    Hypoxia is a central problem in tumor treatment because hypoxic cells are less sensitive to chemo- and radiotherapy than normoxic cells. Radioresistance of hypoxic tumor cells is due to reduced sensitivity towards low Linear Energy Transfer (LET) radiation. High LET α-emitters are thought to eradicate tumor cells independent of cellular oxygenation. Therefore, the aim of this study was to demonstrate that the cell-bound α-particle emitting 213Bi immunoconjugates efficiently kill hypoxic just like normoxic CAL33 tumor cells. For that purpose CAL33 cells were incubated with 213Bianti- EGFR-MAb or irradiated with photons with a nominal energy of 6 MeV both under hypoxic and normoxic conditions. Oxygenation of cells was checked via the hypoxia-associated marker HIF-1α. Survival of cells was analysed using the clonogenic assay. Cell viability was monitored with the WST colorimetric assay. Results were evaluated statistically using a t-test and a Generalized Linear Mixed Model (GLMM). Survival and viability of CAL33 cells decreased both after incubation with increasing 213Bi-anti-EGFR-MAb activity concentrations (9.25 kBq/ml – 1.48 MBq/ml) and irradiation with increasing doses of photons (0.5 – 12 Gy). Following photon irradiation survival and viability of normoxic cells were significantly lower than those of hypoxic cells at all doses analysed. In contrast, cell death induced by 213Bianti- EGFR-MAb turned out to be independent of cellular oxygenation. These results demonstrate for the first time that α-particle emitting 213Bi-immunoconjugates eradicate hypoxic tumor cells as effective as normoxic cells.     Direct observation of a liquid film under a vapor environment in a pool boiling using a nanofluid

    The existence of a liquid film separating a vapor bubble from a heated solid surface is confirmed using a nanofluid. The existence of such a liquid film had been a theoretical premise of the critical heat flux mechanism, significantly difficult to verify through experimental observations. Here, we show that a liquid film under a massive vapor bubble adheres to a heated solid surface. The liquid film comes into being trapped in a dynamic coalescence environment of nucleate bubbles, which grow and depart continuously from the heated surface. In its dryout process, the liquid film displays vapor "holes" originating from the rupture of discrete nucleating bubbles. The dryout process of the liquid film can be understood from the vaporization of rims of the holes and of smooth film region.open51

    Group status drives majority and minority integration preferences

We targeted attitudes at two universities, one where 85% of the student body is White, and another where 76% of students are Black. The results revealed that when a group constituted the majority, its members generally preferred assimilationist policies, and when a group constituted the minority, its members generally preferred pluralistic policies. The results support a functional perspective: Both majority and minority groups seek to protect and enhance their collective identities

    The Borexino detector at the Laboratori Nazionali del Gran Sasso

    Borexino, a large volume detector for low energy neutrino spectroscopy, is currently running underground at the Laboratori Nazionali del Gran Sasso, Italy. The main goal of the experiment is the real-time measurement of sub MeV solar neutrinos, and particularly of the mono energetic (862 keV) Be7 electron capture neutrinos, via neutrino-electron scattering in an ultra-pure liquid scintillator. This paper is mostly devoted to the description of the detector structure, the photomultipliers, the electronics, and the trigger and calibration systems. The real performance of the detector, which always meets, and sometimes exceeds, design expectations, is also shown. Some important aspects of the Borexino project, i.e. the fluid handling plants, the purification techniques and the filling procedures, are not covered in this paper and are, or will be, published elsewhere (see Introduction and Bibliography).Comment: 37 pages, 43 figures, to be submitted to NI

    Abstract: The self as a psychological construct, and the self in relation to the other has been discussed in psychological and sociological literature for decades, but not much attention has been given to the psychological development of the self in relation to the social construction of prejudice. The primary aim of this article is to explore the self in prejudice and thus the psychological processes involved in the development of self within the social context. Consequently, the aim is to explore the self in the construction and expression of prejudice from both a social and psychological approach, and to explain selfhood influences at the individual, group and community levels. I use the conceptual framework of Kohut’s self psychology as a lens to present the development of the self and thus the idea of the development of the self in relation to the other. In such exploration of self in prejudice, I present some of my ideas which include prejudice as an outcome of self-definition in the context of the other, as well as linking self in prejudice and group dynamics to attachment theory and the notion of “selfgroup’ in terms of overidentification with the in-group. While the social and the psychological in terms of the development of the self cannot be separated, I have therefore attempted to merge at some point the two bodies of thought in relation to the self in prejudice

    A human ESC-based screen identifies a role for the translated lncRNA LINC00261 in pancreatic endocrine differentiation

    Long noncoding RNAs (lncRNAs) are a heterogenous group of RNAs, which can encode small proteins. The extent to which developmentally regulated lncRNAs are translated and whether the produced microproteins are relevant for human development is unknown. Using a human embryonic stem cell (hESC)-based pancreatic differentiation system, we show that many lncRNAs in direct vicinity of lineage-determining transcription factors (TFs) are dynamically regulated, predominantly cytosolic, and highly translated. We genetically ablated ten such lncRNAs, most of them translated, and found that nine are dispensable for pancreatic endocrine cell development. However, deletion of LINC00261 diminishes insulin(+) cells, in a manner independent of the nearby TF FOXA2. One-by-one deletion of each of LINC00261's open reading frames suggests that the RNA, rather than the produced microproteins, is required for endocrine development. Our work highlights extensive translation of lncRNAs during hESC pancreatic differentiation and provides a blueprint for dissection of their coding and noncoding roles

    Large-Scale Spatio-Temporal Patterns of Mediterranean Cephalopod Diversity

    Species diversity is widely recognized as an important trait of ecosystems’ functioning and resilience. Understanding the causes of diversity patterns and their interaction with the environmental conditions is essential in order to effectively assess and preserve existing diversity. While diversity patterns of most recurrent groups such as fish are commonly studied, other important taxa such as cephalopods have received less attention. In this work we present spatio-temporal trends of cephalopod diversity across the entire Mediterranean Sea during the last 19 years, analysing data from the annual bottom trawl survey MEDITS conducted by 5 different Mediterranean countries using standardized gears and sampling protocols. The influence of local and regional environmental variability in different Mediterranean regions is analysed applying generalized additive models, using species richness and the Shannon Wiener index as diversity descriptors. While the western basin showed a high diversity, our analyses do not support a steady eastward decrease of diversity as proposed in some previous studies. Instead, high Shannon diversity was also found in the Adriatic and Aegean Seas, and high species richness in the eastern Ionian Sea. Overall diversity did not show any consistent trend over the last two decades. Except in the Adriatic Sea, diversity showed a hump-shaped trend with depth in all regions, being highest between 200–400 m depth. Our results indicate that high Chlorophyll a concentrations and warmer temperatures seem to enhance species diversity, and the influence of these parameters is stronger for richness than for Shannon diversityVersión del editor4,411
