    Burning material behaviour in hypoxic environments: An experimental study examining a representative storage arrangement of acrylonitrile butadiene styrene, polyethylene bubble wrap, and cardboard layers as a composite system

    Cone calorimeter and controlled atmosphere cone calorimeter experiments were conducted on various samples. The intent of the tests was to examine the behavior of uniform and composite samples in a range of thicknesses, irradiances, and oxygen concentrations. Single, uniform layers of acrylonitrile butadiene styrene (ABS) were compared to a composite mix, comprising of ABS with a surface layer of cardboard and a secondary layer of polyethylene bubble wrap (intended to represent a potential storage arrangement). The horizontal samples have been tested at irradiances of 25 and 50 kW/m2 and oxygen concentrations of 20.95%, 17%, and 15% to examine a range of significant variables. Results for the uniform arrangement indicated various correlations, previously observed in the works of others, such as the relationships typically described between applied heat flux, ignitability, heat release rate and the effect of the introduction of hypoxic conditions. However, results were shown to change significantly when samples were arranged to feature composite layers. A hypothesized cause of the behavioral change, namely the soot and char residual introduced from the incomplete combustion of the cardboard layer, highlights further important variables that require consideration in material testing under hypoxic conditions. Such variables, namely specific material behaviors and sample orientation, must be sufficiently captured in the design methodologies of systems reliant upon the introduction of hypoxic conditions. It is concluded that sufficiently capturing a wider range of variables in burning materials under hypoxic conditions will introduce further design resilience and help optimize fire protection/prevention methods

    Quantitative proteomics in resected renal cancer tissue for biomarker discovery and profiling

    <b>Background:</b>  Proteomics-based approaches for biomarker discovery are promising strategies used in cancer research. We present state-of-art label-free quantitative proteomics method to assess proteome of renal cell carcinoma (RCC) compared with noncancer renal tissues.<p></p> <b>Methods:</b>  Fresh frozen tissue samples from eight primary RCC lesions and autologous adjacent normal renal tissues were obtained from surgically resected tumour-bearing kidneys. Proteins were extracted by complete solubilisation of tissues using filter-aided sample preparation (FASP) method. Trypsin digested proteins were analysed using quantitative label-free proteomics approach followed by data interpretation and pathways analysis.<p></p> <b>Results:</b>  A total of 1761 proteins were identified and quantified with high confidence (MASCOT ion score threshold of 35 and P-value <0.05). Of these, 596 proteins were identified as differentially expressed between cancer and noncancer tissues. Two upregulated proteins in tumour samples (adipose differentiation-related protein and Coronin 1A) were further validated by immunohistochemistry. Pathway analysis using IPA, KOBAS 2.0, DAVID functional annotation and FLink tools showed enrichment of many cancer-related biological processes and pathways such as oxidative phosphorylation, glycolysis and amino acid synthetic pathways.<p></p> <b>Conclusions:<b>  Our study identified a number of differentially expressed proteins and pathways using label-free proteomics approach in RCC compared with normal tissue samples. Two proteins validated in this study are the focus of on-going research in a large cohort of patients.<p></p&gt

    Protein logic: a statistical mechanical study of signal integration at the single-molecule level

    Information processing and decision making is based upon logic operations, which in cellular networks has been well characterized at the level of transcription. In recent years however, both experimentalists and theorists have begun to appreciate that cellular decision making can also be performed at the level of a single protein, giving rise to the notion of protein logic. Here we systematically explore protein logic using a well known statistical mechanical model. As an example system, we focus on receptors which bind either one or two ligands, and their associated dimers. Notably, we find that a single heterodimer can realize any of the 16 possible logic gates, including the XOR gate, by variation of biochemical parameters. We then introduce the novel idea that a set of receptors with fixed parameters can encode functionally unique logic gates simply by forming different dimeric combinations. An exhaustive search reveals that the simplest set of receptors (two single-ligand receptors and one double-ligand receptor) can realize several different groups of three unique gates, a result for which the parametric analysis of single receptors and dimers provides a clear interpretation.     A novel method for comparing topological models of protein structures enhanced with ligand information

Copyright @ 2008 The Authors.We introduce TOPS+ strings, a highly abstract string-based model of protein topology that permits efficient computation of structure comparison, and can optionally represent ligand information. In this model, we consider loops as secondary structure elements (SSEs) as well as helices and strands; in addition we represent ligands as first class objects. Interactions between SSEs and between SSEs and ligands are described by incoming/outgoing arcs and ligand arcs, respectively; and SSEs are annotated with arc interaction direction and type. We are able to abstract away from the ligands themselves, to give a model characterized by a regular grammar rather than the context sensitive grammar of the original TOPS model. Our TOPS+ strings model is sufficiently descriptive to obtain biologically meaningful results and has the advantage of permitting fast string-based structure matching and comparison as well as avoiding issues of Non-deterministic Polynomial time (NP)-completeness associated with graph problems. Our structure comparison method is computationally more efficient in identifying distantly related proteins than BLAST, CLUSTALW, SSAP and TOPS because of the compact and abstract string-based representation of protein structure which records both topological and biochemical information including the functionally important loop regions of the protein structures. The accuracy of our comparison method is comparable with that of TOPS. Also, we have demonstrated that our TOPS+ strings method out-performs the TOPS method for the ligand-dependent protein structures and provides biologically meaningful results.     Tropical Deforestation, Community Forests, and Protected Areas in the Maya Forest

    Community forests and protected areas have each been proposed as strategies to stop deforestation. These management strategies should be regarded as hypotheses to be evaluated for their effectiveness in particular places. We evaluated the community-forestry hypothesis and the protected-area hypothesis in community forests with commercial timber production and strict protected areas in the Maya Forest of Guatemala and Mexico. From land-use and land cover change (LUCC) maps derived from satellite images, we compared deforestation in 19 community forests and 11 protected areas in both countries in varying periods from 1988 to 2005. Deforestation rates were higher in protected areas than in community forests, but the differences were not significant. An analysis of human presence showed similar deforestation rates in inhabited protected areas and recently inhabited community forests, but the differences were not significant. There was also no significant difference in deforestation between uninhabited protected areas, uninhabited community forests, and long-inhabited community forests. A logistic regression analysis indicated that the factors correlated with deforestation varied by country. Distance to human settlements, seasonal wetlands, and degree and length of human residence were significant in Guatemala, and distance to previous deforestation and tropical semideciduous forest were significant in Mexico. Varying contexts and especially colonization histories are highlighted as likely factors that influence different outcomes. Poorly governed protected areas perform no better as a conservation strategy than poorly governed community forests with recent colonists in active colonization fronts. Long-inhabited extractive communities perform as well as uninhabited strict protected areas under low colonization pressure. A review of costs and benefits suggests that community forests may generate more local income with lower costs. Small sample sizes may have limited the statistical power of our comparisons, but descriptive statistics on deforestation rates, logistic regression analyses, LUCC maps, data available on local economic impacts, and long-term ethnographic and action-research constitute a web of evidence supporting our conclusions. Long-inhabited community forest management for timber can be as effective as uninhabited parks at delivering long-term forest protection under certain circumstances and more effective at delivering local benefits

    Use of image based sports case studies for teaching mechanics

    Mechanics is a fundamental topic required for both undergraduate and postgraduate students on engineering and technology courses. It can be difficult to motivate and engage students in the theoretical aspects of the topic, especially if they are without a strong mathematical background. There are many sporting examples that can be used to explain some of the basic concepts in mechanics. As many sport interactions are highspeed, visualizing the relation to mechanics can be challenging. From our own research, and that published in the field, we now have access to a range of high quality images that have been generated from high-speed video and photogrammetry work, computational simulations or flow visualizations. Two case studies in which images from ball sport research have been used to explain two key engineering subjects: solid and fluid mechanics. A strategy for future collaboration of academics to share and have access to a range of high quality experimental images was also proposed

    How many new cancer patients in Europe will require radiotherapy by 2025? An ESTRO-HERO analysis

    Background: The objective of this HERO study was to assess the number of new cancer patients that will 29 require at least one course of radiotherapy by 2025. 30 Methods: European cancer incidence data by tumor site and country for 2012 and 2025 was extracted 31 from the GLOBOCAN database. The projection of the number of new cases took into account demographic 32 factors (age and size of the population). Population based stages at diagnosis were taken from four 33 European countries. Incidence and stage data were introduced in the Australian Collaboration for 34 Cancer Outcomes Research and Evaluation (CCORE) model. 35 Results: Among the different tumor sites, the highest expected relative increase by 2025 in treatment 36 courses was prostate cancer (24%) while lymphoma (13%), head and neck (12%) and breast cancer 37 (10%) were below the average. Based on the projected cancer distributions in 2025, a 16% expected 38 increase in the number of radiotherapy treatment courses was estimated. This increase varied across 39 European countries from less than 5% to more than 30%. 40 Conclusion: With the already existing disparity in radiotherapy resources in mind, the data provided here 41 should act as a leverage point to raise awareness among European health policy makers of the need for 42 investment in radiotherapy

    The GALNTL6 Gene rs558129 Polymorphism Is Associated With Power Performance

J Strength Cond Res 34(11): 3031-3036, 2020-The largest genome-wide association study to date in sports genomics showed that endurance athletes were 1.23 times more likely to possess the C allele of the single nucleotide polymorphism rs558129 of N-acetylgalactosaminyltransferase-like 6 gene (GALNTL6), compared with controls. Nevertheless, no further study has investigated GALNTL6 gene in relation to physical performance. Considering that previous research has shown that the same polymorphism can be associated with both endurance and power phenotypes (ACTN3, ACE, and PPARA), we investigated the association between GALNTL6 rs558129 polymorphism and power performance. According to this objective we conducted 2 global studies regarding 2 different communities of athletes in Spain and Russia. The first study involved 85 Caucasian physically active men from the north of Spain to perform a Wingate anaerobic test (WAnT). In the second study we compared allelic frequencies between 173 Russian power athletes (49 strength and 124 speed-strength athletes), 169 endurance athletes, and 201 controls. We found that physically active men with the T allele of GALNTL6 rs558129 had 5.03-6.97% higher power values compared with those with the CC genotype (p < 0.05). Consistent with these findings, we have shown that the T allele was over-represented in power athletes (37.0%) compared with endurance athletes (29.3%; OR = 1.4, p = 0.032) and controls (28.6%; OR = 1.5, p = 0.015). Furthermore, the highest frequency of the T allele was observed in strength athletes (43.9%; odds ratio [OR] = 1.9, p = 0.0067 compared with endurance athletes; OR = 2.0, p = 0.0036 compared with controls). In conclusion, our data suggest that the GALNTL6 rs558129 T allele can be favorable for anaerobic performance and strength athletes. In addition, we propose a new possible functional role of GALNTL6 rs558129, gut microbiome regarding short-chain fatty acid regulation and their anti-inflammatory and resynthesis functions.     A population-based study of the epidemiology of oral squamous cell carcinoma in the Netherlands 1989-2018, with emphasis on young adults

    There has been an increasing trend in oral squamous cell carcinoma (OSCC) in patients under 45 years of age. The aim of this study was to evaluate the burden of OSCC in the Netherlands between 1989 and 2018 among young adults (age 20–34 years) when compared to adults (age 35–44 years), and to describe the burden in older groups as well, utilizing cancer registry data to characterize incidence patterns by age, sex, and risk factors. A total of 18,963 cases of OSCC were reported. The overall incidence rate, as measured by annual percentage change (APC), increased significantly from 1989 to 2010 by 1.3% per year (95% confidence interval (CI) 0.9–1.7%) but decreased thereafter by −0.9% (95% CI −2.5% to 0.7%). Annual incidence increased significantly by 2.4% (95% CI 1.1–3.8%) for patients aged 20–34 years, while it decreased for those aged 35–44 years by −0.9% (95% CI −1.7% to 0.0%). In patients older than 60 years, incidence rates increased overall (60–74 years: APC 1.8%, 95% CI 1.5–2.1%; ≥75 years: APC 1.5%, 95% CI 1.2–1.9%). Overall, 66.5% of patients were smokers and 65.3% were alcohol consumers. The marked differences in incidence within the young age subgroups warrants further investigation to elucidate any likely disparity in biological process and clinical outcomes in these populations
