people aged 12–18 years, newly referred and accepted for structured interventions for drug and/or alcohol problems. Interventions: A remote, web-based computer randomisation system allocated young people to adapted youth social behaviour and network therapy (Y-SBNT) or treatment as usual (TAU). Y-SBNT participants were intended to receive up to six 50-minute sessions over a maximum of 12 weeks. TAU participants continued to receive usual care delivered by their service. Main outcome measures: Feasibility was measured by recruitment rates, retention in treatment and follow-up completion rates. The main clinical outcome was the proportion of days on which the main problem substance was used in the preceding 90-day period as captured by the Timeline Follow-Back interview at 3 and 12 months. Results: In total, 53 young people were randomised (Y-SBNT, n = 26; TAU, n = 27) against a target of 60 (88.3%). Forty-two young people attended at least one treatment session [Y-SBNT 22/26 (84.6%); TAU 20/27 (74.1%)]; follow-up rates were 77.4% at month 3 and 73.6% at month 12. Data for nine young people were missing at both months 3 and 12, so the main clinical outcome analysis was based on 24 young people (92.3%) in the Y-SBNT group and 20 young people (74.1%) in the TAU group. At month 12, the average proportion of days that the main problem substance was used in the preceding 90 days was higher in the Y-SBNT group than in the TAU group (0.54 vs. 0.41; adjusted mean difference 0.13, 95% confidence interval –0.12 to 0.39; p = 0.30). No adverse events were reported. Seventeen young people with experience of substance misuse services were actively involved throughout the study. They informed key elements of the intervention and research process, ensuring that the intervention was acceptable and relevant to our target groups; contributing to the design of key trial documents, ideas for a new model of public involvement and this report. Two parents were also involved. Conclusions: The adapted intervention could be delivered in young people’s services, and qualitative interviews found that Y-SBNT was acceptable to young people, family members and staff. Engagement of family and network members proved difficult within the intervention and research aspects. The study proved the feasibility of this work in routine services but outcome measurement based on narrow substance use variables may be limited and may fail to capture other important changes in wider areas of functioning for young people. Validation of the EuroQol-5 Dimensions for young people aged 12–18 years should be considered and flexible models for involvement of young people in research are required to achieve inclusive representation throughout all aspects of the research process. Although recommendation of a full trial of the Y-SBNT intervention compared with TAU is not supported, this study can inform future intervention development and UK research within routine addiction services.     Microbiological disinfection of water and air by photocatalysis

In water disinfection, two model strains of Escherichia coli (K12 PHL849 and K12 PHL1273) were selected and a comparison of the efficiencies of TiO2 Degussa P-25 versus TiO2 Millennium PC500 were estimated. A more important inactivation of E. coli PHL1273 was obtained on TiO2 Millennium PC500, in line with its better adherence on this solid. An experimental study was performed using a dialysis membrane to investigate the impact of the contact between the microorganisms and the photocatalyst and to determine the role of H2O2 generated in situ. In air disinfection, a total inactivation of virus A/H5N2, close to avian influenza virus A/H5N2, was obtained in a single pass in the Buxair(D gas phase dynamic photoreactor using a contaminated air flow rate of 40 m(3)/h

    Contribution to the study of the beneficial fauna of olive orchards in Porto Martins, Terceira Island, Azores

    Proceedings of the 9th Meeting at Lisboa (Portugal) 26-29 October, 2021.The olive tree culture in Terceira Island occupies a strip in Porto Martins area whose fruit is very sought, mainly by tourists and restaurants. The olive tree is very affected by pests, such as the olive fly, which causes high losses and a drastic loss of production. Therefore, large quantities of pesticides are applied whose applications have a great impact, in particular, on beneficial organisms. In this work, the beneficial fauna present in olive groves was identified through the technique of beatings. The impact of the application of pesticides on these beneficial insects present in olive trees with different levels of treatments was also evaluated. For this, orchards with intensive applications, with less intensive applications and one organic orchard were selected to studied and monitoring. It was verified that in the olive groves with intensive and semi-intensive pesticide applications the abundance of beneficial insects was unbalanced, with a low abundance of predatory insects and still high population densities of the olive cottonweed pest. The results obtained point to the fact that chemical treatments do not destroy the target pests and worst contribute to a reduction in auxiliary organisms. In the organic orchard was observed the highest abundance of predatory insects and the lowest abundance of herbivorous insects. These results showed that this type of orchard is in a natural balance and that this is the way to follow in preserving the presence of beneficial insects in the olive parcels because these are very important to keep low pest populations and permit its better control. The conclusions of this study point to the fact that producers can further increase their yield by taking the biological option in their daily practices that will have a lower impact on the beneficial fauna present in the olive groves. At the same time will also permit to preserve the environment and the health of the applicators, producers and of the consumer by obtaining a product without any pesticide residues.To the producers involved in this study.     Endemic versus epidemic viral spreads display distinct patterns of intraindividual HTLV-2b replication.

    As the replication pattern of leukemogenic PTLVs possesses a strong pathogenic impact, we investigated HTLV-2 replication in vivo in asymptomatic carriers belonging into 2 distinct populations infected by the same HTLV-2b subtype. They include epidemically infected American blood donors, in whom HTLV-2b has been present for only 30 years, and endemically infected Bakola Pygmies from Cameroon, characterized by a long viral endemicity (at least few generations). In blood donors, both the circulating proviral loads and the degree of infected cell proliferation were largely lower than those characterizing asymptomatic carriers infected with leukemogenic PTLVs (HTLV-1, STLV-1). This might contribute to explain the lack of known link between HTLV-2b infection and the development of malignancies in this population. In contrast, endemically infected individuals displayed high proviral loads resulting from the extensive proliferation of infected cells. The route and/or the duration of infection, viral genetic drift, host immune response, genetic background, co-infections or a combination thereof might have contributed to these differences between endemically and epidemically infected subjects. As the clonality pattern observed in endemically infected individuals is very reminiscent of that of leukemogenic PTLVs at the pre-leukemic stage, our results highlight the possible oncogenic effect of HTLV-2b infection in such population
