Los resultados obtenidos animan a usar ABP en las titulaciones de Grado, pero confirman que su aplicación no es trivial cuando los proyectos son complejos y es alto el nivel de autoaprendizaje requerido al estudiante. Se discute también la importancia que tendría el desarrollo de las competencias transversales para el correcto funcionamiento del ABPUniversidad de Málaga.     miRanalyzer: a microRNA detection and analysis tool for next-generation sequencing experiments

    Next-generation sequencing allows now the sequencing of small RNA molecules and the estimation of their expression levels. Consequently, there will be a high demand of bioinformatics tools to cope with the several gigabytes of sequence data generated in each single deep-sequencing experiment. Given this scene, we developed miRanalyzer, a web server tool for the analysis of deep-sequencing experiments for small RNAs. The web server tool requires a simple input file containing a list of unique reads and its copy numbers (expression levels). Using these data, miRanalyzer (i) detects all known microRNA sequences annotated in miRBase, (ii) finds all perfect matches against other libraries of transcribed sequences and (iii) predicts new microRNAs. The prediction of new microRNAs is an especially important point as there are many species with very few known microRNAs. Therefore, we implemented a highly accurate machine learning algorithm for the prediction of new microRNAs that reaches AUC values of 97.9% and recall values of up to 75% on unseen data. The web tool summarizes all the described steps in a single output page, which provides a comprehensive overview of the analysis, adding links to more detailed output pages for each analysis module. miRanalyzer is available at http://web.bioinformatics.cicbiogune.es/microRNA/

    The statistical analysis plan for the unification of treatments and interventions for tinnitus patients randomized clinical trial (UNITI-RCT)

    Background Tinnitus is a leading cause of disease burden globally. Several therapeutic strategies are recommended in guidelines for the reduction of tinnitus distress; however, little is known about the potentially increased effectiveness of a combination of treatments and personalized treatments for each tinnitus patient. Methods Within the Unification of Treatments and Interventions for Tinnitus Patients project, a multicenter, randomized clinical trial is conducted with the aim to compare the effectiveness of single treatments and combined treatments on tinnitus distress (UNITI-RCT). Five different tinnitus centers across Europe aim to treat chronic tinnitus patients with either cognitive behavioral therapy, sound therapy, structured counseling, or hearing aids alone, or with a combination of two of these treatments, resulting in four treatment arms with single treatment and six treatment arms with combinational treatment. This statistical analysis plan describes the statistical methods to be deployed in the UNITI-RCT. Discussion The UNITI-RCT trial will provide important evidence about whether a combination of treatments is superior to a single treatment alone in the management of chronic tinnitus patients. This pre-specified statistical analysis plan details the methodology for the analysis of the UNITI trial results. Trial registration ClinicalTrials.gov NCT04663828. The trial is ongoing. Date of registration: December 11, 2020. All patients that finished their treatment before 19 December 2022 are included in the main RCT analysis

    Global urban environmental change drives adaptation in white clover

    Urbanization transforms environments in ways that alter biological evolution. We examined whether urban environmental change drives parallel evolution by sampling 110,019 white clover plants from 6169 populations in 160 cities globally. Plants were assayed for a Mendelian antiherbivore defense that also affects tolerance to abiotic stressors. Urban-rural gradients were associated with the evolution of clines in defense in 47% of cities throughout the world. Variation in the strength of clines was explained by environmental changes in drought stress and vegetation cover that varied among cities. Sequencing 2074 genomes from 26 cities revealed that the evolution of urban-rural clines was best explained by adaptive evolution, but the degree of parallel adaptation varied among cities. Our results demonstrate that urbanization leads to adaptation at a global scale

    El área protegida Laguna de Rocha es un importante sitio para la conservación de la biodiversidad del Uruguay y alberga una de las comunidades de pescadores artesanales más antiguas del pais.En el marco del Manejo Costero Integrado, este trabajo analiza las dinámicas y vínculos socio-institucionales en el area protegida, en su trayectoria de conformación y en torno a la pesca artesanal del lugar. Asimismo, se discute como estos procesos y las características del sistema de gobernanza, permiten evaluar las capacidades del sector para el desarrollo de instrumentos de gestión integrada, tales como la certificación de pesquerías.La conformación del área protegida muestra un proceso de innovación socionstitucional que permitió la consolidación de una red de actores robustecidos para su gestión. El sistema de gobernanza del área, los vínculos de alta intensidad y los aprendizajes de 20 años de experiencias en torno a la conservación del área, potencian las capacidades para promover un proceso de certificación pesquera adecuado al contexto local. Sin embargo, las mayores limitantes se encuentran en la falta de voluntad a nivel gubernamental para politicas de largo plazo, debilidad en los mecanismos de comunicación y el desconocimiento del potencial de este tipo de instrumentos.Laguna de Rocha is one of the most important biodiversity conservation sites of Uruguay, also is home to a traditional fisherfolk community. These natural and cultural values, led to her being categorized as Protected Landscape, on entry into the National System of Protected Areas (SNAP) in 2010. The aim of this paper is to analyze the dynamics and socio-institutional ties in the Laguna de Rocha Protected Area in shaping his career and currently around artisanal fishing place. It also discussed how these processes and characteristics of the governance system, to assess the capabilities of the sector for the development of integrated tools, such as certification of fisheries management. This research shows, such as the creation of protected area development in a process of innovation that allowed socionstitucional consolidation of a network of actors ruggedized for management. The area governance system, high intensity linkages between key actors of artisanal fisheries and learning 20 years of experience, enhance capabilities to promote fishery certification process appropriate to the local context. However, the main constraints are the lack of will to public policy short and long term, the weakness of the mechanisms of communication between some actors and ignorance of how to implement these instruments

    On biophysical properties and sensitivity to gap junction blockers of connexin 39 hemichannels expressed in hela cells

Although connexins (Cxs) are broadly expressed by cells of mammalian organisms, Cx39 has a very restricted pattern of expression and the biophysical properties of Cx39-based channels [hemichannels (HCs) and gap junction channels (GJCs)] remain largely unknown. Here, we used HeLa cells transfected with Cx39 (HeLa-Cx39 cells) in which intercellular electrical coupling was not detected, indicating the absence of GJCs. However, functional HCs were found on the surface of cells exposed to conditions known to increase the open probability of other Cx HCs (e.g., extracellular divalent cationic-free solution (DCFS), extracellular alkaline pH, mechanical stimulus and depolarization to positive membrane potentials).
