    Culture portability from origin to destination country: The gender division of domestic work among migrants in Italy

    Background: The relative importance of a cultural component in the gender division of unpaid labour is still debated. Objective: Drawing on the epidemiological approach to the study of culture, we investigate the cultural component of gender inequality by examining whether gender asymmetries in housework and childcare in families with a migrant background relate to gender equity in the country of origin. Methods: Through multilevel models based on microlevel data (Istat SCIF survey), we examine the extent to which the division of household labour between immigrant partners living in Italy relates to gender equity in their origin country, proxied by the Global Gender Gap Index. We further analyse the changing importance of gender equity in the country of origin at different lengths of stay in the destination country. Conclusions: Immigrants from more (less) gender-equal countries display greater (lower) equality in the division of routine housework and childcare activities. However, gender equity in the origin country loses its importance for couples living in the destination country for a longer time. These findings point to a significant contribution of culture of origin to gender inequality in the intra-couple division of unpaid labour. Yet nonnegligible differences exist between specific housework and childcare tasks and depending on the time spent in the hosting country.     Cutting edge: IgE plays an active role in tumor immunosurveillance in mice

    Exogenous IgE acts as an adjuvant in tumor vaccination in mice, and therefore a direct role of endogenous IgE in tumor immunosurveillance was investigated. By using genetically engineered mice, we found that IgE ablation rendered mice more susceptible to the growth of transplantable tumors. Conversely, a strengthened IgE response provided mice with partial or complete resistance to tumor growth, depending on the tumor type. By genetic crosses, we showed that IgE-mediated tumor protection was mostly lost in mice lacking FceRI. Tumor protection was also lost after depletion of CD8+ T cells, highlighting a cross-Talk between IgE and T cell- mediated tumor immunosurveillance. Our findings provide the rationale for clinical observations that relate atopy with a lower risk for developing cancer and open new avenues for the design of immunotherapeutics relevant for clinical oncology.     Interaction Between Dietary Lipid Level and Seasonal Temperature Changes in Gilthead Sea Bream Sparus aurata: Effects on Growth, Fat Deposition, Plasma Biochemistry, Digestive Enzyme Activity, and Gut Bacterial Community

    A 121-day feeding trial was undertaken to test the effects of two dietary lipid levels (16 and 21% L16, L21) in triplicated gilthead sea bream groups (initial weight: 67.5 g) reared at two different water temperatures (high, H 23°C and low, L 17°C) in the same recirculation system but exposed to a switch in temperature after 58 days. Fish kept at H were transferred to L (HL transition, autumn shift), and the fish kept at L were exposed to H (LH transition, summer shift), while continuing to receive the same diet to apparent satiation in each group. At the end of the trial, no significant diet effect on specific growth rate (SGR), feed intake (FI), and feed conversion rate (FCR) were detected in fish exposed to HL transition compared with those exposed to LH transition, while gross lipid efficiency (GLE) and lipid efficiency ratio (LER) were higher in L16. After temperature changes, L16 displayed higher SGR, FI, GLE, and LER, while mesenteric fat index was reduced. After temperature changes, the combined effects of low lipid diet and low temperature conditions resulted in higher pepsin activity, while trypsin, chymotrypsin, and lipase activities were generally higher at high lipid content. The combined effect of diet and temperature did not alter the metabolic plasma profile, except for the observed final higher aspartate aminotransferase (AST) and alkaline phosphatase (ALP) values when combining high dietary lipid (L21) and temperature changes. Different diets showed a significantly different gut microbiome layout, only at high temperature with L16 diet resulting in a higher load of Lactobacillus. On the contrary, no dietary impact on ecosystem diversity was observed, independently from the temperature. In addition, L16 diet in the HL transition favored an increase in Weissella and Bradyrhizobium genera in the gut microbiome, while in the final condition of LH transition, L21 diet favored a significant increase in Streptococcus and Bacillus.     Protein synthesis inhibition and loss of homeostatic functions in astrocytes from an Alzheimer's disease mouse model: a role for ER-mitochondria interaction

    Deregulation of protein synthesis and ER stress/unfolded protein response (ER stress/UPR) have been reported in astrocytes. However, the relationships between protein synthesis deregulation and ER stress/UPR, as well as their role in the altered homeostatic support of Alzheimer's disease (AD) astrocytes remain poorly understood. Previously, we reported that in astrocytic cell lines from 3xTg-AD mice (3Tg-iAstro) protein synthesis was impaired and ER-mitochondria distance was reduced. Here we show that impaired protein synthesis in 3Tg-iAstro is associated with an increase of p-eIF2α and downregulation of GADD34. Although mRNA levels of ER stress/UPR markers were increased two-three-fold, we found neither activation of PERK nor downstream induction of ATF4 protein. Strikingly, the overexpression of a synthetic ER-mitochondrial linker (EML) resulted in a reduced protein synthesis and augmented p-eIF2α without any effect on ER stress/UPR marker genes. In vivo, in hippocampi of 3xTg-AD mice, reduced protein synthesis, increased p-eIF2α and downregulated GADD34 protein were found, while no increase of p-PERK or ATF4 proteins was observed, suggesting that in AD astrocytes, both in vitro and in vivo, phosphorylation of eIF2α and impairment of protein synthesis are PERK-independent. Next, we investigated the ability of 3xTg-AD astrocytes to support metabolism and function of other cells of the central nervous system. Astrocyte-conditioned medium (ACM) from 3Tg-iAstro cells significantly reduced protein synthesis rate in primary hippocampal neurons. When added as a part of pericyte/endothelial cell (EC)/astrocyte 3D co-culture, 3Tg-iAstro, but not WT-iAstro, severely impaired formation and ramification of tubules, the effect, replicated by EML overexpression in WT-iAstro cells. Finally, a chemical chaperone 4-phenylbutyric acid (4-PBA) rescued protein synthesis, p-eIF2α levels in 3Tg-iAstro cells and tubulogenesis in pericyte/EC/3Tg-iAstro co-culture. Collectively, our results suggest that a PERK-independent, p-eIF2α-associated impairment of protein synthesis compromises astrocytic homeostatic functions, and this may be caused by the altered ER-mitochondria interaction

    Estimation of the allergenic potential of urban trees and urban parks: Towards the healthy design of urban green spaces of the future

    The impact of allergens emitted by urban green spaces on health is one of the main disservices of ecosystems. The objective of this work is to establish the potential allergenic value of some tree species in urban environments, so that the allergenicity of green spaces can be estimated through application of the Index of Urban Green Zones Allergenicity (IUGZA). Multiple types of green spaces in Mediterranean cities were selected for the estimation of IUGZ. The results show that some of the ornamental species native to the Mediterranean are among the main causative agents of allergy in the population; in particular, Oleaceae, Cupressaceae, Fagaceae, and Platanus hispanica. Variables of the strongest impact on IUGZA were the bioclimatic characteristics of the territory and design aspects, such as the density of trees and the number of species. We concluded that the methodology to assess the allergenicity associated with urban trees and urban areas presented in this work opens new perspectives in the design and planning of urban green spaces, pointing out the need to consider the potential allergenicity of a species when selecting plant material to be used in cities.     Na+, K+-ATPase Subunit Composition in a Human Chondrocyte Cell Line; Evidence for the Presence of α1, α3, ÎČ1, ÎČ2 and ÎČ3 Isoforms

    Membrane transport systems participate in fundamental activities such as cell cycle control, proliferation, survival, volume regulation, pH maintenance and regulation of extracellular matrix synthesis. Multiple isoforms of Na+, K+-ATPase are expressed in primary chondrocytes. Some of these isoforms have previously been reported to be expressed exclusively in electrically excitable cells (i.e., cardiomyocytes and neurons). Studying the distribution of Na+, K+-ATPase isoforms in chondrocytes makes it possible to document the diversity of isozyme pairing and to clarify issues concerning Na+, K+-ATPase isoform abundance and the physiological relevance of their expression. In this study, we investigated the expression of Na+, K+-ATPase in a human chondrocyte cell line (C-20/A4) using a combination of immunological and biochemical techniques. A panel of well-characterized antibodies revealed abundant expression of the α1, ÎČ1 and ÎČ2 isoforms. Western blot analysis of plasma membranes confirmed the above findings. Na+, K+-ATPase consists of multiple isozyme variants that endow chondrocytes with additional homeostatic control capabilities. In terms of Na+, K+-ATPase expression, the C-20/A4 cell line is phenotypically similar to primary and in situ chondrocytes. However, unlike freshly isolated chondrocytes, C-20/A4 cells are an easily accessible and convenient in vitro model for the study of Na+, K+-ATPase expression and regulation in chondrocytes

    With or without you. Intentions, Constraints and Consequences of Childlessness in European Countries.

    This dissertation focuses on the experience of childlessness, which is an increasing one across European Countries. Namely, it deals with the socio-economic drivers and consequences that distinguish the life of childless individuals from that of parents. Mirroring each phase of a hypothetical individual lifecycle, it starts by analysing how intentions to be childless are later realised their realizations (Chapter 2). It then goes on to investigate the individual and contextual determinants of childlessness and how they have changed over time (Chapter 3 and Chapter 4). Last it considers the consequences that childlessness might induce in adulthood and later life (Chapter 5). This is done by combining demographic and sociological perspectives in a joint interpretative framework. Overall the thesis shows that both economic contraints and individual values has a role in determining the choice of a childless life, and highlights how these decisions are taken in contexts where a purality of macro factors interacts in moderating the indivdidual risk of being childless. Understanding the drives to childlessness is relevant also when interpreting the potential effects that being childless has on the life of individuals. All in all, the study shows that the consequences of being childless also depend on how people came to be without children

    Childlessness and low fertility in context: evidence from a multilevel analysis on 20 European countries

    Abstract Low fertility and childlessness have been largely interpreted as being driven by the same mechanisms, although they may be qualitatively different phenomena. The present article discusses this assumption and studies determinants of childlessness by comparing them with determinants of low fertility. Drawing on micro-level data from the European Labour Force Survey (2005–2010) and macro-level data from external data sources, it enters the debate on the micro- and macro-determinants of fertility and childlessness by analysing women who live with or without children when they are 35–39 years old. By means of a series of multilevel models, it discusses the moderating role of the institutional and normative context in the link between individual characteristics and childlessness, and it analyses the extent to which micro- and macro-level determinants of childlessness differ from factors associated with low-fertility. The results indicate that the macro-level factors associated with individual childlessness are similar to those identified in the literature on low fertility. However, interesting differences emerge in how reconciliation and family support policies relate on childlessness and low-fertility across different groups of women. Overall, these findings highlight the importance to study childlessness as a phenomenon on its own, distinguishing its determinants from determinants of (low) fertility

    Formal childcare services and fertility: the case of Italy

    Abstract This study aims at examining the fertility impact of early childhood education and care (ECEC) services for children under three in Italy. ECEC is a social investment-oriented family policy that might have more beneficial effects on fertility than passive support in terms of transfers. We first present a systematic review of studies regarding the impact of ECEC on fertility in high-income countries and then provide an empirical analysis for Italy, a country with lowest-low fertility rates and a welfare system that has traditionally provided limited support to families, especially through ECEC. We combine micro-level data from the Labour Force Survey for Italy from years 2003–2020 with information on regional indicators of public childcare and private childcare. The study employs within-region variation in ECEC over time to assess its effects on the transition to parenthood and parity progression for different groups of women and men. The present contribution indicates that both public and private childcare services have limited but positive short-term effects on fertility behaviour in Italy, contingent on specific socio-demographic groups. We discuss the possible reasons for the constrained effect of childcare on fertility and emphasise the necessity for more substantial and concerted interventions in Italy’s family policies if the aim is to invert demographic trends of lowest fertility and population-ageing
