    Testing Categorical Moderators in Mixed-Effects Meta-analysis in the Presence of Heteroscedasticity

    Mixed-effects models can be used to examine the association between a categorical moderator and the magnitude of the effect size. Two approaches are available to estimate the residual between-studies variance, τ2res—namely, separate estimation within each category of the moderator versus pooled estimation across all categories. We examine, by means of a Monte Carlo simulation study, both approaches for τ2res estimation in combination with two methods, the Wald-type χ2 and F tests, to test the statistical significance of the moderator. Results suggest that the F test using a pooled estimate of τ2res across categories is the best option in most conditions, although the F test using separate estimates of τ2res is preferable if the residual heterogeneity variances are heteroscedastic

    Somatic embryogenesis and plant regeneration of Vitis vinifera cultivars 'Macabeo' and 'Tempranillo'

    Different experimental conditions have been compared to achieve a high efficiency in embryogenic calli initiation from 'Macabeo' and 'Tempranillo' anthers. Specifically, two stages of anther development were tested (corresponding to tetrad cells or uninucleate pollen), and direct culture of anthers was compared to culture after a cold treatment of inflorescences (4 °C during 48 h). In addition, two induction media (C1 P and B2), mainly differing by microelement and cytokinin levels, were evaluated. Experiment repeatability was also examined with a repetition of anther culture one week later. Callus initiation was similar in all media and treatments for both cultivars, usually starting from the anther filament. A simple protocol for efficient induction of embryogenesis in 'Macabeo' and 'Tempranillo' consisted in:selecting the first inflorescence from hardwood cutting,excising anthers at uninucleate pollen stage without cold treatment of the inflorescences,incubating anthers on C1 P medium.The procedure used for embryo germination and plant regeneration, allowed to obtain a conversion rate up to 75 % in 'Macabeo' and 60 % in 'Tempranillo'. The protocol proposed represents the first regeneration system developed for the Spanish cultivars 'Macabeo' and 'Tempranillo'.

    Concerted Transport and Phosphorylation of Diacylglycerol at ER-PM Contacts Sites Regulates Phospholipid Dynamics During Stress

    A universal response of plants to environmental stresses is the activation of plasma membrane (PM) phospholipase C (PLC) that hydrolyzes phosphatidylinositol phosphate (PIP) to produce soluble inositol phosphate (IP) and diacylglycerol (DAG). DAG produced in this way can be either phosphorylated by PM diacylglycerol kinases (DGKs) to produce the second messenger phosphatidic acid (PA) or transferred to the endoplasmic reticulum (ER) by the Synaptotagmin 1 (SYT1) protein at ER-PM Contact Sites (CS). In Arabidopsis, the clearance of DAG at the PM (avoiding deleterious accumulation) by the transfer activity of SYT is essential to maintain PM stability after stress. In this study we identify that DGK1 and DGK2 form a module with SYT1 at ER-PM CS through interaction of their C1 and C2 domains respectively. Global transcriptomic and metabolomic analyses confirms that SYT1 and DGK1/DGK2 are functionally related and lipidomic analysis supports the hypothesis that DGK1 and DGK2 function at the ER by phosphorylating DAG transferred by SYT1 from the PM. DGK1 and DGK2 show structural similarity to human DGKε, the DGK isoform that function at ER-PM CS in the phosphoinositide (PI) cycle. Our data indicate that components of the PI cycle are conserved between animals and plants and provide a novel mechanism leading to an increase in the efficiency of the PI cycle by channeling the transport and hydrolysis of DAG at the ER-PM CS

    Chemical, Structural, and Morphological Changes of a MoVTeNb Catalyst during Oxidative Dehydrogenation of Ethane

    MoVTeNb mixed oxide, a highly active and selective catalyst for the oxidative dehydrogenation of ethane to produce ethylene, exhibits the so-called M1 and M2 crystalline phases. The thermal stability of the MoVTeNb catalytic system was assessed under varying reaction conditions; to this end, the catalyst was exposed to several reaction temperatures spanning from 440 to 550 °C. Both the pristine and spent materials were analyzed by several characterization techniques. The catalyst was stable below 500 °C; a reaction temperature of ≥500 °C brings about the removal of tellurium from the intercalated framework channels of the M1 crystalline phase. Rietveld refinement of X-ray diffraction patterns and microscopy results showed that the tellurium loss causes the progressive partial destruction of the M1 phase, thus decreasing the number of active sites and forming a MoO2 crystalline phase, which is inactive for this reaction. Raman spectroscopy confirmed the MoO2 phase development as a function of reaction temperature. From highresolution transmission electron microscopy and energy-dispersive X-ray spectroscopy analyses it was noticed that tellurium departure occurs preferentially from the end sides of the needlelike M1 crystals, across the [001] plane. Detailed analysis of a solid deposited at the reactor outlet showrf that it consisted mainly of metallic tellurium, suggesting that the tellurium detachment occurs via reduction of Te4+ to Te0 due to a combination of reaction temperature and feed composition.     Synaptotagmins at the endoplasmic reticulum-plasma membrane contact sites maintain diacylglycerol homeostasis during abiotic stress

    Endoplasmic Reticulum-Plasma Membrane contact sites (ER-PM CS) play fundamental roles in all eukaryotic cells. Arabidopsis mutants lacking the ER-PM protein tether synaptotagmin1 (SYT1) exhibit decreased plasma membrane (PM) integrity under multiple abiotic stresses such as freezing, high salt, osmotic stress and mechanical damage. Here, we show that, together with SYT1, the stress-induced SYT3 is an ER-PM tether that also functions in maintaining PM integrity. The ER-PM CS localization of SYT1 and SYT3 is dependent on PM phosphatidylinositol-4-phosphate and is regulated by abiotic stress. Lipidomic analysis revealed that cold stress increased the accumulation of diacylglycerol at the PM in a syt1/3 double mutant relative to WT while the levels of most glycerolipid species remain unchanged. Additionally, SYT1-GFP preferentially binds diacylglycerol in vivo with little affinity for polar glycerolipids. Our work uncovers a SYT-dependent mechanism of stress adaptation counteracting the detrimental accumulation of diacylglycerol at the PM produced during episodes of abiotic stress

    Charged-Higgs phenomenology in the Aligned two-Higgs-doublet model

    The alignment in flavour space of the Yukawa matrices of a general two-Higgs-doublet model results in the absence of tree-level flavour-changing neutral currents. In addition to the usual fermion masses and mixings, the aligned Yukawa structure only contains three complex parameters, which are potential new sources of CP violation. For particular values of these three parameters all known specific implementations of the model based on discrete Z_2 symmetries are recovered. One of the most distinctive features of the two-Higgs-doublet model is the presence of a charged scalar. In this work, we discuss its main phenomenological consequences in flavour-changing processes at low energies and derive the corresponding constraints on the parameters of the aligned two-Higgs-doublet model.Comment: 46 pages, 19 figures.     Indication for the disappearance of reactor electron antineutrinos in the Double Chooz experiment

    The Double Chooz Experiment presents an indication of reactor electron antineutrino disappearance consistent with neutrino oscillations. A ratio of 0.944 ±\pm 0.016 (stat) ±\pm 0.040 (syst) observed to predicted events was obtained in 101 days of running at the Chooz Nuclear Power Plant in France, with two 4.25 GWth_{th} reactors. The results were obtained from a single 10 m3^3 fiducial volume detector located 1050 m from the two reactor cores. The reactor antineutrino flux prediction used the Bugey4 measurement as an anchor point. The deficit can be interpreted as an indication of a non-zero value of the still unmeasured neutrino mixing parameter \sang. Analyzing both the rate of the prompt positrons and their energy spectrum we find \sang = 0.086 ±\pm 0.041 (stat) ±\pm 0.030 (syst), or, at 90% CL, 0.015 << \sang  <\ < 0.16.Comment: 7 pages, 4 figures, (new version after PRL referee's comments

    Cocoa coproducts-based and walnut oil gelled emulsion as animal fat replacer and healthy bioactive source in beef burgers

    The aim of this work was to evaluate the effects on the chemical, physic-chemical, tech-nological, and sensory properties of beef burger when replacing different quantities of fat (50 and 100%) with different levels of oil-in-water-gelled emulsion elaborated with walnut oil and cocoa bean shell flour (GECW). The chemical composition of the samples was affected by the fat replace-ment. The reformulation increased the moisture and ash content while the fat and protein content decreased with respect to the control sample. The linolenic and linolenic acid content of the beef burgers increased as the GECW replacement was augmented. The polyunsaturated fatty/saturated fatty acid ratio increased in both raw and cooked burgers, whereas the atherogenicity index and thrombogenicity index were reduced in both raw and cooked burgers with respect to the control sample. The use of GECW as a fat replacer was found to be effective in improving the cooking loss. Similarly, there were positive effects on reductions in the diameter and the increases in the thickness of the beef burgers. Regarding lipid stability, in both the raw and cooked burgers, the reformulation increased the 2-thiobarbituric acid reactive substance (TBARs) values with respect to the control sample. In both types of reformulated burgers, three bound polyphenols (mainly catechin and epicatechin) and two free polyphenols were identified, as were methylxanthines theobromine and caffeine. The sensory properties for the control and partial pork backfat replacement treatments were similar, while the sample with the total pork backfat replacement treatment showed the lowest scores. The blend of cocoa bean shell flour and walnut oil could be used as new ingredients for the development of beef burgers with a healthier nutritional profile without demeriting their sensory or cooking characteristics and physic-chemical properties.This research was funded by Programa Iberoamericano de Ciencia y TecnologĂ­a para el Desarrollo CYTED: 119RT0568.info:eu-repo/semantics/publishedVersio

    KrĂĽppel-Like Factor 6 Expression Changes during Trophoblast Syncytialization and Transactivates ĂźhCG and PSG Placental Genes

    BACKGROUND: Krüppel-like factor-6 (KLF6) is a widely expressed member of the Sp1/KLF family of transcriptional regulators involved in differentiation, cell cycle control and proliferation in several cell systems. Even though the highest expression level of KLF6 has been detected in human and mice placenta, its function in trophoblast physiology is still unknown. METHODOLOGY/PRINCIPAL FINDINGS: Herein, we explored KLF6 expression and sub-cellular distribution in human trophoblast cells differentiating into the syncytial pathway, and its role in the regulation of genes associated with placental development and pregnancy maintenance. Confocal immunofluorescence microscopy demonstrated that KLF6 is expressed throughout human cytotrophoblast differentiation showing no evident modifications in its nuclear and cytoplasmic localization pattern. KLF6 transcript and protein peaked early during the syncytialization process as determined by qRT-PCR and western blot assays. Overexpression of KLF6 in trophoblast-derived JEG-3 cells showed a preferential nuclear signal correlating with enhanced expression of human β-chorionic gonadotropin (βhCG) and pregnancy-specific glycoprotein (PSG) genes. Moreover, KLF6 transactivated βhCG5, PSG5 and PSG3 gene promoters. Deletion of KLF6 Zn-finger DNA binding domain or mutation of the consensus KLF6 binding site abolished transactivation of the PSG5 promoter. CONCLUSIONS/SIGNIFICANCE: Results are consistent with KLF6 playing a role as transcriptional regulator of relevant genes for placental differentiation and physiology such as βhCG and PSG, in agreement with an early and transient increase of KLF6 expression during trophoblast syncytialization
