    Comparison of the effect of raw and blanched-frozen broccoli on DNA damage in colonocytes

    Consumption of cruciferous vegetables may protect against colorectal cancer. Cruciferous vegetables are rich in a number of bioactive constituents including polyphenols, vitamins and glucosinolates. Before consumption, cruciferous vegetables often undergo some form of processing that reduces their content of bioactive constituents and may determine whether they exert protective effects. The aim of this study was to compare the ability of raw and blanched-frozen broccoli to protect colonocytes against DNA damage, improve antioxidant status and induce xenobiotic metabolising enzymes (XME). Fifteen Landrace x Large White male pigs were divided into five age and weight matched sets (79 days, SD 3, and 34.7 kg, SD 3.9 respectively). Each set consisted of siblings to minimise genetic variation. Within each set, pigs received a cereal-based diet, unsupplemented (control) or supplemented with 600 g/d of raw or blanched-frozen broccoli for 12 d. The consumption of raw broccoli caused a significant 27% increase in DNA damage in colonocytes (P=0.03) relative to the control diet, whereas blanched-frozen broccoli had no significant effect. Both broccoli diets had no significant effect on plasma antioxidant status or hepatic and colonic XME. This study is the first to report that the consumption of raw broccoli can damage DNA in porcine colonocytes

    Dietary reference values for vitamin K

    Following a request from the European Commission, the EFSA Panel on Dietetic Products, Nutrition and Allergies (NDA) derives dietary reference values (DRVs) for vitamin K. In this Opinion, the Panel considers vitamin K to comprise both phylloquinone and menaquinones. The Panel considers that none of the biomarkers of vitamin K intake or status is suitable by itself to derive DRVs for vitamin K. Several health outcomes possibly associated with vitamin K intake were also considered but data could not be used to establish DRVs. The Panel considers that average requirements and population reference intakes for vitamin K cannot be derived for adults, infants and children, and therefore sets adequate intakes (AIs). The Panel considers that available evidence on occurrence, absorption, function and content in the body or organs of menaquinones is insufficient, and, therefore, sets AIs for phylloquinone only. Having assessed additional evidence available since 1993 in particular related to biomarkers, intake data and the factorial approach, which all are associated with considerable uncertainties, the Panel maintains the reference value proposed by the Scientific Committee for Food (SCF) in 1993. An AI of 1 mu g phylloquinone/kg body weight per day is set for all age and sex population groups. Considering the respective reference body weights, AIs for phylloquinone are set at 70 mu g/day for all adults including pregnant and lactating women, at 10 mu g/day for infants aged 7-11 months, and between 12 mu g/day for children aged 1-3 years and 65 mu g/day for children aged 15-17 years.     Cardiovascular Function During Supine Rest in Endurance Trained Males with New Zealand Blackcurrant: A Dose-Response Study

    Purpose Blackcurrant contains anthocyanins that could alter cardiovascular function and reduce cardiovascular disease risk. We examined dose responses of New Zealand blackcurrant (NZBC) extract on cardiovascular function during supine rest. Methods Fifteen endurance trained male cyclists (age: 38±12 years, height: 178±5 cm, body mass: 76±10 kg, V?O2max: 56±8 mL?kg-1?min-1, mean±SD) were randomly assigned using a counterbalanced Latin square design to complete four conditions, a control of no NZBC, or one of three doses (300, 600 or 900 mg?day-1) of NZBC extract (CurraNZTM) for seven-days with a fourteen-day washout. Cardiovascular function (i.e. blood pressure, heart rate, ejection time, cardiac output, stroke volume and total peripheral resistance) during supine rest was examined (Portapres® Model 2). Results Systolic and diastolic blood pressure, heart rate and ejection time were unchanged by NZBC. A dose effect (P<0.05) was observed for cardiac output, stroke volume and total peripheral resistance. A trend for a dose effect was observed for mean arterial blood pressure. Cardiac output increased by 0.6±0.6 L·min-1 (15%) and 1.0±1.0 L·min-1 (28%) and stroke volume by 5±8 mL (7%) and 6±17 mL (18%) between control and 600, and 900 mg?day-1, respectively. Total peripheral resistance decreased by 4±3 mmHg·L-1·min-1 (20%) and 5±9 mmHg·L-1·min-1 (20%) for 600, and 900 mg?day-1. Conclusion Seven-days intake of New Zealand blackcurrant extract demonstrated dose-dependent changes on some cardiovascular parameters during supine rest in endurance-trained male cyclists
