    AFM Force Spectroscopy and Steered Molecular Dynamics Simulation of Protein Contactin 4

    We use a single molecule atomic force spectroscopy combined with the steered molecular dynamics simulation to determine a mechanical behavior of neural cell adhesion protein contactin during its unfolding. Force curves typical for modular proteins were observed, showing at most four unfolding peaks. The analysis of force spectra performed within worm-like chain model of polymer elasticity showed the presence of three unfolding lengths. Small plateaus, most likely resulting from forced transitions within domains were observed for the first time.     COVID-19 and stem cell transplantation; results from an EBMT and GETH multicenter prospective survey

    Altres ajuts: British Society for Blood and Marrow Transplantation and Cellular Therapy (BSBMTCT); UK NIHR Imperial College Biomedical Research Centre.This study reports on 382 COVID-19 patients having undergone allogeneic (n = 236) or autologous (n = 146) hematopoietic cell transplantation (HCT) reported to the European Society for Blood and Marrow Transplantation (EBMT) or to the Spanish Group of Hematopoietic Stem Cell Transplantation (GETH). The median age was 54.1 years (1.0-80.3) for allogeneic, and 60.6 years (7.7-81.6) for autologous HCT patients. The median time from HCT to COVID-19 was 15.8 months (0.2-292.7) in allogeneic and 24.6 months (−0.9 to 350.3) in autologous recipients. 83.5% developed lower respiratory tract disease and 22.5% were admitted to an ICU. Overall survival at 6 weeks from diagnosis was 77.9% and 72.1% in allogeneic and autologous recipients, respectively. Children had a survival of 93.4%. In multivariate analysis, older age (p = 0.02), need for ICU (p < 0.0001) and moderate/high immunodeficiency index (p = 0.04) increased the risk while better performance status (p = 0.001) decreased the risk for mortality. Other factors such as underlying diagnosis, time from HCT, GVHD, or ongoing immunosuppression did not significantly impact overall survival. We conclude that HCT patients are at high risk of developing LRTD, require admission to ICU, and have increased mortality in COVID-19

    Phospholipase iPLA2β averts ferroptosis by eliminating a redox lipid death signal

    Ferroptosis, triggered by discoordination of iron, thiols and lipids, leads to the accumulation of 15-hydroperoxy (Hp)-arachidonoyl-phosphatidylethanolamine (15-HpETE-PE), generated by complexes of 15-lipoxygenase (15-LOX) and a scaffold protein, phosphatidylethanolamine (PE)-binding protein (PEBP)1. As the Ca^{2+} -independent phospholipase A2β (iPLA2β, PLA2G6 or PNPLA9 gene) can preferentially hydrolyze peroxidized phospholipids, it may eliminate the ferroptotic 15-HpETE-PE death signal. Here, we demonstrate that by hydrolyzing 15-HpETE-PE, iPLA_{2}β averts ferroptosis, whereas its genetic or pharmacological inactivation sensitizes cells to ferroptosis. Given that PLA2G6 mutations relate to neurodegeneration, we examined fibroblasts from a patient with a Parkinson’s disease (PD)-associated mutation (fPD^{R747W}) and found selectively decreased 15-HpETE-PE-hydrolyzing activity, 15-HpETE-PE accumulation and elevated sensitivity to ferroptosis. CRISPR-Cas9-engineered Pnpla9^{R748W/R748W} mice exhibited progressive parkinsonian motor deficits and 15-HpETE-PE accumulation. Elevated 15-HpETE-PE levels were also detected in midbrains of rotenone-infused parkinsonian rats and α-synuclein-mutant Snca^{A53T} mice, with decreased iPLA2β expression and a PD-relevant phenotype. Thus, iPLA_{2}β is a new ferroptosis regulator, and its mutations may be implicated in PD pathogenesis

    The European Hematology Association Roadmap for European Hematology Research: a consensus document

    The European Hematology Association (EHA) Roadmap for European Hematology Research highlights major achievements in diagnosis and treatment of blood disorders and identifies the greatest unmet clinical and scientific needs in those areas to enable better funded, more focused European hematology research. Initiated by the EHA, around 300 experts contributed to the consensus document, which will help European policy makers, research funders, research organizations, researchers, and patient groups make better informed decisions on hematology research. It also aims to raise public awareness of the burden of blood disorders on European society, which purely in economic terms is estimated at €23 billion per year, a level of cost that is not matched in current European hematology research funding. In recent decades, hematology research has improved our fundamental understanding of the biology of blood disorders, and has improved diagnostics and treatments, sometimes in revolutionary ways. This progress highlights the potential of focused basic research programs such as this EHA Roadmap. The EHA Roadmap identifies nine ‘sections’ in hematology: normal hematopoiesis, malignant lymphoid and myeloid diseases, anemias and related diseases, platelet disorders, blood coagulation and hemostatic disorders, transfusion medicine, infections in hematology, and hematopoietic stem cell transplantation. These sections span 60 smaller groups of diseases or disorders. The EHA Roadmap identifies priorities and needs across the field of hematology, including those to develop targeted therapies based on genomic profiling and chemical biology, to eradicate minimal residual malignant disease, and to develop cellular immunotherapies, combination treatments, gene therapies, hematopoietic stem cell treatments, and treatments that are better tolerated by elderly patients

    Hematopoietic stem cell transplantation for autoimmune diseases in the time of COVID-19: EBMT guidelines and recommendations

    Coronavirus disease-19 (COVID-19), caused by Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2), represents one of the biggest challenges of 21st century, threatening public health around the globe. Increasing age and presence of co-morbidities are reported risk factors for severe disease and mortality, along with autoimmune diseases (ADs) and immunosuppressive treatments such as haematopoietic stem cell transplantation (HSCT), which are also associated with adverse outcomes. We review the impact of the pandemic on specific groups of patients with neurological, rheumatological, and gastroenterological indications, along with the challenges delivering HSCT in adult and pediatric populations. Moving forward, we developed consensus-based guidelines and recommendations for best practice and quality of patient care in order to support clinicians, scientists, and their multidisciplinary teams, as well as patients and their carers. These guidelines aim to support national and international organizations related to autoimmune diseases and local clinical teams delivering HSCT. Areas of unmet need and future research questions are also highlighted. The waves of the COVID-19 pandemic are predicted to be followed by an "endemic" phase and therefore an ongoing risk within a "new normality". These recommendations reflect currently available evidence, coupled with expert opinion, and will be revised according to necessary modifications in practice.Pathophysiology and treatment of rheumatic disease

    W chwili obecnej w Polsce są realizowane liczne inwestycje o charakterze liniowym i powierzchniowym, których lokalizacja dotyczy obszarów wiejskich. Charakterystyczna w naszym kraju wadliwa struktura przestrzenna gospodarstw rolnych na obszarach realizacji inwestycji jest dezorganizowana jeszcze bardziej. Studia uwarunkował i kierunków zagospodarowania gmin są wyznacznikiem trendów, które gminy chcą realizował na swoim terenie i wynikają z analiz możliwości rozwojowych oraz planowania na szczeblu krajowym, który dotyczy autostrad. W opracowaniu zostanie przedstawiony wpływ, jaki wywarła budowa fragmentu autostrady A-4 w województwie małopolskim na jeden z obrębów ewidencyjnych z gminy Głogów Małopolski. Przedstawiono kierunki zmian użytkowania ziemi tak, aby realizacja scaleń infrastrukturalnych pozwalała na naprawę zdezorganizowanego rozłogu gruntów w gospodarstwach rolnych oraz na realizacje idei wielofunkcyjnego rozwoju wsi. Wskazano na konieczność rozwiązał projektu scalenia gruntów w aspekcie szerszym niż tylko poprawa układu działek w gospodarstwach rolnych.Numerous investments of linear and spatial nature whose location relates to rural areas are being carried out at the present moment in Poland. The characteristic in our country defective spatial structure of agricultural farms in the areas of the realization of the investments being is disorganized much more. The studies of spatial development and the directions of the development of communes are the determinants of the trends which communes want to realize on their terrain and they result from the analyses of developmental possibilities and planning on the national level which relates to highways. It will be introduced in the study the influence, what the building of the fragment of the highway made A-4 in the of Little Poland province on one from list grounds from the commune of Gogów Malopolska. The directions of the changes of the use of the land were also introduced so that the realization of infrastructural land consolidation shoud allow to improve the disorganized deployment of parcels on agricultural farms the realization of the idea of the multifunction development of village. The necessity of the solutions to the project of infrastructural land consolidation was also touched upon in a wider aspect than only the improvement in the development of land parcels on agricultural farms
