    Searching for anthropogenic contaminants in human breast adipose tissues using gas chromatography-time-of-flight mass spectrometry

    The potential of GC-TOF MS for screening anthropogenic organic contaminants in human breast adipose tissues has been investigated. Initially a target screening was performed for a list of 125 compounds which included persistent halogen pollutants (OC pesticides, PCBs, PBDEs), PAHs, alkylphenols, and a notable number of pesticides from the different fungicide, herbicide and insecticide families. Searching for target pollutants was done by evaluating the presence of up to five representative ions for every analyte, all measured at accurate mass (20 mDa mass window). The experimental ion abundance ratios were then compared to those of reference standards for confirmation. Sample treatment consisted of an extraction with hexane and subsequent normal-phase HPLC or SPE clean-up. The fat-free LC fractions were then investigated by GC-TOF MS. Full-spectral acquisition and accurate mass data generated by GC-TOF MS also allowed the investigation of non-target compounds using appropriate processing software to manage MS data. Identification was initially based on library fit using commercial nominal mass libraries. This was followed by comparing the experimental accurate masses of the most relevant ions with the theoretical exact masses with calculations made using the elemental composition calculator included in the software. The application of both target and non-target approaches to around 40 real samples allowed the detection and confirmation of several target pollutants including p,p’- DDE, HCB, and some PCBs and PAHs. Several non-target compounds that could be considered anthropogenic pollutants were also detected. These included 3,5-di-tert-butyl-4-hydroxy-toluene (BHT) and its metabolite 3,5-di-tert-butyl-4-hydroxybenzaldehyde (BHT-CHO), dibenzylamine, n-butyl benzenesulfonamide, some naphthalene-related compounds and several PCBs isomers not 2included in the target list.     Advancing towards universal screening for organic pollutants in waters

    Environmental analytical chemists face the challenge of investigating thousands of potential organic pollutants that may be present in the aquatic environment. High resolution mass spectrometry (HRMS) hyphenated to chromatography offers the possibility of detecting a large number of contaminants without pre-selection of analytes due to its accurate-mass full-spectrum acquisition at good sensitivity. Interestingly, large screening can be made even without reference standards, as the valuable information provided by HRMS allows the tentative identification of the compound detected. In this work, hybrid quadrupole time-of-flight (QTOF) MS was combined with both liquid and gas chromatography (using a single instrument) for screening of around 2000 compounds in waters. This was feasible thanks to the use of atmospheric pressure chemical ionization source in GC. The screening was qualitatively validated for around 300 compounds at three levels (0.02, 0.1, 0.5 μg/L), and screening detection limits were established. Surface, ground water and effluent wastewater samples were analyzed, detecting and identifying a notable number of pesticides and transformation products, pharmaceuticals, personal care products, and illicit drugs, among others.     Comprehensive monitoring of organic micro-pollutants in surface and groundwater in the surrounding of a solid-waste treatment plant of Castellón, Spain

    The solid-waste treatment plant of RECIPLASA is located in the municipality of Onda (Castellón province), which is an important agricultural area of Spain, with predominance of citrus crops. In this plant, all urban solid wastes from the town of Castellón (around 200.000 inhabitants) and other smaller towns as Almassora, Benicàssim, Betxí, Borriana, L’Alcora, Onda and Vila-Real are treated. In order to evaluate the potential impact of this plant on the surrounding water, both surface and groundwater, a comprehensive monitoring of organic pollutants has been carried out along 2011, 2012 and 2013. To this aim, an advanced analytical strategy was applied for wide-scope screening, consisting on the complementary use of liquid chromatography (LC) and gas chromatography (GC) coupled to mass spectrometry (MS) with quadrupole (Q)-time of flight analyser (TOF). A generic solid-phase extraction with Oasis HLB cartridges was applied prior to the chromatographic analysis. The screening included more than 1,500 organic pollutants as target compounds, such as pesticides, pharmaceuticals, veterinary drugs, drugs of abuse, UV-filters, polycyclic aromatic hydrocarbons (PAHs), polychlorinated biphenyls (PCBs), polybrominated diphenyl ethers (PBDEs), among others. Pesticides, mainly herbicides, were the compounds more frequently detected. Other compounds as antioxidants, cosmetics, drugs of abuse, PAHs, pharmaceuticals and UV filters, were also identified in the screening though at much lower frequency. Once the screening was made, quantitative analysis focused on the compounds more frequently detected was subsequently applied using LC coupled to tandem MS with triple quadrupole analyzer. In this way, up to 24 pesticides and transformation products (TPs), 7 pharmaceuticals, one drug of abuse and its metabolite could be quantified at sub-ppb concentrations. Along the three years of study, ten compounds were found at 3 concentrations higher than 0.1 g/L.     Analytical strategy based on the combination of gas chromatography coupled to time-of-flight and hybrid quadrupole time-of-flight mass analyzers for non-target analysis in food packaging

    The potential of an advanced analytical strategy based on the use of gas chromatography (GC) coupled to high resolution mass spectrometry (HRMS) with two different analyzers and ionization sources has been investigated and applied to the non-target analysis of food packaging contaminants. Initially, the approach based on GC–time-of-flight (TOF) MS with electron ionization (EI) source allowed performing a library search and mass accurate measurements of selected ions. Then, a second analysis was performed using hybrid quadrupole (Q) TOF MS with an atmospheric pressure chemical ionization (APCI) source in order to search for the molecular ion or the protonated molecule and study the fragmentation behavior. This analytical strategy was applied to the analysis of four polypropylene/ethylene vinyl alcohol/polypropylene (PP/EVOH/PP) multilayer trays and one PP/Al foil/PP film, each one subjected to migration assays with the food simulants isooctane and Tenax®, in order to investigate its potential on the determination of migrant substances.Generalitat Valenciana (research group of excellence): PROMETEO/2009/054; ISIC 2012/01

    El propósito de este trabajo es abordar, analizar y reflexionar acerca de los diversos escenarios de violencia, desde los relatos, y con diferentes actores, formulando preguntas y planteando estrategias que permitan un acercamiento psicosocial con la comunidad. Dentro de este marco y como lo afirma Franco, A (2016) encontramos “la ley 1448 de 2011se ocupa de ofrecer una serie de mecanismos que buscan superar el estado de vulneración al que fueron sometidas las víctimas y garantizar su rehabilitación integral” (pag.3), teniendo en cuenta que el conflicto armado impacta tanto a la población urbana como a la rural y por lo tanto afecta aspectos sociales, económicos, políticos del individuo, la familia y la sociedad. De esta manera, se debe hacer un reconocimiento que, “es importante reconocer que en conflicto armado ha dejado grandes secuelas en el contexto colombiano y en las comunidades, tanto de manera directa como de manera indirecta; teniendo en cuenta de que el conflicto armado también recae sobre las comunidades rurales y urbanas que están alejados de las realidades y los conflictos qué se desarrollan en primera persona” (Rodríguez, J.; De la Torre, A.; Miranda, C. pg. 9. 2002). Por medio de lo anterior, se establece el presente informe, considerando cada uno de los elementos teóricos y conceptuales de las unidades para dar respuesta a los interrogantes de la guía y poder profundizar en cada aspecto relacionado con los procesos de resiliencia, así como el proceso de afrontamiento y empoderamiento.The purpose of this work is to address, analyze and reflect on the various scenarios of violence, from the stories, and with different actors, asking questions and proposing strategies that allow a psychosocial approach with the community. Within this framework and as stated by Franco, A (2016) we find "law 1448 of 2011 deals with offering a series of mechanisms that seek to overcome the state of vulnerability to which the victims were subjected and guarantee their comprehensive rehabilitation" (p. 3), taking into account that the armed conflict impacts both the urban and rural population and therefore affects social, economic, and political aspects of the individual, the family, and society. In this way, it must be recognized that “it is important to recognize that the armed conflict has left great consequences in the Colombian context and in the communities, both directly and indirectly; taking into account that the armed conflict also falls on rural and urban communities that are far from the realities and conflicts that take place in the first person" (Rodríguez, J.; De la Torre, A.; Miranda, C. pg 9. 2002). Through the foregoing, this report is established, considering each of the theoretical and conceptual elements of the units to answer the questions of the guide and to be able to delve into each aspect related to the resilience processes, as well as the process coping and empowerment

    Megalencephalic leukoencephalopathy with subcortical cysts: a personal biochemical retrospective

    Megalencephalic leukoencephalopathy with subcortical cysts (MLC) is a rare type of leukodystrophy characterized by dysfunction of the role of glial cells in controlling brain fluid and ion homeostasis. Patients affected by MLC present macrocephaly, cysts and white matter vacuolation, which lead to motor and cognitive impairments. To date, there is no treatment for MLC, only supportive care. MLC is caused by mutations in the MLC1 and GLIALCAM genes. MLC1 is a membrane protein with low identity to the Kv1.1 potassium channel and GlialCAM belongs to an adhesion molecule family. Both proteins form a complex with an as-yet-unknown function that is expressed mainly in the astrocytes surrounding the blood-brain barrier and in Bergmann glia. GlialCAM also acts as an auxiliary subunit of the chloride channel ClC-2, thus regulating its localization at cell-cell junctions and modifying its functional properties by affecting the common gate of ClC-2. Recent studies in Mlc1-,GlialCAM-and Clcn2-knockout mice or Mlc1- knockout zebrafish have provided fresh insight into the pathophysiology of MLC and further details about the molecular interactions between these three proteins. Additional studies have shown that GlialCAM/MLC1 also regulates other ion channels (TRPV4, VRAC) or transporters (Na+/K+-ATPase) in a not-understood manner. Furthermore, it has been shown that GlialCAM/ MLC1 may influence signal transduction mechanisms, thereby affecting other proteins not related with transport such as the EGFreceptor. Here, we offer a personal biochemical retrospective of the work that has been performed to gain knowledge of the pathophysiology of MLC, and we discuss future strategies that may be used to identify therapeutic solutions for MLC patients

    Mitochondrial physiology

    As the knowledge base and importance of mitochondrial physiology to evolution, health and disease expands, the necessity for harmonizing the terminology concerning mitochondrial respiratory states and rates has become increasingly apparent. The chemiosmotic theory establishes the mechanism of energy transformation and coupling in oxidative phosphorylation. The unifying concept of the protonmotive force provides the framework for developing a consistent theoretical foundation of mitochondrial physiology and bioenergetics. We follow the latest SI guidelines and those of the International Union of Pure and Applied Chemistry (IUPAC) on terminology in physical chemistry, extended by considerations of open systems and thermodynamics of irreversible processes. The concept-driven constructive terminology incorporates the meaning of each quantity and aligns concepts and symbols with the nomenclature of classical bioenergetics. We endeavour to provide a balanced view of mitochondrial respiratory control and a critical discussion on reporting data of mitochondrial respiration in terms of metabolic flows and fluxes. Uniform standards for evaluation of respiratory states and rates will ultimately contribute to reproducibility between laboratories and thus support the development of data repositories of mitochondrial respiratory function in species, tissues, and cells. Clarity of concept and consistency of nomenclature facilitate effective transdisciplinary communication, education, and ultimately further discovery

    Cabbage and fermented vegetables : From death rate heterogeneity in countries to candidates for mitigation strategies of severe COVID-19

    Large differences in COVID-19 death rates exist between countries and between regions of the same country. Some very low death rate countries such as Eastern Asia, Central Europe, or the Balkans have a common feature of eating large quantities of fermented foods. Although biases exist when examining ecological studies, fermented vegetables or cabbage have been associated with low death rates in European countries. SARS-CoV-2 binds to its receptor, the angiotensin-converting enzyme 2 (ACE2). As a result of SARS-CoV-2 binding, ACE2 downregulation enhances the angiotensin II receptor type 1 (AT(1)R) axis associated with oxidative stress. This leads to insulin resistance as well as lung and endothelial damage, two severe outcomes of COVID-19. The nuclear factor (erythroid-derived 2)-like 2 (Nrf2) is the most potent antioxidant in humans and can block in particular the AT(1)R axis. Cabbage contains precursors of sulforaphane, the most active natural activator of Nrf2. Fermented vegetables contain many lactobacilli, which are also potent Nrf2 activators. Three examples are: kimchi in Korea, westernized foods, and the slum paradox. It is proposed that fermented cabbage is a proof-of-concept of dietary manipulations that may enhance Nrf2-associated antioxidant effects, helpful in mitigating COVID-19 severity.Peer reviewe
