Background: MicroRNA is a type of small RNA of about 22 nucleotide length. Most popular miRNAs are found in the liver and are extensively important in determining the biological and clinical functions. MicroRNA, in host cells, may impact the replication of viruses either positively or negatively. In this study the miR-122 expression was examined and compared in three groups, two sample groups of patients with chronic hepatitis and hepatitis B virus-associated cirrhosis, and a healthy control group.Materials and Methods: In this study, 108 samples of whole blood were taken from each participant. Then the miRNA expression evaluation was conducted through relative real time PCR.Results: The results indicated that miR-122 expression was elevated in patients with chronic hepatitis B and HBV related cirrhosis about 1.8 times (P<0.05) more than control group which is statistically significant. Conclusion: According to the results of this study, measuring the miR-122 expression levels may be used as a biomarker and an indicator of the disease progression of chronic hepatitis B to HBV related cirrhosis to HCC, but needs more investigations and more samples

Nanostructured ZnO thin film has been synthesized via sol-gel method. In this study, effect of stabilizer, as a vital part of sol with different molar ratios of stabilizer to Zn (stabilizer/Zn 0.25, 0.5, 1, 2), on structural, morphological and optoelectronic properties of ZnO thin film has been investigated. Triehtylamine (TeA) and triethanolamine (TEA), as two important tertiary amines for synthesize of ZnO, has been used. Spin coating technique performed to deposition of sol on glass substrate and after deposition process, the samples clacined at 500 C. X-ray diffraction method conducted in order to find structural properties of the films. The results showed the formation of hexagonal wurtzite ZnO as well as increasing the unit cell parameters by increasing TeA content. Field emission scanning electron microscopy (FESEM) used in order to see morphological changes for different molar ratios of stabilizer to Zn. The images demonstrate grain segregation in TeA samples by increasing TeA molar ratio. Also, in TEA samples, formation of micro holes in TEA/Zn 0.5 and smaller grain size for higher TEA ratios has been observed. UV-Vis spectroscopy was employed to obtain optoelectronic properties and the results have shown dependence of optical band gap to stabilizer’s type and content. Key points 1. Combining several parameters suggested in different papers in one experiment to get the active enzyme. 2. Decreasing the temperature of induction and the amount of inducer and using a chaperone-like tag simultaneously to help the protein folding system of the bacteria. 3. Using a statistical method (RSM) to analyze a number of treatments and prediction of the most effective ones to optimize the expression system

Gene expression profiling revealed overexpression of vesicle amine transport protein-1 (<i>VAT-1</i>) as a potential oncogene in gastric cancer

161-165Gastric cancer (GC) is one of the most common types of cancer worldwide. Owing to the distinct molecular pathology and the increasing progression rate of GC in Asia, suppression subtractive hybridization (SSH) was used as a high-throughput gene expression analysis method to find genes associated with GC pathogenesis. Total RNA was extracted from the clinical samples, and mRNA was isolated and used in SSH method. The subtracted library was subjected to cloning and the randomly selected clones were sequenced. qRT-PCR was used for expression analysis. The overexpression of vesicle amine transport protein-1 (VAT-1) gene was observed and its expression was analyzed by qRT-PCR in clinical tissue samples. According to the potential oncogenic activity of VAT-1 and its probable surface-occurrence in GC cells, it might be involved in GC pathogenesis and invasion, and it is suggested to be investigated in the diagnosis and therapeutics of GC
