    Implications of solar flare hard X-ray "knee" spectra observed by RHESSI

    We analyse the RHESSI photon spectra of four flares that exhibit significant deviations from power laws - i.e. changes in the "local" Hard X-ray spectral index. These spectra are characterised by two regions of constant power law index connected by a region of changing spectral index - the "knee". We develop theoretical and numerical methods of describing such knees in terms of variable photon spectral indices and we study the results of their inversions for source mean thin target and collisional thick target injection electron spectra. We show that a particularly sharp knee can produce unphysical negative values in the electron spectra, and we derive inequalities that can be used to test for this without the need for an inversion to be performed. Such unphysical features would indicate that source model assumptions were being violated, particularly strongly for the collisional thick target model which assumes a specific form for electron energy loss. For all four flares considered here we find that the knees do not correspond to unphysical electron spectra. In the three flares that have downward knees we conclude that the knee can be explained in terms of transport effects through a region of non-uniform ionisation. In the other flare, which has an upward knee, we conclude that it is most likely a feature of the accelerated spectrum

    Correlated X-ray and Optical Variability in V404 Cyg in Quiescence

    We report simultaneous X-ray and optical observations of V404 Cyg in quiescence. The X-ray flux varied dramatically by a factor of >20 during a 60ks observation. X-ray variations were well correlated with those in Halpha, although the latter include an approximately constant component as well. Correlations can also be seen with the optical continuum, although these are less clear. We see no large lag between X-ray and optical line variations; this implies they are causally connected on short timescales. As in previous observations, Halpha flares exhibit a double-peaked profile suggesting emission distributed across the accretion disk. The peak separation is consistent with material extending outwards to at least the circularization radius. The prompt response in the entire Halpha line confirms that the variability is powered by X-ray (and/or EUV) irradiation.Comment: 5 pages; Accepted for publication in the Astrophysical Journal Letter

    Integrating the framing of clinical questions via PICO into the retrieval of medical literature for systematic reviews

    The PICO process is a technique used in evidence based practice to frame and answer clinical questions. It involves structuring the question around four types of clinical information: Population, Intervention, Control or comparison and Outcome. The PICO framework is used extensively in the compilation of systematic reviews as the means of framing research questions. However, when a search strategy (comprising of a large Boolean query) is formulated to retrieve studies for inclusion in the review, PICO is offen ignored. This paper evaluates how PICO annotations can be applied and integrated into retrieval to improve the screening of studies for inclusion in systematic reviews. The task is to increase precision while maintaining the high level of recall essential to ensure systematic reviews are representative and unbiased. Our results show that restricting the search strategies to match studies using PICO annotations improves precision, however recall is slightly reduced, when compared to the non-PICO baseline. This can lead to both time and cost savings when compiling systematic reviews

    Conceptions of basic education teachers about math proof: influence of professional experience

    A prova é uma atividade que desempenha um papel fundamental na construção do conhecimento matemático, razão pela qual adquire relevância nos programas escolares de Matemática. Admitindo que as conceções dos professores sobre a prova afetam a forma como ela é tratada em sala de aula, procuramos averiguar as conceções de professores portugueses de Matemática do 3.º ciclo do Ensino Básico (do 7.º ao 9.º ano) sobre diferentes aspetos da prova matemática e a influência que a experiência profissional tem nessas conceções. Adotando uma abordagem metodológica mista, recolhemos os dados através de um questionário, respondido por 72 professores, e de uma entrevista a duas professoras com experiências profissionais diferentes. Os resultados revelam que os professores, sobretudo os que têm menos tempo de docência, consideram que a prova matemática tem uma natureza distinta da de outras disciplinas, é uma atividade essencial para a construção do conhecimento matemático, tem como função verificar e explicar a veracidade de uma afirmação e permite desenvolver o raciocínio e a comunicação matemática. No que respeita à participação dos alunos na atividade de provar, são os professores com mais tempo de docência que mais o destacam, o que permite aos alunos perceberem a natureza desta atividade. Em termos curriculares, são os professores com menos experiência docente que mais concordam com a presença da prova logo nos primeiros anos, embora salientem que esta atividade só faz sentido em alguns tópicos programáticos.Proof is an activity that plays a key role in the construction of mathematical knowledge, which is why it acquires relevance in mathematics programs. Admitting that teachers' conceptions about proof influence how it is handled in the classroom, we tried to investigate the conceptions of Portuguese mathematics teachers from the 3rd cycle of basic education (grade 7 to 9) on different aspects of mathematical proof and the relationship that the teachers’ experience has in these conceptions. Adopting a mixed methodological approach, we collected data through a questionnaire answered by 72 teachers, and an interview with two teachers with different professional experience. The results show that teachers, especially those with less experience, consider that mathematical proof has a distinct nature from other disciplines, it is an essential activity for the construction of mathematical knowledge and its function is to verify and explain the truth of a statement by developing reasoning and mathematical communication. With respect to student participation in the activity of proving, teachers with more teaching experience are the ones that most highlight it, which allows students to realize the nature of this activity.     The comprehensive English National Lynch Syndrome Registry: development and description of a new genomics data resource

    \ua9 2024 The Author(s). Background: Lynch Syndrome (LS) is a cancer predisposition syndrome caused by constitutional pathogenic variants in the mismatch repair (MMR) genes. To date, fragmentation of clinical and genomic data has restricted understanding of national LS ascertainment and outcomes, and precluded evaluation of NICE guidance on testing and management. To address this, via collaboration between researchers, the National Disease Registration Service (NDRS), NHS Genomic Medicine Service Alliances (GMSAs), and NHS Regional Clinical Genetics Services, a comprehensive registry of LS carriers in England has been established. Methods: For comprehensive ascertainment of retrospectively identified MMR pathogenic variant (PV) carriers (diagnosed prior to January 1, 2023), information was retrieved from all clinical genetics services across England, then restructured, amalgamated, and validated via a team of trained experts in NDRS. An online submission portal was established for prospective ascertainment from January 1, 2023. The resulting data, stored in a secure database in NDRS, were used to investigate the demographic and genetic characteristics of the cohort, censored at July 25, 2023. Cancer outcomes were investigated via linkage to the National Cancer Registration Dataset (NCRD). Findings: A total of 11,722 retrospective and 570 prospective data submissions were received, resulting in a comprehensive English National Lynch Syndrome Registry (ENLSR) comprising 9030 unique individuals. The most frequently identified pathogenic MMR genes were MSH2 and MLH1 at 37.2% (n = 3362) and 29.1% (n = 2624), respectively. 35.9% (n = 3239) of the ENLSR cohort received their LS diagnosis before their first cancer diagnosis (presumptive predictive germline test). Of these, 6.3% (n = 204) developed colorectal cancer, at a median age of initial diagnosis of 51 (IQR 40–62), compared to 73 years (IQR 64–80) in the general population (p < 0.0001). Interpretation: The ENLSR represents the first comprehensive national registry of PV carriers in England and one of the largest cohorts of MMR PV carriers worldwide. The establishment of a secure, centralised infrastructure and mechanism for routine registration of newly identified carriers ensures sustainability of the data resource.     Faith-based Institutions as Venues for Obesity Prevention

    Purpose of review: To critique the scope and value of recent studies with a focus on obesity-related health promotion in faith organizations. Recent findings: Electronic database searches, scanning of the reference lists of identified articles, and hand searching of journals for articles written in English and published in 2013-16, revealed 16 studies. Half of the studies involved African-Americans, in churches and with predominantly female participants. Research among other ethnic groups was more likely to be exploratory. All of the 11 studies reporting the impact of programs on weight-related measures showed favourable outcomes. However due to study limitations (small sample size; short duration; attrition), significant unbiased effects cannot yet be concluded for most of the interventions reviewed. Study strengths included application of theory in community engagement, and detailed description of cultural tailoring. Summary: Faith organizations show promise as settings for obesity prevention among high-risk groups, particularly African-Americans. Support for progressing formative work to adequately powered, randomized controlled trials is vital. Wider involvement of diverse faith settings, and targeting obesity in men and childhood, would be valuable developments

    The <i>Pratylenchus penetrans</i> transcriptome as a source for the development of alternative control strategies:mining for putative genes involved in parasitism and evaluation of <i>in planta</i> RNAi

    The root lesion nematode Pratylenchus penetrans is considered one of the most economically important species within the genus. Host range studies have shown that nearly 400 plant species can be parasitized by this species. To obtain insight into the transcriptome of this migratory plant-parasitic nematode, we used Illumina mRNA sequencing analysis of a mixed population, as well as nematode reads detected in infected soybean roots 3 and 7 days after nematode infection. Over 140 million paired end reads were obtained for this species, and de novo assembly resulted in a total of 23,715 transcripts. Homology searches showed significant hit matches to 58% of the total number of transcripts using different protein and EST databases. In general, the transcriptome of P. penetrans follows common features reported for other root lesion nematode species. We also explored the efficacy of RNAi, delivered from the host, as a strategy to control P. penetrans, by targeted knock-down of selected nematode genes. Different comparisons were performed to identify putative nematode genes with a role in parasitism, resulting in the identification of transcripts with similarities to other nematode parasitism genes. Focusing on the predicted nematode secreted proteins found in this transcriptome, we observed specific members to be up-regulated at the early time points of infection. In the present study, we observed an enrichment of predicted secreted proteins along the early time points of parasitism by this species, with a significant number being pioneer candidate genes. A representative set of genes examined using RT-PCR confirms their expression during the host infection. The expression patterns of the different candidate genes raise the possibility that they might be involved in critical steps of P. penetrans parasitism. This analysis sheds light on the transcriptional changes that accompany plant infection by P. penetrans, and will aid in identifying potential gene targets for selection and use to design effective control strategies against root lesion nematodes

    Passing the Panda Standard: A TAD Off the Mark?

    Tilapia, a tropical freshwater fish native to Africa, is an increasingly important global food commodity. The World Wide Fund for Nature (WWF), a major environmental nongovernmental organization, has established stakeholder dialogues to formulate farm certification standards that promote ‘‘responsible’’ culture practices. As a preface to its ‘‘tilapia aquaculture dialogue,’’ the WWF for Nature commissioned a review of potential certification issues, later published as a peer-reviewed article. This article contends that both the review and the draft certification standards subsequently developed fail to adequately integrate critical factors governing the relative sustainability of tilapia production and thereby miss more significant issues related to resource-use efficiency and the appropriation of ecosystem space and services. This raises a distinct possibility that subsequent certification will promote intensive systems of tilapia production that are far less ecologically benign than existing widely practiced semiintensive alternatives. Given the likely future significance of this emergent standard, it is contended that a more holistic approach to certification is essential

    The genome of the yellow potato cyst nematode, Globodera rostochiensis, reveals insights into the basis of parasitism and virulence

    BACKGROUND: The yellow potato cyst nematode, Globodera rostochiensis, is a devastating plant pathogen of global economic importance. This biotrophic parasite secretes effectors from pharyngeal glands, some of which were acquired by horizontal gene transfer, to manipulate host processes and promote parasitism. G. rostochiensis is classified into pathotypes with different plant resistance-breaking phenotypes. RESULTS: We generate a high quality genome assembly for G. rostochiensis pathotype Ro1, identify putative effectors and horizontal gene transfer events, map gene expression through the life cycle focusing on key parasitic transitions and sequence the genomes of eight populations including four additional pathotypes to identify variation. Horizontal gene transfer contributes 3.5 % of the predicted genes, of which approximately 8.5 % are deployed as effectors. Over one-third of all effector genes are clustered in 21 putative ‘effector islands’ in the genome. We identify a dorsal gland promoter element motif (termed DOG Box) present upstream in representatives from 26 out of 28 dorsal gland effector families, and predict a putative effector superset associated with this motif. We validate gland cell expression in two novel genes by in situ hybridisation and catalogue dorsal gland promoter element-containing effectors from available cyst nematode genomes. Comparison of effector diversity between pathotypes highlights correlation with plant resistance-breaking. CONCLUSIONS: These G. rostochiensis genome resources will facilitate major advances in understanding nematode plant-parasitism. Dorsal gland promoter element-containing effectors are at the front line of the evolutionary arms race between plant and parasite and the ability to predict gland cell expression a priori promises rapid advances in understanding their roles and mechanisms of action.SE-vdA is supported by BBSRC grant BB/M014207/1.
