    Diplomová práce nazvaná Softwarové aplikace pro diagnostiku Parkinsonovy choroby s využitím počítačových periferií se zabývá možnostmi hodnocení tremoru z křivek zaznamenaných digitálním perem touchpadu, případně počítačovou myší. Cílem práce bylo navrhnout a realizovat minimálně dvě softwarové aplikace využívající počítačové periferie ke sběru data a následně je otestovat na skupině minimálně 10 pacientů. K matematickému hodnocení získaných dat byla použita Fourierova transformace, která sloužila k výpočtu frekvenčního spektra a výkonové spektrální hustoty, dále směrodatná odchylka a aritmetický průměr. K návrhu měřících aplikací bylo využito prostředí Microsoft Visual Studio a programovací jazyk C# a k hodnocení naměřených dat programové prostředí MATLAB.The thesis titled Software Applications for the Diagnosis of Parkinson's Disease Using Computer Peripherals deals with the possibility of evaluating tremors from the curves recorded by the digital touchpad pen, or a computer mouse. The aim of the thesis was to design and implement at least two software applications using computer peripherals to collect data and then test them on a group of at least 10 patients. The Fourier Transform was used for the mathematical evaluation of the obtained data, which used to calculate the frequency spectrum and the power spectral density, the standard deviation and the arithmetic mean.     Import 04/11/2015Bakalářská práce se zabývá realizací výukového modelu pro měření biologických signálů v rozsahu výstupních hodnot zobrazitelných na PLC zařízení. Zaměřuje se převážně na dvě nejsnáze proveditelné měření – záznam pulzní křivky a měření dechové frekvence. Pro realizaci je zvoleno měření dechové frekvence. Postupně práce popisuje návrh a realizaci výukového modelu. Dále jsou porovnávány získané výsledky s ideálními fyziologickými průběhy signálu. Hotový výukový model bude sloužit jako vzdělávací pomůcka do kroužku kybernetiky probíhající na VŠB – TUO.This bacherol´s thesis deals with the implementation of educational model for measuring biological signals with output values displayable on the PLC devices. It focuses mainly on the two easiest feasible measurements - recording pulse waveform and the measurement of respiratory rate. For implementation of measurement is selected breathing frequency. The paper describes the design and implementation of educational model. Furthermore, the results obtained are compared with the ideal physiological waveforms.     Předmětem diplomové práce „Financování z ESF“ je studium a popis politiky hospodářské a sociální soudržnosti, dále Evropského sociálního fondu, jeho začlenění ve struktuře fondů Evropské unie, jeho cíle a užitky. Druhá část této práce má čtenářům poskytnout cenné informace a užitečné rady, jak postupovat při přípravě projektu čerpající dotace z Evropského sociálního fondu. V praktické části práce jsou veškeré poznatky uplatněny na konkrétním projektu a to na projektu Městského úřadu ve Žďáru nad Sázavou.The thesis „Financing from the ESF“ deals with a study and description of a policy of economic and social cohesion, further the European Social Fund, its incorporation in a structure of European Union funds, its objectives and benefits. The second part of this thesis is to supply readers with valuable information and useful advice on how to act when preparing a project drawing subsidies from the European Social Fund. In the practical part of the thesis, all knowledge is applied to a concrete project namely a project of the Municipal Office in Žďár nad Sázavou.

    Cílem diplomové práce na téma „Optimalizace financování Žďáru nad Sázavou“ je zjistit potřeby obyvatel města Žďáru nad Sázavou a na základě výsledků navrhnout nová řešení financování dle rozpočtových možností města. Dílčím cílem je analyzovat příjmovou a výdajovou stránku rozpočtu města Žďár nad Sázavou a jeho plnění. Práce je rozdělena na tři části, teoretickou, analytickou a praktickou. První kapitola vymezuje stěžejní pojmy a principy související se studovanou problematikou. V analytické části je analyzováno hospodaření města za posledních pět let. V poslední části práce jsou navržena řešení v jednotlivých činnostech města Žďár nad Sázavou dle výsledků z dotazníkového šetření. Závěr obsahuje syntézu a závěrečná shrnutí zjištěných poznatků a nedostatků v hospodaření města Žďár nad Sázavou.The goal of my dissertation, on the theme financing optimization, is to find out the needs of dwellers in town Žďár nad Sázavou. On the basis of my results I will suggest new financing solutions according to town budget expenditure. Partial target is to analyse income and expenditure budget of Žďár nad Sázavou and its fulfilment. My dissertation is divided into three parts theoretical, analytical and practical. The first part specifies crucial terms and principles related to studied points at issue. In the second part is analysed financial management for the past five years. In the last part are suggested solutions for separated activities in Žďár nad Sázavou according to questionnaire survey. Conclusion consists of synthesis and final summaries of detected information and deficits in financial management in Žďár nad Sázavou.

    Niche differentiation of bacteria and fungi in carbon and nitrogen cycling of different habitats in a temperate coniferous forest: A metaproteomic approach

    Temperate coniferous forests sustain the highest levels of biomass of all terrestrial ecosystems and belong to the major carbon sinks on Earth. However, the community composition and its functional diversity depending on the habitat have yet to be unveiled. Here, we analyzed the proteomes from litter, plant roots, rhizosphere, and bulk soil in a temperate coniferous forest at two time points to improve the understanding of the interplay between bacterial and eukaryotic communities in different habitats. Our metaproteomic approach yielded a total of 139,127 proteins that allowed to differentiate the contribution of microbial taxa to protein expression as well as the general functionality based on KEGG Orthology in each habitat. The pool of expressed carbohydrate-active enzymes (CAZymes) was dominated by fungal proteins. While CAZymes in roots and litter targeted mostly the structural biopolymers of plant origin such as lignin and cellulose, the majority of CAZymes in bulk and rhizosphere soil targeted oligosaccharides, starch, and glycogen. Proteins involved in nitrogen cycling were mainly of bacterial origin. Most nitrogen cycling proteins in litter and roots participated in ammonium assimilation while those performing nitrification were the most abundant in bulk and rhizosphere soil. Together, our results indicated niche differentiation of the microbial involvement in carbon and nitrogen cycling in a temperate coniferous forest topsoil

    Large-scale genome sequencing of mycorrhizal fungi provides insights into the early evolution of symbiotic traits

    Mycorrhizal fungi are mutualists that play crucial roles in nutrient acquisition in terrestrial ecosystems. Mycorrhizal symbioses arose repeatedly across multiple lineages of Mucoromycotina, Ascomycota, and Basidiomycota. Considerable variation exists in the capacity of mycorrhizal fungi to acquire carbon from soil organic matter. Here, we present a combined analysis of 135 fungal genomes from 73 saprotrophic, endophytic and pathogenic species, and 62 mycorrhizal species, including 29 new mycorrhizal genomes. This study samples ecologically dominant fungal guilds for which there were previously no symbiotic genomes available, including ectomycorrhizal Russulales, Thelephorales and Cantharellales. Our analyses show that transitions from saprotrophy to symbiosis involve (1) widespread losses of degrading enzymes acting on lignin and cellulose, (2) co-option of genes present in saprotrophic ancestors to fulfill new symbiotic functions, (3) diversification of novel, lineage-specific symbiosis-induced genes, (4) proliferation of transposable elements and (5) divergent genetic innovations underlying the convergent origins of the ectomycorrhizal guild. Mycorrhizal symbioses have evolved repeatedly in diverse fungal lineages. A large phylogenomic analysis sheds light on genomic changes associated with transitions from saprotrophy to symbiosis, including divergent genetic innovations underlying the convergent origins of the ectomycorrhizal guild.Peer reviewe

    Soil Salinity and pH Drive Soil Bacterial Community Composition and Diversity Along a Lateritic Slope in the Avon River Critical Zone Observatory, Western Australia

    Soils are crucial in regulating ecosystem processes, such as nutrient cycling, and supporting plant growth. To a large extent, these functions are carried out by highly diverse and dynamic soil microbiomes that are in turn governed by numerous environmental factors including weathering profile and vegetation. In this study, we investigate geophysical and vegetation effects on the microbial communities of iron-rich lateritic soils in the highly weathered landscapes of Western Australia (WA). The study site was a lateritic hillslope in southwestern Australia, where gradual erosion of the duricrust has resulted in the exposure of the different weathering zones. High-throughput amplicon sequencing of the 16S rRNA gene was used to investigate soil bacterial community diversity, composition and functioning. We predicted that shifts in the microbial community would reflect variations in certain edaphic properties associated with the different layers of the lateritic profile and vegetation cover. Our results supported this hypothesis, with electrical conductivity, pH and clay content having the strongest correlation with beta diversity, and many of the differentially abundant taxa belonging to the phyla Actinobacteria and Proteobacteria. Soil water repellence, which is associated with Eucalyptus vegetation, also affected beta diversity. This enhanced understanding of the natural system could help to improve future crop management in WA since the physicochemical properties of the agricultural soils in this region are inherited from laterites via the weathering and pedogenesis processes
