Late phonological development in Spanish children with bilateral hearing loss / Desarrollo fonologico tardio en ninos espanoles con perdidas auditivas bilaterales
This study has a twofold objective: to analyse and compare the phonological processes in a sample of Spanish children with hearing loss, both with a cochlear implant and with a hearing aid, with a group with normal hearing; and to determine whether there are differences between the participants with a cochlear implant and with a hearing aid in the frequency and nature of the phonological processes. The sample is made up of 168 participants, eight with hearing loss (four with an implant and four with a hearing aid) and 160 with normal hearing. Samples of spontaneous speech were collected and transcribed using the tools from the CHILDES project. For the analysis, the phonological processes paradigm was adopted, evaluating phonological development based on normative error rates. The participants with a hearing loss show slower phonological development in terms of phonological processes, along with atypical processes. Furthermore, the participants with cochlear implants committed more phonological errors than those that wear a hearing aid. The implications of the results are discussed, and it is recommended that auditory stimulation should be done early in children with hearing loss regardless of their technical aid
HIV-1 Vpu Protein Mediates the Transport of Potassium in Saccharomyces cerevisiae
Human immunodeficiency virus type 1 (HIV-1) Vpu is an integral membrane protein that belongs to the viroporin family. Viroporins interact with cell membranes, triggering membrane permeabilization and promoting release of viral particles. In vitro electrophysiological methods have revealed changes in membrane ion currents when Vpu is present; however, in vivo the molecular mechanism of Vpu at the plasma membrane is still uncertain. We used the yeast Saccharomyces cerevisiae as a genetic model system to analyze how Vpu ion channel impacts cellular homeostasis. Inducible expression of Vpu impaired cell growth, suggesting that this viral protein is toxic to yeast cultures. This toxicity decreased with extracellular acidic pH. Also, Vpu toxicity diminished as the extracellular K(+) concentration was increased. However, expression of the Vpu protein suppresses the growth defect of K(+) uptake-deficient yeast (Îtrk1,2). The phenotype rescue of these highly hyperpolarized cells was almost total when they were grown in medium supplemented with high concentrations of KCl (100 mM) at pH 7.0 but was significantly reduced when the extracellular K(+) concentration or pH was decreased. These results indicate that Vpu has the ability to modify K(+) transport in both yeast strains. Here, we show also that Vpu confers tolerance to the aminoglycoside antibiotic hygromycin B in Îtrk1,2 yeast. Our results suggest that Vpu interferes with cell growth of wild-type yeast but improves proliferation of the hyperpolarized trk1,2 mutant by inducing plasma membrane depolarization. Furthermore, evaluation of the ion channel activity of the Vpu protein in Îtrk1,2 yeast could aid in the development of a high-throughput screening assay for molecules that target the retroviral protein.This study was supported by Grants PI PI05/00013 and PI08/0912 from Fondo de InvestigaciĂłn Sanitaria. Choice of the initial antiretroviral treatment for HIV-positive individuals in the era of integrase inhibitors
BACKGROUND: We aimed to describe the most frequently prescribed initial antiretroviral therapy (ART) regimens in recent years in HIV-positive persons in the Cohort of the Spanish HIV/AIDS Research Network (CoRIS) and to investigate factors associated with the choice of each regimen. METHODS: We analyzed initial ART regimens prescribed in adults participating in CoRIS from 2014 to 2017. Only regimens prescribed in >5% of patients were considered. We used multivariable multinomial regression to estimate Relative Risk Ratios (RRRs) for the association between sociodemographic and clinical characteristics and the choice of the initial regimen. RESULTS: Among 2874 participants, abacavir(ABC)/lamivudine(3TC)/dolutegavir(DTG) was the most frequently prescribed regimen (32.1%), followed by tenofovir disoproxil fumarate (TDF)/emtricitabine (FTC)/elvitegravir(EVG)/cobicistat(COBI) (14.9%), TDF/FTC/rilpivirine (RPV) (14.0%), tenofovir alafenamide (TAF)/FTC/EVG/COBI (13.7%), TDF/FTC+DTG (10.0%), TDF/FTC+darunavir/ritonavir or darunavir/cobicistat (bDRV) (9.8%) and TDF/FTC+raltegravir (RAL) (5.6%). Compared with ABC/3TC/DTG, starting TDF/FTC/RPV was less likely in patients with CD4100.000 copies/mL. TDF/FTC+DTG was more frequent in those with CD4100.000 copies/mL. TDF/FTC+RAL and TDF/FTC+bDRV were also more frequent among patients with CD4<200 cells//muL and with transmission categories other than men who have sex with men. Compared with ABC/3TC/DTG, the prescription of other initial ART regimens decreased from 2014-2015 to 2016-2017 with the exception of TDF/FTC+DTG. Differences in the choice of the initial ART regimen were observed by hospitals' location. CONCLUSIONS: The choice of initial ART regimens is consistent with Spanish guidelines' recommendations, but is also clearly influenced by physician's perception based on patient's clinical and sociodemographic variables and by the prescribing hospital location
The 2010 very high energy gamma-ray flare & 10 years of multi-wavelength observations of M 87
Abridged: The giant radio galaxy M 87 with its proximity, famous jet, and
very massive black hole provides a unique opportunity to investigate the origin
of very high energy (VHE; E>100 GeV) gamma-ray emission generated in
relativistic outflows and the surroundings of super-massive black holes. M 87
has been established as a VHE gamma-ray emitter since 2006. The VHE gamma-ray
emission displays strong variability on timescales as short as a day. In this
paper, results from a joint VHE monitoring campaign on M 87 by the MAGIC and
VERITAS instruments in 2010 are reported. During the campaign, a flare at VHE
was detected triggering further observations at VHE (H.E.S.S.), X-rays
(Chandra), and radio (43 GHz VLBA). The excellent sampling of the VHE gamma-ray
light curve enables one to derive a precise temporal characterization of the
flare: the single, isolated flare is well described by a two-sided exponential
function with significantly different flux rise and decay times. While the
overall variability pattern of the 2010 flare appears somewhat different from
that of previous VHE flares in 2005 and 2008, they share very similar
timescales (~day), peak fluxes (Phi(>0.35 TeV) ~= (1-3) x 10^-11 ph cm^-2
s^-1), and VHE spectra. 43 GHz VLBA radio observations of the inner jet regions
indicate no enhanced flux in 2010 in contrast to observations in 2008, where an
increase of the radio flux of the innermost core regions coincided with a VHE
flare. On the other hand, Chandra X-ray observations taken ~3 days after the
peak of the VHE gamma-ray emission reveal an enhanced flux from the core. The
long-term (2001-2010) multi-wavelength light curve of M 87, spanning from radio
to VHE and including data from HST, LT, VLA and EVN, is used to further
investigate the origin of the VHE gamma-ray emission. No unique, common MWL
