    Effects of oxytocin administration on salivary sex hormone levels in autistic and neurotypical women

    Abstract: Background: Oxytocin administration, which may be of therapeutic value for individuals with social difficulties, is likely to affect endogenous levels of other socially relevant hormones. However, to date, the effects of oxytocin administration on endogenous hormones have only been examined in neurotypical individuals. The need to consider multi-hormone interactions is particularly warranted in oxytocin trials for autism due to evidence of irregularities in both oxytocin and sex steroid systems. Methods: In this double-blind cross-over study, saliva samples were collected from 16 autistic and 29 neurotypical women before and after intranasal administration of 24 IU oxytocin or placebo. Oestradiol, testosterone, and oxytocin levels were quantified in saliva samples. Participants also completed the Autism-Spectrum Quotient (AQ) and Empathy Quotient (EQ) questionnaires. Results: Distinct patterns of change in testosterone and oestradiol levels pre- to-post-administration were observed in autistic relative to neurotypical women (ANCOVA, p < 0.05 main effect of Group), controlling for sample collection time. The mean percent change oestradiol was + 8.8% for the autism group and − 13.0% for the neurotypical group (t = 1.81, p = 0.08), while the mean percent change testosterone was + 1.1% in the autism group and − 12.6% in the neurotypical group (t = 1.26, p = 0.22). In the oxytocin condition, the mean percent change oestradiol was + 12.6% in the autism group and − 6.9% in the neurotypical group (t = 1.78, p = 0.08), while the mean percent change testosterone was + 14.4% in the autism group and − 15.2% in the neurotypical group (t = 3.00, p = 0.006). Robust regression confirmed that group differences in percent change hormone levels were not driven by a small number of influential individuals. Baseline hormone levels did not differ between groups when considered individually. However, baseline testosterone relative to oestradiol (T:E2 ratio) was higher in autistic women (p = 0.023, Cohen’s d = 0.63), and this ratio correlated positively and negatively with AQ and EQ scores, respectively, in the combined sample. Limitations: Further studies with larger and more diverse autistic sample are warranted to confirm these effects. Conclusions: This study provides the first evidence that oxytocin influences endogenous testosterone levels in autistic individuals, with autistic women showing increases similar to previous reports of neurotypical men. These findings highlight the need to consider sex steroid hormones as a variable in future oxytocin trials

    Genetic risk for neurodegenerative disorders, and its overlap with cognitive ability and physical function

    Neurodegenerative disorders are associated with impaired cognitive function and worse physical health outcomes. This study aims to test whether polygenic risk for Alzheimer’s disease, Amyotrophic Lateral Sclerosis (ALS), or frontotemporal dementia (FTD) is associated with cognitive function and physical health in the UK Biobank, a cohort of healthy individuals. Group-based analyses were then performed to compare the top and bottom 10% for the three neurodegenerative polygenic risk scores; these groups were compared on the cognitive and physical health variables. Higher polygenic risk for AD, ALS, and FTD was associated with lower cognitive performance. Higher polygenic risk for FTD was also associated with increased forced expiratory volume in 1s and peak expiratory flow. A significant group difference was observed on the symbol digit substitution task between individuals with high polygenic risk for FTD and high polygenic risk for ALS. The results suggest some overlap between polygenic risk for neurodegenerative disorders, cognitive function and physical health

    A genome-wide association study of testicular germ cell tumor

    We conducted a genome-wide association study for testicular germ cell tumor (TGCT), genotyping 307,666 SNPs in 730 cases and 1,435 controls from the UK and replicating associations in a further 571 cases and 1,806 controls. We found strong evidence for susceptibility loci on chromosome 5 (per allele OR = 1.37 (95% CI = 1.19–1.58), P = 3 times 10-13), chromosome 6 (OR = 1.50 (95% CI = 1.28–1.75), P = 10-13) and chromosome 12 (OR = 2.55 (95% CI = 2.05–3.19), P = 10-31). KITLG, encoding the ligand for the receptor tyrosine kinase KIT, which has previously been implicated in the pathogenesis of TGCT and the biology of germ cells, may explain the association on chromosome 12

    The repertoire and intentionality of gestural communication in wild chimpanzees

    A growing body of evidence suggests that human language may have emerged primarily in the gestural rather than vocal domain, and that studying gestural communication in great apes is crucial to understanding language evolution. Although manual and bodily gestures are considered distinct at a neural level, there has been very limited consideration of potential differences at a behavioural level. In this study, we conducted naturalistic observations of adult wild East African chimpanzees (Pan troglodytes schweinfurthii) in order to establish a repertoire of gestures, and examine gesture use and comprehension, comparing across manual and bodily gestures. At the population level, 120 distinct gesture types were identified, consisting of 65 manual gestures and 55 bodily gestures. Both bodily and manual gestures were used intentionally and effectively to attain specific goals, by signallers who are sensitive to recipient attention. However, manual gestures differed from bodily gestures in terms of communicative persistence, indicating a qualitatively different form of behavioural flexibility in achieving goals. Both repertoire size and frequency of manual gesturing were more affiliative than bodily gestures, while bodily gestures were more antagonistic. These results indicate that manual gestures may have played a significant role in the emergence of increased flexibility in great ape communication and social bonding

    Late Artinskian–Early Kungurian (Early Permian) warming and maximum marine flooding in the East Gondwana interior rift, Timor and Western Australia, and comparisons across East Gondwana

    © 2016Substantial new information is presented on upper Artinskian–Kungurian deposits in Timor-Leste and in the Canning, Southern Carnarvon and northern Perth basins of Western Australia. These basins, situated between about 35°S and 55°S palaeolatitude, formed part of the East Gondwana interior rift, a precursor to the rift that 100 my later formed the Indian Ocean in this region. Timor lay near the main axis of the East Gondwana interior rift, whereas the Western Australian basins were marginal splays from the rift axis. The main depocentres developed as a result of faulting that was initiated during the Late Pennsylvanian. Detailed lithostratigraphic and biostratigraphic analyses have been made on the newly recognized Bua-bai limestone and the type Cribas Group in Timor, the Noonkanbah Formation in the Canning Basin, the Byro Group in the Merlinleigh Sub-basin of the Southern Carnarvon Basin, and the Carynginia Formation in the northern Perth Basin. In Timor the succession, which is highly disrupted by faulting, was deposited under open-marine conditions probably in a shelf–basin setting. Restricted, very shallow-water seas flooded the Canning Basin and the Merlinleigh–Byro–Irwin sub-basins of the Southern Carnarvon and northern Perth basins and had highly variable oxygen levels and salinities typical of estuarine environments. A similar pattern of warming and bathymetric change is recognized in all studied basins. During the early part of the late Artinskian cool conditions prevailed, with water temperatures 0–4 °C forming sea ice in the Merlinleigh–Byro–Irwin rift. Rapid warming during the latter part of the late Artinskian was accompanied by maximum marine flooding close to the Artinskian–Kungurian boundary. Climatic and bathymetric conditions then allowed carbonate mounds, with larger fusulines and a variety of algae, to develop in the northern part of the rift system, and Tubiphytes, conodonts, and brachiopods with Tethyan affinities to migrate into the marginal-rift basins despite the generally adverse water quality at these depositional sites. Comparison between the stratigraphic record from the East Gondwana interior rift and coeval records from Lhasa and Sibumasu indicate a similar pattern of climate change during the Carboniferous to end Cisuralian. Similar trends probably are present in Eastern Australia although there is confusion over the correlation of some units
