Accordingly, we divide the sample of local and regional food marketers into quartiles segmented by profitability performance as an initial exploration of how strong and weak performance may vary across scale, location, and choice of direct and intermediated channels. Moreover, other financial metrics that vary across types of producers and performance-based quartiles are analyzed.     Post-Occupancy Evaluation and IEQ Measurements from 64 Office Buildings: Critical Factors and Thresholds for User Satisfaction on Thermal Quality

    The indoor environmental quality (IEQ) of buildings can have a strong influence on occupantsā€™ comfort, productivity, and health. Post-occupancy evaluation (POE) is necessary in assessing the IEQ of the built environment, and it typically relies on the subjective surveys of thermal quality, air quality, visual quality, and acoustic quality. In this research, we expanded POE to include both objective IEQ measurements and the technical attributes of building systems (TABS) that may affect indoor environment and user satisfaction. The suite of three tools, including user satisfaction survey, workstation IEQ measurements, and TABS in the National Environmental Assessment Toolkit (NEAT) has been deployed in 1601 workstations in 64 office buildings, generating a rich database for statistical evaluation of possible correlations between the physical attributes of workstations, environmental conditions, and user satisfaction. Multivariate regression and multiple correlation coefficient statistical analysis revealed the relationship between measured and perceived IEQ indices, interdependencies between IEQ indices, and other satisfaction variables of significance. The results showed that overall, 55% of occupants responded as ā€œsatisfiedā€ or ā€œneutralā€, and 45% reported being ā€œdissatisfiedā€ in their thermal quality. Given the dataset, air temperature in work area, size of thermal zone, window quality, level of temperature control, and radiant temperature asymmetry with faƧade are the critical factors for thermal quality satisfaction in the field. As a result, the outcome of this research contributes to identifying correlations between occupant satisfaction, measured data, and technical attributes of building systems. The presented integrated IEQ assessment method can further afford robust predictions of building performance against metrics and guidelines for IEQ standards to capture revised IEQ thresholds that impact building occupantsā€™ satisfaction.</jats:p

    Non-invasive Eye Tracking Methods for New World and Old World Monkeys

    Eye-tracking methods measure what humans and other animals visually attend to in the environment. In nonhuman primates, eye tracking can be used to test hypotheses about how primates process social information. This information can further our understanding of primate behavior as well as offer unique translational potential to explore causes of or treatments for altered social processing as seen in people with neurodevelopmental disorders such as autism spectrum disorder and schizophrenia. However, previous methods for collecting eye-tracking data in nonhuman primates required some form of head restraint, which limits the opportunities for research with respect to the number of or kinds of primates that can undergo an eye-tracking study. We developed a novel, noninvasive method for collecting eye tracking data that can be used both in animals that are difficult to restrain without sedation as well as animals that are of different ages and sizes as the box size can be adjusted. Using a transport box modified with a viewing window, we collected eye-tracking data in both New (Callicebus cupreus) and Old World monkeys (Macaca mulatta) across multiple developmental time points. These monkeys had the option to move around the box and avert their eyes from the screen, yet, they demonstrated a natural interest in viewing species-specific imagery with no previous habituation to the eye-tracking paradigm. Provided with opportunistic data from voluntary viewing of stimuli, we found that juveniles viewed stimuli more than other age groups, videos were viewed more than static photo imagery, and that monkeys increased their viewing time when presented with multiple eye tracking sessions. This noninvasive approach opens new opportunities to integrate eye-tracking studies into nonhuman primate research

    Nanostructured Pt(NH3)4Cl2/SiO2 for nanomedicine: catalytic degradation of DNA in cancer cells

    In vivo suppression of glioblastoma multiforme (GBM) in Wistar rats using silica-shelled biocatalytic Pt(NH3)4Cl2 nanoparticles is reported. These nanoparticles were synthesized by a sol-gel technique and characterized by SEM and HRTEM imaging. We confirmed morphological uniformity (30 nm) and surface acidity of the nanoparticles, respectively, by TEM imaging and FTIR spectral analysis. Interestingly, treatment of Wistar rats intraperitoneally inoculated with C6 cells using the biocatalysts resulted in considerable tumor shrinkage. Efficiency of the biocatalyst to shrink a tumor is superior to that by the commercial cytotoxic agent cisplatin. The tumor suppression property of Pt(NH3)4Cl2 nanoparticles is attributed to catalytic damage of DNA in C6 cells

    Methionine Antagonizes para-Aminosalicylic Acid Activity via Affecting Folate Precursor Biosynthesis in Mycobacterium tuberculosis

    para-Aminosalicylic acid (PAS) is a second-line anti-tubercular drug that is used for the treatment of drug-resistant tuberculosis (TB). PAS efficacy in the treatment of TB is limited by its lower potency against Mycobacterium tuberculosis relative to many other drugs in the TB treatment arsenal. It is known that intrinsic metabolites, such as, para-aminobenzoic acid (PABA) and methionine, antagonize PAS and structurally related anti-folate drugs. While the basis for PABA-mediated antagonism of anti-folates is understood, the mechanism for methionine-based antagonism remains undefined. In the present study, we used both targeted and untargeted approaches to identify factors associated with methionine-mediated antagonism of PAS activity. We found that synthesis of folate precursors as well as a putative amino acid transporter, designated MetM, play crucial roles in this process. Disruption of metM by transposon insertion resulted in a ā‰„30-fold decrease in uptake of methionine in M. bovis BCG, indicating that metM is the major facilitator of methionine transport. We also discovered that intracellular biotin confers intrinsic PAS resistance in a methionine-independent manner. Collectively, our results demonstrate that methionine-mediated antagonism of anti-folate drugs occurs through sustained production of folate precursors

    Age at menarche and current substance use among Canadian adolescent girls: results of a cross-sectional study

    <p>Abstract</p> <p>Background</p> <p>Substance use is among the key public health threats that find its genesis during adolescence. Timing of puberty has been lately researched as a potential predictor of subsequent substance abuse. The present study, therefore, aims to assess the effect of age at menarche on current practices of smoking, alcohol drinking and drug use among 14-15 year old Canadian girls.</p> <p>Methods</p> <p>The analysis of the study was based on all female respondents aged 14 to 15 years during Cycle 4 (2000/2001) of the National Longitudinal Survey of Children & Youth (NLSCY). The main independent variable was age at menarche assessed as the month and year of the occurrence of the first menstrual cycle. The dependent variables were current smoking, heavy alcohol drinking in the past 12 months and drug use in the past 12 months. Three logistic regression models were performed to investigate the association between age at menarche and each of the substance use outcomes, adjusting for possible confounders. Bootstrapping was performed to account for the complex sampling design.</p> <p>Results</p> <p>The total weighted sample included in the analysis represented 295,042 Canadian girls. The prevalence of current smokers, heavy drinkers (drunk in the past 12 months) and drug users in the past 12 months was approximately 22%, 38% and 26%, respectively. After adjusting of all potential confounders, no association was found between age at menarche and any of the substance use outcomes. School performance and relationship with the father, however, stood out as the main variables to be associated with smoking, heavy drinking and drug use.</p> <p>Conclusions</p> <p>Qualitative studies understanding the social and psychological changes experienced by early maturing Canadian adolescents are warranted to identify other correlates or pathways to substance use in this higher risk population.</p

    Alcohol expectancies in childhood: Change with the onset of drinking and ability to predict adolescent drunkenness and binge drinking

    Aims: Childhood expectancies about alcohol are present long before drinking begins. We examined the relationship between alcohol expectancies in childhood and onset of drinking, binge drinking, and drunkenness in adolescence and the influence of drinking onset on development of alcohol expectancies. Design: A prospective, longitudinal study of children assessed for alcohol expectancies and drinking at 4 time points between ages 6 and 17. Setting: Community study of families at high risk for alcoholism conducted in a 4-county area in the Midwest. Participants: The study involved 614 children; 460 were children of alcoholics and 70% were male. Measurements: Expectancies about effects of alcohol were measured using the Beverage Opinion Questionnaire and childā€™s drinking was measured using the Drinking and Drug History - Youth Form. Findings: Partial factor invariance was found for expectancy factors from age 6 to age 17. Survival analysis showed that social/relaxation expectancies in childhood predicted time to onset of binge drinking and first time drunk (Wald chi-square, 1 d.f. = 3.8, p < .05 and 5.1, p < .05, respectively). The reciprocal effect was also present; when adolescents began drinking, there was an increase in social/relaxation expectancy and a concomitant increase in slope of the expectancy change lasting throughout adolescence. Conclusions: A reciprocal relationship exists between childhood alcohol expectancies and involvement with alcohol. Higher expectancies for positive effects predict earlier onset of problem drinking.     Abundance and Distribution of Enteric Bacteria and Viruses in Coastal and Estuarine Sedimentsā€”a Review

    The long term survival of fecal indicator organisms (FIOs) and human pathogenic microorganisms in sediments is important from a water quality, human health and ecological perspective. Typically, both bacteria and viruses strongly associate with particulate matter present in freshwater, estuarine and marine environments. This association tends to be stronger in finer textured sediments and is strongly influenced by the type and quantity of clay minerals and organic matter present. Binding to particle surfaces promotes the persistence of bacteria in the environment by offering physical and chemical protection from biotic and abiotic stresses. How bacterial and viral viability and pathogenicity is influenced by surface attachment requires further study. Typically, long-term association with surfaces including sediments induces bacteria to enter a viable-but-non-culturable (VBNC) state. Inherent methodological challenges of quantifying VBNC bacteria may lead to the frequent under-reporting of their abundance in sediments. The implications of this in a quantitative risk assessment context remain unclear. Similarly, sediments can harbor significant amounts of enteric viruses, however, the factors regulating their persistence remains poorly understood. Quantification of viruses in sediment remains problematic due to our poor ability to recover intact viral particles from sediment surfaces (typically <10%), our inability to distinguish between infective and damaged (non-infective) viral particles, aggregation of viral particles, and inhibition during qPCR. This suggests that the true viral titre in sediments may be being vastly underestimated. In turn, this is limiting our ability to understand the fate and transport of viruses in sediments. Model systems (e.g., human cell culture) are also lacking for some key viruses, preventing our ability to evaluate the infectivity of viruses recovered from sediments (e.g., norovirus). The release of particle-bound bacteria and viruses into the water column during sediment resuspension also represents a risk to water quality. In conclusion, our poor process level understanding of viral/bacterial-sediment interactions combined with methodological challenges is limiting the accurate source apportionment and quantitative microbial risk assessment for pathogenic organisms associated with sediments in aquatic environments

    A Reappraisal of Childrenā€™s ā€˜Potentialā€™

    What does it mean for a child to fulfil his or her potential? This article explores the contexts and implications of the much-used concept of potential in educational discourses. We claim that many of the popular, political and educational uses of the term in relation to childhood have a problematic blind spot: interpersonality, and the necessary coexistence for the concept to be receivable of all childrenā€™s ā€˜potentialsā€™. Rather than advocating abandoning the termā€”a futile gesture given its emotive forceā€”we argue that the concept of childrenā€™s potential must be profoundly rethought to be workable as a philosophical notion in education. In an era marked by the unspoken assumption that ā€˜unlimited potentialā€™ is always a good thing, we argue that it might be necessary to think about the limitations of the notion of individual potential; namely, the moment when it comes into contact with other peopleā€™s projects. We propose a conceptualisation of potential as the negotiated, situated, ever-changing creation of a group of individuals, in a process marked by conflict, and which remains essentially difficult.This is the final version of the article.     Smoking onset and the time-varying effects of self-efficacy, environmental smoking, and smoking-specific parenting by using discrete-time survival analysis

    This study examined the timing of smoking onset during mid- or late adolescence and the time-varying effects of refusal self-efficacy, parental and sibling smoking behavior, smoking behavior of friends and best friend, and parental smoking-specific communication. We used data from five annual waves of the ā€˜Family and Healthā€™ project. In total, 428 adolescents and their parents participated at baseline. Only never smokers were included at baseline (nĀ =Ā 272). A life table and Kaplanā€“Meier survival curve showed that 51% of all adolescents who did not smoke at baseline did not start smoking within 4Ā years. The risk for smoking onset during mid- or late adolescence is rather stable (hazard ratio between 16 and 19). Discrete-time survival analyses revealed that low refusal self-efficacy, high frequency of communication, and sibling smoking were associated with smoking onset one year later. No interaction effects were found. Conclusively, the findings revealed that refusal self-efficacy is an important predictor of smoking onset during mid- or late adolescence and is independent of smoking-specific communication and smoking behavior of parents, siblings, and (best) friend(s). Findings emphasize the importance of family prevention programs focusing on self-efficacy skills
