    Mutational Analysis of the Active Site and Antibody Epitopes of the Complement-inhibitory Glycoprotein, CD59

    The Ly-6 superfamily of cell surface molecules includes CD59, a potent regulator of the complement system that protects host cells from the cytolytic action of the membrane attack complex (MAC). Although its mechanism of action is not well understood, CD59 is thought to prevent assembly of the MAC by binding to the C8 and/or C9 proteins of the nascent complex. Here a systematic, structure-based mutational approach has been used to determine the region(s) of CD59 required for its protective activity. Analysis of 16 CD59 mutants with single, highly nonconservative substitutions suggests that CD59 has a single active site that includes Trp-40, Arg-53, and Glu-56 of the glycosylated, membrane-distal face of the disk-like extracellular domain and, possibly, Asp-24 positioned at the edge of the domain. The putative active site includes residues conserved across species, consistent with the lack of strict homologous restriction previously observed in studies of CD59 function. Competition and mutational analyses of the epitopes of eight CD59-blocking and non-blocking monoclonal antibodies confirmed the location of the active site.     Sequential activation of NKT cells and NK cells provides effective innate immunotherapy of cancer

    The CD1d reactive glycolipid, α-galactosylceramide (α-GalCer), potently activates T cell receptor-α type I invariant NKT cells that secondarily stimulate the proliferation and activation of other leukocytes, including NK cells. Here we report a rational approach to improving the antitumor activity of α-GalCer by using delayed interleukin (IL)-21 treatment to mature the α-GalCer–expanded pool of NK cells into highly cytotoxic effector cells. In a series of experimental and spontaneous metastases models in mice, we demonstrate far superior antitumor activity of the α-GalCer/IL-21 combination above either agent alone. Superior antitumor activity was critically dependent upon the increased perforin-mediated cytolytic activity of NK cells. Transfer of α-GalCer–pulsed dendritic cells (DCs) followed by systemic IL-21 caused an even more significant reduction in established (day 8) metastatic burden and prolonged survival. In addition, this combination prevented chemical carcinogenesis more effectively. Combinations of IL-21 with other NK cell–activating cytokines, such as IL-2 and IL-12, were much less effective in the same experimental metastases models, and these cytokines did not substitute effectively for IL-21 in combination with α-GalCer. Overall, the data suggest that NK cell antitumor function can be enhanced greatly by strategies that are designed to expand and differentiate NK cells via DC activation of NKT cells

    We study the relationship between the field star formation and cluster formation properties in a large sample of nearby dwarf galaxies. We use optical data from the Hubble Space Telescope and from ground-based telescopes to derive the ages and masses of the young (t_age < 100Myr) cluster sample. Our data provides the first constraints on two proposed relationships between the star formation rate of galaxies and the properties of their cluster systems in the low star formation rate regime. The data show broad agreement with these relationships, but significant galaxy-to-galaxy scatter exists. In part, this scatter can be accounted for by simulating the small number of clusters detected from stochastically sampling the cluster mass function. However, this stochasticity does not fully account for the observed scatter in our data suggesting there may be true variations in the fraction of stars formed in clusters in dwarf galaxies.     Small Big Data: Using multiple data-sets to explore unfolding social and economic change

    Bold approaches to data collection and large-scale quantitative advances have long been a preoccupation for social science researchers. In this commentary we further debate over the use of large-scale survey data and official statistics with ‘Big Data’ methodologists, and emphasise the ability of these resources to incorporate the essential social and cultural heredity that is intrinsic to the human sciences. In doing so, we introduce a series of new data-sets that integrate approximately 30 years of survey data on victimisation, fear of crime and disorder and social attitudes with indicators of socio-economic conditions and policy outcomes in Britain. The data-sets that we outline below do not conform to typical conceptions of ‘Big Data’. But, we would contend, they are ‘big’ in terms of the volume, variety and complexity of data which has been collated (and to which additional data can be linked) and ‘big’ also in that they allow us to explore key questions pertaining to how social and economic policy change at the national level alters the attitudes and experiences of citizens. Importantly, they are also ‘small’ in the sense that the task of rendering the data usable, linking it and decoding it, required both manual processing and tacit knowledge of the context of the data and intentions of its creators

    The statistics of triggered star formation: an overdensity of massive YSOs around Spitzer bubbles

    We present a detailed statistical study of massive star formation in the environment of 322 Spitzer mid-infrared bubbles by using the RMS survey for massive Young Stellar Objects (YSOs). Using a combination of simple surface density plots and a more sophisticated angular cross-correlation function analysis we show that there is a statistically significant overdensity of RMS YSOs towards the bubbles. There is a clear peak in the surface density and angular cross-correlation function of YSOs projected against the rim of the bubbles. By investigating the autocorrelation function of the RMS YSOs we show that this is not due to intrinsic clustering of the RMS YSO sample. RMS YSOs and Spitzer bubbles are essentially uncorrelated with each other beyond a normalised angular distance of two bubble radii. The bubbles associated with RMS YSOs tend to be both smaller and thinner than those that are not associated with YSOs. We interpret this tendency to be due to an age effect, with YSOs being preferentially found around smaller and younger bubbles. We find no evidence to suggest that the YSOs associated with the bubbles are any more luminous than the rest of the RMS YSO population, which suggests that the triggering process does not produce a top heavy luminosity function or initial mass function. We suggest that it is likely that the YSOs were triggered by the expansion of the bubbles and estimate that the fraction of massive stars in the Milky Way formed by this process could be between 14 and 30%.Comment: 12 pages, 8 figures.     Decay-accelerating factor expression in the rat kidney is restricted to the apical surface of podocytes.BackgroundDecay-accelerating factor (DAF) has inhibitory activity toward complement C3 and C5 convertases. DAF is present in human glomeruli and on cultured human glomerular visceral epithelial cells (GEC). We studied the distribution and function of rat DAF.MethodsFunction-neutralizing antibodies (Abs) were raised against DAF. The distribution of DAF in vivo was determined by immunoelectron microscopy. Functional studies were performed in cultured GEC and following IV injection of anti-DAF Abs into rats.ResultsDAF was present exclusively on the apical surfaces of GEC, and was not present on the basal surfaces of GEC, nor other glomerular or kidney cells. DAF was functionally active on cultured GEC, and served to limit complement activation in concert with CD59, an inhibitor of C5b-9 formation. Upon injection into normal rats, anti-DAF F(ab′)2 Abs bound to GEC in vivo, yet there was no evidence for complement activation and animals did not develop abnormal albuminuria. Anti-megalin complement-activating IgG Abs were “planted” on GEC, which activated complement as evidenced by the presence of C3d on GEC. Attempts to inhibit DAF function with anti-DAF Abs did not affect the quantity of complement activation by these anti-megalin Abs, nor did it lead to development of abnormal albuminuria. In contrast, in the puromycin aminonucleoside model of GEC injury and proteinuria, anti-DAF Abs slowed the recovery from renal failure that occurs in this model.ConclusionIn cultured rat GEC, DAF is an effective complement regulator. In vivo, DAF is present on GEC apical surfaces. Yet, it appears that DAF is not essential to prevent complement activation from occurring under normal circumstances and in those cases in which complement-activating Abs are present on the basal surfaces of GEC in vivo. However, in proteinuric conditions, DAF appears to be protective to GEC

    Herschel-ATLAS: Multi-wavelength SEDs and physical properties of 250 micron-selected galaxies at z < 0.5

    We present a pan-chromatic analysis of an unprecedented sample of 1402 250 micron-selected galaxies at z < 0.5 (mean z = 0.24) from the Herschel-ATLAS survey. We complement our Herschel 100-500 micron data with UV-K-band photometry from the Galaxy And Mass Assembly (GAMA) survey and apply the MAGPHYS energy-balance technique to produce pan-chromatic SEDs for a representative sample of 250 micron selected galaxies spanning the most recent 5 Gyr of cosmic history. We derive estimates of physical parameters, including star formation rates, stellar masses, dust masses and infrared luminosities. The typical H-ATLAS galaxy at z < 0.5 has a far-infrared luminosity in the range 10^10 - 10^12 Lsolar (SFR: 1-50 Msolar/yr) thus is broadly representative of normal star forming galaxies over this redshift range. We show that 250 micron-selected galaxies contain a larger mass of dust at a given infra-red luminosity or star formation rate than previous samples selected at 60 micron from IRAS. We derive typical SEDs for H-ATLAS galaxies, and show that the emergent SED shape is most sensitive to specific star formation rate. The optical-UV SEDs also become more reddened due to dust at higher redshifts. Our template SEDs are significantly cooler than existing infra-red templates. They may therefore be most appropriate for inferring total IR luminosities from moderate redshift submillimetre selected samples and for inclusion in models of the lower redshift submillimetre galaxy populations.Comment: 26 pages, 24 figures, Accepted by MNRA
