    P53 Codon 72 (Arg72Pro) Polymorphism and Prostate Cancer Risk: Association between Disease Onset and Proline Genotype

    The tumor suppressor gene p53 plays an important role in the stress response of the cell and is mutated in 50% of all human tumors. The p53 Arg72Pro single-nucleotide polymorphism (SNP) was found to be associated with an increased risk of various malignancies. Biochemical and biological differences between the 2 polymorphic variants of wild-type P53 might lead to distinct susceptibility to HPV- and non-HPV-induced tumors. For prostate cancer, only limited data are available, especially in the Caucasian pop-ulation. Therefore, we determined the distribution of the Arg72Pro SNP in a Caucasian case-control study including 118 prostate cancer patients and 194 male controls without any malignancy using restriction fragment length polymorphism analysis. A subset of 33 tumors was tested for HPV infection, and no HPV DNA was found. Cases and controls showed similar distributions of alleles in the SNP (p = 0.720). Regarding the onset of the disease, patients diagnosed at ≤60 years of age and older patients (>60 years of age) showed a significant difference in genotype distribution (p = 0.035); there was also an increased occurrence of risk allele Pro72 in cases aged ≤60 years (p = 0.045). A subset of 64 prostate tumors was stained immunohistochemically for P53. 5 of 64 prostate tumors (7.8%) were positive for P53 expression, indicating integrity of the protein in the majority of cases. Genotype distribution showed no association with the Gleason score or additional histopathological characteristics. This study shows that the overall risk of prostate cancer was not associated with Arg72Pro SNP and HPV infection in our cohort. However, disease onset might be modulated by the p53 Pro72 allele, suggesting an important role of apoptosis regulation in prostate carcinogenesis

    Last Glacial Period Cryptotephra Deposits in an Eastern North Atlantic Marine Sequence: Exploring Linkages to the Greenland Ice-Cores

    The establishment of a tephra framework for the Greenland ice-cores spanning the last glacial period, particularly between 25 and 45 ka b2k, provides strong potential for precisely correlating other palaeoclimatic records to these key archives. Tephra-based synchronisation allows the relative timing of past climatic changes recorded within different depositional environments and potential causal mechanisms to be assessed. Recent studies of North Atlantic marine records have demonstrated the potential of tracing cryptotephra horizons in these sequences and the development of protocols now allows a careful assessment of the isochronous nature of such horizons. Here we report on tephrochronological investigations of a marine sequence retrieved from the Goban Spur, Eastern North Atlantic, covering ?25–60 ka b2k. Density and magnetic separation techniques and an assessment of potential transport and depositional mechanisms have identified three previously unknown isochronous tephra horizons along with deposits of the widespread North Atlantic Ash Zone II and Faroe Marine Ash Zone III. Correlations between the new horizons and the Greenland ice-core tephra framework are explored and despite no tie-lines being identified the key roles that high-resolution climatostratigraphy and shard-specific trace element analysis can play within the assessment of correlations is demonstrated. The previously unknown horizons are new additions to the overall North Atlantic tephra framework for the last glacial period and could be key horizons for future correlations

    A Molecular Study on the Prevalence and Virulence Potential of Aeromonas spp. Recovered from Patients Suffering from Diarrhea in Israel

    Background: Species of the genus Aeromonas are native inhabitants of aquatic environments and have recently been considered emerging human pathogens. Although the gastrointestinal tract is by far the most common anatomic site from which aeromonads are recovered, their role as etiologic agents of bacterial diarrhea is still disputed. Aeromonas-associated diarrhea is a phenomenon occurring worldwide; however, the exact prevalence of Aeromonas infections on a global scale is unknown. Methodology/Principal Findings: The prevalence and virulence potential of Aeromonas in patients suffering from diarrhea in Israel was studied using molecular methods. 1,033 diarrheal stools were sampled between April and September 2010 and Aeromonas species were identified in 17 (,2%) patients by sequencing the rpoD gene. Aeromonas species identity and abundance was: A. caviae (65%), A. veronii (29%) and Aeromonas taiwanensis (6%). This is the first clinical record of A. taiwanensis as a diarrheal causative since its recent discovery from a wound infection in a patient in Taiwan. Most of the patients (77%) from which Aeromonas species were isolated were negative for any other pathogens. The patients ranged from 1 to 92 years in age. Aeromonas isolates were found to possess different virulence-associated genes: ahpB (88%), pla/ lip/lipH3/apl-1 (71%), act/hlyA/aerA (35%), alt (18%), ast (6%), fla (65%), lafA (41%), TTSS ascV (12%), TTSS ascF-ascG (12%), TTSS-dependent ADP-ribosylating toxins aexU (41%) and aexT (6%) in various combinations.     Tephra without borders: Far-reaching clues into past explosive eruptions

    This review is intended to highlight recent exciting advances in the study of distal (>100 km from the source) tephra and cryptotephra deposits and their potential application for volcanology. Geochemical correlations of tephra between proximal and distal locations have extended the geographical distribution of tephra over tens of millions square kilometers. Such correlations embark on the potential to reappraise volume and magnitude estimates of known eruptions. Cryptotephra investigations in marine, lake and ice-core records also give rise to continuous chronicles of large explosive eruptions many of which were hitherto unknown. Tephra preservation within distal ice sheets and varved lake sediments permit precise dating of parent eruptions and provide new insight into the frequency of eruptions. Recent advances in analytical methods permit an examination of magmatic processes and the evolution of the whole volcanic belts at distances of hundreds and thousands of kilometers from source. Distal tephrochronology has much to offer volcanology and has the potential to significantly contribute to our understanding of sizes, recurrence intervals and geochemical make-up of the large explosive eruptions

    Mutational Activation of FGFR3 is Not Involved in the Development of Prostate Cancer

    Objective: The mutational constitutive activation of FGFR3 has been discovered in several malignancies but only limited data on FGFR3 mutations in prostate cancer are available. Most recently, activating FGFR3 mutations were described as being associated with low-grade prostate tumors. Therefore, we investigated the FGFR3 mutation status in a comprehensive series of prostate tumors. Methods: 102 archival formalin- fixed paraffin-embedded prostate tumors of patients treated with radical prostatectomy [with a low-grade subgroup (Gleason score ^ 6) of 29 patients] as well as 29 incidental prostate tumors [low-grade tumors (Gleason score ^ 6); n = 22] and 16 benign prostatic hyperplasia samples obtained by transurethral resection of the prostate were investigated. After microdissection and DNA isolation, all FGFR3 mutation hotspots discovered in human malignancies were analyzed using the SNaPshot © approach or restriction fragment length polymorphism (RFLP) analysis. Results: All cases could successfully be analyzed by SNaPshot; 80 cases were investigated using RFLP. No mutation in FGFR3 could be detected in any of the analyzed cases. Conclusion: The most recently reported FGFR3 mutations in low-grade prostate tumors could not be verified in our series. There were also no mutations in prostate tumors from patients with concomitant bladder tumors as reported previously. These data suggest that the mutational activation of FGFR3 plays no important role in prostate carcinogenesis, which is in accordance with previous studies performed on smaller tumor cohorts

    Should nurses be role models for healthy lifestyles? Results from a modified Delphi study

    Aim To explore the expectation that nurses should be role models for healthy behaviours. Background Nurses are expected to be role models for healthy behaviours. Whether this is a realistic and acceptable expectation has not been explored. Design Modified Policy Delphi study with two rounds of data collection. Method Purposive sampling was used to explore areas of agreement and disagreement among six stakeholder groups who influence nursing roles: practising nurses, nursing students, service users, policy makers, workforce development leads and stakeholders working in nurse education. Two rounds of a modified Policy Delphi study were conducted between February - June 2015. The first round used telephone interviews for an open exploration of opinions. The second round used attitude statements to explore convergence and divergence of opinions across stakeholder groups. Responses were analysed thematically. Results Policy and professional discourse that asserts that nurses should be healthy role models was seen as unrealistic and unhelpful. Contrary to the view that nurses should epitomize and demonstrate healthy behaviours to encourage patients and to be credible in advice, stakeholders agreed that it was more important to be seen as ‘human’ and understand the challenges of health behaviour change. Student and practising nurses did not see role modelling healthy behaviours as a reasonable professional expectation. Conclusions The findings challenge the assumptions underpinning the argument that nurses be healthy role models. Further research is needed to understand the views of frontline nurses and to further explore avenues by which health services staff health can be improved
