    Mechanisms and biomarker candidates in pterygium development

    Pterygium pathogenesis has been mainly associated with UV light exposure; however, this association remains quite controversial. The complete mechanism of pterygium also remains to be clarified. Factors such as inflammation, viral infection, oxidative stress, DNA methylation, inflammatory mediators, extracellular matrix modulators, apoptotic and oncogenic proteins, loss of heterozygosity, microsatellite instability, lymphangiogenesis, epithelial-mesenchymal cell transition, and alterations in cholesterol metabolism have been identified as causes. Several studies aimed to clarify the molecular mechanisms underlying the growth and proliferation of pterygium. Understanding its molecular basis provides new potential therapeutic targets for its prevention and treatment. A comprehensive search of the databases, namely, MedLine, EMBASE, and LILACS, was conducted with the following key words: pterygium, epidemiology, pathogenesis, biomarkers, and review.     A escala de waterlow aplicada em pessoas com lesão medular

    Objetivos: Avaliar os fatores de risco para úlceras por pressão em pessoas hospitalizadas por lesão medular, utilizando a Escala de Waterlow e as orientações prestadas para os pacientes antes e depois do aparecimento das úlceras.Método: Estudo transversal, com 60 pacientes, realizado em um hospital da cidade de Fortaleza (Ceará-Brasil). A coleta de dados efetuou-se de maio a outubro de 2008 por meio da utilização da escala de Waterlow. Para a análise foram utilizados: Teste de Kolmogorov-Smirnov, Coeficiente de correlação de Pearson e Coeficiente de correlação de Spearman.Resultados: O fator de risco mais significante para o desenvolvimento da úlcera por pressão foi a mobilidade física prejudicada associada ao longo tempo de internação. A presença de acompanhante foi outro achado importante, pois dos 19 pacientes que não desenvolveram úlcera por pressão todos tinham acompanhantes. Foi constatado também que 90% dos pacientes não receberam nenhum tipo de orientação antes do advento da úlcera por pressão, e que apenas 31% receberam orientações depois do aparecimento da mesma.Conclusões: A variável mais significativa em relação ao risco para desenvolver úlcera por pressão foi a mobilidade física prejudicada, visto que os pacientes eram dependentes de cadeira de rodas ou inertes no leito. A maioria não recebeu orientação antes do aparecimento da úlcera por pressão e somente alguns receberam orientação. Assim o estudo revelou-se eficaz na avaliação dos fatores de risco bem como para o conhecimento das orientações de enfermagem recebidas

    Exploring the cellular uptake of hectorite clay mineral and its drug carrier capabilities

    In the last years, the use of clay minerals for pharmaceutical purposes has increased due to their interesting properties. Hectorite (Ht) is a clay belonging to the smectite group which has attracted attention for applications in biology, tissue engineering and as drug carrier and delivery system. However, the mechanisms involved in Ht cellular uptake and transport into cells, are still unclear. Herein, we used a labeled Ht (Ht/1Cl) to study both the cellular uptake, by confocal laser scanning microscopy, and internalization pathways involved in the cellular uptake, by various endocytosis-inhibiting studies and fluorescence microscopy. These studies highlighted that Ht can penetrate the cellular membrane, localizing mainly in the cytoplasm. The main intracellular transport mechanisms are the ATP-dependent ones and those where filaments and microtubules are involved. Finally, as proof of concept for the potential of Ht as carrier system, we envisaged the covalent grafting of the anticancer molecule methotrexate (MTX), chosen as model, to obtain the Ht-MTX nanomaterial. The covalent linkage was confirmed by several techniques and the morphology of the obtained nanomaterial was imaged by SEM and TEM investigations. The kinetic release of the drug from the Ht-MTX nanomaterial in physiological conditions was studied as well. Furthermore, cytotoxic studies on different cell lines, namely, HL-60, HL-60R, MCF-7, 5637, UMUC3 and RT112 showed that Ht could be a promising material for anticancer therapy.The work was financially supported by the University of Palermo.     Physiological and productive performance of papaya plants irrigated in a semiarid environment

    Papaya tree has great economic importance and potential of cultivation in semiarid environment. However, water scarcity in this region may inhibits photosynthetic activity and limit the production of this crop, being necessary to use strategies of irrigation management that rationalize water and allow the production of the plants without affecting photosynthetic activity. In this sense, this study aimed to evaluate the physiological and productive performance of papaya “Calimosa” under different water replacement rates in semiarid environment. Papaya plants were irrigated with four water replacement rates (50, 75, 100 and 125% ETo) from 30 days after transplant of seedlings to the experimental area.  At 10 months after transplant were realized evaluations of gas exchange and chlorophyll fluorescence, and at 12 months after transplant, it was started harvesting fruits to determine the yield. Water replacement rates significantly influenced the parameters of gas exchange of papaya with better results on rates of 125% ETo. However, application of water replacement rates of 100% ETo provide conditions adequate for the activity of gas exchange and better quantum efficiency of photosystem II, that contribute to fruit production in papaya. With rates less than 75% ETo occur drastic reductions in gas exchange, quantum efficiency of photosystem II, and fruit production of papaya, which impairs its cultivation in semiarid environments

    Formação Continuada de Docentes no Ensino Superior: experiências e contradições em tempos de neoliberalismo

    Este artigo tem por objetivo discutir a formação continuada de docentes do ensino superior, considerando seus atuais desafios e dilemas e como estes têm sido convergidos e superados nos espaços coletivos construídos em algumas Instituições de Ensino Superior (IES). Compreende-se a educação como operadora de mudança, como espaço de formação cidadã, que considera os afetos e as subjetividades, nesse sentido, ela está impossibilitada de qualquer tentativa de neutralidade, seja nos espaços institucionais, nos objetivos das discussões, nas formações promovidas ou na forma de vínculos trabalhistas. Assim, a formação continuada de professores do ensino superior, entendida como continuidade da formação profissional, tem possibilidades de promover novas reflexões e meios de desenvolver e aprimorar o fazer profissional, de modo a repensar as necessidades institucionais e sociais. Notamos que as inovações didático-pedagógicas contribuem em determinados contextos, mas não se sustentam sem a reflexão sobre a função da educação na sociedade. A valorização da experiência prática que considere a importância de complementação didático-pedagógica e a convivência de profissionais experientes com iniciantes também são práticas que favorecem o processo da formação continuada dos docentes.

    Genetic comparison of sickle cell anaemia cohorts from Brazil and the United States reveals high levels of divergence

    Genetic analysis of admixed populations raises special concerns with regard to study design and data processing, particularly to avoid population stratification biases. The point mutation responsible for sickle cell anaemia codes for a variant hemoglobin, sickle hemoglobin or HbS, whose presence drives the pathophysiology of disease. Here we propose to explore ancestry and population structure in a genome-wide study with particular emphasis on chromosome 11 in two SCA admixed cohorts obtained from urban populations of Brazil (Pernambuco and Sao Paulo) and the United States (Pennsylvania). Ancestry inference showed different proportions of European, African and American backgrounds in the composition of our samples. Brazilians were more admixed, had a lower African background (43% vs. 78% on the genomic level and 44% vs. 76% on chromosome 11) and presented a signature of positive selection and Iberian introgression in the HbS region, driving a high differentiation of this locus between the two cohorts.     Genotype-phenotype Correlations In Cyp1b1-associated Primary Congenital Glaucoma Patients Representing Two Large Cohorts From India And Brazil

    Conselho Nacional de Desenvolvimento Científico e Tecnológico (CNPq)Background Primary congenital glaucoma (PCG), occurs due to the developmental defects in the trabecular meshwork and anterior chamber angle in children. PCG exhibits genetic heterogeneity and the CYP1B1 gene has been widely implicated worldwide. Despite the diverse mutation spectra, the clinical implications of these mutations are yet unclear. The present study attempted to delineate the clinical profile of PCG in the background of CYP1B1 mutations from a large cohort of 901 subjects from India (n=601) and Brazil (n=300). Methods Genotype-phenotype correlations was undertaken on clinically well characterized PCG cases from India (n=301) and Brazil (n=150) to assess the contributions of CYP1B1 mutation on a set of demographic and clinical parameters. The demographic (gender, and history of consanguinity) and quantitative clinical (presenting intraocular pressure [IOP] and corneal diameter [CD]) parameters were considered as binary and continuous variables, respectively, for PCG patients in the background of the overall mutation spectra and also with respect to the prevalent mutations in India (R368H) and Brazil (4340delG). All these variables were fitted in a multivariate logistic regression model using the Akaike Information Criterion (AIC) to estimate the adjusted odds ratio (OR) using the R software (version 2.14.1). Results The overall mutation spectrum were similar across the Indian and Brazilian PCG cases, despite significantly higher number of homozygous mutations in the former (p=0.024) and compound heterozygous mutations in the later (p=0.012). A wide allelic heterogeneity was observed and only 6 mutations were infrequently shared between these two populations. The adjusted ORs for the binary (demographic) and continuous (clinical) variables did not indicate any susceptibility to the observed mutations (p>0.05). Conclusions The present study demonstrated a lack of genotype-phenotype correlation of the demographic and clinical traits to CYP1B1 mutations in PCG at presentation.     Health-related quality of life in patients with type 1 diabetes mellitus in the different geographical regions of Brazil : data from the Brazilian Type 1 Diabetes Study Group

    Background: In type 1 diabetes mellitus (T1DM) management, enhancing health-related quality of life (HRQoL) is as important as good metabolic control and prevention of secondary complications. This study aims to evaluate possible regional differences in HRQoL, demographic features and clinical characteristics of patients with T1DM in Brazil, a country of continental proportions, as well as investigate which variables could influence the HRQoL of these individuals and contribute to these regional disparities. Methods: This was a retrospective, cross-sectional, multicenter study performed by the Brazilian Type 1 Diabetes Study Group (BrazDiab1SG), by analyzing EuroQol scores from 3005 participants with T1DM, in 28 public clinics, among all geographical regions of Brazil. Data on demography, economic status, chronic complications, glycemic control and lipid profile were also collected. Results: We have found that the North-Northeast region presents a higher index in the assessment of the overall health status (EQ-VAS) compared to the Southeast (74.6 ± 30 and 70.4 ± 19, respectively; p < 0.05). In addition, North- Northeast presented a lower frequency of self-reported anxiety-depression compared to all regions of the country (North-Northeast: 1.53 ± 0.6; Southeast: 1.65 ± 0.7; South: 1.72 ± 0.7; Midwest: 1.67 ± 0.7; p < 0.05). These findings could not be entirely explained by the HbA1c levels or the other variables examined. Conclusions: Our study points to the existence of additional factors not yet evaluated that could be determinant in the HRQoL of people with T1DM and contribute to these regional disparities

    Cabbage and fermented vegetables : From death rate heterogeneity in countries to candidates for mitigation strategies of severe COVID-19

    Large differences in COVID-19 death rates exist between countries and between regions of the same country. Some very low death rate countries such as Eastern Asia, Central Europe, or the Balkans have a common feature of eating large quantities of fermented foods. Although biases exist when examining ecological studies, fermented vegetables or cabbage have been associated with low death rates in European countries. SARS-CoV-2 binds to its receptor, the angiotensin-converting enzyme 2 (ACE2). As a result of SARS-CoV-2 binding, ACE2 downregulation enhances the angiotensin II receptor type 1 (AT(1)R) axis associated with oxidative stress. This leads to insulin resistance as well as lung and endothelial damage, two severe outcomes of COVID-19. The nuclear factor (erythroid-derived 2)-like 2 (Nrf2) is the most potent antioxidant in humans and can block in particular the AT(1)R axis. Cabbage contains precursors of sulforaphane, the most active natural activator of Nrf2. Fermented vegetables contain many lactobacilli, which are also potent Nrf2 activators. Three examples are: kimchi in Korea, westernized foods, and the slum paradox. It is proposed that fermented cabbage is a proof-of-concept of dietary manipulations that may enhance Nrf2-associated antioxidant effects, helpful in mitigating COVID-19 severity.Peer reviewe
