    Wind, waves, and acoustic background levels at Station ALOHA

The definitive version was published in Journal of Geophysical Research 117 (2012): C03017, doi:10.1029/2011JC007267.Frequency spectra from deep-ocean near-bottom acoustic measurements obtained contemporaneously with wind, wave, and seismic data are described and used to determine the correlations among these data and to discuss possible causal relationships. Microseism energy appears to originate in four distinct regions relative to the hydrophone: wind waves above the sensors contribute microseism energy observed on the ocean floor; a fraction of this local wave energy propagates as seismic waves laterally, and provides a spatially integrated contribution to microseisms observed both in the ocean and on land; waves in storms generate microseism energy in deep water that travels as seismic waves to the sensor; and waves reflected from shorelines provide opposing waves that add to the microseism energy. Correlations of local wind speed with acoustic and seismic spectral time series suggest that the local Longuet-Higgins mechanism is visible in the acoustic spectrum from about 0.4 Hz to 80 Hz. Wind speed and acoustic levels at the hydrophone are poorly correlated below 0.4 Hz, implying that the microseism energy below 0.4 Hz is not typically generated by local winds. Correlation of ocean floor acoustic energy with seismic spectra from Oahu and with wave spectra near Oahu imply that wave reflections from Hawaiian coasts, wave interactions in the deep ocean near Hawaii, and storms far from Hawaii contribute energy to the seismic and acoustic spectra below 0.4 Hz. Wavefield directionality strongly influences the acoustic spectrum at frequencies below about 2 Hz, above which the acoustic levels imply near-isotropic surface wave directionality.Funding for the ALOHA Cabled Observatory was provided by the National Science Foundation and the State of Hawaii through the School of Ocean and Earth Sciences and Technology at the University of Hawaii-Manoa (F.     SNX12 Role in Endosome Membrane Transport

    In this paper, we investigated the role of sorting nexin 12 (SNX12) in the endocytic pathway. SNX12 is a member of the PX domain-containing sorting nexin family and shares high homology with SNX3, which plays a central role in the formation of intralumenal vesicles within multivesicular endosomes. We found that SNX12 is expressed at very low levels compared to SNX3. SNX12 is primarily associated with early endosomes and this endosomal localization depends on the binding to 3-phosphoinositides. We find that overexpression of SNX12 prevents the detachment (or maturation) of multivesicular endosomes from early endosomes. This in turn inhibits the degradative pathway from early to late endosomes/lysosomes, much like SNX3 overexpression, without affecting endocytosis, recycling and retrograde transport. In addition, while previous studies showed that Hrs knockdown prevents EGF receptor sorting into multivesicular endosomes, we find that overexpression of SNX12 restores the sorting process in an Hrs knockdown background. Altogether, our data show that despite lower expression level, SNX12 shares redundant functions with SNX3 in the biogenesis of multivesicular endosomes

    Nothing is more difficult today than deciding what to do about abortion, gay marriage, economic injustice, war, torture, global warming, euthanasia, capital punishment, and a host of other controversies, particularly in a world in which people of varying religious, cultural, and ethnic backgrounds commonly live side by side. Can we draw on the wisdom of the past to address these contemporary ethical dilemmas? Can we see more clearly how we should consider what is right and wrong, and good and bad, and then work through these divisive problems toward decisions that make sense to us?While challenging moral relativism, Doing Ethics in a Diverse World uses a pluralist approach that draws on religious as well as secular positions and on Eastern as well as Western traditions. The book’s approach reasons by analogy from the rule of law, including international human rights law, as a means to constructing ethical presumptions about duty, character, relationships, and rights. These presumptions are weighed against the predicted consequences of acting on them, which either confirm the presumptions or support alternative actions.     Identification of Interleukin-1 by Functional Screening as a Key Mediator of Cellular Expansion and Disease Progression in Acute Myeloid Leukemia

    Secreted proteins in the bone marrow microenvironment play critical roles in acute myeloid leukemia (AML). Through an ex vivo functional screen of 94 cytokines, we identified that the pro-inflammatory cytokine interleukin-1 (IL-1) elicited profound expansion of myeloid progenitors in ∌67% of AML patients while suppressing the growth of normal progenitors. Levels of IL-1ÎČ and IL-1 receptors were increased in AML patients, and silencing of the IL-1 receptor led to significant suppression of clonogenicity and in vivo disease progression. IL-1 promoted AML cell growth by enhancing p38MAPK phosphorylation and promoting secretion of various other growth factors and inflammatory cytokines. Treatment with p38MAPK inhibitors reversed these effects and recovered normal CD34+ cells from IL-1-mediated growth suppression. These results highlight the importance of ex vivo functional screening to identify common and actionable extrinsic pathways in genetically heterogeneous malignancies and provide impetus for clinical development of IL-1/IL1R1/p38MAPK pathway-targeted therapies in AML
