    Further analysis of previously implicated linkage regions for Alzheimer's disease in affected relative pairs

    Background Genome-wide linkage studies for Alzheimer's disease have implicated several chromosomal regions as potential loci for susceptibility genes. Methods In the present study, we have combined a selection of affected relative pairs (ARPs) from the UK and the USA included in a previous linkage study by Myers et al. (Am J Med Genet, 2002), with ARPs from Sweden and Washington University. In this total sample collection of 397 ARPs, we have analyzed linkage to chromosomes 1, 9, 10, 12, 19 and 21, implicated in the previous scan. Results The analysis revealed that linkage to chromosome 19q13 close to the APOE locus increased considerably as compared to the earlier scan. However, linkage to chromosome 10q21, which provided the strongest linkage in the previous scan could not be detected. Conclusion The present investigation provides yet further evidence that 19q13 is the only chromosomal region consistently linked to Alzheimer's disease

    Homocysteine and Familial Longevity: The Leiden Longevity Study

    Homocysteine concentrations are a read-out of methionine metabolism and have been related to changes in lifespan in animal models. In humans, high homocysteine concentrations are an important predictor of age related disease. We aimed to explore the association of homocysteine with familial longevity by testing whether homocysteine is lower in individuals that are genetically enriched for longevity. We measured concentrations of total homocysteine in 1907 subjects from the Leiden Longevity Study consisting of 1309 offspring of nonagenarian siblings, who are enriched with familial factors promoting longevity, and 598 partners thereof as population controls. We found that homocysteine was related to age, creatinine, folate, vitamin B levels and medical history of hypertension and stroke in both groups (all p<0.001). However, levels of homocysteine did not differ between offspring enriched for longevity and their partners, and no differences in the age-related rise in homocysteine levels were found between groups (p for interaction 0.63). The results suggest that homocysteine metabolism is not likely to predict familial longevity

    Bidialectalism and bilingualism: Exploring the role of language similarity as a link between linguistic ability and executive control

    The notion of bilingual advantages in executive functions (EF) is based on the assumption that the demands posed by cross-language interference serve as EF training. These training effects should be more pronounced the more cross-language interference bilinguals have to overcome when managing their two languages. In the present study, we investigated the proposed link between linguistic and EF performance using the similarity between the two languages spoken since childhood as a proxy for different levels of cross-language interference. We assessed the effect of linearly increasing language dissimilarity on linguistic and EF performance in multiple tasks in four groups of young adults (aged 18–33): German monolinguals (n = 24), bidialectals (n = 25; German and Swiss German dialect), bilinguals speaking two languages of the same Indo-European ancestry (n = 24; e.g., German-English), or bilinguals speaking two languages of different ancestry (n = 24; e.g., German-Turkish). Bayesian linear-mixed effects modeling revealed substantial evidence for a linear effect of language similarity on linguistic accuracy, with better performance for participants with more similar languages and monolinguals. However, we did not obtain evidence for the presence of a similarity effect on EF performance. Furthermore, language experience did not modulate EF performance, even when testing the effect of continuous indicators of bilingualism (e.g., age of acquisition, proficiency, daily foreign language usage). These findings question the theoretical assumption that life-long experience in managing cross-language interference serves as EF training

    Critical success factors for governing farmer-managed public goods in rural areas in the Netherlands

    Multifunctional land use has become a widely supported pathway for Europe's countryside. Brussels and the national governments stimulate farmers to integrate primary production with non-agricultural practices from which they can also benefit. In favour of this development different stakeholders are encouraged to collaborate to produce the so-called farmer-managed public goods. This paper explores critical success factors for the production and maintenance of these public goods. In two Dutch case study regions we show a chronology of major events from 1975 leading to a shift from primary agricultural production towards a successful multifunctional land use. We evaluated which critical success factors crucial for managing public goods manifest themselves in our case study regions. Besides six critical success factors described in literature (Property rights, Asymmetric information, Administrative complexity, Governance, Time lags and Market structure) four other factors appear to be crucial in governing farmer-managed public goods: (1) establishing a system of rewards; (2) a mix of governance forms; (3) visionary leadership with networks in both the public and the private sectors and (4) time for new ideas to mature and to build commitment among the actors.</p

    Mycobacterium avium (M. a.) subsp. paratuberculosis (MAP) is a worldwide-distributed obligate pathogen in ruminants causing Johne&rsquo;s disease. Due to a lack of complete subtype III genome sequences, there is not yet conclusive information about genetic differences between strains of cattle (MAP-C, type II) and sheep (MAP-S) type, and especially between MAP-S subtypes I, and III. Here we present the complete, circular genome of MAP-S/type III strain JIII-386 (DE) closed by Nanopore-technology and its comparison with MAP-S/type I closed genome of strain Telford (AUS), MAP-S/type III draft genome of strain S397 (U.S.), twelve closed MAP-C strains, and eight closed M.-a.-complex-strains. Structural comparative alignments revealed clearly the mosaic nature of MAP, emphasized differences between the subtypes and the higher diversity of MAP-S genomes. The comparison of various genomic elements including transposases and genomic islands provide new insights in MAP genomics. MAP type specific phenotypic features may be attributed to genes of known large sequence polymorphisms (LSPSs) regions I&ndash;IV and deletions #1 and #2, confirmed here, but could also result from identified frameshifts or interruptions of various virulence-associated genes (e.g., mbtC in MAP-S). Comprehensive core and pan genome analysis uncovered unique genes (e.g., cytochromes) and genes probably acquired by horizontal gene transfer in different MAP-types and subtypes, but also emphasized the highly conserved and close relationship, and the complex evolution of M.-a.-strains
