    HUWE1 E3 ligase promotes PINK1/PARKINindependent mitophagy by regulating AMBRA1 activation via IKKa

    The selective removal of undesired or damaged mitochondria by autophagy, known as mitophagy, is crucial for cellular homoeostasis, and prevents tumour diffusion, neurodegeneration and ageing. The pro-autophagic molecule AMBRA1 (autophagy/beclin-1 regulator-1) has been defined as a novel regulator of mitophagy in both PINK1/PARKIN-dependent and -independent systems. Here, we identified the E3 ubiquitin ligase HUWE1 as a key inducing factor in AMBRA1-mediated mitophagy, a process that takes place independently of the main mitophagy receptors. Furthermore, we show that mitophagy function of AMBRA1 is post-translationally controlled, upon HUWE1 activity, by a positive phosphorylation on its serine 1014. This modification is mediated by the IKKα kinase and induces structural changes in AMBRA1, thus promoting its interaction with LC3/GABARAP (mATG8) proteins and its mitophagic activity. Altogether, these results demonstrate that AMBRA1 regulates mitophagy through a novel pathway, in which HUWE1 and IKKα are key factors, shedding new lights on the regulation of mitochondrial quality control and homoeostasis in mammalian cells

    Considerable enhancement of the critical current in a superconducting film by magnetized magnetic strip

    We show that a magnetic strip on top of a superconducting strip magnetized in a specified direction may considerably enhance the critical current in the sample. At fixed magnetization of the magnet we observed diode effect - the value of the critical current depends on the direction of the transport current. We explain these effects by a influence of the nonuniform magnetic field induced by the magnet on the current distribution in the superconducting strip. The experiment on a hybrid Nb/Co structure confirmed the predicted variation of the critical current with a changing value of magnetization and direction of the transport current.Comment: 6 pages, 7 figure

    Feasibility studies of the time-like proton electromagnetic form factor measurements with PANDA at FAIR

    The possibility of measuring the proton electromagnetic form factors in the time-like region at FAIR with the \PANDA detector is discussed. Detailed simulations on signal efficiency for the annihilation of pˉ+p\bar p +p into a lepton pair as well as for the most important background channels have been performed. It is shown that precision measurements of the differential cross section of the reaction pˉ+pe++e\bar p +p \to e^++ e^- can be obtained in a wide angular and kinematical range. The individual determination of the moduli of the electric and magnetic proton form factors will be possible up to a value of momentum transfer squared of q214q^2\simeq 14 (GeV/c)2^2. The total pˉ+pe++e\bar p +p\to e^++e^- cross section will be measured up to q228q^2\simeq 28 (GeV/c)2^2. The results obtained from simulated events are compared to the existing data.     Scanning Probe Microscopy of silicon layers after Ag+ implantation

    Описан способ формирования нанопористого кремния на поверхности монокристаллического Si при имплантации ионами Ag+. Методами СЗМ и СЭМ показано, что в результате на поверхности Si формируются аморфные слои нанопористого кремния со средними размерами пор ~130 нм, в структуре которых синтезируются наночастицы Ag диаметром от 5 до 20 нм.An idea to create nanoporous silicon layers by low-energy high-dose Ag-ion implantation was realized. Surface structures were analyzed by scanning electron microscopy and scanning probe microscopy.     Feasibility studies of time-like proton electromagnetic form factors at PANDA at FAIR

    Simulation results for future measurements of electromagnetic proton form factors at \PANDA (FAIR) within the PandaRoot software framework are reported. The statistical precision with which the proton form factors can be determined is estimated. The signal channel pˉpe+e\bar p p \to e^+ e^- is studied on the basis of two different but consistent procedures. The suppression of the main background channel, i.e.\textit{i.e.} pˉpπ+π\bar p p \to \pi^+ \pi^-, is studied. Furthermore, the background versus signal efficiency, statistical and systematical uncertainties on the extracted proton form factors are evaluated using two different procedures. The results are consistent with those of a previous simulation study using an older, simplified framework. However, a slightly better precision is achieved in the PandaRoot study in a large range of momentum transfer, assuming the nominal beam conditions and detector performance

    Continued 26S proteasome dysfunction in mouse brain cortical neurons impairs autophagy and the Keap1-Nrf2 oxidative defence pathway

    The ubiquitin–proteasome system (UPS) and macroautophagy (autophagy) are central to normal proteostasis and interdependent in that autophagy is known to compensate for the UPS to alleviate ensuing proteotoxic stress that impairs cell function. UPS and autophagy dysfunctions are believed to have a major role in the pathomechanisms of neurodegenerative disease. Here we show that continued 26S proteasome dysfunction in mouse brain cortical neurons causes paranuclear accumulation of fragmented dysfunctional mitochondria, associated with earlier recruitment of Parkin and lysine 48-linked ubiquitination of mitochondrial outer membrane (MOM) proteins, including Mitofusin-2. Early events also include phosphorylation of p62/SQSTM1 (p62) and increased optineurin, as well as autophagosomal LC3B and removal of some mitochondria, supporting the induction of selective autophagy. Inhibition of the degradation of ubiquitinated MOM proteins with continued 26S proteasome dysfunction at later stages may impede efficient mitophagy. However, continued 26S proteasome dysfunction also decreases the levels of essential autophagy proteins ATG9 and LC3B, which is characterised by decreases in their gene expression, ultimately leading to impaired autophagy. Intriguingly, serine 351 phosphorylation of p62 did not enhance its binding to Keap1 or stabilise the nuclear factor erythroid 2-related factor 2 (Nrf2) transcription factor in this neuronal context. Nrf2 protein levels were markedly decreased despite transcriptional activation of the Nrf2 gene. Our study reveals novel insights into the interplay between the UPS and autophagy in neurons and is imperative to understanding neurodegenerative disease where long-term proteasome inhibition has been implicated

    Описан способ получения структурированного композиционного материала на подложке из халькогенидного стекла (GeSe) при имплантации ионами Ag+. Методами СЗМ и СЭМ показано, что в результате на поверхности GeSe образуется дифракционная решетка с периодом ~ 25 мкм и высотой ступенек ~ 170-200 нм.An idea to create microstructured composite material on a GeSe glass substrate by Ag+ ion implantation is realized.
