    Dendritic cell deficiencies persist seven months after SARS-CoV-2 infection

    Severe Acute Respiratory Syndrome Coronavirus (SARS-CoV)-2 infection induces an exacerbated inflammation driven by innate immunity components. Dendritic cells (DCs) play a key role in the defense against viral infections, for instance plasmacytoid DCs (pDCs), have the capacity to produce vast amounts of interferon-alpha (IFN-α). In COVID-19 there is a deficit in DC numbers and IFN-α production, which has been associated with disease severity. In this work, we described that in addition to the DC deficiency, several DC activation and homing markers were altered in acute COVID-19 patients, which were associated with multiple inflammatory markers. Remarkably, previously hospitalized and nonhospitalized patients remained with decreased numbers of CD1c+ myeloid DCs and pDCs seven months after SARS-CoV-2 infection. Moreover, the expression of DC markers such as CD86 and CD4 were only restored in previously nonhospitalized patients, while no restoration of integrin β7 and indoleamine 2,3-dyoxigenase (IDO) levels were observed. These findings contribute to a better understanding of the immunological sequelae of COVID-19

    Incidence, Clinical Characteristics and Management of Inflammatory Bowel Disease in Spain : Large-Scale Epidemiological Study

    (1) Aims: To assess the incidence of inflammatory bowel disease (IBD) in Spain, to describe the main epidemiological and clinical characteristics at diagnosis and the evolution of the disease, and to explore the use of drug treatments. (2) Methods: Prospective, population-based nationwide registry. Adult patients diagnosed with IBD-Crohn's disease (CD), ulcerative colitis (UC) or IBD unclassified (IBD-U)-during 2017 in Spain were included and were followed-up for 1 year. (3) Results: We identified 3611 incident cases of IBD diagnosed during 2017 in 108 hospitals covering over 22 million inhabitants. The overall incidence (cases/100,000 person-years) was 16 for IBD, 7.5 for CD, 8 for UC, and 0.5 for IBD-U; 53% of patients were male and median age was 43 years (interquartile range = 31-56 years). During a median 12-month follow-up, 34% of patients were treated with systemic steroids, 25% with immunomodulators, 15% with biologics and 5.6% underwent surgery. The percentage of patients under these treatments was significantly higher in CD than UC and IBD-U. Use of systemic steroids and biologics was significantly higher in hospitals with high resources. In total, 28% of patients were hospitalized (35% CD and 22% UC patients, p < 0.01). (4) Conclusion: The incidence of IBD in Spain is rather high and similar to that reported in Northern Europe. IBD patients require substantial therapeutic resources, which are greater in CD and in hospitals with high resources, and much higher than previously reported. One third of patients are hospitalized in the first year after diagnosis and a relevant proportion undergo surgery

    The genetic architecture of the human cerebral cortex

    The cerebral cortex underlies our complex cognitive capabilities, yet little is known about the specific genetic loci that influence human cortical structure. To identify genetic variants that affect cortical structure, we conducted a genome-wide association meta-analysis of brain magnetic resonance imaging data from 51,665 individuals. We analyzed the surface area and average thickness of the whole cortex and 34 regions with known functional specializations. We identified 199 significant loci and found significant enrichment for loci influencing total surface area within regulatory elements that are active during prenatal cortical development, supporting the radial unit hypothesis. Loci that affect regional surface area cluster near genes in Wnt signaling pathways, which influence progenitor expansion and areal identity. Variation in cortical structure is genetically correlated with cognitive function, Parkinson's disease, insomnia, depression, neuroticism, and attention deficit hyperactivity disorder

    Omecamtiv mecarbil in chronic heart failure with reduced ejection fraction, GALACTIC‐HF: baseline characteristics and comparison with contemporary clinical trials

    Aims: The safety and efficacy of the novel selective cardiac myosin activator, omecamtiv mecarbil, in patients with heart failure with reduced ejection fraction (HFrEF) is tested in the Global Approach to Lowering Adverse Cardiac outcomes Through Improving Contractility in Heart Failure (GALACTIC‐HF) trial. Here we describe the baseline characteristics of participants in GALACTIC‐HF and how these compare with other contemporary trials. Methods and Results: Adults with established HFrEF, New York Heart Association functional class (NYHA) ≥ II, EF ≤35%, elevated natriuretic peptides and either current hospitalization for HF or history of hospitalization/ emergency department visit for HF within a year were randomized to either placebo or omecamtiv mecarbil (pharmacokinetic‐guided dosing: 25, 37.5 or 50 mg bid). 8256 patients [male (79%), non‐white (22%), mean age 65 years] were enrolled with a mean EF 27%, ischemic etiology in 54%, NYHA II 53% and III/IV 47%, and median NT‐proBNP 1971 pg/mL. HF therapies at baseline were among the most effectively employed in contemporary HF trials. GALACTIC‐HF randomized patients representative of recent HF registries and trials with substantial numbers of patients also having characteristics understudied in previous trials including more from North America (n = 1386), enrolled as inpatients (n = 2084), systolic blood pressure &lt; 100 mmHg (n = 1127), estimated glomerular filtration rate &lt; 30 mL/min/1.73 m2 (n = 528), and treated with sacubitril‐valsartan at baseline (n = 1594). Conclusions: GALACTIC‐HF enrolled a well‐treated, high‐risk population from both inpatient and outpatient settings, which will provide a definitive evaluation of the efficacy and safety of this novel therapy, as well as informing its potential future implementation

    Status, population trend and genetic structure of South American fur seals, Arctocephalus australis , in southwestern Atlantic waters

    The South American fur seal (SAFS), Arctocephalus australis, was severely depleted during the 18th century. This work updates information on distribution, abundance, seasonal movements, and genetic structure along the southwestern South Atlantic. Its distribution in Argentine waters includes nineteen rookeries on the Patagonian coast, Isla de los Estados, and islets of the Beagle Channel. Censuses were carried out in rookeries from Isla Escondida to Isla de los Estados from 1996 to 2014. The largest concentrations are found in Chubut, (4,500–15,500 seals) and Isla de los Estados (4,500). Pups were recorded on Isla Escondida, Rasa, and Isla de los Estados. An increasing number of SAFS were recorded in northern Patagonia, including seals marked in Uruguay. Skin samples were taken from southern Brazil to Tierra del Fuego in order to evaluate population structure and demographic trends. Genetic results indicated no population subdivisions (Φst = −0.0292, Fst = −0.00716, P > 0.05). Bayesian skyline plots constructed for the entire data set indicated evidence of rapid population expansion discernible in Nef between the last 20,000 and 17,000 yr. Genetics results and observation data from marked fur seals support the hypothesis of ancient gene flow and a single Atlantic population.     RESUMEN En los últimos dos años, la Universidad Privada del Norte (UPN) ha venido desarrollando actividades de Responsabilidad Social en diversos aspectos, buscando fomentar en los jóvenes universitarios un espíritu de solidaridad con su entorno y su compromiso con la sociedad. Por ello, en este proyecto de responsabilidad social se propone la realización de una serie de actividades tanto en el Asentamiento Humano Las Torres de San Borja, Distrito de Moche, como en la Universidad, teniendo como finalidad, concientizar a la comunidad sobre el cuidado del medio ambiente, salud, calidad de vida, etc. El proyecto no había previsto factores claves que permitirían su ejecución, cómo es el presupuesto otorgado por los organismos cooperantes y los estudiantes. Esta falencia, relacionada con la gestión, trajo como consecuencia retrasos en la ejecución establecida. Por otro lado, el equipo de voluntariado estableció estrategias y acciones concretas para que toda la comunidad UPN participe activamente en el desarrollo sostenible del enfoque de la Responsabilidad Social Universitaria. Gracias al proyecto UNIONES, el trabajo en conjunto de estudiantes de voluntariado de UPN en coordinación con el Centro de Atención y Educación a la Familia (CAEF), además del apoyo de la población del AA.HH. Las Torres de San Borja, se logró desarrollar una serie de actividades acordadas en nuestro proyecto.ABSTRACT For the last few years, UPN has developed various social responsibility activities on diverse aspects, hoping to foment in our students a spirit of solidarity and commitment to our society. Thanks to the UNIONES project, the collective efforts of both students and volunteers from UPN and in cooperation with the Family Education and attention center (CAEF), as well as the support of the very people of the human settlement LAS TORRES DE SAN BORJA, every listed activity for the project was successfully executed
