    The relationship between epilithic biofilm stability and its associated meiofauna under two patterns of flood disturbance

    Habitat stability is an important driver of ecological community composition and development. River epilithic biofilms are particularly unstable habitats for the establishment of benthic communities because they are regularly disturbed by floods. Our aim was to determine the influence of habitat instability on meiobenthic organisms. We hypothesized that hydrologic variables are the most important predictors of meiofauna distribution. We monitored epilithic communities (meiofauna and microalgae) with a high sampling frequency during 2 sampling periods with contrasting hydrodynamic patterns in a temperate river (the Garonne, France). Nematodes and rotifers dominated meiofaunal assemblages. The critical flow velocity threshold for their maintenance in the biofilm was ,30 cm/s, a result suggesting that meiofauna can resist higher flow velocity within the biofilm than within sediments. Nematode distribution was primarily influenced by the duration of undisturbed periods, whereas rotifer distribution was also correlated with the thickness of the biofilm. During the periods after floods, rotifers were faster colonizers than nematodes. Collectively, our results show that flow regime was an essential driver for biofilm community development

    Dynamics of reactive distillation for the production of ethyl acetate: experiments at a pilot plant and modelling

    In order to understand the complex behaviour of the reactive distillation process and to be able to provide an accurate design of a reactive column, detailed analyses on both continuous and transient regime become necessary. The objective is the definition of a reliable simulation model, based on experimental data obtained from a real pilot-scale plant device for the heterogeneously catalysed esterification of acetic acid and ethanol to form ethyl acetate and water. The choice of the parameters for the continuous equilibrium model was discussed and the simulation results provided good agreement with experimental data, revealing an interesting sensitivity of the catalyst activity to the feed composition. Once column configuration and operational parameters were validated, dynamic experiments were realized so as to interpret the sensitivity of different disturbances. Feed flow rates, reflux ratio and heat duty were perturbed and the consequent open loop transient responses were identified. The assessment of hydrodynamic parameters and the validation of the transient data allow the definition of a reliable dynamic model that represents tendencies and behaviours of the process well.     Fate and Biological Activity of the Antimicrobial Lasso Peptide Microcin J25 Under Gastrointestinal Tract Conditions

    The bacteriocin microcin J25 (MccJ25) inhibits the growth of Gram-negative pathogens including Salmonella and Shigella species, and Escherichia coli. This 21-amino acid peptide has remarkable stability to heat and extreme pH values and resistance to many proteases, thanks to a characteristic lasso structure. In this study, we used the dynamic simulator TIM-1 as gastro-intestinal tract model to evaluate the stability and antibacterial activity of MccJ25 during passage through the proximal portion of the human gastrointestinal tract. MccJ25 concentration was measured in the different simulator sections by HPLC, and inhibition of Salmonella enterica serotype Enteritidis was evaluated using qualitative and quantitative assays. LC-MS/MS analysis and subsequent molecular networking analysis on the Global Natural Product Social Molecular Networking platform (GNPS) and analysis of the peptide degradation in the presence of proteolytic enzymes mimicking the gastro-intestinal conditions permitted to delineate the fate of MccJ25 through identification of the main degradation products. MccJ25 was relatively stable under gastric conditions, but degraded rapidly in the compartment mimicking the duodenum, notably in the presence of pancreatin. Among pancreatin components, elastase I appeared primarily responsible for MccJ25 breakdown, while α-chymotrypsin was less efficient

    Cytotoxicity of Pyrazine-Based Cyclometalated (C^Npz^C)Au(III) Carbene Complexes: Impact of the Nature of the Ancillary Ligand on the Biological Properties

    The synthesis of a series of cyclometalated gold(III) complexes supported by pyrazine-based (C^N^C)-type pincer ligands is reported, including the crystal structure of a cationic example. The compounds provide a new platform for the study of antiproliferative properties of gold(III) complexes. Seven complexes were tested: the neutral series (C^Npz^C)AuX [X = Cl (1), 6-thioguanine (4), C≡CPh (5), SPh (6)] and an ionic series that included the N-methyl complex [(C^NpzMe^C)AuCl]BF4 (7) and the N-heterocyclic carbene complexes [(C^Npz^C)AuL]+ with L = 1,3-dimethylbenzimidazol-2-ylidene (2) or 1,3,7,9-tetramethylxanthin-8-ylidene (3). Tests against human leukemia cells identified 1, 2, 3, and 4 as particularly promising, whereas protecting the noncoordinated N atom on the pyrazine ring by methylation (as in 7) reduced the cytotoxicity. Complex 2 proved to be the most effective of the entire series against the HL60 leukemia, MCF-7 breast cancer, and A549 lung cancer cell lines, with IC50 values down to submicromolar levels, associated with a lower toxicity toward healthy human lung fibroblast cells. The benzimidazolylidene complex 2 accumulated more effectively in human lung cancer cells than its caffeine-based analogue 3 and the gold(III) chloride 1. Compound 2 proved to be unaffected by glutathione under physiological conditions for periods of up to 6 days and stabilizes the DNA G-quadruplex and i-motif structures; the latter is the first such report for gold compounds. We also show the first evidence of inhibition of MDM2–p53 protein–protein interactions by a gold-based compound and identified the binding mode of the compound with MDM2 using saturation transfer difference NMR spectroscopy combined with docking calculations

    Great effort has been applied to model and simulate the dynamic behavior of the reactive distillation as a successfulprocess intensification example. However, very little experimental work has been carried out in transient conditions.The work presents a series of experiments for the production of ethyl acetate from esterification of acetic acid andethanol in a reactive distillation pilot column. The steady-state approach performed experiments with both excessof alcohol and stoichiometric feed configuration. Predicted and measured results show good agreement and reveala strong dependency of the structured packing catalyst activity on the pilot geometry and its operating conditions.The transient process behavior of the heterogeneously catalyzed system was deeply investigated and continuousand dynamic data were collected for an equilibrium model validation, after different perturbations on parameters.The experimental validation is shown to be essential to provide realistic hydrodynamic parameters, to understandthe sensitive parameters such as heat losses and to adapt values for the catalyst holdup as a function of the system

    Exploring a Tetrahydroquinoline Antimalarial Hit from the Medicines for Malaria Pathogen Box and Identification of its Mode of Resistance as PfeEF2

    New antimalarial treatments with novel mechanism of action are needed to tackle Plasmodium falciparum infections that are resistant to first-line therapeutics. Here we report the exploration of MMV692140 ( 2 ) from the Pathogen Box, a collection of 400 compounds that was made available by Medicines for Malaria Venture (MMV) in 2015. Compound 2 was profiled in in vitro models of malaria and was found to be active against multiple life-cycle stages of Plasmodium parasites. The mode of resistance, and putatively its mode of action, was identified as Plasmodium falciparum translation elongation factor 2 ( Pf eEF2), which is responsible for the GTP-dependent translocation of the ribosome along mRNA. The compound maintains activity against a series of drug-resistant parasite strains. The structural motif of the tetrahydroquinoline ( 2 ) was explored in a chemistry program with its structure-activity relationships examined, resulting in the identification of an analog with 30-fold improvement of antimalarial asexual blood stage potency

    Acridine-decorated cyclometallated gold(III) complexes: synthesis and anti-tumour investigations

    (C^N) and (C^N^C) cyclometalated Au(III) represent a highly promising class of potential anticancer agents. We report here the synthesis of seven new cyclometalated Au(III) complexes with five of them bearing an acridine moiety attached via (N^O) or (N^N) chelates, acyclic amino carbenes (AAC) and N-heterocyclic carbenes (NHC). The antiproliferative properties of the different complexes were evaluated in vitro on a panel of cancer cells including leukaemia, lung and breast cancer cells. We observed a trend between the cytotoxicity and the intracellular gold uptake of some representative compounds of the series. Some of the acridine-decorated complexes were demonstrated to interact with ds-DNA using FRET-melting techniques

    Highlighting a New Morphospecies within the Dialium Genus Using Leaves and Wood Traits

    peer reviewedDuring inventories of lesser-known timber species in eastern Gabon, a new Dialium morphospecies was discovered. To discriminate it from the two other 2–5 leaflets Dialium species, 25 leaf traits were measured on 45 trees (16 Dialium pachyphyllum, 14 Dialium lopense, 15 Dialium sp. nov.). Nine wood chemical traits, as well as infrared spectra, were also examined on harvestable trees (four Dialium pachyphyllum and four Dialium sp. nov.). This study revealed seven discriminant leaf traits that allowed to create a field identification key. Nine significant differences (five in sapwood and four in heartwood) in terms of wood composition were highlighted. The use of the PLS-DA technique on FT-IR wood spectra allowed to accurately identify the new morphospecies. These results provide strong support for describing a new species in this genus.
