    Benchmarking of techniques used to assess the freeze damage in potatoes

    International audienceIn this study, benchmarking of methods used for assessing freeze damage in potatoes was carried out. Initially, the samples were frozen by subjecting them to three different temperatures (i.e. at -18 °C, -30 °C, and at -74 °C). Then, different analytical techniques comprising of focused methods (i.e. cryo-Scanning elctron microscopy-cryo-SEM, confocal laser scanning microscopy-CLSM) and global methods (i.e. texture analysis, low field nuclear magnetic resonance (NMR), exudate loss and colour change) were used to assess the impact of the freezing treatment from the different point of view addressed by each method. As a result, each of these methods were able to distinguish significantly fresh samples from the frozen-thawed samples. Focused methods like cryo-SEM and CLSM methods could differentiate the impact of all three different protocols. Meanwhile, texture analysis (including conventional method and novel method based on a touchless laser puff firmness tester), NMR and exudate loss could only determine the quality difference between -18 °C and -74 °C freezing conditions. Colour analysis was found as an inappropriate parameter for comparing the three freezing protocols. Among all analytical techniques, cryo-SEM provides the most authentic information about the product as the analysis is performed in frozen state, while for other techniques the product is thawed prior to analysis

    Influence of intraspecific variability and abiotic factors on mycotoxin production in Penicillium roqueforti

    International audiencePenicillium roqueforti has the ability to produce secondary metabolites, including roquefortine C (ROQC) and mycophenolic acid (MPA). In a previous study, the presence of these mycotoxins, alone or in co-occurrence, has been reported in blue-veined cheese. A high variability of mycotoxin content has also been observed, although the majority of samples exhibited relatively low concentrations. The observed variability raises the question of the factors impacting ROQC and MPA production. In this context, the mycotoxigenic potential of 96 P. roqueforti strains (biotic factor) and the effect of some abiotic factors (pH, temperature, NaCl and O2 contents, and C/N ratio) on mycotoxin production were evaluated. A high intraspecific diversity, established via genotypic (RAPD) and phenotypic (FTIR) approaches, was observed. It was associated with mycotoxigenic potential variability and may thus explain part of the observed variability in mycotoxin content of blue-veined cheese. Moreover, a significant decrease of ROQC and MPA production was observed for conditions (temperature, C/N ratio, O2 and NaCl concentrations) encountered during cheese-making compared with optimal growth conditions. The results also highlighted that there was no significant effect of addition of ROQC amino-acid precursor on the production of both mycotoxins whereas a pH increase from 4.5 to 6.5 slightly reduced MPA but not ROQC production

    Mayotte seismic crisis: building knowledge in near real-time by combining land and ocean-bottom seismometers, first results

    International audienceSummary The brutal onset of seismicity offshore Mayotte island North of the Mozambique Channel, Indian Ocean, that occurred in May 2018 caught the population, authorities, and scientific community off guard. Around 20 potentially felt earthquakes were recorded in the first 5 days, up to magnitude Mw 5.9. The scientific community had little pre-existing knowledge of the seismic activity in the region due to poor seismic network coverage. During 2018 and 2019, the MAYOBS/REVOSIMA seismology group was progressively built between four French research institutions to improve instrumentation and data sets to monitor what we know now as an on-going exceptional sub-marine basaltic eruption. After the addition of 3 medium-band stations on Mayotte island and 1 on Grande Glorieuse island in early 2019, the data recovered from the Ocean Bottom Seismometers were regularly processed by the group to improve the location of the earthquakes detected daily by the land network. We first built a new local 1D velocity model and established specific data processing procedures. The local 1.66 low VP/VS ratio we estimated is compatible with a volcanic island context. We manually picked about 125,000 P and S phases on land and sea bottom stations to locate more than 5,000 events between February 2019 and May 2020. The earthquakes outline two separate seismic clusters offshore that we named Proximal and Distal. The Proximal cluster, located 10km offshore Mayotte eastern coastlines, is 20 to 50 km deep and has a cylindrical shape. The Distal cluster start 5 km to the east of the Proximal cluster and extends below Mayotte's new volcanic edifice, from 50 km up to 25 km depth. The two clusters appear seismically separated, however our dataset is insufficient to firmly demonstrate this
