    Una nueva especie de Andiceras Krantz (Cephalopoda: Ammonoidea) del Jurásico Tardío-Cretácico Temprano de la Cuenca Neuquina, Mendoza, Argentina: Sistemática y bioestratigrafía

    Se describe una nueva especie de amonoideo perteneciente al género andino endémico Andiceras Krantz. Los restos asignados a Andiceras planulatus sp. nov. se disponen estratigráficamente entre los últimos niveles con representantes de la Biozona de Asociación de Substeueroceras koeneni de edad tithoniana tardía, y los primeros elementos de la Biozona de Asociación de Argentiniceras noduliferum de edad berriasiana temprana, de acuerdo al esquema clásico de biozonación andina. La posición estratigráfica particular de esta nueva especie, junto con su morfología de fácil identificación, la convierten en un potencial buen marcador bioestratigráfico para el reconocimiento del límite Jurásico-Cretácico en la región andina.A new ammonoid species of the Andean endemic genus Andiceras Krantz is here described from upper Tithonian-lower Berriasian levels of the Vaca Muerta and Chachao Formations of the Mendoza Group, in north-western Mendoza Province, Argentina. Andiceras planulatus sp. nov. lies between the last beds with representatives of the Substeueroceras koeneni Assemblage Zone of late Tithonian age and the first bed yielding elements of the Argentiniceras noduliferum Assemblage Zone of early Berriasian age, according to the traditional Andean ammonoid zonation scheme. The particular stratigraphic position of Andiceras planulatus, together with its easily recognizable morphology makes this species a good potential biostratigraphic marker for the Jurassic-Cretaceous boundary in the Andean region.Fil: Vennari, Verónica Vanesa.     Molecular characterization of dissolved organic matter linked to microbial (Bacteria and Archaea) diversity in the main water masses of the Eastern North Atlantic Ocean

    The microbe-dissolved organic matter (DOM) interactions include microbial uptake and DOM reworking and release, affecting the composition of the heterogeneous DOM pool. In turns, this distinct DOM composition can select for microbial assemblages. We investigated the diversity of microbial (both Bacteria and Archaea) communities (combining Illumina tag sequencing of 16S rRNA gen -amplicon sequencing variants, ASVs- and metagenomics) and the chemodiversity of dissolved organic molecules (extracted by solid phase extraction and analyzed by Fourier Transform Ion Cyclotron Resonance Mass Spectrometry, FT-ICR-MS) in the Eastern North Atlantic Ocean off the Galician coast (43oN, 9o-15oW). Sampling ranged from 100 m to 5000 m, thereby encompassing a wide variety of water masses with contrasting origins and different aging. Applying diversity ecological metrics to both organic compounds and ASVs, we found that microbial diversity and richness were negatively correlated with DOM concentration and chemo-diversity. Besides, our results demonstrated the link between this trend and water mass aging, which enhances biosphere taxonomic diversity but reduces molecular variety. DOM diversity, decreasing along the water mass aging gradient, would likely reflect the persistence of the most refractory molecules, generated as by- product of the DOM degradation by microbes. In two PCoAs based on the metagenomic data, combined PCoA axis 1 and PCoA axis 2, explained the 80% and 20% of the microbial gene ́s structural variability among water masses. The oldest water masses, originated at high latitudes, such as NADW and LDW, were associated to higher abundance of genes involved in metabolism of aromatic compound. Intermediate waters such as ENACWst were related to sulphur/iron/phosphorous metabolism related genes. Surface waters were linked to genes involved in photosynthesis, autotrophy and cell division. Taken together, the observed increase of DOM homogenization along water mass aging was associated to differences in the functional diversity of microbial communities

    Loss of TMEM106B exacerbates C9ALS/FTD DPR pathology by disrupting autophagosome maturation

    Disruption to protein homeostasis caused by lysosomal dysfunction and associated impairment of autophagy is a prominent pathology in amyotrophic lateral sclerosis and frontotemporal dementia (ALS/FTD). The most common genetic cause of ALS/FTD is a G4C2 hexanucleotide repeat expansion in C9orf72 (C9ALS/FTD). Repeat-associated non-AUG (RAN) translation of G4C2 repeat transcripts gives rise to dipeptide repeat (DPR) proteins that have been shown to be toxic and may contribute to disease etiology. Genetic variants in TMEM106B have been associated with frontotemporal lobar degeneration with TDP-43 pathology and disease progression in C9ALS/FTD. TMEM106B encodes a lysosomal transmembrane protein of unknown function that is involved in various aspects of lysosomal biology. How TMEM106B variants affect C9ALS/FTD is not well understood but has been linked to changes in TMEM106B protein levels. Here, we investigated TMEM106B function in the context of C9ALS/FTD DPR pathology. We report that knockdown of TMEM106B expression exacerbates the accumulation of C9ALS/FTD-associated cytotoxic DPR proteins in cell models expressing RAN-translated or AUG-driven DPRs as well as in C9ALS/FTD-derived iAstrocytes with an endogenous G4C2 expansion by impairing autophagy. Loss of TMEM106B caused a block late in autophagy by disrupting autophagosome to autolysosome maturation which coincided with impaired lysosomal acidification, reduced cathepsin activity, and juxtanuclear clustering of lysosomes. Lysosomal clustering required Rab7A and coincided with reduced Arl8b-mediated anterograde transport of lysosomes to the cell periphery. Increasing Arl8b activity in TMEM106B-deficient cells not only restored the distribution of lysosomes, but also fully rescued autophagy and DPR protein accumulation. Thus, we identified a novel function of TMEM106B in autophagosome maturation via Arl8b. Our findings indicate that TMEM106B variants may modify C9ALS/FTD by regulating autophagic clearance of DPR proteins. Caution should therefore be taken when considering modifying TMEM106B expression levels as a therapeutic approach in ALS/FTD

    Increased Expression of Fatty-Acid and Calcium Metabolism Genes in Failing Human Heart

    Heart failure (HF) involves alterations in metabolism, but little is known about cardiomyopathy-(CM)-specific or diabetes-independent alterations in gene expression of proteins involved in fatty-acid (FA) uptake and oxidation or in calcium-(Ca(2+))-handling in the human heart.RT-qPCR was used to quantify mRNA expression and immunoblotting to confirm protein expression in left-ventricular myocardium from patients with HF (n = 36) without diabetes mellitus of ischaemic (ICM, n = 16) or dilated (DCM, n = 20) cardiomyopathy aetiology, and non-diseased donors (CTL, n = 6).Significant increases in mRNA of genes regulating FA uptake (CD36) and intracellular transport (Heart-FA-Binding Protein (HFABP)) were observed in HF patients vs CTL. Significance was maintained in DCM and confirmed at protein level, but not in ICM. mRNA was higher in DCM than ICM for peroxisome-proliferator-activated-receptor-alpha (PPARA), PPAR-gamma coactivator-1-alpha (PGC1A) and CD36, and confirmed at the protein level for PPARA and CD36. Transcript and protein expression of Ca(2+)-handling genes (Two-Pore-Channel 1 (TPCN1), Two-Pore-Channel 2 (TPCN2), and Inositol 1,4,5-triphosphate Receptor type-1 (IP3R1)) increased in HF patients relative to CTL. Increases remained significant for TPCN2 in all groups but for TPCN1 only in DCM. There were correlations between FA metabolism and Ca(2+)-handling genes expression. In ICM there were six correlations, all distinct from those found in CTL. In DCM there were also six (all also different from those found in CTL): three were common to and three distinct from ICM.DCM-specific increases were found in expression of several genes that regulate FA metabolism, which might help in the design of aetiology-specific metabolic therapies in HF. Ca(2+)-handling genes TPCN1 and TPCN2 also showed increased expression in HF, while HF- and CM-specific positive correlations were found among several FA and Ca(2+)-handling genes

    Post-Operative Functional Outcomes in Early Age Onset Rectal Cancer

    Background: Impairment of bowel, urogenital and fertility-related function in patients treated for rectal cancer is common. While the rate of rectal cancer in the young (<50 years) is rising, there is little data on functional outcomes in this group. Methods: The REACCT international collaborative database was reviewed and data on eligible patients analysed. Inclusion criteria comprised patients with a histologically confirmed rectal cancer, <50 years of age at time of diagnosis and with documented follow-up including functional outcomes. Results: A total of 1428 (n=1428) patients met the eligibility criteria and were included in the final analysis. Metastatic disease was present at diagnosis in 13%. Of these, 40% received neoadjuvant therapy and 50% adjuvant chemotherapy. The incidence of post-operative major morbidity was 10%. A defunctioning stoma was placed for 621 patients (43%); 534 of these proceeded to elective restoration of bowel continuity. The median follow-up time was 42 months. Of this cohort, a total of 415 (29%) reported persistent impairment of functional outcomes, the most frequent of which was bowel dysfunction (16%), followed by bladder dysfunction (7%), sexual dysfunction (4.5%) and infertility (1%). Conclusion: A substantial proportion of patients with early-onset rectal cancer who undergo surgery report persistent impairment of functional status. Patients should be involved in the discussion regarding their treatment options and potential impact on quality of life. Functional outcomes should be routinely recorded as part of follow up alongside oncological parameters

    The James Webb Space Telescope Mission

    Twenty-six years ago a small committee report, building on earlier studies, expounded a compelling and poetic vision for the future of astronomy, calling for an infrared-optimized space telescope with an aperture of at least 4m4m. With the support of their governments in the US, Europe, and Canada, 20,000 people realized that vision as the 6.5m6.5m James Webb Space Telescope. A generation of astronomers will celebrate their accomplishments for the life of the mission, potentially as long as 20 years, and beyond. This report and the scientific discoveries that follow are extended thank-you notes to the 20,000 team members. The telescope is working perfectly, with much better image quality than expected. In this and accompanying papers, we give a brief history, describe the observatory, outline its objectives and current observing program, and discuss the inventions and people who made it possible.     MUSiC : a model-unspecific search for new physics in proton-proton collisions at root s=13TeV

    Results of the Model Unspecific Search in CMS (MUSiC), using proton-proton collision data recorded at the LHC at a centre-of-mass energy of 13 TeV, corresponding to an integrated luminosity of 35.9 fb(-1), are presented. The MUSiC analysis searches for anomalies that could be signatures of physics beyond the standard model. The analysis is based on the comparison of observed data with the standard model prediction, as determined from simulation, in several hundred final states and multiple kinematic distributions. Events containing at least one electron or muon are classified based on their final state topology, and an automated search algorithm surveys the observed data for deviations from the prediction. The sensitivity of the search is validated using multiple methods. No significant deviations from the predictions have been observed. For a wide range of final state topologies, agreement is found between the data and the standard model simulation. This analysis complements dedicated search analyses by significantly expanding the range of final states covered using a model independent approach with the largest data set to date to probe phase space regions beyond the reach of previous general searches.Peer reviewe

    Measurement of prompt open-charm production cross sections in proton-proton collisions at root s=13 TeV

    The production cross sections for prompt open-charm mesons in proton-proton collisions at a center-of-mass energy of 13TeV are reported. The measurement is performed using a data sample collected by the CMS experiment corresponding to an integrated luminosity of 29 nb(-1). The differential production cross sections of the D*(+/-), D-+/-, and D-0 ((D) over bar (0)) mesons are presented in ranges of transverse momentum and pseudorapidity 4 < p(T) < 100 GeV and vertical bar eta vertical bar < 2.1, respectively. The results are compared to several theoretical calculations and to previous measurements.Peer reviewe

    Development and validation of HERWIG 7 tunes from CMS underlying-event measurements

    This paper presents new sets of parameters (“tunes”) for the underlying-event model of the HERWIG7 event generator. These parameters control the description of multiple-parton interactions (MPI) and colour reconnection in HERWIG7, and are obtained from a fit to minimum-bias data collected by the CMS experiment at s=0.9, 7, and 13Te. The tunes are based on the NNPDF 3.1 next-to-next-to-leading-order parton distribution function (PDF) set for the parton shower, and either a leading-order or next-to-next-to-leading-order PDF set for the simulation of MPI and the beam remnants. Predictions utilizing the tunes are produced for event shape observables in electron-positron collisions, and for minimum-bias, inclusive jet, top quark pair, and Z and W boson events in proton-proton collisions, and are compared with data. Each of the new tunes describes the data at a reasonable level, and the tunes using a leading-order PDF for the simulation of MPI provide the best description of the dat
