    Lolium perenne, as one of the most important forage grasses of temperate regions, combines a number of very useful characteristics, e.g., good seedling establishment, with a low resistance to drought and limited winter hardiness. Trait selection and introgression can be greatly enhanced by the use of molecular markers in a genetic linkage map. The aim of this project was the generation of a genomic microsatellite library which when combined with microsatellites developed from a Genethresher database would give good genome coverage coupled to high levels of marker polymorphism

    Biotin carboxyl carrier protein and carboxyltransferase subunits of the multi-subunit form of acetyl-CoA carboxylase from \u3ci\u3eBrassica napus\u3c/i\u3e: cloning and analysis of expression during oilseed rape embryogenesis

    In the oilseed rape Brassica napus there are two forms of acetyl- CoA carboxylase (ACCase). As in other dicotyledonous plants there is a type I ACCase, the single polypeptide 220 kDa form, and a type II multi-subunit complex analogous to that of Escherichia coli and Anabaena. This paper describes the cloning and characterization of a plant biotin carboxyl carrier protein (BCCP) from the type II ACCase complex that shows 61% identity/79% similarity with Anabaena BCCP at the amino acid level. Six classes of nuclear encoded oilseed rape BCCP cDNA were cloned, two of which contained the entire coding region. The BCCP sequences allowed the assignment of function to two previously unassigned Arabidopsis expressed sequence tag (EST) sequences. We also report the cloning of a second type II ACCase component from oilseed rape, the ÎČ-carboxyltransferase subunit (ÎČCT), which is chloroplast-encoded. Northern analysis showed that although the relative levels of BCCP and ÎČCT mRNA differed between different oilseed rape tissues, their temporal patterns of expression were identical during embryo development. At the protein level, expression of BCCP during embryo development was studied by Western blotting, using affinity-purified anti-biotin polyclonal sera. With this technique a 35 kDa protein thought to be BCCP was shown to reside within the chloroplast. This analysis also permitted us to view the differential expression of several unidentified biotinylated proteins during embryogenesis

    Project Half Double: Preliminary Results for Phase 1, June 2016

    Project Half Double has a clear mission to succeed in finding a project methodology that can increase the success rate of our projects while increasing the speed at which we generate new ideas and develop new products and services. Chaos and complexity should be seen as a basic condition and as an opportunity rather than a threat and a risk. We are convinced that by doing so, we can strengthen Denmark’s competitiveness and play an important role in the battle for jobs and future welfare. The overall goal is to deliver “projects in half the time with double the impact”, where projects in half the time should be understood as half the time to impact (benefit realisation, effect is achieved) and not as half the time for project execution.The purpose of Project Half Double is to improve Danish industrial competitiveness by radically increasing the pace and impact of the development and innovation activities carried out within the framework of the projects.The formal part of Project Half Double was initiated in June 2015. We started out by developing, refining and testing the Half Double methodology on seven pilot projects in the first phase of the project, which will end June 2016.The current status of responding to the above overall Project Half Double goal for the seven pilot projects can be summarised as follows:- The LantmĂ€nnen Unibake pilot project was able to launch the first stores after 5 months, which is considerably shorter lead time than comparable reference projects, which have had a lead time of 10 months or more. This is in line with the overall goal of Project Half Double of delivering impact faster.- Four pilot projects have the potential to deliver impact faster, but it is too early to evaluate. Some results might be evaluated in the second half of 2016, while other results take longer to evaluate (Coloplast, Novo Nordisk, GN Audio and VELUX).- Two pilot projects will probably not be able to deliver impact faster, although it is too early to evaluate them. The evaluation of these pilot projects takes place over a longer period of time as it will take years before many of the key performance indicators associated with them can be evaluated (Grundfos and Siemens Wind Power).In addition to the current status of delivering impact faster for the seven pilot projects, it is important to highlight that Project Half Double phase 1 has planted many seeds in the pilot organisations concerning project methodology and beyond. The many learning points from each pilot project show that Project Half Double has left its clear footprint in the pilot organisations, and that the Half Double methodology has evolved and developed very much during Project Half Double phase 1

    Crystal Structure of a Eukaryotic GEN1 Resolving Enzyme Bound to DNA

    SummaryWe present the crystal structure of the junction-resolving enzyme GEN1 bound to DNA at 2.5 Å resolution. The structure of the GEN1 protein reveals it to have an elaborated FEN-XPG family fold that is modified for its role in four-way junction resolution. The functional unit in the crystal is a monomer of active GEN1 bound to the product of resolution cleavage, with an extensive DNA binding interface for both helical arms. Within the crystal lattice, a GEN1 dimer interface juxtaposes two products, whereby they can be reconnected into a four-way junction, the structure of which agrees with that determined in solution. The reconnection requires some opening of the DNA structure at the center, in agreement with permanganate probing and 2-aminopurine fluorescence. The structure shows that a relaxation of the DNA structure accompanies cleavage, suggesting how second-strand cleavage is accelerated to ensure productive resolution of the junction

    Exploring the tourist destination as a mosaic: The alternative lifecycles of the seaside amusement arcade sector in Britain

    One criticism of the tourism area lifecycle model is that it treats destinations as homogeneous entities. Instead destinations can be conceptualised as a mosaic of elements, each of which can follow a lifecycle that is different from that of the destination overall. This paper examines this issue with reference to amusement arcades in British seaside resorts and triangulates secondary sources and in-depth interviews to examine the historical evolution of this sector. It argues that the arcade sector has followed a lifecycle trajectory that is independent of the resorts in which they are located. A range of internal/external factors and global, national and local influences have affected the lifecycle of the arcade sector, including global developments in the entertainment industries; the influence of state policies and legislation; and the responses of local entrepreneurs to resort restructuring. The paper ends by arguing that destinations can be conceptualised as 'assemblages' of interacting elements

    The ‘heritagisation’ of the British seaside resort: The rise of the ‘old penny’ arcade.

    Amusement arcades have long been a key component of the British seaside resort. For almost a century, they enjoyed popularity and success and became established as a quintessential feature of the British seaside holiday. However, the advent of home-based video games along with recent gambling legislation has led to a decline of the seaside amusement arcade sector. Arcades gained a reputation as unsavoury places and their appearance and fortunes often mirrored those of the resorts in which they were located. However, over the past decade, a new variant of the seaside amusement arcade has appeared, featuring mechanical machines working on pre-decimal currency. Such ‘old penny arcades’ frequently describe themselves as museums or heritage centres and they offer an experience based on a nostalgic affection for the ‘traditional’ seaside holiday. They have appeared in the context of an increasing interest in the heritage of the British seaside resort and constitute one element of the ‘heritagisation’ of such resorts. This paper argues that such arcades can be important elements of strategies to reposition and rebrand resorts for the heritage tourism market
