    Sensitivity of the Immunohistochemistry technique in central nervous system fragments of cattle and horses naturally infected by rabies virus

    A raiva é uma zoonose viral que acomete o sistema nervoso central (SNC) de mamíferos, considerada um grave problema de saúde pública. Herbívoros (bovinos e equinos) são frequentemente acometidos pela in-fecção após serem atacados por morcegos hematófagos (Desmodus rotundus). A técnica de imunofluorescência direta (IFD) realizada em tecidos frescos, recomendada pela Organização Mundial de Saúde (OMS), é utilizada para o diagnóstico da raiva. A técnica de imuno-histoquímica (IHQ) é utilizada para detectar antígenos em tecidos fixados, pelo uso de anticorpos monoclonais/policlonais. O objetivo deste trabalho foi avaliar a sensibilidade da IHQ na detecção de antígenos do vírus da raiva em amostras de SNC de herbívoros fixadas em formol, analisando a distribuição antigênica em diferentes fragmentos do SNC. Os resultados demonstraram concordância das técnicas de IFD e IHQ. A IHQ mostrou maior sensibilidade em amostras de bovinos em relação às de equinos, especialmente quando realizada em fragmentos de cerebelo e tronco encefálico. A detecção de antígeno nestes fragmentos foi mais consistente para ambas as técnicas, nas duas espécies. Estes resultados demonstram que a IHQ pode ser empregada para a vigilância epidemiológica da raiva, entretanto, recomenda-se cautela ao se empregar a IHQ para diagnóstico de doença em herbívoros, especialmente quando o fragmento encaminhado ao laboratório for apenas o hipocampo.Rabies is a viral zoonosis that causes disease in the central nervous system (CNS) of mammals and it is considered a serious problem of public health. Herbivorous (bovines and equines) are often infected after being attacked by vampire bats (Desmodus rotundus). The direct fluorescent antibody technique is used as a diagnostic test to detect viral antigens in fresh tissues and is recommended by the World Health Organization. The immunohistochemistry technique (IHC) is used to detect the viral antigen through the use of monoclonal/policlonal antibodies in formalin-fixed tissues. The aim of this work was to evaluate the sensitivity of the IHC in samples of CNS of herbivorous fixed in formol, analyzing the antigenic distribution in different fragments of the CNS. The results demonstrated good agreement between the two techniques for the rabies diagnosis. The IHC presented higher sensitivity in samples of cattle comparing to horse samples, especially in fragments of cerebellum and brain stem. These fragments demonstrated to be more suitable for antigen detection by both techniques in the two species. These data demonstrate that the IHC is suitable for rabies vigilance yet cautions should be taken in examining cattle and horses samples, when the submitted specimen is only the hippocampus

A implementação de filosofia Lean na manutenção visa melhorar o planeamento e a programação das atividades, aumentar a produtividade, motivar os trabalhadores, ajudar na introdução de novos hábitos e de novas atitudes no trabalho e ainda identificar os desperdícios que devem ser eliminados. A competitividade e o preço excessivo dos produtos (bens ou serviços) que o cliente final não quer pagar, faz com que as empresas públicas e privadas tomem medidas para evitar todo o tipo de desperdícios, rentabilizando ao máximo a sua cadeia de valor. A filosofia Lean foi uma das formas encontradas para identificar e eliminar alguns desses desperdícios, melhorando o processo produtivo e, desta forma, tornando as organizações mais competitivas. O Lean tem várias ferramentas podendo ser aplicadas em diversas áreas. Uma dessas áreas é a manutenção, onde se pretende que os equipamentos operem sem interrupções e com uma produção de qualidade. Algumas das ferramentas do Lean encaixam-se perfeitamente na área da manutenção para identificação de desperdícios, criação de valor, realização de planeamento, melhoria do sistema de trabalho na organização e até no próprio desenvolvimento dos trabalhadores. A presente dissertação visa mostrar algumas das ferramentas mais comuns da filosofia Lean com aplicabilidade na manutenção de equipamentos de apoio à atividade de um serviço publico e ao desenvolvimento de um modelo de avaliação com apresentação através de estudo de caso. No trabalho infra são apresentadas diversas ferramentas Lean, nomeadamente 5s, trabalho padronizado, (standardwork uniformização), gestão visual (andon), melhoria continua (kaisen), mapeamento fluxo de valor na manutenção (MVSM), SIPOC, gestão visual, 5w, ciclo PDCA. O modelo de avaliação está estruturado em 3 grandes áreas “Clientes”, “Organização” e “Fornecedores”, vai subdividir-se em 7 critérios e 60 atributos. Dos 60 atributos, foram desenvolvidos 6 e que compõem mais de 70 atributos, perfazendo mais de 130 no final o modelo proposto neste trabalho.The creation of a lean evaluation model is important to create good practice in the field of maintenance as the principle of an activity that uses lean tools aware of all its advantages. Implementing Lean's philosophy of maintenance aims to improve planning and scheduling of activities, increase productivity, motivate employees, assist in introducing new habits and new attitudes at work, and identify the wastes that need to be eliminated. Competitiveness and the excessive price of products (goods or services) that the final customer does not want to pay, makes public and private companies take measures to avoid all types of waste, making the most of its value chain. The Lean philosophy was one of the ways found to identify and eliminate some of these wastes, improving the production process and thus making companies more competitive. Lean has several tools that can be applied in several areas. One of these areas is maintenance, where the equipment is intended to operate without interruption and with a quality production. Some of Lean's tools fit perfectly into the maintenance area for waste identification, value creation. The present dissertation aims to show some of the most common tools of the Lean philosophy with applicability in the maintenance of equipment to support the activity of a public service and the development of an evaluation model with presentation through case study. In the work below are presented several Lean tools, namely 5s, standardized work, (standardwork standardization), visual management (andon), continuous improvement (kaisen), maintenance value flow mapping (MVSM), SIPOC, visual management, 5w, cycle PDCA. The avaliation model is structured in 3 large areas "Customers", "Organization" and "Suppliers", will be subdivided into 7 criteria and 60 attributes.     The term basal plate of the human placenta as a source of functional extravillous trophoblast cells

    Background\ud Extravillous trophoblast (EVT) cells are of pivotal importance in human embryo implantation and homeostasis of the maternal fetal interface. Invasion of the endometrium by EVT contributes to placental anchorage, spiral artery remodeling, immunological defense, tolerogenic responses, and several collaborative cross talks involved in establishing and maintaining a successful pregnancy. We report here an improved protocol for the isolation of fully differentiated EVT cells from the basal plate of the human term placenta.\ud \ud \ud Methods\ud The basal plate was carefully dissected from the villous tissue and the amniochorion membrane prior to enzymatic digestion. Term basal EVT cells were isolated using a 30 and 60% Percoll gradient. A panel of markers and characteristics of the isolated cells were used to confirm the specificity and efficiency of the method so that their potential as an investigative tool for placental research could be ascertained.\ud \ud \ud Results\ud Isolated cells were immunoreactive for cytokeratin-7 (CK-7), placental growth factor, placental alkaline phosphatase, human leukocyte antigen G1 (HLA-G1), and α1 and α5 integrins, similarly to the EVT markers from first trimester placental villi. Around 95% of the isolated cells labeled positively for CK-7 and 82% for HLA-G1. No significant change in viability was observed during 48 h of EVT culture as indicated by propidium iodide incorporation and trypan blue test exclusion. Genes for metalloproteinases MMP-2 and MMP9 (positive regulators of trophoblast invasiveness) were expressed up to 48 h of culturing, as also the gelatinolytic activity of the isolated cells. Transforming growth factor (TGF)-beta, which inhibits proliferation, migration, and invasiveness of first-trimester EVT cells, also reduced invasion of isolated term EVT cells in transwell assays, whereas epidermal growth factor was a positive modulator.\ud \ud \ud Conclusions\ud Term basal plate may be a viable source of functional EVT cells that is an alternative to villous explant-derived EVT cells and cell lines.     Lying in Wait: The Resurgence of Dengue Virus After the Zika Epidemic in Brazil

    After the Zika virus (ZIKV) epidemic in the Americas in 2016, both Zika and dengue incidence declined to record lows in many countries in 2017-2018, but in 2019 dengue resurged in Brazil, causing ~2.1 million cases. In this study we use epidemiological, climatological and genomic data to investigate dengue dynamics in recent years in Brazil. First, we estimate dengue virus force of infection (FOI) and model mosquito-borne transmission suitability since the early 2000s. Our estimates reveal that DENV transmission was low in 2017-2018, despite conditions being suitable for viral spread. Our study also shows a marked decline in dengue susceptibility between 2002 and 2019, which could explain the synchronous decline of dengue in the country, partially as a result of protective immunity from prior ZIKV and/or DENV infections. Furthermore, we performed phylogeographic analyses using 69 newly sequenced genomes of dengue virus serotype 1 and 2 from Brazil, and found that the outbreaks in 2018-2019 were caused by local DENV lineages that persisted for 5-10 years, circulating cryptically before and after the Zika epidemic. We hypothesize that DENV lineages may circulate at low transmission levels for many years, until local conditions are suitable for higher transmission, when they cause major outbreaks

    The Long-Baseline Neutrino Experiment: Exploring Fundamental Symmetries of the Universe

    The preponderance of matter over antimatter in the early Universe, the dynamics of the supernova bursts that produced the heavy elements necessary for life and whether protons eventually decay --- these mysteries at the forefront of particle physics and astrophysics are key to understanding the early evolution of our Universe, its current state and its eventual fate. The Long-Baseline Neutrino Experiment (LBNE) represents an extensively developed plan for a world-class experiment dedicated to addressing these questions. LBNE is conceived around three central components: (1) a new, high-intensity neutrino source generated from a megawatt-class proton accelerator at Fermi National Accelerator Laboratory, (2) a near neutrino detector just downstream of the source, and (3) a massive liquid argon time-projection chamber deployed as a far detector deep underground at the Sanford Underground Research Facility. This facility, located at the site of the former Homestake Mine in Lead, South Dakota, is approximately 1,300 km from the neutrino source at Fermilab -- a distance (baseline) that delivers optimal sensitivity to neutrino charge-parity symmetry violation and mass ordering effects. This ambitious yet cost-effective design incorporates scalability and flexibility and can accommodate a variety of upgrades and contributions. With its exceptional combination of experimental configuration, technical capabilities, and potential for transformative discoveries, LBNE promises to be a vital facility for the field of particle physics worldwide, providing physicists from around the globe with opportunities to collaborate in a twenty to thirty year program of exciting science.     Antimicrobial resistance among migrants in Europe: a systematic review and meta-analysis

    BACKGROUND: Rates of antimicrobial resistance (AMR) are rising globally and there is concern that increased migration is contributing to the burden of antibiotic resistance in Europe. However, the effect of migration on the burden of AMR in Europe has not yet been comprehensively examined. Therefore, we did a systematic review and meta-analysis to identify and synthesise data for AMR carriage or infection in migrants to Europe to examine differences in patterns of AMR across migrant groups and in different settings. METHODS: For this systematic review and meta-analysis, we searched MEDLINE, Embase, PubMed, and Scopus with no language restrictions from Jan 1, 2000, to Jan 18, 2017, for primary data from observational studies reporting antibacterial resistance in common bacterial pathogens among migrants to 21 European Union-15 and European Economic Area countries. To be eligible for inclusion, studies had to report data on carriage or infection with laboratory-confirmed antibiotic-resistant organisms in migrant populations. We extracted data from eligible studies and assessed quality using piloted, standardised forms. We did not examine drug resistance in tuberculosis and excluded articles solely reporting on this parameter. We also excluded articles in which migrant status was determined by ethnicity, country of birth of participants' parents, or was not defined, and articles in which data were not disaggregated by migrant status. Outcomes were carriage of or infection with antibiotic-resistant organisms. We used random-effects models to calculate the pooled prevalence of each outcome. The study protocol is registered with PROSPERO, number CRD42016043681. FINDINGS: We identified 2274 articles, of which 23 observational studies reporting on antibiotic resistance in 2319 migrants were included. The pooled prevalence of any AMR carriage or AMR infection in migrants was 25·4% (95% CI 19·1-31·8; I2 =98%), including meticillin-resistant Staphylococcus aureus (7·8%, 4·8-10·7; I2 =92%) and antibiotic-resistant Gram-negative bacteria (27·2%, 17·6-36·8; I2 =94%). The pooled prevalence of any AMR carriage or infection was higher in refugees and asylum seekers (33·0%, 18·3-47·6; I2 =98%) than in other migrant groups (6·6%, 1·8-11·3; I2 =92%). The pooled prevalence of antibiotic-resistant organisms was slightly higher in high-migrant community settings (33·1%, 11·1-55·1; I2 =96%) than in migrants in hospitals (24·3%, 16·1-32·6; I2 =98%). We did not find evidence of high rates of transmission of AMR from migrant to host populations. INTERPRETATION: Migrants are exposed to conditions favouring the emergence of drug resistance during transit and in host countries in Europe. Increased antibiotic resistance among refugees and asylum seekers and in high-migrant community settings (such as refugee camps and detention facilities) highlights the need for improved living conditions, access to health care, and initiatives to facilitate detection of and appropriate high-quality treatment for antibiotic-resistant infections during transit and in host countries. Protocols for the prevention and control of infection and for antibiotic surveillance need to be integrated in all aspects of health care, which should be accessible for all migrant groups, and should target determinants of AMR before, during, and after migration.     Hidden among bromeliads in the Brazilian semiarid: first records of Phyllopezus lutzae for the Caatinga domain and its predation by Tropidurus hispidus

    During wildlife rescue and monitoring activities, we recorded 142 individuals of Phyllopezus lutzae in the municipalities of Tucano and Nova Soure, state of Bahia, Northeastern Brazil. These records are the first of this species in the Caatinga domain. Moreover, an adult individual of Tropidurus hispidus was recorded attempting to subdue an adult P. lutzae. Beyond to expand the known distribution range of the species, our records show that P. lutzae inhabits an ecological and climate domain different from Atlantic Forest where it was previously known, and that it is a potential prey of T. hispidus.Asociación Herpetológica Argentin
