    Selected cultivars of cornelian cherry (Cornus mas L.) as a new food source for human nutrition

    The aim of this work was to determine antioxidant activity in 12 cultivars of cornelian cherry (Cornusmas L.). Two assays based on ion reduction of ABTS (2,2´-azinobis(3-ethylbenzothiazoline-6-sulphonate) and DPPH (2,2-diphenyl-1-picrylhydrazyl) radicals were used for antioxidant activity. Totalphenolic content of the fruit was analysed by Folin-Ciocelteau colorimetric method and ascorbic acidcontent was analysed using column chromatography - electrochemical detector (Coulochem III). Thehighest amounts of total phenolic content were found in cultivars ´Vydubeckij´ and ´Titus´ as 7.96 and8.11 g gallic acid kg-1 of fresh mass (FM). High correlation between polyphenols and antioxidant activityin fruits of the cultivars was observed (r2 = 0.970 for DPPH test and r2 = 0.978 for ABTS test). Thehighest total content of ascorbic acid was determined in cultivar ´Olomoucky´, with the value of 3.11 gkg-1 FM. This study attempts to contribute to the knowledge of human nutritional properties of thesecornelian cherry cultivars and may be useful for the evaluation of dietary information and furtherpropagation of cultivation and utilization of this fruit in the world

    Highlights from the 11-Year Record of Tropospheric Ozone from OMI/MLS and Continuation of that Long Record Using OMPS Measurements

    Since October 2004 the Ozone Monitoring Instrument (OMI) and Microwave Limb Sounder (MLS) onboard the Aura satellite have provided over 11 years of continuous tropospheric ozone measurements. These OMI/MLS measurements have been used in many studies to evaluate dynamical and photochemical effects caused by ENSO, the Madden-Julian Oscillation (MJO) and shorter timescales, as well as long-term trends and the effects of deep convection on tropospheric ozone. Given that the OMI and MLS instruments have now extended well beyond their expected lifetimes, our goal is to continue their long record of tropospheric ozone using recent Ozone Mapping Profiler Suite (OMPS) measurements. The OMPS onboard the Suomi National Polar-orbiting Partnership NPP satellite was launched on October 28, 2011 and is comprised of three instruments: the nadir mapper, the nadir profiler, and the limb profiler. Our study combines total column ozone from the OMPS nadir mapper with stratospheric column ozone from the OMPS limb profiler to measure tropospheric ozone residual. The time period for the OMPS measurements is March 2012 present. For the OMPS limb profiler retrievals, the OMPS v2 algorithm from Goddard is tested against the University of Saskatchewan (USask) Algorithm. The retrieved ozone profiles from each of these algorithms are evaluated with ozone profiles from both ozonesondes and the Aura Microwave Limb Sounder (MLS). Effects on derived OMPS tropospheric ozone caused by the 2015-2016 El Nino event are highlighted. This recent El Nino produced anomalies in tropospheric ozone throughout the tropical Pacific involving increases of approximately 10 DU over Indonesia and decreases approximately 5-10 DU in the eastern Pacific. These changes in ozone due to El Nino were predominantly dynamically-induced, caused by the eastward shift in sea-surface temperature and convection from the western to the eastern Pacific

    Merged SAGE II, Ozone_cci and OMPS ozone profile dataset and evaluation of ozone trends in the stratosphere

    In this paper, we present a merged dataset of ozone profiles from several satellite instruments: SAGE II on ERBS, GOMOS, SCIAMACHY and MIPAS on Envisat, OSIRIS on Odin, ACE-FTS on SCISAT, and OMPS on Suomi-NPP. The merged dataset is created in the framework of the European Space Agency Climate Change Initiative (Ozone_cci) with the aim of analyzing stratospheric ozone trends. For the merged dataset, we used the latest versions of the original ozone datasets. The datasets from the individual instruments have been extensively validated and intercompared; only those datasets which are in good agreement, and do not exhibit significant drifts with respect to collocated ground-based observations and with respect to each other, are used for merging. The long-term SAGE–CCI–OMPS dataset is created by computation and merging of deseasonalized anomalies from individual instruments. The merged SAGE–CCI–OMPS dataset consists of deseasonalized anomalies of ozone in 10° latitude bands from 90° S to 90° N and from 10 to 50 km in steps of 1 km covering the period from October 1984 to July 2016. This newly created dataset is used for evaluating ozone trends in the stratosphere through multiple linear regression. Negative ozone trends in the upper stratosphere are observed before 1997 and positive trends are found after 1997. The upper stratospheric trends are statistically significant at midlatitudes and indicate ozone recovery, as expected from the decrease of stratospheric halogens that started in the middle of the 1990s and stratospheric cooling

    Neuronal methylome reveals CREB-associated neuro-axonal impairment in multiple sclerosis

    BACKGROUND: Due to limited access to brain tissue, the precise mechanisms underlying neuro-axonal dysfunction in neurological disorders such as multiple sclerosis (MS) are largely unknown. In that context, profiling DNA methylation, which is a stable and cell type-specific regulatory epigenetic mark of genome activity, offers a unique opportunity to characterize the molecular mechanisms underpinning brain pathology in situ. We examined DNA methylation patterns of neuronal nuclei isolated from post-mortem brain tissue to infer processes that occur in neurons of MS patients. RESULTS: We isolated subcortical neuronal nuclei from post-mortem white matter tissue of MS patients and non-neurological controls using flow cytometry. We examined bulk DNA methylation changes (total n = 29) and further disentangled true DNA methylation (5mC) from neuron-specific DNA hydroxymethylation (5hmC) (n = 17), using Illumina Infinium 450K arrays. We performed neuronal sub-type deconvolution using glutamate and GABA methylation profiles to further reduce neuronal sample heterogeneity. In total, we identified 2811 and 1534 significant (genome-wide adjusted P value < 0.05) differentially methylated and hydroxymethylated positions between MS patients and controls. We found striking hypo-5mC and hyper-5hmC changes occurring mainly within gene bodies, which correlated with reduced transcriptional activity, assessed using published RNAseq data from bulk brain tissue of MS patients and controls. Pathway analyses of the two cohorts implicated dysregulation of genes involved in axonal guidance and synaptic plasticity, with meta-analysis confirming CREB signalling as the most highly enriched pathway underlying these processes. We functionally investigated DNA methylation changes of CREB signalling-related genes by immunohistofluoresence of phosphorylated CREB in neurons from brain sections of a subcohort of MS patients and controls (n = 15). Notably, DNA methylation changes associated with a reduction of CREB activity in white matter neurons of MS patients compared to controls. CONCLUSIONS: Our data demonstrate that investigating 5mC and 5hmC modifications separately allows the discovery of a substantial fraction of changes occurring in neurons, which can escape traditional bisulfite-based DNA methylation analysis. Collectively, our findings indicate that neurons of MS patients acquire sustained hypo-5mC and hyper-5hmC, which may impair CREB-mediated neuro-axonal integrity, in turn relating to clinical symptoms

    Childhood solid tumours in relation to population mixing around the time of birth

    In a retrospective cohort study of 673 787 live births in the Northern Region of England, 1975 - 1994, we investigated whether a higher level of population mixing around birth was a risk factor for solid tumours, by diagnostic group (Hodgkin's disease, brain and spinal tumours, neuroblastoma, other solid tumours), diagnosed during 1975-2001 under age 15 years. Logistic regression was used to relate risk to population mixing, based on (i) all movers and (ii) incomers from outside the region. Both ward and county district level analyses were performed. There was a decreased risk of brain and spinal tumours with increasing population mixing based on incomers from outside the region (OR for trend across three categories = 0.79, 95% CI: 0.66-0.95, P = 0.01 in the ward level analysis). Although this may be because of chance, it is consistent with a role of exposure to infection and immunological response in the aetiology of these tumours. For other tumour groups, there was no consistent evidence of an association between risk and population mixing

    Trends in Global Tropospheric Ozone Inferred from a Composite Record of TOMS/OMI/MLS/OMPS Satellite Measurements and the MERRA-2 GMI Simulation

    Past studies have suggested that ozone in the troposphere has increased globally throughout much of the 20th century due to increases in anthropogenic emissions and transport. We show, by combining satellite measurements with a chemical transport model, that during the last four decades tropospheric ozone does indeed indicate increases that are global in nature, yet still highly regional. Satellite ozone measurements from Nimbus-7 and Earth Probe Total Ozone Mapping Spectrometer (TOMS) are merged with ozone measurements from the Aura Ozone Monitoring Instrument/Microwave Limb Sounder (OMI/MLS) to determine trends in tropospheric ozone for 19792016. Both TOMS (19792005) and OMI/MLS (20052016) depict large increases in tropospheric ozone from the Near East to India and East Asia and further eastward over the Pacific Ocean. The 38-year merged satellite record shows total net change over this region of about +6 to +7 Dobson units (DU) (i.e., 15 %20 % of average background ozone), with the largest increase (4 DU) occurring during the 20052016 Aura period. The Global Modeling Initiative (GMI) chemical transport model with time-varying emissions is used to aid in the interpretation of tropospheric ozone trends for 19802016. The GMI simulation for the combined record also depicts the greatest increases of +6 to +7 DU over India and East Asia, very similar to the satellite measurements. In regions of significant increases in tropospheric column ozone (TCO) the trends are a factor of 22.5 larger for the Aura record when compared to the earlier TOMS record; for India and East Asia the trends in TCO for both GMI and satellite measurements are +3 DU decade(exp 1) or greater during 20052016 compared to about +1.2 to +1.4 DU decade(exp 1) for 19792005. The GMI simulation and satellite data also reveal a tropospheric ozone increases in +4 to +5 DU for the 38-year record over central Africa and the tropical Atlantic Ocean. Both the GMI simulation and satellite-measured tropospheric ozone during the latter Aura time period show increases of +3 DU decade1 over the N Atlantic and NE Pacific

    Salvage high-dose chemotherapy for children with extragonadal germ-cell tumours

    We reviewed the European Group for Blood and Marrow Transplantation (EBMT) experience with salvage high-dose chemotherapy (HDC) in paediatric patients with extragonadal germ-cell tumour (GCT). A total of 23 children with extragonadal GCT, median age 12 years (range 1–20), were treated with salvage HDC with haematopoietic progenitor cell support. The GCT primary location was intracranial site in nine cases, sacrococcyx in eight, retroperitoneum in four, and mediastinum in two. In all, 22 patients had a nongerminomatous GCT and one germinoma. Nine patients received HDC in first- and 14 in second- or third-relapse situation. No toxic deaths occurred. Overall, 16 of 23 patients (70%) achieved a complete remission. With a median follow-up of 66 months (range 31–173 months), 10 (43%) are continuously disease-free. Of six patients who had a disease recurrence after HDC, one achieved a disease-free status with surgical resection followed by chemotherapy and radiotherapy. In total, 11 patients (48%) are currently disease-free. Eight of 14 patients (57%) with extracranial primary and three of nine patients (33%) with intracranial primary GCT are currently disease-free. HDC induced impressive long-term remissions as salvage treatment in children with extragonadal extracranial GCTs. Salvage HDC should be investigated in prospective trials in these patients

The average annual surface air temperature (SAT) anomaly for 2016 for land stations north of starting in 1900, and is a significant increase over the previous highest value of +1.2°C, which was observed in 2007, 2011, and 2015. Average global annual temperatures also showed record values in 2015 and 2016. Currently, the Arctic is warming at more than twice the rate of lower latitudes

    Increasing incidence of childhood tumours of the central nervous system in Denmark, 1980–1996

    The registered incidence rate of childhood central nervous system (CNS) tumours has increased in several countries. It is uncertain whether these increases are biologically real or owing to improved diagnostic methods. We explored the medical records of 626 CNS tumours diagnosed in Danish children between 1980 and 1996. Population-based registers were used to extract data on mortality and background population. Temporal patterns were analysed by regression techniques. Most tumours were verified by computed tomography (78%) or magnetic resonance imaging (14%). Overall, the incidence rate increased by 2.9% per year (95% confidence interval (CI): 1.3;4.5) and the mortality rate increased by 1.4% per year (95% CI: −0.4;3.3). Among children aged 0–4 years, the survival rate after diagnosis remained almost unchanged, whereas among children aged 5–14 years, the 10-year survival rate improved from 59 to 74%. These data suggest that the incidence rate of CNS tumours among Danish children has truly increased, although alternative explanations cannot be excluded
