    Association of synaptosomal-associated protein 25 (SNAP-25) gene polymorphism with temperament and character traits in women with fibromyalgia syndrome

    Background: Synaptosomal-associated protein 25 (SNAP-25) may be contribute to the pathogenesis of fibromyalgia Syndrome (FMS) by affecting the release of neurotransmitters. Objectives: We aimed to investigate the relationship between the SNAP-25 gen (DdeI = rs1051312 and MnlI = rs3746544) polymorphism and the temperament and character traits. Methods: A total of 85 female patients diagnosed with FMS and 70 age-matched healthy female subjects were enrolled into the study. The Temperament and Character Inventory (TCI) were performed on all the patients. SNAP-25 gene polymorphism was determined in the patients group and controls group. Results: No significant difference between groups was found regarding the distribution of SNAP-25 MnlI polymorphism (p > 0.05), but it was seen that the frequency of TC genotype for DdeI gene was higher in the patients group (p < 0.05). Increased hazard avoidance was found in the patients group (p < 0.05). When TCI scores were assessed in terms of SNAP-25 gene polymorphism, no statistically significant relationship was detected between the TT, TG, GG genotypes for MnlI gen and TCI scores (p > 0.05). However, increased hazard avoidance was detected in patients with TC genotype for DdeI gene compared to patients without such genotype. Discussion: SNAP-25 might be an etiological factor in FMS pathogenesis and might affect personality traits of FMS patients by mediating neurotransmitter release

    Ciljevi: U ovoj studiji istraživalo se antibakterijsko djelovanje ojedinačne i kombinirane primjene otopina za irigaciju u slučaju kontaminacije bakterijom Enterococcus faecalis. i to 5,25-postotng natrijeva hipoklorita (NaOCl), 2-postotnog klorheksidina (CHX), 17-postotne etilendiamintetraoctene kiseline (EDTA), 3-postotnog vodikova peroksida (H 2O2 ), MTAD-a, SmearCleara ( SC) i 13,8-postotnog klorova dioksida (ClO 2 ) Materijal i metode: Nasumično je grupirano 280 jednokorijenskih ljudskih pretkutnjaka (premolara) u 26 testnih skupna i u 2 kontrolne (negativna i pozitivna) te su 24 sata inkubirane bakterijom E. faecalis, osim negativne kontrolne skupine. Testirani rezultati su sljedeći: NaOCl; CHX; ClO2 ; MTAD; SC; EDTA; H2O2; NaOCl + CHX; NaOCl + MTAD; SC + NaOCl; EDTA + NaOCl; H2O2 + NaOCl; ClO2 + CHX; CHX + MTAD; SC + CHX; EDTA + CHX; CHX + H2O2; ClO2 + MTAD; SC + ClO2; EDTA + ClO2; ClO2 + H2O2; SC + MTAD; EDTA + MTAD; MTAD + H2O2; SC + H2O2 i EDTA + H2O2. Zabilježene vrijednosti optičke gustoće su 0, 6, 12, 18, 24, 30, 36, 42 i 4 h, a za svaku otopinu stvorena je krivulja rasta bakterija.Rezultati: CHX, MTAD i ClO2 pokazali su visok potencijal smanjenja E. faecalis samostalno i u svim kombinacijama. U skupinama EDTA, H2O2, H2O2 + EDTA, H2O2 + NaOCl i SC + NaOCl postignuto je manje antibakterijsko djelovanje od onoga u ostalima. U skupini SC + CHX zabilježen je najbolji antibakterijski učinak na E. faecalis.Zaključak: Kombinacija SC + CHX može se preporučiti kao najučinkovitiji protokol za irigaciju u slučaju dugotrajnih endodontskih infekcija s E. faecalis.Objectives: This study assessed the antibacterial activity of both separate and combined uses of 5.25% sodium hypochlorite (NaOCl), 2% chlorhexidine (CHX), 17% ethylenediaminetetraacetic acid (EDTA), 3% hydrogen peroxide (H2O2), MTAD, SmearClear (SC) and 13.8% chlorine dioxide (ClO2) irrigation solutions against Enterococcus faecalis. Material and Methods: Two hundred eighty single rooted human premolars were randomly grouped into 26 test and 2 control (negative and positive) groups and were incubated for 24 h with E. faecalis, except for the negative control group. The tested solutions were as follow: NaOCl; CHX; ClO2; MTAD; SC; EDTA; H2O2; NaOCl + CHX; NaOCl + MTAD; SC + NaOCl; EDTA + NaOCl; H2O2 + NaOCl; ClO2 + CHX; CHX + MTAD; SC + CHX; EDTA + CHX; CHX + H2O2; ClO2 + MTAD; SC + ClO2; EDTA + ClO2; ClO2 + H2O2; SC+MTAD; EDTA+MTAD; MTAD + H2O2; SC + H2O2; and EDTA + H2O2. Optic density values were recorded at 0, 6, 12, 18, 24, 30, 36, 42 and 48 h and bac-terial growth curve created for each solution. Results: The CHX, MTAD and ClO2 showed a high poten-tial for the elimination of E. faecalis, both alone and in all combinations. The EDTA, H2O2, H2O2 + EDTA, H2O2 + NaOCl and SC + NaOCl groups showed less antibacterial activity than the other groups. The SC + CHX group showed the best antibacterial effect against E. faecalis. Conclusion: The SC + CHX com-bination can be recommended as the most effective irrigation regimen against E. faecalisin persis-tent endodontic infections

    Root canal hydrophobization by dentinal silanization: Improvement of silicon-based endodontic treatment tightness

    A new strategy to improve silicon-based endodontic treatment tightness by dentine hydrophobization is presented in this work: root dentine was silanized to obtain a hydrophobic dentine-sealer interface that limits fluid penetration. This strategy was based on the grafting of aliphatic carbon chains on the dentine through a silanization with the silane end groups [octadecyltrichlorosilane (OTS) and octadecyltriethoxysilane]. Dentine surface was previously pretreated, applying ethylenediaminetetraacetic acid and sodium hypochlorite, to expose hydroxyl groups of collagen for the silane grafting. Collagen fibers exposure after pretreatment was visible with scanning electron microscopy, and Fourier transform infrared (FTIR) spectroscopy showed their correct exposition for the silanization (amide I and II, with 1630, 1580, and 1538 cm⁻¹ peaks corresponding to the vibration of C=O and C--N bonds). The grafting of aliphatic carbon chains was confirmed by FTIR (peaks at 2952 and 2923 cm⁻¹ corresponding to the stretching of C--H bonds) and by the increasing of the water contact angle. The most efficient hydrophobization was obtained with OTS in ethyl acetate, with a water contact angle turning from 51° to 109°. Gas and liquid permeability tests showed an increased seal tightness after silanization: the mean gas and water flows dropped from 2.02 × 10⁻⁸ to 1.62 × 10⁻⁸ mol s⁻¹ and from 10.8 × 10⁻³ to 5.4 × 10⁻³ µL min⁻¹, respectively. These results show clear evidences to turn hydrophilic dentine surface into a hydrophobic surface that may improve endodontic sealing. Copyright © 2013 Wiley Periodicals, Inc

    Evaluation of three instrumentation techniques at the precision of apical stop and apical sealing of obturation

    OBJECTIVE: The aim of this study was to investigate the ability of two NiTi rotary apical preparation techniques used with an electronic apex locator-integrated endodontic motor and a manual technique to create an apical stop at a predetermined level (0.5 mm short of the apical foramen) in teeth with disrupted apical constriction, and to evaluate microleakage following obturation in such prepared teeth. MATERIAL AND METHODS: 85 intact human mandibular permanent incisors with single root canal were accessed and the apical constriction was disrupted using a #25 K-file. The teeth were embedded in alginate and instrumented to #40 using rotary Lightspeed or S-Apex techniques or stainless-steel K-files. Distance between the apical foramen and the created apical stop was measured to an accuracy of 0.01 mm. In another set of instrumented teeth, root canals were obturated using gutta-percha and sealer, and leakage was tested at 1 week and 3 months using a fluid filtration device. RESULTS: All techniques performed slightly short of the predetermined level. Closest preparation to the predetermined level was with the manual technique and the farthest was with S-Apex.     In vitro analysis of the cytotoxicity and the antimicrobial effect of four endodontic sealers

    <p>Abstract</p> <p>Introduction</p> <p>The aim of this study was to investigate <it>in vitro </it>the cytotoxicity and antibacterial properties of four different endodontic sealers using human periodontal ligament fibroblast cell proliferation and visual analysis of growth inhibition.</p> <p>Methods</p> <p>A silicone (GuttaFlow), silicate (EndoSequence BC), zinc oxide eugenol (Pulp Canal Sealer EWT) and epoxy resin (AH Plus Jet) based sealer were incubated with PDL fibroblasts (10<sup>4 </sup>cells/ml, n = 6) up to 96 h. Cell proliferation (RFU) was determined by means of the Alamar Blue assay. Cell growth and morphology was visualized by means of fluorescent dyes. Possible antibacterial properties of the different sealers were visualized by means of SEM (<it>Enterococcus faecalis; Parvimonas micra</it>).</p> <p>Results</p> <p>Fibroblast proliferation depended on sealer and cultivation time. After 72 and 96 h GuttaFlow and EndoSequence BC showed relatively non-cytotoxic reactions, while Pulp Canal Sealer EWT and AH Plus Jet caused a significant decrease of cell proliferation (p < 0.001). Visualization of cell growth and morphology with various fluorescent dyes supplemented the results. No antibacterial effect of EndoSequence BC to <it>P. micra </it>was found, whereas GuttaFlow showed a weak, Pulp Canal Sealer EWT and AH Plus Jet extensive growth inhibition. Also, no antibacterial effect of GuttaFlow, EndoSequence BC or AH Plus Jet to <it>E. faecalis </it>could be detected.</p> <p>Conclusions</p> <p>These <it>in vitro </it>findings reveal that GuttaFlow and EndoSequence BC can be considered as biocompatible sealing materials. However, prior to their clinical employment, studies regarding their sealing properties also need to be considered.</p

    Effects of eluted components from 4-META/MMA-TBB adhesive resin sealer on osteoblastic cell proliferation

    Background/purpose: Adhesive resin sealer systems are applied to seal root-canal systems more effectively through the formation of a resin impregnation layer. The purpose of this study was to investigate the effects of eluted components of the adhesive, Super-Bond sealer (SBS), on the proliferation of osteoblastic cells in vitro. Materials and methods: The standard powder:liquid ratio according to the manufacturer\u27s instructions was used to produce a cylindrical block of SBS (5 mm in diameter, 10 mm long) for this elution study. The resin block was placed on a 100-mm culture dish. Osteoblastic cells were seeded at a density of 4 × 10 6 in α-minimum essential medium containing 10% fetal bovine serum and cultured in a humidified incubator. After 3 days of culture with or without SBS, cells were retrieved and lysed according to the manufacturer\u27s instructions. The cellular events induced by the eluted components from SBS were analyzed using an antibody assay for mitogen-activated protein kinases (MAPKs) and a 3-(4,5-dimethylthyazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay. Furthermore, the concentration of boron, a component of the catalyst, tri-n-butyl borane (TBB), was analyzed using inductively coupled plasma optical emission spectrometry. Results: Expression of MAPKs increased after SBS application. The MTT assay indicated that TBB, one of the components of SBS, accelerated the proliferation of osteoblastic cells. Values of boron were 1.66 ± 0.37 and 1.74 ± 0.30 ppm in cells cultured with and without FBS, respectively. Conclusions: The eluted components from SBS can increase the expression of some MAPKs related to osteoblastic cell proliferation and differentiation in vitro. Both the elution experiment and treatment of cell culture with SBS components indicated that the boron originating from TBB is likely to be responsible for activation of the proliferation of osteoblastic cells

    Analysis of the common genetic component of large-vessel vasculitides through a meta- Immunochip strategy

    Giant cell arteritis (GCA) and Takayasu's arteritis (TAK) are major forms of large-vessel vasculitis (LVV) that share clinical features. To evaluate their genetic similarities, we analysed Immunochip genotyping data from 1,434 LVV patients and 3,814 unaffected controls. Genetic pleiotropy was also estimated. The HLA region harboured the main disease-specific associations. GCA was mostly associated with class II genes (HLA-DRB1/HLA-DQA1) whereas TAK was mostly associated with class I genes (HLA-B/MICA). Both the statistical significance and effect size of the HLA signals were considerably reduced in the cross-disease meta-analysis in comparison with the analysis of GCA and TAK separately. Consequently, no significant genetic correlation between these two diseases was observed when HLA variants were tested. Outside the HLA region, only one polymorphism located nearby the IL12B gene surpassed the study-wide significance threshold in the meta-analysis of the discovery datasets (rs755374, P?=?7.54E-07; ORGCA?=?1.19, ORTAK?=?1.50). This marker was confirmed as novel GCA risk factor using four additional cohorts (PGCA?=?5.52E-04, ORGCA?=?1.16). Taken together, our results provide evidence of strong genetic differences between GCA and TAK in the HLA. Outside this region, common susceptibility factors were suggested, especially within the IL12B locus
