    Associations of cannabis use disorder with cognition, brain structure, and brain function in African Americans

    Although previous studies have highlighted associations of cannabis use with cognition and brain morphometry, critical questions remain with regard to the association between cannabis use and brain structural and functional connectivity. In a cross-sectional community sample of 205 African Americans (age 18–70) we tested for associations of cannabis use disorder (CUD, n = 57) with multi-domain cognitive measures and structural, diffusion, and resting state brain-imaging phenotypes. Post hoc model evidence was computed with Bayes factors (BF) and posterior probabilities of association (PPA) to account for multiple testing. General cognitive functioning, verbal intelligence, verbal memory, working memory, and motor speed were lower in the CUD group compared with nonusers (p \u3c .011; 1.9 \u3c BF \u3c 3,217). CUD was associated with altered functional connectivity in a network comprising the motor-hand region in the superior parietal gyri and the anterior insula (p \u3c .04). These differences were not explained by alcohol, other drug use, or education. No associations with CUD were observed in cortical thickness, cortical surface area, subcortical or cerebellar volumes (0.12 \u3c BF \u3c 1.5), or graph-theoretical metrics of resting state connectivity (PPA \u3c 0.01). In a large sample collected irrespective of cannabis used to minimize recruitment bias, we confirm the literature on poorer cognitive functioning in CUD, and an absence of volumetric brain differences between CUD and non-CUD. We did not find evidence for or against a disruption of structural connectivity, whereas we did find localized resting state functional dysconnectivity in CUD. There was sufficient proof, however, that organization of functional connectivity as determined via graph metrics does not differ between CUD and non-user group

    Membrane curvature in cell biology: An integration of molecular mechanisms.

    Curving biological membranes establishes the complex architecture of the cell and mediates membrane traffic to control flux through subcellular compartments. Common molecular mechanisms for bending membranes are evident in different cell biological contexts across eukaryotic phyla. These mechanisms can be intrinsic to the membrane bilayer (either the lipid or protein components) or can be brought about by extrinsic factors, including the cytoskeleton. Here, we review examples of membrane curvature generation in animals, fungi, and plants. We showcase the molecular mechanisms involved and how they collaborate and go on to highlight contexts of curvature that are exciting areas of future research. Lessons from how membranes are bent in yeast and mammals give hints as to the molecular mechanisms we expect to see used by plants and protists

    Investigations into a putative role for the novel BRASSIKIN pseudokinases in compatible pollen-stigma interactions in Arabidopsis thaliana.

    BACKGROUND: In the Brassicaceae, the early stages of compatible pollen-stigma interactions are tightly controlled with early checkpoints regulating pollen adhesion, hydration and germination, and pollen tube entry into the stigmatic surface. However, the early signalling events in the stigma which trigger these compatible interactions remain unknown. RESULTS: A set of stigma-expressed pseudokinase genes, termed BRASSIKINs (BKNs), were identified and found to be present in only core Brassicaceae genomes. In Arabidopsis thaliana Col-0, BKN1 displayed stigma-specific expression while the BKN2 gene was expressed in other tissues as well. CRISPR deletion mutations were generated for the two tandemly linked BKNs, and very mild hydration defects were observed for wild-type Col-0 pollen when placed on the bkn1/2 mutant stigmas. In further analyses, the predominant transcript for the stigma-specific BKN1 was found to have a premature stop codon in the Col-0 ecotype, but a survey of the 1001 Arabidopsis genomes uncovered three ecotypes that encoded a full-length BKN1 protein. Furthermore, phylogenetic analyses identified intact BKN1 orthologues in the closely related outcrossing Arabidopsis species, A. lyrata and A. halleri. Finally, the BKN pseudokinases were found to be plasma-membrane localized through the dual lipid modification of myristoylation and palmitoylation, and this localization would be consistent with a role in signaling complexes. CONCLUSION: In this study, we have characterized the novel Brassicaceae-specific family of BKN pseudokinase genes, and examined the function of BKN1 and BKN2 in the context of pollen-stigma interactions in A. thaliana Col-0. Additionally, premature stop codons were identified in the predicted stigma specific BKN1 gene in a number of the 1001 A. thaliana ecotype genomes, and this was in contrast to the out-crossing Arabidopsis species which carried intact copies of BKN1. Thus, understanding the function of BKN1 in other Brassicaceae species will be a key direction for future studies

    The SUN Protein Mps3 Is Required for Spindle Pole Body Insertion into the Nuclear Membrane and Nuclear Envelope Homeostasis

    The budding yeast spindle pole body (SPB) is anchored in the nuclear envelope so that it can simultaneously nucleate both nuclear and cytoplasmic microtubules. During SPB duplication, the newly formed SPB is inserted into the nuclear membrane. The mechanism of SPB insertion is poorly understood but likely involves the action of integral membrane proteins to mediate changes in the nuclear envelope itself, such as fusion of the inner and outer nuclear membranes. Analysis of the functional domains of the budding yeast SUN protein and SPB component Mps3 revealed that most regions are not essential for growth or SPB duplication under wild-type conditions. However, a novel dominant allele in the P-loop region, MPS3-G186K, displays defects in multiple steps in SPB duplication, including SPB insertion, indicating a previously unknown role for Mps3 in this step of SPB assembly. Characterization of the MPS3-G186K mutant by electron microscopy revealed severe over-proliferation of the inner nuclear membrane, which could be rescued by altering the characteristics of the nuclear envelope using both chemical and genetic methods. Lipid profiling revealed that cells lacking MPS3 contain abnormal amounts of certain types of polar and neutral lipids, and deletion or mutation of MPS3 can suppress growth defects associated with inhibition of sterol biosynthesis, suggesting that Mps3 directly affects lipid homeostasis. Therefore, we propose that Mps3 facilitates insertion of SPBs in the nuclear membrane by modulating nuclear envelope composition

    An estimate of the number of tropical tree species

    The high species richness of tropical forests has long been recognized, yet there remains substantial uncertainty regarding the actual number of tropical tree species. Using a pantropical tree inventory database from closed canopy forests, consisting of 657,630 trees belonging to 11,371 species, we use a fitted value of Fisher’s alpha and an approximate pantropical stem total to estimate the minimum number of tropical forest tree species to fall between ∼40,000 and ∼53,000, i.e. at the high end of previous estimates. Contrary to common assumption, the Indo-Pacific region was found to be as species-rich as the Neotropics, with both regions having a minimum of ∼19,000–25,000 tree species. Continental Africa is relatively depauperate with a minimum of ∼4,500–6,000 tree species. Very few species are shared among the African, American, and the Indo-Pacific regions. We provide a methodological framework for estimating species richness in trees that may help refine species richness estimates of tree-dependent taxa

    Compatible pollen responses in Arabidopsis thaliana: Investigations into stigma-expressed genes

    The Brassicaceae “dry” stigmas lack surface secretions, meaning there is strict control of compatible pollen responses. This consists of processes which facilitate pollen water uptake and germination, and cell wall modifications for pollen tube growth through the stigma towards the ovules. This basal compatible pollen response pathway is triggered by a Brassicaceae family-specific recognition system that leads to an exocyst complex-mediated regulated secretion of factors from the stigmatic papillae towards the compatible pollen grain. Physiological requirements for compatible pollen germination on the stigma, such as polarized secretion, are well-studied; however, the signalling components upstream of these processes during the compatible pollen response are unknown. The work presented in this thesis demonstrates use of a reverse-genetics approach to identify candidate genes that may be involved in the compatible pollen response pathway. Candidate genes were chosen based on stigma-specific expression patterns using available transcriptome datasets. The pseudokinase BRASSIKIN1 (BKN1) and its closely-related paralogue, BKN2, were identified as candidate signalling proteins. An E6-like protein (E6L1) was also identified as a protein that may be involved in early pollen reception. Mutant analyses for each of these genes in Arabidopsis thaliana uncovered a role for each during early compatible pollen responses. BKN1 and BKN2 appear to function redundantly, where double amiRNA lines showed reduced pollen adhesion, pollen hydration and seed set, while knockout lines for E6L1 displayed comparatively mild fertility defects. The evidence presented in this thesis supports that the BKNs and E6L1 are involved in early compatible pollen responses in the stigma, and the reverse-genetics approach presented here represents a useful tool for identifying candidate genes involved in compatible pollen responses in the stigma.Ph.D.2020-11-16 00:00:0

    This study explored the concept of a spiritual retreat for frontline employees of a large corporate call centre. During a 1 day retreat, 4 call centre employees were introduced to various meditation and retreat activities. Follovsdng the retreat the participants were asked to incorporate the various meditations and activities into their workplace. The participants kept journals throughout the study in an effort to determine what occurred when these practices were transferred from the retreat setting to the workplace. This study examined how a working spirituality enhances one's sense of fulfillment, defined by certain critical elements: relationship, awareness, ritual, internal commitment, and choice. Although the retreat was a successful means of exploring these elements, the degree to which each employee could benefit from them was determined by the extent of their internal commitment not only to themselves, but also to their jobs
